Este trabalho teve por objetivo descrever a morfologia dos diásporos e as suas fases de germinação, bem como determinar o substrato mais adequado para a germinação das sementes e o crescimento das plântulas de palmeira-da-rainha (Archontophoenix alexandrae (F. Mueller) H. Wendl. e Drude). Periodicamente, unidades representativas de cada fase de germinação foram retiradas para as descrições morfológicas. Os substratos utilizados foram Plantmax®, areia, terra (latossolo roxo) e outro com proporções iguais de terra, areia e esterco (TAE). Foi instalado um experimento em delineamento inteiramente casualizado. Avaliaram-se a porcentagem, o tempo médio de germinação e o crescimento inicial das plântulas aos 135 dias após a emergência, com base na altura, diâmetro do colo e número, comprimento e largura das folhas. As sementes são albuminosas, com endosperma ruminado e oleaginoso, e o embrião é lateral, periférico e relativamente indiferenciado. A germinação é do tipo criptocotiledonar hipógea, iniciando-se com a formação de uma massa de células indiferenciadas na depressão micropilar. Essa massa de células torna-se cilíndrica, com a diferenciação dos primórdios caulinares e radiculares. Concomitantemente, ocorre o desenvolvimento de raízes adventícias no eixo embrionário. O substrato mais indicado para a germinação de sementes de palmeira-da-rainha é o Plantmax®, porém, para o crescimento inicial, indica-se Plantmax® e TAE.

Avaliou-se o efeito da adição de subprodutos de oleaginosas na dieta de ovinos em substituição ao farelo de soja. Foram distribuídos quatro ovinos Santa Inês, com peso corporal de, aproximadamente, 40kg, canulados no rúmen, em quadrado latino (4x4), com quatro dietas e quatro períodos, analisados por regressão para a avaliação da degradação ruminal, dos valores de pH e das concentrações de nitrogênio amoniacal in vivo. Os subprodutos foram as tortas de amendoim, girassol e soja, em dietas isonitrogenadas, com 70% de concentrado e 30% de volumoso (feno de tifton). Não foi observado efeito da interação tempo x dieta para os valores de pH e concentração ruminal de nitrogênio amoniacal (P>0,05). O pH apresentou valor médio de 6,2. As concentrações ruminais de nitrogênio amoniacal não foram afetadas, com valor médio de 29,9mg/dL. Não foram observadas diferenças (P>0,05) nas taxas de degradação da matéria seca e na degradabilidade potencial.

The aimm of this study was to evaluate the influence of washing water temperature and pressure on mesophilic and psycluotrophic aerobic bacterial populations, yeasts and molds, total conforms and fecal coliforms population, whereas the cattle carcass surface may become contaminated during the different slaughter procedures and the final carcass washing may reduce microbial population Samples were taken by sponge swabbing in four areas of carcass surface (flank, neck, chest and rump), from which 20 were washed with write, at 25 degrees C and without artificial pressure, 20 with water under a pressure of 3atm, 20 with water at 40 degrees C and without artificial pressure and 20 with water at 40 degrees C under a pressure of 3atm For control purpose and using the same method to collect more 20 carcasses samples were taken without washing., loading 100 samples The results showed that the water at 25 degrees C undo 3atm pressure was more efficient to remove microorganisms from carcass surface than the heated water, although this latter promoter a mote visible clean carcasses

Yeasts are attractive hosts for heterologous protein production as they follow the general eukaryotic post-translational modification pattern. The well-known Saccharomyces cerevisiae has been used to produce a large variety of foreign proteins. The proper function of muscle tropomyosin depends on a specific modification at its N-terminus. Although tropomyosin has been produced in different expression systems, only the recombinant protein produced in the yeast Pichia pastoris has native-like functional properties. In this paper we describe the production of functional skeletal muscle tropomyosin in the yeast S. cerevisiae. The recombinant protein was produced in high amounts and production was strongly affected by genetic and environmental factors, including plasmid copy number, promoter strength, and growth media composition. The characteristics of fermentation and aerobic stability were evaluated in corn silage inoculated with different doses of Lactobacillus buchneri. The whole corn plant (300 g/kg DM) was ensiled in quadruplicate laboratory silos (7L). L. buchneri 40788 was applied at 5×10(4), 1×10(5), 5×10(5) and 1×10(6) cfu/g of fresh forage. Silages with no additive were used as controls. After 130 d of ensiling, the silages were subjected to an aerobic stability evaluation for 12 days, in which chemical and microbiological parameters as well as the temperature of the silage were measured to determine the aerobic deterioration. The addition of L. buchneri resulted in increased acetic acid concentrations. The number of yeast colonies was low in all treated silages. The pH, lactic and propionic acid concentrations did not differ between silages. Under aerobic conditions, all the treated silages showed a low number of yeasts and a great aerobic stability. Therefore, L. buchneri is effective against yeasts and improves the aerobic stability of corn silage in laboratory silos. However, doses equal or superior to 1×10(5) cfu/g of fresh forage were more efficient in the control of aerobic spoilage.

Objetivou-se avaliar os efeitos da inoculação de Bacillus subtilis sobre as características e perdas ocorridas na fermentação, no desenvolvimento de leveduras e fungos filamentosos e na estabilidade aeróbia de silagens de milho. Estudou-se o milho híbrido 2B655, no qual avaliaram-se os seguintes tratamentos: silagem sem inoculação de B. subtilis e silagens inoculadas com B. subtilis nas concentrações de 5x10(4); 1x10(5) e 5x10(5)UFC/g de forragem. O delineamento experimental utilizado foi o inteiramente casualizado, em esquema de parcelas subdivididas, em que as silagens constituíram as parcelas e os tempos de exposição aeróbia as subparcelas, com quatro repetições. Os dados obtidos foram submetidos à análise de variância por meio do software SISVAR®, bem como aplicou-se a análise de regressão a 5% de significância. A aplicação de B. subtilis não alterou as características químicas e as perdas no processo de fermentação da silagem de milho. A contagem de leveduras na abertura dos silos foi reduzida, assim como a população de fungos filamentosos diminuiu durante a exposição aeróbia, o que implicou em menores valores de pH e resultou em maior estabilidade aeróbia, devido à utilização da maior dose de B. subtilis. A inoculação de Bacillus subtilis na concentração de 5x10(5)UFC/g de forragem controla o crescimento dos micro-organismos deterioradores e melhora a estabilidade aeróbia da silagem de milho, a manter os valores de pH mais estáveis na fase de pós-abertura dos silos.

This work study of solar distillation feasibility in effluent of petroleum industry: produced water, making possible your reuse for irrigation of oleaginous cultures or fodder crops or in steam generation, as well the transport phenomena involved. The methodology for development of this project was to characterize the effluent to be treated and to accomplish physical and chemical analysis in the distilled, to build distillation equipment, concomitant operation of both equipments and implementation of data processing and economical evaluation. The methodology used for all parameters is outlined in APHA (1998) and sampling of the type compound. The feeding of distillation equipment was performed with treated effluent from UTPF of Guamaré. The temperature was monitored throughout the distillers and during the time of operation. The distillers feed occur, as a rule, for sifon. The distillers were operated by a period of 17 months between July 2007 and February 2009, in which 40 experiments were performed. The radiation and temperature datas were acquired in the INPE s site and the temperature inside of the distillers was registered by DATALOGGER Novus. The rates of condensation (mL / min) were determined by measuring of the flow in a graduate test tube of 10 mL and a chronometer. We used two simple solar effect distillers of passive type with different angles in coverage: 20 ° and 45 °. The results obtained in this study and the relevant discussions are divided into six topics: sample characterization and quality of distilled; construction of distillers; operation (data, temperature profile), climatic aspects, treatment of data and economical analysis. Results obtained can be inferred that: the energy loss by the adoption of vessel glass was not significant, however, complicates the logistics of maintenance the equipment on a large scale. In the other hand, the surface of the tub with a glass shield on the equipment deterioration, both devices showed similar performance, so there is not justified for use of equipment 450. With regard to the climatological study it was verified that the Natal city presents monthly medium radiation varying in a range between 350 and 600 W/m2, and medium of wind speed of 5 m / s. The medium humidity is around 70% and rainfall is very small. The regime of the system is transient and although it has been treated as a stationary system shows that the model accurately represents the distillers system's 20 degrees. The quality of the distilled with regard to the parameters evaluated in this study is consistent with the Class 3 waters of CONAMA (Resolution 357). Therefore we can conclude that solar distillation has viability for treat oilfield produced water when considered the technical and environmental aspects, although it is not economically viable

OBJETIVO: Monitorar e caracterizar fungos anemófilos e leveduras de fontes bióticas e abióticas de uma unidade hospitalar. MÉTODOS: As coletas foram realizadas mensalmente e em dois períodos, do centro cirúrgico e unidades de terapia intensiva adulto e neonatal em hospital de Araraquara, Estado de São Paulo. Para coleta de fungos anemófilos foi utilizado amostrador tipo Andersen de simples estágio. A pesquisa de leveduras foi feita das mãos e de orofaringe de profissionais de saúde, bem como de superfícies de leitos e de maçanetas das áreas críticas. RESULTADOS: Foram recuperados do centro cirúrgico 32 gêneros de fungos anemófilos e 31 das unidades de terapia intensiva. Os gêneros mais freqüentemente isolados foram Cladophialophora spp., Fusarium spp., Penicillium spp., Chrysosporium spp. e Aspergillus spp. Durante o período de estudo, houve reforma e implantação de uma unidade dentro do hospital, que coincidiu com o aumento na contagem de colônias de Cladophialophora spp., Aspergillus spp. e Fusarium spp. Leveduras foram encontradas em 39,4% dos profissionais de saúde (16,7% das amostras dos espaços interdigitais, 12,1% do leito subungueal e 10,6% da orofaringe) e, em 44% das amostras do mobiliário, com predomínio do gênero Candida (C. albicans, C. guilliermondii, C. parapsilosis e C. lusitaniae) seguido por Trichosporon spp. CONCLUSÕES: Observou-se número relativamente elevado de fungos anemófilos (potencialmente patogênicos) em áreas especiais e níveis expressivos de leveduras em fontes bióticas e abióticas. O monitoramento microbiológico ambiental deve ser realizado, principalmente em salas especiais com pacientes imunocomprometidos, sujeitos à exposição de patógenos do meio ambiente, assim como, advindos de profissionais de saúde.

The pathogenic fungus, Histoplasma capsulatum, causes the respiratory and systemic disease 'histoplasmosis'. This disease is primarily acquired via inhalation of aerosolized microconidia or hyphal fragments of H. capsulatum. Evolution of this respiratory disease depends on the ability of H. capsulatum yeasts to survive and replicate within alveolar macrophages. It is known that adhesion to host cells is the first step in colonization and biofilm formation. Some microorganisms become attached to biological and non-biological surfaces due to the formation of biofilms. Based on the importance of biofilms and their persistence on host tissues and cell surfaces, the present study was designed to investigate biofilm formation by H. capsulatum yeasts, as well as their ability to adhere to pneumocyte cells. H. capsulatum biofilm assays were performed in vitro using two different clinical strains of the fungus and biofilms were characterized using scanning electron microscopy. The biofilms were measured using a 2,3-bis(2-methoxy-4-nitro-5-sulfophenyl)-5-[(phenylamino)carbonyl]-2H-tetrazolium-hydroxide (XTT) reduction assay. The results showed that both the H. capsulatum strains tested were very efficient at adhering to host cells and forming biofilm. Therefore, this is a possible survival strategy adopted by this fungus.

Aims: The aim of this study was to identify and determine the diversity, occurrence and distribution of fungi in water used at a haemodialysis centre.Methods and Results: Samples in the hydraulic circuit for the distribution of the water, dialysate samples and samples of sterilization solution from dialysers were collected over a 3-month period, and 500 ml of each sample was filtered through membranes. All together 116 isolates of fungi were recovered from 89% of all water samples collected inside the haemodialysis unit, with prevalence of moulds in tap water samples and of yeasts in dialysate samples. Fusarium spp. was the most abundant genus found, whereas Candida parapsilosis was the predominant yeast species.Conclusions: This study demonstrated that various fungi were present in the water system. These data suggest the inclusion of the detection and quantification of fungi in the water of haemodialysis.Significance and Impact of the Study: The recovery of fungi from aqueous haemodialysis environments implies a potential risk for haemodialysis patients and indicates the need for continuous maintenance and monitoring. Further studies on fungi in haemodialysis water systems are required to investigate the organism ability to persist, their role in biofilm formation and their clinical significance.

In this study, water and eight sanitizing solutions (vinegar at 6, 25, and 50%; acetic acid at 2 and 4%; peracetic acid at 80 ppm, sodium hypochlorite at 200 ppm, and sodium dichloroisocyanurate at 200 ppm) were compared in terms of their effectiveness against the natural microbiota of lettuce. All of the samples were kept in contact with the sanitizing solutions for 15 min, and the effectiveness of a sanitizing agent was evaluated on the basis of the number of decimal reductions of the total aerobic mesophilic count, the mold and yeast count, the total coliform count, and the Escherichia coli count. The average initial levels of these organisms in the samples were 6.94 log(10) CFU/g for aerobic mesophilic microorganisms, 5.62 log(10) CFU/g for molds and yeasts, and 3.25 log(10) CFU/g for total coliforms. of 10 samples analyzed, only 4 contained E. coli, and the average initial level of this microorganism in these 4 samples was 1.64 log(10) CFU/g. Salmonella was not detected in any of the samples tested. The decimal reductions of the populations of aerobic mesophilic microorganisms, molds and yeasts, total coliforms, and E. coli were 0.78, 0.87, 0.82, and >0.14 log(10) CFU/g, respectively, in water; 2.89, >3.41, >2.21, and >0.26 log(10) CFU/g, respectively, in 50% vinegar; 2.42, >3.20, >1.99, and >0.26 log(10) CFU/g, respectively, in 25% vinegar; 1.83, 2.57, 1.58, and >0.26 log(10) CFU/g, respectively, in 6% vinegar; 3.91, >3.58, >2.25, and >0.26 log(10) CFU/g, respectively, in 4% acetic acid; 3.37, >3.53, >2.25, and >0.26 log(10) CFU/g, respectively, in 2% acetic acid; 1.85, 2,32, 1.44, and >0.20 log(10) CFU/g, respectively, in 80 ppm of peracetic acid; 2.63, 2.75, 1.91, and >0.26 log(10) CFU/g, respectively, in 200 ppm of sodium hypochlorite; and 3.23, >3.08, >1.95, and >0.26 log(10) CFU/g, respectively, in 200 ppm of sodium dichloroisocyanurate. Statistical analysis of the results showed that the effectiveness levels for all of the sanitizing agents tested were equivalent to or higher than that for sodium hypochlorite at 200 ppm.

Paracoccidioides brasiliensis is a dimorphic fungus known to produce invasive systemic disease in humans. The 43-kDa glycoprotein of P, brasiliensis is the major diagnostic antigen of paracoccidioidomycosis and may act as a virulence factor, since it is a receptor for laminin. Very little is known about early interact-ions between this fungus and the host cells, so we developed in vitro a model system employing cultured mammalian cells (Vero cells), in order to investigate the factors and virulence mechanisms of P. brasiliensis related to the adhesion and invasion process. We found that there is a permanent interaction after 30 min of contact between the fungus and the cells. The yeasts multiply in the cells for between 5 and 24 h. Different strains of P, brasiliensis were compared, and strain 18 thigh virulence) was the most strongly adherent, followed by strain 113 (virulent), 265 (considered of low virulence) and 113M(mutant obtained by ultraviolet radiation, deficient in gp43). P. brasiliensis adhered to the epithelial cells by a narrow tube, while depressions were noticed in the cell surface, suggesting an active cavitation process. An inhibition assay was performed and it was verified that anti-gp43 serum and a pool of sera from individuals with paracoccidioidomycosis were able to inhibit the adhesion of P. brasiliensis to the Vero cells. Glycoprotein 43 (gp43) antiserum abolished 85 % of the binding activity of P. brasiliensis. This fungus can also invade the Vero cells, and intraepithelial parasitism could be an escape mechanism in paracoccidioidomycosis. (C) 2000 Editions scientifiques et medicales Elsevier SAS.
