The tsunami deposits of the valley of Agaete (Pérez-Torrado et al., 2006), north-western Gran Canaria, attributed to the Guimar flank collapse in Tenerife, have been revisited and new data are presented here. Besides the occurrences reported by Pérez-Torrado et al. (2006) a new outcrop was found and named “La Ruina” (at 28º 05’ 47,41” N; 15º 41’ 52,04” W; 71 m asl). The above-mentioned authors suggested the possibility that more than one marine conglomerate deposit could be present in the outcrops of “Llanos de Turmán” and “Berrazales”. At “La Gasolinera” and “La Aldea 1” the conglomerates are formed by a single layer representing one depositional event; at “La Aldea 2”, the conglomerates are composed of two layers directly contacting with each other, but evidence of a time hiatus between them was not found. Although the hypothesis of stacking of two depositional units within the same episode versus deposition of two distinct layers in different time-moments is debatable at the present state of knowledge, the first possibility is favoured. The field evidence at “Llanos de Turman” and “Berrazales” unquestionably shows that terrestrial sediments (colluvia; paleosols) are present and separate two marine conglomerate deposits, indicating that at least two distinct tsunami inundations are needed to explain the stratigraphy. However, at the new “La Ruina” outcrop, besides the two deposits mentioned above, a third and older marine conglomerate was found, clearly separated in time from the ones cited above. The existence of marine conglomerates emplaced in different moments is evidenced by the occurrence of intercalated paleosols, colluvia and other subaerial materials, implying significant time intervals between the emplacement of marine conglomeratic layers. A number of gastropod operculae from the tsunamiites were sent for U-Th dating to try to further constrain the age span of these deposits. The field evidence presented above shows that the emplacement of the deposits is related to, at least, three tsunami events. The lateral correlation between different outcrops is difficult due to variable number of deposits in each outcrop, lateral discontinuity and variability, and to compositional and textural similarity between distinct tsunami sediments. The occurrence of three Pleistocene tsunami deposits in the same area points to a relatively high frequency of tsunamis (generated by landslides, surface rupturing earthquakes, fast entry of voluminous volcanic deposits into the sea or large submarine eruptions). It is possible that this recurrence of tsunami inundations may reflect multiple-phased landslides responsible for the mega-landslide scars prominent in the geomorphology of the neighbouring island of Tenerife. La sostenibilidad de los sistemas olivareros situados en zonas de pendiente y montaña (SMOPS) en Andalucía se encuentra actualmente amenazada por las elevadas tasas de abandono que afectan a estos sistemas productivos. Así, la consumación de este proceso de abandono, no sólo pondría en peligro a las propias explotaciones, sino a todo el conjunto de bienes y servicios no productivos y al patrimonio cultural generado por este sistema productivo. En este contexto, la búsqueda de alternativas políticas enfocadas a revertir este proceso se erige como una necesidad categórica en aras de garantizar en el largo plazo la sostenibilidad de los olivares de montaña. Esta tesis pretende hacer frente a esta necesidad a través de la construcción de un marco político alternativo para los SMOPS, que permita la integración simultánea de todas las dimensiones que pueden influir en su desarrollo; esto es: el marco político actual, principalmente determinado por la Política Agraria Común (PAC) de la Unión Europea (UE); las preferencias de la sociedad hacia la oferta de bienes y servicios públicos generados por los SMOPS; y las preferencias y voluntad de innovación hacia nuevos manejos y sistemas de gestión de los agricultores y propietarios de las explotaciones. Para ello, se emplea una metodología de investigación mixta, que abarca la realización de cuatro encuestas (personales y online) llevadas a cabo a los agentes o grupos de interés involucrados directa o indirectamente en la gestión de los SMOPS –ciudadanos, agricultores y propietarios y expertos-; una profunda revisión de las herramientas de política agroambiental actuales y posibles alternativas a las mismas; y el desarrollo de nuevas estrategias metodológicas para dotar de mayor precisión y fiabilidad las estimaciones obtenidas a partir del Método del Experimento de Elección (MEE) en el campo de la valoración medioambiental. En general, los resultados muestran que una estrategia de política agroambiental basada en la combinación de los Contratos Territoriales de Zona Rural (CTZR) y el manejo ecológico supondría una mejora en la sostenibilidad de los sistemas olivareros de montaña andaluces, que, al mismo tiempo, propiciaría una mejor consideración de las necesidades y demandas de los agentes implicados en su gestión. Asimismo, los hallazgos obtenidos en esta investigación demandan un cambio de paradigma en los actuales pagos agroambientales, que han de pasar de una estrategia basada en la implementación de acciones, a otra enfocada al logro de objetivos, la cual, en el caso del olivar, se podría centrar en el aumento del secuestro de carbono en el suelo. Desde un punto de vista metodológico, los resultados han contribuido notablemente a mejorar la fiabilidad y precisión de las conclusiones estimadas a partir del MEE, mediante el diseño de un novedoso proceso iterativo para detectar posibles comportamientos inconsistentes por parte de los entrevistados con respecto a su máxima Disposición al Pago (DAP) para lograr la situación considerada como “óptima” en los olivares ecológicos de montaña andaluces. En líneas generales, el actual marco institucional favorece la puesta en práctica de la mayoría de las estrategias propuestas en esta tesis; sin embargo son necesarios mayores esfuerzos para reconducir los actuales Pagos Agroambientales y Climáticos de la PAC, hacia una estrategia de política agroambiental adaptada a las necesidades y requisitos del territorio en el que se aplica, enfocada al logro de objetivos y que sea capaz de integrar y coordinar al conjunto de agentes y grupos de interés involucrados -directa o indirectamente- en la gestión de los olivares de montaña. En este contexto, se espera que la puesta en práctica de nuevas estructuras y acuerdos de gobernanza territorial juegue un importante papel en el desarrollo de una política agroambiental realmente adaptada a las necesidades de los sistemas olivareros de montaña andaluces. ABSTRACT The long-term sustainability of Andalusian sloping and mountainous olive production systems (SMOPS) is currently threatened by the high abandonment rates that affect these production systems. The effective occurrence of this abandonment process is indeed menacing not only farms themselves, but also the wide array of public goods and services provided by SMOPS and the cultural heritage held by this production system. The search of policy alternatives aimed at tackling this process is thus a central necessity. This thesis aims to undertake this necessity by building an alternative policy framework for SMOPS that simultaneously integrates the several dimensions that are susceptible to influence it, namely: the current policy framework, mainly determined by the European Union’s (EU) Common Agricultural Policy (CAP); the social preferences toward the supply of SMOPS’ public goods and services; and farmers’ preferences and willingness to innovate toward new management practices in their farms. For this purpose, we put into practice a mixed-method strategy that combines four face-to-face and online surveys carried out with SMOPS’ stakeholders -including citizens, farmers and experts-; in-depth analysis of current and alternative agrienvironmental policy (AEP) instruments; and the development of novel methodological approaches to advance toward more reliable Discrete Choice Experiment’s (DCE) outcomes in the field of environmental valuation. Overall, results show that a policy strategy based on the combination of Territorial Management Contracts (TMC) and organic management would further enhance Andalusian SMOPS’ sustainability by simultaneously taking into account stakeholders’ demands and needs. Findings also call for paradigm shift of the current action-oriented design of Agri-Environmental-Climate Schemes (AECS), toward a result-based approach, that in the case of olive orchards should particularly be focused on enhancing soil carbon sequestration. From a methodological perspective, results have contributed to improve the accuracy and feasibility of DCE outcomes by designing a novel and iterative procedure focused in ascertaining respondents’ inconsistent behaviour with respect to their stated maximum WTP for the attainment of an ideal situation to be achieved in organic Andalusian SMOPS. Generally, the present institutional framework favours the implementation of the main policy strategies proposed in this thesis, albeit further efforts are required to better conduct current CAP’s agri-environmental instruments toward a territorially targeted result-oriented strategy capable to integrate and coordinate the whole set of stakeholders involved in the management of SMOPS. In this regard, alternative governance structures and arrangements are expected to play a major role on the process of tackling SMOPS’ agri-environmental policy challenge.

The several hundred members of the eukaryotic protein kinase superfamily characterized to date share a similar catalytic domain structure, consisting of 12 conserved subdomains. Here we report the existence and wide occurrence in eukaryotes of a protein kinase with a completely different structure. We cloned and sequenced the human, mouse, rat, and Caenorhabditis elegans eukaryotic elongation factor-2 kinase (eEF-2 kinase) and found that with the exception of the ATP-binding site, they do not contain any sequence motifs characteristic of the eukaryotic protein kinase superfamily. Comparison of different eEF-2 kinase sequences reveals a highly conserved region of ≈200 amino acids which was found to be homologous to the catalytic domain of the recently described myosin heavy chain kinase A (MHCK A) from Dictyostelium. This suggests that eEF-2 kinase and MHCK A are members of a new class of protein kinases with a novel catalytic domain structure.

The reduction of 12-oxophytodienoic acid (OPDA) to 3-oxo-2(2′[Z]-pentenyl)-cyclopentane-1-octanoic acid is catalyzed by 12-oxophytodienoate-10,11-reductase (OPR). Analysis of the isomer preference of OPR has indicated that the activity is composed of two isoenzymes exhibiting different stereoselectivities. The two isoforms of OPR have been separated, using protein extracts of Rock Harlequin (Corydalis sempervirens) as the starting material. OPRI, the enzyme reported earlier from the same species and corresponding to the cloned OPR from Arabidopsis, utilized 9R,13R-OPDA >> 9S,13R-OPDA but not the 13S-configured isomers, whereas the new activity, OPRII, effectively reduced all four OPDA isomers, including the natural 9S,13S-OPDA (cis-[+]-OPDA). OPRII activity is characterized in detail. The enzyme's enzymatic, biochemical, and immunological properties prove that it is a close relative of OPRI. The roles of OPRI and OPRII in octadecanoid biology are discussed.

The relationship between the production of reactive oxygen species and the hypersensitive response (HR) of tobacco (Nicotiana tabacum L.) toward an incompatible race of the Oomycete Phytophthora parasitica var nicotianae has been investigated. A new assay for superoxide radical (O2−) production based on reduction of the tetrazolium dye sodium,3′-(1-[phenylamino-carbonyl]-3,4-tetrazolium)-bis(4-methoxy-6-nitro) benzene-sulfonic acid hydrate (XTT) has enabled the quantitative estimation of perhydroxyl/superoxide radical acid-base pair (HO2·/O2−) production during the resistant response. Tobacco suspension cells were inoculated with zoospores from compatible or incompatible races of the pathogen. Subsequent HO2·/O2− production was monitored by following the formation of XTT formazan. In the incompatible interaction only, HO2·/O2− was produced in a minor burst between 0 and 2 h and then in a major burst between 8 and 10 h postinoculation. During this second burst, rates of XTT reduction equivalent to a radical flux of 9.9 × 10−15 mol min−1 cell−1 were observed. The HO2·/O2− scavengers O2− dismutase and Mn(III)desferal each inhibited dye reduction. An HR was observed in challenged, resistant cells immediately following the second burst of radical production. Both scavengers inhibited the HR when added prior to the occurrence of either radical burst, indicating that O2− production is a necessary precursor to the HR.

The Bajo Segura Basin (eastern Betic Cordillera) is a Mediterranean marginal basin where the Messinian Erosional Surface (MES), formed during the Messinian Salinity Crisis sea-level fall, is well developed. Overlying this major discontinuity the lower Pliocene transgressive sediments record the reflooding of the Mediterranean and the return to an open marine environment, the continental shelf being rebuilt after the Messinian erosion. The stratigraphic and biostratigraphic study of six sections allows two transgressive-regressive sequences filling the MES to be distinguished, correlated with the previously distinguished Mediterranean offshore seismic units. Ten calcareous nannofossil bioevents have been identified. The lower sequence can be dated according to nannofossil biozones NN12 to NN14 and the upper sequence by NN15 to NN16. The boundary between both lower Pliocene sedimentary sequences occur after the first common occurrence (FCO) of Discoaster asymmetricus found in the uppermost sediments of the lower sequence and before the first occurrence (FO) of Discoaster tamalis in the lowermost part of the upper sequence. Thus this sequence boundary can be estimated at between 4.1 and 4.0Ma ago.

New age models for twelve Deep Sea Drilling Project sites in the North Pacific have been produced, based on (in order of importance in our dataset) a recompilation of previously published diatom, calcareous nannofossil and foraminifer first and last occurrences, and magnetostratigraphy. The projected ages of radiolarian first and last occurrences derived from the line of correlation of the age/depth plots have been computed from these sites, and 28 radiolarian events have thereby been newly cross calibrated to North Pacific diatom and other stratigraphy. Several of the North Pacific radiolarian events are older than in previously published equatorial Pacific calibrations, and some may be diachronous within the North Pacific. These patterns may be due to complex latitudinal patterns of clinal variation in morphotypes within lineages, or to migration events from the North Pacific towards the Equator.

A new planktic foraminifer transfer function (GSF18) related 5 North Atlantic assemblages to winter and summer sea surface temperature. GSF18, based on recombined and simplified core top census data, preserves most environmental information and reproduces modern North Atlantic conditions with approximately the same accuracy as previous transfer functions, but can be more readily applied to faunal samples ranging in age from Pliocene to Holocene. Transfer function GSF18 has been applied to faunal data from Deep Sea Drilling Project Hole 552A to produce a 2.5 m.y. sea-surface temperature (SST) time series. Estimates show several periods between 2.3 and 4.6 Ma during which mean SST's were both several degrees warmer and several degrees cooler than modern conditions. Between 2.9 and 4.0 Ma SST was generally warmer than modern except for a 250 k.y. interval centered at 3.3 Ma. Maximum SST, with respect to modern conditions, occurred after the cool interval near 3.1 Ma when SST was approximately 3.6° C warmer than present conditions. Comparison of SST estimates with stable isotope data suggest that after peak warming at 3.1 Ma, there was an overall surface water cooling with concomitant build up of global ice volume, culminating in Northern Hemisphere glaciation. This event is also indicated by the presence of ice rafted detritus in 552A sediments at about 2.45 Ma.

Chemical analysis of an Australian Streptomyces species yielded a range of known anthracyclines and biosynthetically related metabolites, including daunomycin (1), E-rhodomycinone (2), 11-hydroxyauramycinone (3), 11-hydroxysulfurmycinone (4), aklavinone (5), bisanhydro-gamma-rhodomycinone (6), and the anthraquinone 7, as well as the hitherto unreported blanchaquinone (8). The structure assigned to 8 was secured by detailed spectroscopic analysis and correlation to known analogues, such as the anthraquinone 7. This account also represents the first natural occurrence of 3, 4, and 7 and the first spectroscopic characterization of 11-hydroxysulfurmycinone (4).

In this study, a new method was developed based on aqueous phenylation, purge-and-trap preconcentration, gas chromatography (GC) separation, and detection by atomic fluorescence spectrometry (AFS) or inductively coupled plasma mass spectrometry (ICPMS). This technique is suitable for simultaneous determination of trace or ultratrace levels of CH3Hg+ and CH3CH2Hg+ in environmental samples. Method detection limits were 0.03 ng/L for both CH3Hg+ and CH3CH2Hg+ when AFS was used as the detector and 0.02 and 0.01 ng/L for CH3Hg+ and CH 3CH2Hg+ with ICPMS, respectively. The new method has the additional benefits of being free from interference by Cl - and dissolved organic matter. Using the method developed, both CH3Hg+ and CH3CH2Hg+ were detected in a number of soil and sediment samples collected from the Florida Everglades. The identity of CH3CH2Hg+ was verified by purge-and-trap-GC/MS analysis. The possibility of analytical artifact was excluded by using stable isotope tracer technique in combination with ICPMS detection. CH3CH 2Hg+ in the soil samples analyzed was at ng/g level, similar to that of CH3Hg+. The prevalence of CH 3CH2Hg+ in the soil of the Florida Everglades suggests that ethylation plays an important role in the geochemistry of Hg in this wetland. Soil incubation and sawgrass culture experiments using stable isotope tracers revealed that CH3Hg+ was mainly produced by microbial activities under anaerobic conditions, agreeing well with the general understanding of methylation mechanisms of Hg in the environment. Ethylation of Hg was not confirmed in these experiments, indicating that ethylation of Hg most probably follows different mechanisms in comparison to methylation. Further experiments revealed that trace levels of ethyllead species were able to transfer ethyl group to Hg in both deionized water and freshwater matrixes, producing CH3CH2Hg+. This might partially account for the occurrence of CH3CH2Hg+ in the relatively pristine environment of the Florida Everglades.
