The castor bean plant is a tropical species and is subject to various diseases, which cause great losses. Among these diseases, the gray mold (Botryotinia ricini) is one of the most important. The fungus spore production was evaluated in the media of cultures BDA; Oats-Agar; Mazeina-Agar; Rice-Agar; castor-bean crushed leaves-agar (FM), FM-CaCO3; V8 juice to 5% (V8-5%); V8-10%; V8-20% and tomato juice at 5% (TJ-5%); TJ-10% and TJ-20%. The production of spore in different media of cultures was evaluated at 8(th) days of incubation. The data were analyzed using the comparison of means, through the test Tukey a 5% probability, and the data processed for (X + 1) (0.5). All means of crops tested were able to produce conidia, but the best results was obtained with the culture medium V8-20% (5.7 x 10(6) conidia/mL) and BDA (3.5 x 10(6) conidia/mL).

The authors analysed the simultaneous effects of the dimensional and weight alteration of resins utilized for esthetic facets submitted to warehousing conditions and periods of time. The resins showed different behaviour and a neat correlation between dimensional alteration and weight. The condition of immersion promoted dimensional alteration 50% lower and an alteration of weight three times higher.

The purpose of this study was to investigate the peroxidase and polyphenoloxidase activities during the refrigeration at 4oC of yacon tuberous roots with and without polyethylene packing and compared with untreated roots at regular conditions. It was concluded that refrigeration treatment maintains a low activity of these enzymes and keeps good appearance of the roots.

This paper aims to evaluate the physiological quality of Sebastiania commersoniana seeds stored in cloth bag, plastic bag and glass container and kept in two conditions: laboratory workbench and cold chamber (4 ± 2°C e 80% relative humidity - RH) for 531 days. Periodically, samples were taken in order to determine the moisture content and to perform germination tests. The experimental design was completely randomized, in factorial scheme of 3 × 2 (wrappings × conditions), for each period of storage evaluated (0, 158, 271, 389 and 531 days of storage). Supplementary, a polynomial regression was adjusted in order to describe the physiological quality of the seeds for the different conditions during the experimental period. The results lead to the following considerations: a) storage under variable temperature and air humidity (laboratory workbench) can be used for periods of over five and a half months, with the seeds stored in cloth bag, plastic bag or glass container; b) the physiological quality of the seeds was not hardly altered when stored in cold chamber in cloth bag, plastic bag and glass container for over 18 months.

Rosemary (Rosmarinus officinalis L.) is a spice from Lamiaceae family known since ancient times because its medicinal effects and, currently, several studies have pointed its antioxidant and antimicrobian effects. Lipid oxidation is a problem in food production because proceed the lost of organoleptical and nutritional qualities so required in Market. Fish salting is an ancient conservation method that expect reduce water activity and, consequently, microorganism growth in food, except halophillic bacteria. In the meantime, the inconvenient of this procedure is that the salt accelerates tissue's lipid oxidation. The aim of this work was evaluate the antioxidative and antimicrobian effects by treatment and pre treatment with rosemary (Rosmarinus officinalis L.) aqueous extract in dry salted tilapia fillets, storaged in freezing temperatures. To follow the oxidative, dry salted tilapia fillets were treated or pre treated with rosemary natural extract and storage at -18°C for 240 days. Analisys of 2-thiobarbituric acid reactive substances (TBARS), soluble nitrogen in trichloroacetic acid (TCA), water activity and microbiology were done. The pre treatment (3.39±0,53) and the treatment with rosemary (3.31±0.79) had oxidative index twice lower than the control treatment (6.14±1.21) in the last time of the research. The microbiological rosemary analisys showed count levels of resistant microorganisms to salt (2.0×103CFU/g of sample), whom causes the initial fillets contamination. The microbiological counts remained invariable in all groups during storage periods.

Objective: This case-control study analyzed mass spectrometry fingerprinting patterns of culture media samples used for embryo culture to predict embryo implantation. Methods: The culture medium harvested after embryo transfer of 22 embryos from 13 patients was used for the experiments. After embryo transfer, the remaining culture media were collected and samples were split in positive (n=8) and negative (n=14) implantation groups according to implantation outcomes (100% or 0% of implantation). Samples were individually diluted and injected directly to the Electrospray ionization (ESI) MS coupled to a Quadrupole Time-of-flight MS (Q-ToF-MS).Ions relative intensities of each spectrum were considered. Data analysis was conducted in MatLab 7.0 version using Partial Least Squares - Discriminant Analysis toolbox. Results: There were 3027 observed ions at 100% and 0% implantation groups by ESI-Q-ToF-MS. The statistical model could categorize the samples in two clusters, based on their positive and negative implantation outcomes. Less intense ions present in the mass spectra with statistical significance have contributed to the major differences to group distinction. Conclusions: Positive and negative implantation embryos showed a specific biochemical pattern present in culture media, which could be detected as a fast, simple and non-invasive way. This biochemical profile could help the selection of the most viable embryo, improving single embryo transfer and thus eliminating the risk and undesirable outcomes of multiple pregnancies. © Todos os direitos reservados a SBRA - Sociedade Brasileira de Reprodução Assistida.

A laboratory culture of Ankistrodesmus gracilis algae was evaluated by studying the biology of the species and its chemical composition in a traditional medium (CHU 12) and in two alternative culture media, NPK (20-5-20) and macrophyte (Eichhornia crassipes) + NPK, in three different types of recipients (fiberglass, carboy and plastic bag). First peak in the growth curve of Ankistrodesmus gracilis occurred on the ninth day in macrophyte + NPK medium (74.16 x 10 5 cells mL -1) in a fiberglass recipient. However, highest density (p < 0.01) was reported in medium CHU 12 (122.87 x 10 5 cells mL -1) in a plastic bag on the twelfth day. Cell density was over 70 x 10 5 cells mL -1 starting on the twelfth day. Growth rate of A. gracilis was similar (p > 0.05) in culture media in the three recipients. Protein and fiber were similar (p > 0.05) in the treatments, but lipids were higher (p < 0.05) in NPK. Nitrate, ammonia, total phosphorus and orthophosphate contents were over 1 mg L -1 in NPK (p < 0.01). Results show that alternative media, such as NPK and macrophyte + NPK, are possible for large-scale culture of A. gracilis cultured in three types of recipients. Costs are low, occupying less space when cultured in plastic bags and in the laboratory.

The use of culture media produced with commercial fertilizers can represent a simple and low cost alternative for commercial orchid propagation. The aim of this study was to evaluate the in vitro growth of plantlets of Cattleya trianaei in culture medium MS reduced and and formulated with Peters® NPK 10-30-20 in different doses. 90 day-old plantlets with two leaflets were submitted to five treatments (T1 - reduced MS; T2 - Peters® 1 g L -1; T3 - Peters® 2 g L-1; T4 - Peters® 3 g L -1 and T5 - Peters® 5 g L-1) arranged in a completely randomized design with five replicates with 25 plantlets for each treatment and incubated during 180 days, with subcultures at each 60 days, when the number of roots, root length, number of leaves, shoot length and shoot fresh matter were evaluated. The simplified culture medium with fertilizer Peters® 3 g L -1 presented results statistically different as for the number of roots, number of leaves, shoot length and shoot fresh matter and it can be recommended for in vitro growth of this ornamental orchid.

The bacteria Bacillus thuringiensis var. israelensis (Bti) generates certain toxins with pesticide action, which can be used on the control of transmissible diseases by culicides, specially Aedes aegypti, the dengue vector. This biopesticide has been produced by submerged fermentation and, in Brazil, this production has been made by very little research centers and, more recently, by a unique small enterprise. For the implementation of a viable vectors control program through biopesticides, some studies about culture media are essential in order to join efficiency and low costs. In this way, agroindustrial wastes or by-products have been used as a nutrient source for the culture media production. In this study, corn steep liquor, a corn industrial processing by-product and tryptose, both with/without sugar addition, were compared as culture media. Cellular growth was evaluated by optical density at 620 nm, spore production by total viable cell count and LC50 by bioassays against 4th instar larvae. Among the four examined substrates, the medium composed by glucose plus corn steep liquor presented the best spore production and bioassay results.

This study aimed to evaluate the quality of 'Palmer' mangoes previously stored at low temperatures, after their transference to the environmental condition. Fruits harvested at physiological maturity were carefully transported to the Laboratory where they were selected, standardized as the color, size and absence of injuries and treated with fungicide before they were stored at 2°C (75.7% RH), 5°C (73.8% RH) e 12°C (82% RH) for 7, 14 and 21 days. At the end of each period, the fruits were transferred to environmental condition (22.9°C; 62.3% RH), where they were kept for 1, 3, 5 and 7 days, simulating the trading period, and evaluated for the occurrence of injuries and rottenness; peel and pulp color; firmness; contents of soluble solids, titratable acidity and ascorbic acid, as well as, the activities of the enzymes peroxidase, polyphenoloxidase and phenylalanine ammonia-lyase. The results indicated that 'Palmer' mangoes can be stored at 12°C for 21 days without damage to ripening, but with limitations due to the occurrence of decay. The storage at 2°C and 5°C was limited by the occurrence of injuries in the peel, but at the temperature of 2°C these symptoms were more severe and compromised the development of the characteristic color of the peel. However, the ripening of the pulp was not harmed, but this process occurred with less intensity than in mangoes maintained at 12°C.
