BACKGROUND: Australian farmers and their workers are exposed to a wide variety of pesticides. Organophosphate (OP) insecticides are a widely used class of pesticide used for animal husbandry practices (Naphthalophos for sheep dipping, jetting and drench), crop production for pest control (Dimethoate) and in public health (Maldison for head lice). Acute poisonings with this class of insecticide are reported among agricultural workers and children around the globe, due to the inhibition of acetylcholinesterase (AChE). Less is known about chronic exposures. Regular monitoring of erythrocyte AChE will enable farmers to identify potential exposure to organophosphate insecticides and take action to reduce exposures and improve their health and safety practices. This study aims to assess and improve the integration of AChE monitoring into routine point of care health clinics, and provide farming and non-farming people with a link between their AChE activity and their household chemical and agrichemical use. METHODS/DESIGN: The research will target individuals who work on mixed farming enterprises and routinely using OPs (n = 50) and non-farmers (n = 30). Baseline data are collected regarding demographic, health conditions and behaviours, Kessler 10 (K10) scores, chemical use and personal protection. Baseline anthropometric measures include height, weight, hip and waist circumference, body fat analysis and, biochemical analysis of fasted total serum cholesterol, triglycerides, low-density cholesterol (LDL), high-density cholesterol (HDL) and blood glucose. Analysis of erythrocyte cholinesterase (EAChE) activity is also conducted using a finger prick test. Testing of EAChE is then repeated in all participants every 3 weeks for a maximum of three times over a period 10 weeks. Participants are provided with full feedback and counselling about their EAChE activity after each reading and a detailed summary provided to all participants at the completion of the study. Data will be analysed using repeated measures within a general linear model. DISCUSSION: This work will provide an evidence base and recommendations for the integration of EAChE monitoring into Australian rural health clinics, leading to research which will further quantify pesticide exposure both on the farm and in the home, highlighting the importance of sustaining and providing a safe work and home environment for farming communities. Methomyl (Lannate®) is an insecticide from the carbamate group, frequently used in pest control in various types of crops. This compound works inhibiting the activity of the enzyme acetylcholinesterase. The use of physicochemical and ecotoxicological analysis is the most efficient strategy for the correct characterization and control of residues of metomil. The main objectives of this study were to evaluate the acute toxicity of methomyl in 96 hours of exposure and, through a sublethal assay of 5 hours, to assess its effect on the activity of acetylcholinesterase present in brain and squeletic dorsal muscle of the Danio rerio fish. The results showed that the LC50-96 found to D. rerio was 3.4 mg/L and it was found through the average of four definitive tests. In vitro assays were used to test the inhibitory action of methomyl directly over soluble AChE, extracted from the squeletic dorsal muscle, with maximum inhibition of 68.57% to the insecticide concentrations of 0.2 mg/L. In sublethal tests with D. rerio, inhibitory effect of methomyl was found over the soluble form of AChE in the squeletic dorsal muscle, both in one and five hours of fish exposure to the insecticide. In both period, the average values of inhibition were around 61%. In the same condition, no significant inhibitory effect of methomyl soluble and membrane AChE of the D. rerio was observed in the 0.42, 0.85, 1.70 and 2.50 mg/L concentrations and in both times of fish exposure

Plodia interpunctella (Indian meal moth) is a cosmopolitan pest that attacks not only a wide range of stored grain as well other food products. Due to its economic importance several researches have focused in a method with ability to control this pest with few or no damage to the environment. The study of digestive enzymes inhibitors, lectins and chitin-binding proteins, has often been proposed as an alternative to reduce insect damage. In this study we report the major classes of digestive enzymes during larval growth in P. Interpunctella, being those proteinases actives at pH 9.5 and optimum temperature of 50 oC to both larvae of the 3rd instar and pre-pupal stage of development. In vitro and zymogram assays presented the effects of several inhibitors, such as SBTI, TLCK and PMSF to intestinal homogenate of 3rd instar larvae of 62%, 92% and 87% of inhibition and In pre-pupal stage of 87%, 62 % and 55% of inhibition, respectively. Zymograms showed inhibition of two low molecular masses protein bands by TLCK and that in presence of SBTI were retarded. These results are indicative of predominance of digestive serine proteinases in gut homogenate from Plodia interpunctella larvae. This serine proteinase was then used as a target to evaluate the effect of SBTI on larvae in in vivo assay. Effect of SBTI on mortality and larval mass was not observed at until 4% of concentration (w/w) in diets. Chitin, another target to insecticidal proteins, was observed by chemical method. Moreover, optic microscopy confirmed the presence of a peritrophic membrane. Established this target, in vivo effect of EvV, a chitin binding vicilin, evaluated during the larval development of P. interpunctella and was obtained a LD50 of 0,23% and WD50 of 0,27% to this protein. Mechanism of action was proposed through of the in vivo digestibility of EvV methodology. During the passage through the larval digestive tract was observed that EvV was susceptible to digestive enzymes and a reactive fragment, visualized by Western blotting, produced by digestion was recovered after dissociation of the peritrophic membrane. The bound of EvV to peritrophic membrane was confirmed by immunohystochemical assays that showed strong immunofluorescent signal of EvV-FITC binding and peritrophic membrane. These results are a indicative that vicilins could be utilized as potential insecticide to Plodia interpunctella and a control methods using EvV as bioinsecticide should be studied to reduce lost caused by storage insect pests

Thiamethoxam is a systemic insecticide from the neonicotinoid group, nitroguanidin family which affects the nicotinic receptor acetyl choline in the insect membrane, wounding the nervous system and causing the death of the insect. It was used with success in the control of initial pests of several crops. It was considered that thiamethoxam has a bioactivator effect, because in the absence of insects promoted increase in vigor, development and productivity of crops. This work was carried out to verify if thiamethoxam causes histological changes in sugarcane roots. In this work, it was used optical microscopy, images arrest, tissue biometrics and statistical analysis, in young roots of sugarcane RB 83 5486 after the treatments with different thiamethoxam concentrations. It was determined changes in histological structure of tissues 7, 14, 21 and 28 days after the treatments, establishing its effects on root plant anatomy. It was verified that thiamethoxam increased root cortex width, increasing the vascular cylinder and the metaxylem vessel elements number in the vascular tissue until 21 days after application.

Astilbin was isolated in high yield from Dimorphandra mollis, and its insecticidal and growth inhibiting activity by stomach ingestion were evaluated against Anticarsia gemmatalis and Spodoptera frugiperda. The insecticidal activity of astilbin, the weight reduction of the larval phase and the prolongation of the larval and pupal phases were verified for both species. Astilbin was identified on the base of its NMR, MS and physical data. (C) 2002 Society of Chemical Industry.

The guava tree pest that has been requesting larger control frequency, in the area of Jaboticabal - São Paulo in the last years, is the psilideo Triozoida limbata. Thus, the objectives of this research were: a) Compare control tactics T. limbata based on monitoring and selectivity of insecticide to decrease the applications; b) To register and to correlate the natural enemies in habitants in guava tree ecosystem with the key pest and with meteorological factors; c) See if the control of T. limbata affects the population of fruit flies and d) To verify the efficiency of appropriate insecticides to IMP, in control of T. limbata. The experiment was conducted in 2004, in an orchard of Paluma cultivar in Vista Alegre do Alto, SP. The used strategies were (doses in g.a.i./100L water): control; imidacloprid + beta-cyfluthrin (2.5 + 0.3); acetamiprid (4.0); fenpropathrin (15.0) and regional conventional treatment. It was concluding that is possible to reduce the number of applications of less aggressive insecticides to the environment and the man, with the adoption of monitoring of T. limbata and implementing the action level. The natural enemies' populations densities (Scymnus spp., Cycloneda sanguinea, Azia luteipes, Crysoperla spp., Polybia spp., Brachygastra ssp.) present positive correlations with the populations of T. limbata. Population fluctuations of T. limbata and natural enemies are not altered by meteorological factors (precipitation and temperature) in irrigated orchard. The population of Anastrepha spp. is minimized when of the applications is for control T. limbata. The insecticides imidacloprid, imidacloprid + beta-cyfluthrin, acetamiprid and fenpropathrin are efficient in the control of T. limbata.

The great importance of cockroaches as household pest have been on their ability to spread microorganisms harmful to humans and animals. Rest during the day in dark, humid and hot as sewerage. At night go into stores and kitchens or places to deposit and manipulation of food as bakeries, restaurants, hospitals and homes wich requires clinig operations. This work aimed to evaluate mortality of B. germanica ( L., 1767) ( Blattodea: Blattellidae) under different periods and exposure area treated by insecticides in laboratory. The tests were carried out at Department of Fitossanidade at UNESP, Campus of Jaboticabal, SP, Brasil. The insecticides were applied by Potter's tower sprayer on Petri dishes. Three times of exposure (2, 8 and 32 min) and four exposure areas ( 25, 50, 75 and 100%) and volume of 0,5 ml for the second experiment were tested. It was used the insecticides Pyrethroids gammacyalothrine, deltamethrine, lambdacialothrine, alfacipermethrine, cipermethrine and carbamates bendiocarb, in the dosages recommended by the manufacturers, and I was used control without application. Five adult cockroaches was confined in the dishes for both experiments. The mortality evaluation was done 0, 1, 2, 4, 24, 48 and 72 hours after of the confinement on the treated surface. It was concluded that for exposure duration experiment all the insecticides have a good efficiency. The exposure area experiment the insecticide cipermethrine was what the one which had the higher mortality. Regarding to the effect of exposure duration on the accumulated mortality every duration times had a high mortality. In relation to the exposure area the highest mortality reached to 100% of treated area.

The present work aimed at learning the population dynamic of Triozoida limbata (Enderlein, 1918) (Hemiptera: Psyllidae) in a guava orchard submitted to the minimum use of insecticide in Jaboticabal - SP. For the study of population fluctuation and the evolution of damage from the psillid we analyzed 20 plants of the orchard biweekly, during the period from March 2005 to June 2006. For each plant we evaluated 10 leaves containing the symptom of the attack from the psillid. The evaluated parameters were the number of nymphs and the percentage of damage caused in the leaves, in accordance with a scale of notes. These parameters were analyzed in histograms and correlated to the meteorological factors. The population density of the psillid increased considerably with the approach of the spring and presented a positive correlation with the temperature. The level of damage of the psillid accompanied the population density of the pest. According to the data found in this work we may conclude that the increase of the temperature causes an increase in the population density of T. limbata and the largest population densities from psillid occur in the months of September to November and the smallest densities occur between the months of May and July.
