Arabidopsis is a model plant used to study disease resistance; Solanum tuberosum or potato is a crop species. Both plants possess inducible defense mechanisms that are deployed upon recognition of pathogen invasion. Transcriptional reprogramming is crucial to the activation of defense responses. The Pathogenesis-Related (PR) genes are activated in these defense programs. Expression of Arabidopsis PR-l and potato PR-10a serve as markers for the deployment of defense responses in these plants. PR-l expression indicates induction of systemic acquired resistance (SAR). Activation of SAR requires accumulation of salicylic acid (SA), in addition to the interaction of the non-expressor of pathogenesis-related genes I (NPRI), with the TGA transcription factors. The PR-10a is activated in response to pathogen invasion, wounding and elicitor treatment. PR-10a induction requires recruitment of the Whirly I (Whyl) activator to the promoter. This locus is also negatively regulated by the silencer element binding factor (SEBF). We established that both the PR-l and PR-10a are occupied by repressors under non-inducing conditions. TGA2 was found to be a constitutive resident and repressor of PR-l, which mediates repression by forming an oligomeric complex on the promoter. The DNA-binding activity of this oligomer required the TGA2 N-terminus (NT). Under resting conditions we determined that the PR-10a is bound by a repressosome containing SEBF and curiously the activator Pto interacting protein 4 (Pti4). In the context of this repressosome, SEBF is responsible for PR-10a binding, yet rWe also showed that PR-l and PR-10a are activated by different means. In PR-l activation the NPRI NT domain alleviates TGA2-mediated repression by interacting with the TGA2 NT. TGA2 remains at the PR-l but adopts a dimeric conformation and forms an enhanceosome with NPRl. In contrast, the PR-10a is activated by evicting the repressosome and recruiting Why! to the promoter. These results advance our understanding of the mechanisms regulating PR-l and PR-10a expression under resting and inducing conditions. Trichoderma spp are effective competitors against other fungi because they are mycoparasitic and produce hydrolytic enzymes and secondary metabolites that inhibit the growth of their competitors. Inhibitory compounds produced by Trichoderma aggressivum, the causative agent of green mold disease, are more toxic to the hybrid off-white strains of Agaricus bisporus than the commercial brown strains, consistent with the commercial brown strain’s increased resistance to the disease. This project looked at the response of hybrid off-white and commercial brown strains of A. bisporus to the presence of T. aggressivum metabolites with regard to three A. bisporus genes: laccase 1, laccase 2, and manganese peroxidase. The addition of T. aggressivum toxic metabolites had no significant effect on MnP or lcc1 transcript abundance. Alternatively, laccase 2 appears to be involved in resistance to T. aggressivum because the presence of T. aggressivum metabolites results in higher lcc2 transcript abundance and laccase activity, especially in the commercial brown strain. The difference in laccase expression and activity between A. bisporus strains was not a result of regulatory or coding sequence differences. Alteration of laccase transcription by RNAi resulted in transformants with variable levels of laccase transcript abundance. Transformants with a low number of lcc transcripts were very sensitive to T. aggressivum toxins, while those with a high number of lcc transcripts had increased resistance. These results indicated that laccase activity, in particular that encoded by lcc2, serves as a defense response of A. bisporus to T. aggressivum toxins and contributes to green mold disease resistance in commercial brown strains.

Le Costimulateur Inductible (ICOS) est un récepteur exprimé à la surface des cellules T CD4 auxiliaires et T CD8 cytotoxiques. Il fut démontré à l’aide de modèles murins de transplantation de moelle osseuse que ICOS joue un rôle important dans l’induction de la maladie du greffon contre l’hôte aigüe (GVHD). ICOS potentialise deux signaux médiés par le récepteur de cellules T (TCR) : l’activation de la phosphoinositide 3-kinase (PI3K) ainsi que la mobilisation interne de calcium. En conditions in vitro, dans les cellules CD4 et CD8, ICOS réussi à potentialiser le flux de calcium médié par le TCR indépendamment de PI3K. La voie de signalisation de ICOS impliquée dans la GVHD demeure inconnue. Cependant, en utilisant une lignée de souris ‘knock-in’ nommée ICOS-Y181F, dans laquelle le cellules T ont sélectivement perdu la capacité d’activer PI3K par l’entremise d’ICOS, nous avons démontré que les cellules T peuvent utiliser un mécanisme ICOS indépendant de PI3K afin d’induire la GVHD. La mobilisation interne du Ca2+ mène à l’activation de NFAT, un facteur de transcription clé régulant des gènes comme IFN-γ, qui exprime une des cytokines clés impliquées dans la GVHD. Nous émettons comme hypothèse que la capacité pathogénique intacte des cellules T ICOSY181F à induire la GVHD, repose sur la signalisation du Ca2+ indépendante de PI3K. Le but de mon projet est d’identifier les résidus responsables de cette signalisation de Ca2+ médiée par ICOS ainsi que le mécanisme par lequel ce récepteur fonctionne. À l’aide de la mutagénèse dirigée, j’ai généré des mutants d’ICOS et j’ai analysé par cytométrie en flux leur capacité à activer le flux de Ca2+. J’ai ainsi identifié un groupe de lysine sur la queue cytoplasmique d’ICOS situé à proximité de la membrane comme étant essentiel à la fonction de potentialisation du flux de Ca2+. Je fournis également des preuves de l’implication de la kinase Lck, membre de la famille de kinases Src, dans la voie de signalisation de ICOS médiant la potentialisation du flux de Ca2+. Ainsi, ICOS s’associe à Lck et mène à une augmentation de l’activation de PLCγ1, la protéine effectrice clé causant la sortie de Ca2+ de la réserve intracellulaire. En conclusion, notre étude permet de comprendre davantage une des voies de signalisation d’ICOS. L’influx de Ca2+ dans les cellules T implique la voie ICOS-Lck-PLCγ1. Une compréhension plus approfondie de cette voie de signalisation pourrait s’avérer bénéfique afin d’élaborer de nouvelles stratégies menant à la prévention de maladies reliées à ICOS, comme la GVHD.

INTRODUCTION: Emerging evidence indicates that nitric oxide (NO), which is increased in osteoarthritic (OA) cartilage, plays a role in 4-hydroxynonenal (HNE) generation through peroxynitrite formation. HNE is considered as the most reactive product of lipid peroxidation (LPO). We have previously reported that HNE levels in synovial fluids are more elevated in knees of OA patients compared to healthy individuals. We also demonstrated that HNE induces a panoply of inflammatory and catabolic mediators known for their implication in OA cartilage degradation. The aim of the present study was to investigate the ability of inducible NO synthase (iNOS) inhibitor, L-NIL (L-N6-(L-Iminoethyl)Lysine), to prevent HNE generation through NO inhibition in human OA chondrocytes. METHOD: Cells and cartilage explants were treated with or without either an NO generator (SIN or interleukin 1beta (IL-1β)) or HNE in absence or presence of L-NIL. Protein expression of both iNOS and free-radical-generating NOX subunit p47 (phox) were investigated by western blot. iNOS mRNA detection was measured by real-time RT-PCR. HNE production was analysed by ELISA, Western blot and immunohistochemistry. S-nitrosylated proteins were evaluated by Western Blot. Prostaglandin E2 (PGE2) and metalloproteinase 13 (MMP-13) levels as well as glutathione S-transferase (GST) activity were each assessed with commercial kits. NO release was determined using improved Griess method. Reactive oxygen species (ROS) generation was revealed using fluorescent microscopy with the use of commercial kits. RESULTS: L-NIL prevented IL-1β-induced NO release, iNOS expression at protein and mRNA levels, S-nitrosylated proteins and HNE in a dose dependent manner after 24h of incubation. Interestingly, we revealed that L-NIL abolished IL-1β-induced NOX component p47phox as well as ROS release. The HNE-induced PGE2 release and both cyclooxygenase-2 (COX-2) and MMP-13 expression were significantly reduced by L-NIL addition. Furthermore, L-NIL blocked the IL-1β induced inactivation of GST, an HNE-metabolizing enzyme. Also, L-NIL prevented HNE induced cell death at cytotoxic levels. CONCLUSION: Altogether, our findings support a beneficial effect of L-NIL in OA by preventing LPO process in NO-dependent and/or independent mechanisms.

À l’origine, la nouvelle concernant l’occupation américaine d’Haïti en 1915 a suscité peu d’indignation aux États-Unis. En effet, on reproche à la république son instabilité politique et on juge aussi qu’une intervention américaine concourrait à l’édifice de l’autorité de la loi. À partir de 1915 et surtout en 1920, l’Association nationale pour l’avancement des gens de couleur (NAACP), fondée en 1909, critique cette ingérence et milite pour y voir un terme. W.E. B. Du Bois et James Weldon Johnson, deux figures publiques noires importantes travaillant au sein de l’organisation, dénoncent avec conviction l’occupation d’Haïti. Les historiens ont jusqu’ici jugé que la NAACP fut inspirée par des considérations de solidarité raciale en adhérant à la cause de la souveraineté haïtienne. Si la thèse présente ne réfute pas cette possibilité, elle cherche tout de même à démontrer que le cadre conceptuel de la solidarité raciale ne saurait illustrer toute la complexité de la campagne haïtienne érigée par la NAACP. Par conséquent, une attention dirigée davantage sur le contexte social et politique américain entre 1915 et 1922 révèle que pour la NAACP, la dénonciation de l’occupation américaine d’Haïti représentait d’une part une opportunité de discuter des problèmes sociaux touchant les Afro-Américains, et d’autre part, une occasion de renforcer sa position aux États-Unis.

Crises cause social disturbances within their host organisation and the patterns of interpersonal ties that emerge are an important determinant of crisis management efficiency. In this article, social network analysis is used within a construction project context, to demonstrate that efficient crisis management depends upon the design and maintenance of an appropriate social fabric. However, crises have defence mechanisms that make management difficult by inducing forces that encourage people to pursue inappropriate social ties. Purposeful social intervention is therefore an essential part of the crisis management process to confront and avoid disorganisation.
