A method was developed to determine simazine, atrazine and their metabolite, 2-chloro-4,6-diamino-1,3,5-triazine, in urine. The presence of these herbicides in urine may reflect possible exposure to pesticides. Sample preparation involved protein precipitation and solid-phase extraction. The samples were analyzed by high-performance liquid chromatography-mass spectrometry. The detection limits were 0.4 mug/l and the analytes have a linear response in the interval 6-800 mug/l. The precision of the method was reflected in the RSD of <2.4% for the herbicides studied. Based on the detectable herbicide levels from spiked urine samples collected from unexposed volunteers, this method can be used to determine the low levels necessary for establishing reference values of the selected herbicides and the metabolite. A rapid analytical approach suitable to achieve a comprehensive characterization of the compounds present in the infusion prepared from the leaves of Byrsonima fagifolia Niedenzu (Malpighiaceae), a Brazilian plant used as an infusion to treat gastric disorders, was developed. The method was based on high-performance liquid chromatography coupled to electrospray negative ionisation multistage ion trap mass spectrometry (HPLC/ESI-ITMSn). The main ions in the ESI-ITMS spectra were attributed to a quinic acid core containing from one to five galloyl units. Quercetin derivatives containing one and two sugar moieties as well as galloyl esterification were also detected. These results indicated that HPLC/ESI-ITMSn is easily applicable to infusions of this plant and allows the rapid and direct identification of these compounds in crude plant extracts. A fast and reliable method, based on high-performance liquid chromatography coupled to electrospray ionization ion trap tandem mass spectrometry (HPLC/ESI-ITMS), was developed to investigate the infusion prepared from the leaves of Byrsonima crassa Niedenzu (Malpighiaceae), a native plant used in Brazil against gastric disorders. The use of on-line reverse-phase HPLC/ESI-ITMS allowed separation of three major classes of compounds and identification of over 20 very polar compounds characterized as galloylquinic acids, proanthocyanidins, and flavonoid glycosides, as well as the dimeric flavonoid amentoflavone and minor amounts of galloyl hexose and galloyl saccharose. The microbiological bioassay, the UV-spectrophotometry and the high performance liquid chromatography (HPLC) methods for assaying sparfloxacin in tablets were compared. The accuracy, repeatability, and precision of each method was assessed and precise. All methods were reliable within acceptable limits for antibiotic pharmaceutical preparations being accurate and precise. The microbiological bioassay and HPLC are more specific than UV-spectrophotometric analysis. However, the microbiological bioassay requires 20 h to get results, and HPLC is the most expensive analysis. The application of each method as a routine analysis should be investigated considering cost, simplicity, equipment, solvents, speed, and application to large or small workloads.
Separation of the toxic zierin from Zollernia ilicifolia by high speed countercurrent chromatography

Preliminary pharmacological assays of the 70% methanol extract from the leaves of the Brazilian medicinal plant Zollernia ilicifolia Vog. (Fabaceae) showed analgesic and antiulcerogenic effects. Previous analyses have shown that this extract contains, besides flavonoid glycosides and saponins, a toxic cyanogenic glycoside. Flavonoids and saponins are compounds reported in literature with antiulcerogenic activity. In this work, we developed a methodology to separate the cyanogenic glycoside from these compounds in order to obtain enough amount of material to perform pharmacological assays. The cyanogenic glycoside zierin (2S)-β-D-glucopyranosyloxy-(3-hydroxy-phenyl)- acetonitrile was separated from the other components by high speed countercurrent chromatography (HSCCC). The solvent system used was composed of chloroform-methanol-n-propanol-water (5:6:1:4, v/v/v/v). This technique led to the separation of zierin from the possible active compounds of Zollernia ilicifolia.

A rational and selective method using on-line high-performance liquid chromatography (HPLC) coupled with electrospray quadrupole time-of-flight tandem mass spectrometry (ESI-QToF-MS/MS) was established for the dereplication of phenolic derivatives from Qualea grandiflora and Qualea cordata. The selection of the extracts was based on the antioxidant capacity measured by in vitro DPPH assay. The HPLC-ESI-QToF-MS/MS analysis was conducted by on-flow detection, using high-resolution mass/ratio ions as well as collision induced MS/MS experiments for selected protonated ions. The dereplication of the EtOAc fraction from the hydro alcohol extract from the stem bark of Q. grandiflora allowed the detection of the flavonoids: 3',4',5',5,6,7-hexahydroxy- 8 methylflavanone, 8-methyl-naringenine and 3',7-dimethoxy-8 methyl-4',5,7- trihydroxyflavanone, as well as a benzophenone derivatives: bis(4,6-dimethoxy-2- hydroxy-3-methylphenyl)- metanone, 3',4'-dimethoxy-8-methyl-5,6,7 trihydroxyflavanone, 7-methoxy-6-methyl- 3',4',5 trihydroxyflavanone, 6,8-dimethyl-3' methoxy-4',5,7 trihydroxyflavanone and 3',5'-dimethoxy-6,8- dimethyl-4',5,7 trihydroxyflavanone were detected in the EtOAc fraction from the hydro-alcohol extract from the leaves of Q. cordata. © 2013 Sociedade Brasileira de Química.

Validation of analytical methodology for quantification of cefazolin sodium by liquid chromatography

A reversed-phase high performance liquid chromatography method was validated for the determination of cefazolin sodium in lyophilized powder for solution for injection to be applied for quality control in pharmaceutical industry. The liquid chromatography method was conducted on a Zorbax Eclipse Plus C18 column (250 x 4.6 mm, 5 μm), maintained at room temperature. The mobile phase consisted of purified water: acetonitrile (60: 40 v/v), adjusted to pH 8 with triethylamine. The flow rate was of 0.5 mL min-1 and effluents were monitored at 270 nm. The retention time for cefazolin sodium was 3.6 min. The method proved to be linear (r2 =0.9999) over the concentration range of 30-80 µg mL-1. The selectivity of the method was proven through degradation studies. The method demonstrated satisfactory results for precision, accuracy, limits of detection and quantitation. The robustness of this method was evaluated using the Plackett–Burman fractional factorial experimental design with a matrix of 15 experiments and the statistical treatment proposed by Youden and Steiner. Finally, the proposed method could be also an advantageous option for the analysis of cefazolin sodium, contributing to improve the quality control and to assure the therapeutic efficacy

A simple and sensitive analytical method for simultaneous determination of anastrozole, bicalutamide, and tamoxifen as well as their synthetic impurities, anastrozole pentamethyl, bicalutamide 3-fluoro-isomer, and tamoxifen e-isomer, was developed and validated by using high performance liquid chromatography (HPLC). The separation was achieved on a Symmetry (R) C-8 column (100 x 4.6 mm i.d., 3.5 mu m) at room temperature (+/- 24 degrees C), with a mobile phase consisting of acetonitrile/water containing 0.18% N,N dimethyloctylamine and pH adjusted to 3.0 with orthophosphoric acid (46.5/53.5, v/v) at a flow rate of 1.0 mL min(-1) within 20 min. The detection was made at a wavelength of 270 nm by using ultraviolet (UV) detector. No interference peaks from excipients and relative retention time indicated the specificity of the method. The calibration curve showed correlation coefficients (r) > 0.99 calculated by linear regression and analysis of variance (ANOVA). The limit of detection (LOD) and limit of quantitation (LOQ), respectively, were 2.2 and 6.7 mu g mL(-1) for anastrozole, 2.61 and 8.72 mu g mL(-1) for bicalutamide, 2.0 and 6.7 mu g mL(-1) for tamoxifen, 0.06 and 0.22 mu g mL(-1) for anastrozole pentamethyl, 0.02 and 0.07 mu g mL(-1) for bicalutamide 3-fluoro-isomer, and 0.002 and 0.007 mu g mL(-1) for tamoxifen e-isomer. Intraday and interday relative standard deviations (RSDs) were <2.0% (drugs) and <10% (degradation products) as well as the comparison between two different analysts, which were calculated by f test. A universal and robust analytical method for the determination of Δ9-tetrahydrocannabinol (THC) and two of its metabolites Δ9-(11-OH)-tetrahydrocannabinol (11-OH-THC) and 11-nor-Δ9-carboxy-tetrahydrocannabinol (THC-COOH) in human whole blood was developed and validated for use in forensic toxicology. Protein precipitation, integrated solid phase extraction and on-line enrichment followed by high-performance liquid chromatography separation and detection with a triple quadrupole mass spectrometer were combined. The linear ranges used for the three cannabinoids were from 0.5 to 20 ng/mL for THC and 11-OH-THC and from 2.5 to 100 ng/mL for THC-COOH, therefore covering the requirements for forensic use. Correlation coefficients of 0.9980 or better were achieved for all three analytes. No relevant hydrolysis was observed for THC-COOH glucuronide with this procedure--in contrast to our previous GC-MS procedure, which obviously lead to an artificial increase of the THC-COOH concentration due to the hydrolysis of the glucuronide-conjugate occurring at high pH during the phase-transfer catalyzed methylation step.

New directly acting antivirals (DAAs) that inhibit hepatitis C virus (HCV) replication are increasingly used for the treatment of chronic hepatitis C. A marked pharmacokinetic variability and a high potential for drug-drug interactions between DAAs and numerous drug classes have been identified. In addition, ribavirin (RBV), commonly associated with hemolytic anemia, often requires dose adjustment, advocating for therapeutic drug monitoring (TDM) in patients under combined antiviral therapy. However, an assay for the simultaneous analysis of RBV and DAAs constitutes an analytical challenge because of the large differences in polarity among these drugs, ranging from hydrophilic (RBV) to highly lipophilic (telaprevir [TVR]). Moreover, TVR is characterized by erratic behavior on standard octadecyl-based reversed-phase column chromatography and must be separated from VRT-127394, its inactive C-21 epimer metabolite. We have developed a convenient assay employing simple plasma protein precipitation, followed by high-performance liquid chromatography coupled to tandem mass spectrometry (HPLC-MS/MS) for the simultaneous determination of levels of RBV, boceprevir, and TVR, as well as its metabolite VRT-127394, in plasma. This new, simple, rapid, and robust HPLC-MS/MS assay offers an efficient method of real-time TDM aimed at maximizing efficacy while minimizing the toxicity of antiviral therapy.

Purpose: To develop a high-performance liquid chromatography (HPLC) fingerprint method for the quality control and origin discrimination of Gastrodiae rhizoma . Methods: Twelve batches of G. rhizoma collected from Sichuan, Guizhou and Shanxi provinces in china were used to establish the fingerprint. The chromatographic peak (gastrodin) was taken as the reference peak, and all sample separation was performed on a Agilent C18 (250 mm×4.6 mmx5 μm) column with a column temperature of 25 °C. The mobile phase was acetonitrile/0.8 % phosphate water solution (in a gradient elution mode) and the flow rate of 1 mL/min. The detection wavelength was 270 nm. The method was validated as per the guidelines of Chinese Pharmacopoeia. Results: The chromatograms of the samples showed 11 common peaks, of which no. 4 was identified as that of Gastrodin. Data for the samples were analyzed statistically using similarity analysis and hierarchical cluster analysis (HCA). The similarity index between reference chromatogram and samples’ chromatograms were all > 0.80. The similarity index of G. rhizoma from Guizhou, Shanxi and Sichuan is evident as follows: 0.854 - 0.885, 0.915 - 0.930 and 0.820 - 0.848, respectively. The samples could be divided into three clusters at a rescaled distance of 7.5: S1 - S4 as cluster 1; S5 - S8 cluster 2, and others grouped into cluster 3. Conclusion: The findings indicate that HPLC fingerprinting technology is appropriate for quality control and origin discrimination of G. rhizoma.

Os incêndios florestais são uma importante fonte de emissão de compostos gasosos e de aerossóis. Em Portugal, onde a maioria dos incêndios ocorre no norte e centro do país, os incêndios destroem todos os anos milhares de hectares, com importantes perdas em termos económicos, de vidas humanas e qualidade ambiental. As emissões podem alterar consideravelmente a química da atmosfera, degradar a qualidade do ar e alterar o clima. Contudo, a informação sobre as caraterísticas das emissões dos incêndios florestais nos países do Mediterrâneo é limitada. Tanto a nível nacional como internacional, existe um interesse crescente na elaboração de inventários de emissões e de regulamentos sobre as emissões de carbono para a atmosfera. Do ponto de vista atmosférico da monitorização atmosférica, os incêndios são considerados um desafio, dada a sua variabilidade temporal e espacial, sendo de esperar um aumento da sua frequência, dimensão e severidade, e também porque as estimativas de emissões dependem das caraterísticas dos biocombustíveis e da fase de combustão. O objetivo deste estudo foi quantificar e caraterizar as emissões de gases e aerossóis de alguns dos mais representativos incêndios florestais que ocorreram no centro de Portugal nos verões de 2009 e de 2010. Efetuou-se a colheita de amostras de gases e de duas frações de partículas (PM2.5 e PM2.5-10) nas plumas de fumo em sacos Tedlar e em filtros de quartzo acoplados a um amostrador de elevado volume, respetivamente. Os hidrocarbonetos totais (THC) e óxidos de carbono (CO e CO2) nas amostras gasosas foram analisados em instrumentos automáticos de ionização de chama e detetores não dispersivos de infravermelhos, respetivamente. Para algumas amostras, foram também quantificados alguns compostos de carbonilo após reamostragem do gás dos sacos Tedlar em cartuchos de sílica gel revestidos com 2,4-dinitrofenilhidrazina (DNPH), seguida de análise por cromatografia líquida de alta resolução. Nas partículas, analisou-se o carbono orgânico e elementar (técnica termo-óptica), iões solúveis em água (cromatografia iónica) e elementos (espectrometria de massa com plasma acoplado por indução ou análise instrumental por ativação com neutrões). A especiação orgânica foi obtida por cromatografia gasosa acoplada a espectrometria de massa após extração com recurso a vários solventes e separação dos extratos orgânicos em diversas classes de diferentes polaridades através do fracionamento com sílica gel. Os fatores de emissão do CO e do CO2 situaram-se nas gamas 52-482 e 822-1690 g kg-1 (base seca), mostrando, respetivamente, correlação negativa e positiva com a eficiência de combustão. Os fatores de emissão dos THC apresentaram valores mais elevados durante a fase de combustão latente sem chama, oscilando entre 0.33 e 334 g kg-1 (base seca). O composto orgânico volátil oxigenado mais abundante foi o acetaldeído com fatores de emissão que variaram desde 1.0 até 3.2 g kg-1 (base seca), seguido pelo formaldeído e o propionaldeído. Observou-se que as emissões destes compostos são promovidas durante a fase de combustão latente sem chama. Os fatores de emissão de PM2.5 e PM10 registaram valores entre 0.50-68 e 0.86-72 g kg-1 (base seca), respetivamente. A emissão de partículas finas e grosseiras é também promovida em condições de combustão lenta. As PM2.5 representaram cerca de 90% da massa de partículas PM10. A fração carbonosa das partículas amostradas em qualquer dos incêndios foi claramente dominada pelo carbono orgânico. Foi obtida uma ampla gama de rácios entre o carbono orgânico e o carbono elementar, dependendo das condições de combustão. Contudo, todos os rácios refletiram uma maior proporção de carbono orgânico em relação ao carbono elementar, típica das emissões de queima de biomassa. Os iões solúveis em água obtidos nas partículas da pluma de fumo contribuíram com valores até 3.9% da massa de partículas PM2.5 e 2.8% da massa de partículas de PM2.5-10. O potássio contribuiu com valores até 15 g mg-1 PM2.5 e 22 g mg-1 PM2.5-10, embora em massa absoluta estivesse maioritariamente presente nas partículas finas. Os rácios entre potássio e carbono elementar e entre potássio e carbono orgânico obtidos nas partículas da pluma de fumo enquadram-se na gama de valores relatados na literatura para emissões de queima de biomassa. Os elementos detetados nas amostras representaram, em média, valores até 1.2% e 12% da massa de PM2.5 e PM2.5-10, respetivamente. Partículas resultantes de uma combustão mais completa (valores elevados de CO2 e baixos de CO) foram caraterizadas por um elevado teor de constituintes inorgânicos e um menor conteúdo de matéria orgânica. Observou-se que a matéria orgânica particulada é composta principalmente por componentes fenólicos e produtos derivados, séries de compostos homólogos (alcanos, alcenos, ácidos alcanóicos e alcanóis), açúcares, biomarcadores esteróides e terpenóides, e hidrocarbonetos aromáticos policíclicos. O reteno, um biomarcador das emissões da queima de coníferas, foi o hidrocarboneto aromático dominante nas amostras das plumas de fumo amostradas durante a campanha que decorreu em 2009, devido ao predomínio de amostras colhidas em incêndios em florestas de pinheiros. O principal açúcar anidro, e sempre um dos compostos mais abundantes, foi o levoglucosano. O rácio levoglucosano/OC obtido nas partículas das plumas de fumo, em média, registaram valores desde 5.8 a 23 mg g-1 OC. Os rácios levoglucosano/manosano e levoglucosano/(manosano+galactosano) revelaram o predomínio de amostras provenientes da queima de coníferas. Tendo em conta que a estimativa das emissões dos incêndios florestais requer um conhecimento de fatores de emissão apropriados para cada biocombustível, a base de dados abrangente obtida neste estudo é potencialmente útil para atualizar os inventários de emissões. Tem vindo a ser observado que a fase de combustão latente sem chama, a qual pode ocorrer simultaneamente com a fase de chama e durar várias horas ou dias, pode contribuir para uma quantidade considerável de poluentes atmosféricos, pelo que os fatores de emissão correspondentes devem ser considerados no cálculo das emissões globais de incêndios florestais. Devido à falta de informação detalhada sobre perfis químicos de emissão, a base de dados obtida neste estudo pode também ser útil para a aplicação de modelos no recetor no sul da Europa.

Chitosan is a natural polymer, biodegradable, nontoxic, high molecular weight derived from marine animals, insects and microorganisms. Oligomers of glucosamine (GlcN) and N-acetylglucosamine (GlcNAc) have interesting biological activities, including antitumor effects, antimicrobial activity, antioxidant and others. The alternative proposed by this work was to study the viability of producing chitooligosaccharides using a crude enzymes extract produced by the fungus Metarhizium anisopliae. Hydrolysis of chitosan was carried out at different times, from 10 to 60 minutes to produce chitooligosaccharides with detection and quantification performed by High Performace Liquid Chromatography (HPLC). The evaluation of cytotoxicity of chitosan oligomers was carried out in tumor cells (HepG2 and HeLa) and non-tumor (3T3). The cells were treated for 72 hours with the oligomers and cell viability investigated using the method of MTT. The production of chitosan oligomers was higher for 10 minutes of hydrolysis, with pentamers concentration of 0.15 mg/mL, but the hexamers, the molecules showing greater interest in biological properties, were observed only with 30 minutes of hydrolysis with a concentration of 0.004 mg/mL. A study to evaluate the biological activities of COS including cytotoxicity in tumor and normal cells and various tests in vitro antioxidant activity of pure chitosan oligomers and the mixture of oligomers produced by the crude enzyme was performed. Moreover, the compound with the highest cytotoxicity among the oligomers was pure glucosamine, with IC50 values of 0.30; 0.49; 0.44 mg/mL for HepG2 cells, HeLa and 3T3, respectively. Superoxide anion scavenging was the mainly antioxidant activity showed by the COS and oligomers. This activity was also depending on the oligomer composition in the chitosan hydrolysates. The oligomers produced by hydrolysis for 20 minutes was analyzed for the ability to inhibit tumor cells showing inhibition of proliferation only in HeLa cells, did not show any effect in HepG2 cells and fibroblast cells (3T3)
