A major goal in vaccine development is elimination of the 'cold chain', the transport and storage system for maintenance and distribution of the vaccine product. This is particularly pertinent to liquid formulation of vaccines. We have previously described the rod-insert vaginal ring (RiR) device, comprising an elastomeric body into which are inserted lyophilised, rod-shaped, solid drug dosage forms, and having potential for sustained mucosal delivery of biomacromolecules, such as HIV envelope protein-based vaccine candidates. Given the solid, lyophilised nature of these insert dosage forms, we hypothesised that antigen stability may be significantly increased compared with more conventional solubilised vaginal gel format. In this study, we prepared and tested vaginal ring devices fitted with lyophilised rod inserts containing the model antigen bovine serum albumin (BSA). Both the RiRs and the gels that were freeze-dried to prepare the inserts were evaluated for BSA stability using PAGE, turbidimetry, microbial load, MALDI-TOF and qualitative precipitate solubility measurements. When stored at 4°C, but not when stored at 40°C/75% RH, the RiR formulation offered protection against structural and conformational changes to BSA. The insert also retained matrix integrity and release characteristics. The results demonstrate that lypophilised gels can provide relative protection against degradation at lower temperatures compared to semi-solid gels. The major mechanism of degradation at 40°C/75% RH was shown to be protein aggregation. Finally, in a preliminary study, we found that addition of trehalose to the formulation significantly reduces the rate of BSA degradation compared to the original formulation when stored at 40°C/75% RH. Establishing the mechanism of degradation, and finding that degradation is decelerated in the presence of trehalose, will help inform further development of RiRs specifically and polymer based freeze-dried systems in general.

This dissertation attempts to retrieve the integration of prayer and theology in the life of the church. Prayer is a spiritual and bodily theological activity that forms Christian identity and virtuous character. The bodily dimension of Christian prayer plays an essential role in theological understanding and moral formation. However, the embodiment of prayer has been mostly neglected in modern academic theology. This study highlights the significance of the body at prayer in theological studies and spiritual formation.

Chapter 1 presents Karl Barth’s theology of prayer as a model of the integration of prayer, theology, and Christian life (lex orandi, lex credendi, lex agendi). However, Barth’s attempt to overcome the dichotomy between theory and practice in theology did not pay much attention to embodiment of prayer. Through ritual studies and phenomenology (Marcel Mauss, Maurice Merleau-Ponty, and Pierre Bourdieu), chapter 2 shows why the bodily dimension of the practice of prayer should be recovered in theology and ministry; then it explains how Christians in the early and medieval church actually prayed with the body, how their bodily actions were understood in their theological paradigms, and how their actions contributed to the formation of Christian character. Chapter 3 narrows the focus to the formation of the heart in the making of Christian character. The practice of prayer has been emphasized not only as an expression of the inner heart of pray-ers but also as a channel of grace that shapes their affections as enduring dispositions of the heart. Furthermore, historically the bodily practice of prayer gave theological authority to the devout Christians who were marginalized in academic theology or ecclesiastical hierarchy, and Chapter 4 presents the lex orandi of praying women who gained their theological knowledge, wisdom, and authority through their exemplary practices of prayer (Catherine of Siena, Mechthild of Magdeburg, Julian of Norwich, Margery Kempe, and Teresa of Avila). These historical examples reveal how Christian communities appreciated and celebrated the theological voices from the margins, which developed from theological embodiments in prayer.

This dissertation concludes that academic theology needs to heed these diverse theological voices, which are nurtured through everyday practice, as an integral part of theological studies. Therefore, it calls for a new paradigm for understanding the relationship between theory and practice in theological education. The integration between theory and bodily practice is necessary for both academic theology and spiritual formation. A more holistic understanding of Christian practices will not only enhance the training of scholars and clergy but also give the laity their own theological voices that will enrich academic theology.

However, it was pointed out that at the level of technical details the two concepts are significantly different. One possible explanation that we suggest is that the process of falsification is an active process, whereas statistical learning theory is mainly concerned with supervised learningSupervised learning, which is a passive process of learning from examples arriving from a stationary distribution. We show that concepts that are closer (although still distant) to Karl Popper’s definitions of falsifiability can be found in the domain of learning using membership queries, and derive relations between Popper’s dimension, exclusion dimension, and the VC-dimensionVC dimension.

The memoirs were originally written for the Harvard University competition in 1940 and were translated by the author in 2001. Reflections on his childhood in Germany and Austria. His parents were both from Poland. They moved to Vienna in 1921, where his father opened a haberdashery store in the Second district (Leopoldstadt). Otto attended primary school in Czerningasse. Birth of his sister Cecile in 1924. After his failing business endeavors his father decided to move back to Germany, where the family opened a department store in Elbing, East Prussia. Otto attended Gymnasium, where he was one of only two Jewish students in his class. Growing Nazi movement among students. Summer vacations on the Baltic Sea. Private piano lessons. Hitler’s rise in Germany and life under National Socialism. Bar mitzvah in 1933. Anti-Jewish boycotts. His father fled to Vienna in order to escape a rounding up of Jews. The family followed soon after to Austria. Otto attended Gymnasium in the Zirkusgasse and started to work as a tutor. Member of a youth group and hiking tours in the mountains. Recollections of the Anschluss in 1938. Fervent attempts to obtain an exit visa for the United States, where they had a relative in New York. Description of discriminations and frequent attacks on Jewish friends and relatives in the weeks after the Anschluss. Otto was picked up by Nazi stormtroops. He was forced to hold up an anti-Jewish sign and was walked up and down, receiving beatings and spittings in front of a jeering crowd. Detailed account of the atmosphere within the Jewish population. The Gymnasium Zirkusgasse was transferred into a Jewish school. Frequent attacks of Hitler Youths on the students. Preparations for the “Matura” despite the turmoil. The book contains an introduction by Paul Weisz and a collection of family letters written during World War II. The letters were written between February 1938 and September 1945. Some were translated into English and complemented by commentary by the editor, Paul Weisz. Paul Weisz' introduction is 10 pages long and serves as a short memoir by itself. He provides a family chronicle, the living circumstances of his family, and his childhood in Vienna. He ends in 1938 when the family was eager to leave Austria. The following years are covered by the various letters he brought together in this book. The authors are cousin Willie, then already in Palestine, his father Samuel, his mother Stephanie, and his sister Ruth. His father and mother fled to Belgium, but were arrested after the beginning of World War II. They were deported to internment camps in France (St. Cyprien). His sister Ruth tried to escape from Austria to Palestine via the Danube. She got stuck in Yugoslavia, and was interned in Sabac internment camp. Paul's mother died in France in 1942, his father was sent to a concentration camp in Poland and murdered. His sister Ruth was murdered in Yugoslavia. Paul was released in Canada, and was enabled to go to college. He later named his children after his family members who did not survive the Nazi terror: Stephanie, Ruth, and Samuel.

Esta tese tem como objetivo investigar os modos de articulação da produção e socialização do conhecimento na perspectiva do tratamento de problemáticas regionais, e sua relação com as políticas acadêmicas na Universidade Federal Fluminense em função de sua inserção na região onde se constrói, atualmente, o Complexo Petroquímico do Estado do Rio de Janeiro COMPERJ. Parte da concepção de universidade de Karl Jaspers, considerada um modelo neo-humboltiano, para depois situar o pensamento educacional de Anísio Teixeira, Florestan Fernandes e Darcy Ribeiro, como base para a formação da ideia de universidade no Brasil. As contribuições de diferentes autores sobre a formação das universidades brasileiras, o contexto histórico, social e econômico em que se inserem e como discutem os conceitos de indissociabilidade entre ensino e pesquisa, autonomia universitária e legitimidade da universidade são apresentas. Tendo como referência autores que se preocupam com as transformações impostas à sociedade pelo uso intensivo da tecnologia, pelas mudanças na dinâmica territorial, pela reorganização do espaço geográfico, pelo crescimento das desigualdades nas cidades, apresenta-se contribuições para o diálogo com o objeto de estudo. Por se tratar de objeto em construção, escolheu-se a pesquisa qualitativa como metodologia de estudo, por permitir captar, no seu próprio movimento, um conjunto de processos de mudanças institucionais no mesmo momento de sua manifestação. O estudo demonstrou possibilidades de interlocução bastante profícuas, com resultados expressivos na direção da produção do saber novo, contextualizado, ao apresentar a relação desses estudos e pesquisas com o ensino e a extensão praticados pelos docentes. Se, no entanto, para que se crie um clima competição interlocal visando atrair capitais privados e recursos de programas do governo federal, forem impostas condições que coloquem em risco a liberdade intelectual e a autonomia universitária, o projeto de universidade que se defende estará irremediavelmente comprometido. Espera-se que o resultado desta pesquisa possa contribuir com as discussões teóricas referentes ao papel da universidade na sociedade.
