O extrato butílico da Phytolacca dodecandra (tipo 44) obtido de frutos procedentes da Etiópia, foi testado como moluscicida em nosso laboratório e no campo. As concentrações letais, CL90, com 24 horas de exposição, para Biomphalaria glabrata adulta, recém-eclodidas e desovadas foram de 4,5, 23,0 e 102,0 ppm, respectivamente. Para peixes, Lebistes reticulatus, a CL90 foi de 2,0 ppm. Esses resultados foram semelhantes aos obtidos por Lemma em 1984, na Etiópia. Em dois criadouros com água parada, tratados com 10ppm de estrato butílico e 3ppm de niclosamida, ocorreu 84,0 e 100,0% de mortalidade B. glabrata, respectivamente. Os dois produtos foram tóxicos para peixes (L. reticulatus) no campo. É discutida a possibilidade do uso de moluscidas de origem vegetal, como alternativa para o combate a focos de esquistossomose no Brasil.

En aquesta tesis es presenten els resultats de la investigació duta a terme a les comunitats indígenes Tsimane’ de l’Amazònia boliviana. La investigació estudia la percepció dels indígenes sobre l’etnoclassificació del seu territori. S’estableix una clau de classificació i es determina la importància dels elements paisatgístics del territori Tsimane’ segons la percepció local. Aquesta informació permetrà integrar el coneixement local dins dels programes de desenvolupament integral i de planificació territorial en l’Amazònia Boliviana. L’estudi conclou que la població Tsimane’ classifica els elements paisatgístics del seu entorn en 89 taques conformades per una espècies arbòria dominant i que estan incloses en un o més dels nou paisatges identificats: Därsi Därä, Sajras, Sinues Ojñi’, Mayes, Múcúya, Tsäquis Därä, Cum, Tajñi’ i Jaman. A partir d’un anàlisi multicriteri s’ha determinat una importància total per cada paisatge segons els següents criteris d’importància: diversitat de taques, activitats econòmiques realitzables, presència espiritual, percepció individual i importància relativa segons els altres paisatges. Així doncs s’ha trobat que el paisatge més important és el Därsi Därä (bosc primari caracteritzat per un estrat arbori superior a 50 metres d’altura). També s’han analitzat les dades discernint segons el gènere de l’entrevistat i segons la proximitat de les comunitats estudiades a la ciutat més propera.

Malaria remains globally the most important parasitic disease of man. Data on its deleterious effects during pregnancy have been extensively documented in hyperendemic, holoendemic, and mesoendemic areas from Africa and Asia where Plasmodium falciparum is responsible for almost all infections. However, knowledge about malaria during pregnancy in areas where transmission is unstable and P. vivax is the most prevalent species, such as the Brazilian Amazon, is scarce. Here, we report a preliminary cross sectional descriptive study, carried out at the Fundação de Medicina Tropical do Amazonas, a reference centre for diagnosis and treatment of tropical diseases in the west-Amazon (Manaus, Brazil). A total of 1699 febrile childbearing age women had positive thick blood smears to Plasmodium species, between January and November 1997: 1401 (82.5%) were positive for P. vivax , 286 (16.8%) for P. falciparum and 12 (0.07%) carried mixed infections. From the malarious patients, 195 were pregnant. The ratio of P. falciparum to P. vivax infections in the group of non-pregnant infected women was 1:5.6 while it was 1:2.3 in that of pregnant infected ones. Similar rates or even proportionally more vivax infections during pregnancy were expected to occur, in function of the contraindication of primaquine with the resulting increased P. vivax relapse rates. Such an observation suggests that the mechanism of resistance/susceptibility to infection and/or malaria pathogenesis in pregnant women may differ according to Plasmodium species and that the extensively described increase in the frequencies of malaria infection during pregnancy may be specifically due to P. falciparum infection.

In this work we investigated the frequency of polymorphism in exon II of the gene encoding most of the amino-terminal region of the serine rich antigen (SERA) in Plasmodium falciparum field samples. The blood samples were colleted from P. falciparum infected individuals in three areas of the Brazilian Amazon. Two fragments have been characterized by polymerase chain reaction: one of 175 bp corresponding to the repeat region with 5 octamer units and one other of 199 bp related to the 6 repeat octamer units of SERA protein. The 199 bp fragment was the predominant one in all the studied areas. The higher frequency of this fragment has not been described before and could be explained by an immunological selection of the plasmodial population in the infected individuals under study. Since repeat motifs in the amino-terminal region of SERA contain epitopes recognized by parasite-inhibitor antibodies, data reported here suggest that the analysis of the polymorphism of P. falciparum isolates in different geographical areas is a preliminary stage before the final drawing of an universal vaccine against malaria can be reached.

In the present study, a quassinoid, neosergeolide, isolated from the roots and stems of Picrolemma sprucei (Simaroubaceae), the indole alkaloids ellipticine and aspidocarpine, isolated from the bark of Aspidosperma vargasii and A. desmanthum (Apocynaceae), respectively, and 4-nerolidylcatechol, isolated from the roots of Pothomorphe peltata (Piperaceae), all presented significant in vitro inhibition (more active than quinine and chloroquine) of the multi-drug resistant K1 strain of Plasmodium falciparum. Neosergeolide presented activity in the nanomolar range. This is the first report on the antimalarial activity of these known, natural compounds. This is also the first report on the isolation of aspidocarpine from A. desmanthum. These compounds are good candidates for pre-clinical tests as novel lead structures with the aim of finding new antimalarial prototypes and lend support to the traditional use of the plants from which these compounds are derived.

A study of crepuscular and night-biting mosquitoes was conducted at remote settlements along the Padauiri River, middle Negro River, state of Amazonas, Brazil. Collections were performed with human bait and a CDC-light trap on three consecutive days per month from June 2003-May 2004. In total, 1,203 h of collection were performed, of which 384 were outside and 819 were inside houses. At total of 11,612 specimens were captured, and Anophelinae (6.01%) were much less frequent than Culicinae (93.94%). Anopheles darlingi was the most frequent Anophelinae collected. Among the culicines, 2,666 Culex (Ae.) clastrieri Casal & Garcia, 2,394 Culex. (Mel.) vomerifer Komp, and 1,252 Culex (Mel.) eastor Dyar were the most frequent species collected. The diversity of insects found reveals the receptivity of the area towards a variety of diseases facilitated by the presence of vectors involved in the transmission of Plasmodium, arboviruses and other infectious agents.

We report two ciprofloxacin and ofloxacin-resistant Neisseria gonorrhoeae strains that were isolated from the urethral discharge of male patients at the sexually transmitted diseases outpatient clinic of the Alfredo da Matta Foundation (Manaus, state of Amazonas, Brazil). The gonococci displayed minimal inhibitory concentrations (> 32.00 µg/mL) and three mutations in the quinolone resistance-determining region (S91F and D95G in GyrA and S87R in ParC). Both isolates were genotyped using N. gonorrhoeae multi-antigen sequence typing and the analysis showed that the ST225 which represented an emerging widespread multi-resistant clone that has also been associated with reduced susceptibility to ceftriaxone. We recommend continued surveillance of this pathogen to assess the efficacy of anti-gonococcal antibiotics in Brazil.

Artemisinin is the active antimalarial compound obtained from the leaves of Artemisia annua L. Artemisinin, and its semi-synthetic derivatives, are the main drugs used to treat multi-drug-resistant Plasmodium falciparum (one of the human malaria parasite species). The in vitro susceptibility of P. falciparum K1 and 3d7 strains and field isolates from the state of Amazonas, Brazil, to A. annua infusions (5 g dry leaves in 1 L of boiling water) and the drug standards chloroquine, quinine and artemisinin were evaluated. The A. annua used was cultivated in three Amazon ecosystems (várzea, terra preta de índio and terra firme) and in the city of Paulínia, state of São Paulo, Brazil. Artemisinin levels in the A. annua leaves used were 0.90-1.13% (m/m). The concentration of artemisinin in the infusions was 40-46 mg/L. Field P. falciparum isolates were resistant to chloroquine and sensitive to quinine and artemisinin. The average 50% inhibition concentration values for A. annua infusions against field isolates were 0.11-0.14 μL/mL (these infusions exhibited artemisinin concentrations of 4.7-5.6 ng/mL) and were active in vitro against P. falciparum due to their artemisinin concentration. No synergistic effect was observed for artemisinin in the infusions.

Tachia sp. are used as antimalarials in the Amazon Region and in vivo antimalarial activity of a Tachia sp. has been previously reported. Tachia grandiflora Maguire and Weaver is an Amazonian antimalarial plant and herein its cytotoxicity and antimalarial activity were investigated. Spectral analysis of the tetraoxygenated xanthone decussatin and the iridoid aglyone amplexine isolated, respectively, from the chloroform fractions of root methanol and leaf ethanol extracts was performed. In vitro inhibition of the growth of Plasmodium falciparum Welch was evaluated using optical microscopy on blood smears. Crude extracts of leaves and roots were inactive in vitro. However, chloroform fractions of the root and leaf extracts [half-maximal inhibitory concentration (IC50) = 10.5 and 35.8 µg/mL, respectively] and amplexine (IC50= 7.1 µg/mL) were active in vitro. Extracts and fractions were not toxic to type MRC-5 human fibroblasts (IC50> 50 µg/mL). Water extracts of the roots of T. grandiflora administered by mouth were the most active extracts in the Peters 4-day suppression test in Plasmodium berghei-infected mice. At 500 mg/kg/day, these extracts exhibited 45-59% inhibition five to seven days after infection. T. grandiflora infusions, fractions and isolated substance have potential as antimalarials.

Plasmodium vivax is the most widespread parasite causing malaria, being especially prevalent in the Americas and Southeast Asia. Children are one of the most affected populations, especially in highly endemic areas. However, there are few studies evaluating the therapeutic response of infants with vivax malaria. This study retrospectively evaluated the parasitaemia clearance in children diagnosed with vivax malaria during the first five days of exclusive treatment with chloroquine (CQ). Infants aged less than six months old had a significantly slower parasitaemia clearance time compared to the group of infants and children between six months and 12 years old (Kaplan-Meier survival analysis; Wilcoxon test; p = 0.004). The impaired clearance of parasitaemia in younger children with vivax malaria is shown for the first time in Latin America. It is speculated that CQ pharmacokinetics in young children with vivax malaria is distinct, but this specific population may also allow the detection of CQ-resistant parasites during follow-up, due to the lack of previous immunity.

Anaemia is amongst the major complications of malaria, a major public health problem in the Amazon Region in Latin America. We examined the haemoglobin (Hb) concentrations of malaria-infected patients and compared it to that of malaria-negative febrile patients and afebrile controls. The haematological parameters of febrile patients who had a thick-blood-smear performed at an infectious diseases reference centre of the Brazilian Amazon between December 2009-January 2012 were retrieved together with clinical data. An afebrile community control group was composed from a survey performed in a malaria-endemic area. Hb concentrations and anaemia prevalence were analysed according to clinical-epidemiological status and demographic characteristics. In total, 7,831 observations were included. Patients with Plasmodium falciparum infection had lower mean Hb concentrations (10.5 g/dL) followed by P. vivax-infected individuals (12.4 g/dL), community controls (12.8 g/dL) and malaria-negative febrile patients (13.1 g/dL) (p < 0.001). Age, gender and clinical-epidemiological status were strong independent predictors for both outcomes. Amongst malaria-infected individuals, women in the reproductive age had considerably lower Hb concentrations. In this moderate transmission intensity setting, both vivax and falciparum malaria are associated with reduced Hb concentrations and risk of anaemia throughout a wide age range.

Neste artigo, o objetivo é analisar como a pesquisa-ação tem sido aplicada nos estudos da área de Administração no Brasil. Apresenta-se um recorte da pesquisa-ação nos estudos brasileiros em Administração, escrutinando trabalhos de 44 periódicos nacionais de Administração, e 38 encontros ou simpósios da Associação Nacional de Pós-Graduação e Pesquisa em Administração (Anpad), entre os anos de 2000 e 2010. Identificaram-se 22 artigos em periódicos e quatro em eventos da Anpad que referenciavam pesquisa-ação, porém, apenas 21 compuseram o estrato final de artigos examinados. Analisaram-se objetivos, procedimentos, nível de engajamento de outsiders e consequências do ciclo acadêmico e do ciclo da ação, tanto para a ciência quanto para a realidade sob intervenção. Nos resultados da pesquisa, indica-se que, no geral, os autores não distinguem apropriadamente as duas intenções da metodologia, salvo poucas exceções. Especula-se, então, sobre a utilização da pesquisa-ação no campo de estudo da administração no Brasil. Conclui-se que o conceito de pesquisa-ação é reconhecido e internalizado pelos autores, mas os relatos de sua aplicação ainda são deficientes. Por fim, argumenta-se que pesquisa e ação na área de Administração podem e devem caminhar juntas e propõem-se algumas sugestões de pesquisas.

Como parte do Projeto PROVIVE, foram analisadas a riqueza de espécies, a composição taxonômica, a proporção de espécies raras e a constância taxonômica ao nível de família relacionada à riqueza de espécies, em comunidades de Coleoptera, em Vila Velha, Ponta Grossa, Paraná. Os dados foram obtidos a partir de coletas através de armadilha malaise no estrato do sub-bosque de cinco áreas com diferentes graus de intervenção antrópica, de setembro de 1999 a agosto de 2000. As 52 semanas de amostragem nas cinco áreas resultaram na coleta de 10.822 indivíduos de 1659 espécies. Todas as áreas apresentaram alta riqueza de espécies e diversidade, como indicado por vários índices. A área em estágio mais avançado de sucessão vegetal foi menos rica do que aquelas em estágio inicial/intermediária. De acordo com diferentes estimadores de riqueza de espécies, o número de espécies coletadas poderia aumentar de 22-123% com o aumento do esforço de coleta. As áreas menos conservadas foram mais ricas em espécies raras ("singletons", "doubletons" e únicas) do que as mais conservadas. Nas cinco áreas houve uma constância taxonômica entre as famílias mais ricas (Curculionidae, Chrysomelidae, Cerambycidae, Staphylinidae, Mordelidae, Elateridae, Scarabaeidae, Coccinellidae e Tenebrionidae) envolvendo 60% do total de espécies, como observado para a abundância de indivíduos. A existência de um padrão de constância taxonômica de famílias, quando considerados 60% da riqueza de espécies e/ou de abundância de indivíduos por local, poderá tornar mais fácil e rápido o estudo de comunidades de Coleoptera, habilitando a ordem a ser um táxon indicador de condições ambientais de áreas florestadas.

Este trabalho teve como objetivo obter dados sobre a ecologia da nidificação de Centris (Hemisiella) tarsata Smith em três ecossistemas: mata ciliar (MC), mata mesofítica (MM) e eucaliptal (EC), utilizandose ninhos-armadilha confeccionados em gomos de bambu, distribuídos em diferentes alturas: 1,5 m e 5-12 m do solo. Foram obtidos 41 ninhos: 31 no EC e 10 na MM, a maioria no estrato superior e com maior freqüência de nidificações ocorrendo no período de estiagem. A razão sexual foi de 1,9:1 (fêmeas/ machos) no EC e de 1,08:1 na MM. Cerca de 22% dos ninhos do EC e 40% da MM foram parasitados por Mesocheira bicolor (Fabricius, 1804) (Hymenoptera, Apidae) e Coelioxys sp. (Hymenoptera, Megachilidae). A análise polínica revelou predominância de grãos de pólen de Banisteriopsis sp. (Malpighiaceae) e Cassia sp. (Caesalpiniaceae) no EC e de espécies de Caesalpiniaceae Kunth. e Banisteriopsis Robinson na MM.
