O número de genótipos de girassol (Helianthus annuus L.) disponíveis aos agricultores é pequeno e pouco se conhece sobre os genótipos provenientes de outros países que possam ser utilizados no melhoramento genético desta cultura. O objetivo deste trabalho foi analisar a variabilidade genética de 31 acessos e de cinco cultivares nos sistemas isoenzimáticos fosfoglicomutase (PGM), 6fosfogliconato desidrogenase (PGD), fosfoglicoisomerase (PGI) e esterase (EST). Utilizou-se a técnica de eletroforese em gel de amido/penetrose para o fracionamento das isoenzimas da PGM, PGI e PGD e focalização isoelétrica para EST. Foram detectados um total de seis locos e 14 alelos para estes quatro sistemas. Observou-se variantes alélicas para os locos Pgi2, Pgm1, Pgd2, cada um com dois alelos, e para Est1, que apresentou seis alelos. Os testes de adequação aos modelos de equilíbrio de Hardy-Weinberg e de Wright evidenciaram que em dez acessos houve um desvio significativo de endocruzamento.

The objective of this work was to characterize 27 potato genotypes, using molecular markers. Polyacrylamide gel electrophoresis, RAPD techniques and isozymes of esterase, phosphoglucomutase and soluble proteins were analyzed in tubers, and isocitrate dehydrogenase, aspartate transaminase, phosphoglucomutase and peroxidase, in leaves. Eighteen primers were tested and four were chosen, kits OPX (01, 04 and 09) and OPY (07), to analyze RAPD markers in leaf extracts. Similarity and cluster analysis were conducted using Jaccard coefficient and the unweighted pair-group method using arithmetic average. Despite the differences detected in the analysis of proteins and isozymes in the tubers, as well as of isozymes in the leaves, the characterization of all genotypes through gel electrophoresis was not possible, while RAPD markers were efficient to characterize all the 27 genotypes.

En les darreres dècades la necessitat d’una major producció en l’agricultura ha implicat l’ús de productes químics per a millorar la producció. Entre aquests productes trobem els insecticides que, tot i ser específics per a determinades funcions, en molts casos acaben afectant també a altres organismes que no en són la diana. Els assajos d’ecotoxicitat són una eina clau per a determinar el grau d’afectació d’aquests insecticides. En aquest estudi es pretén determinar l’efecte de l’aplicació directa en el sòl de les dosis recomanades de l’insecticida Confidor 20SL (amb imidacloprid com a principi actiu) sobre Eisenia fetida i dels lixiviats d’aquests sòls sobre Daphnia magna i Selenastrum capricornutum simulant fenòmens d’escorrentia o lixiviació naturals. En el cas de E.fetida s’obté una LC50 de 24.71 mg/kg sòl i per la reproducció un valor de EC50 de 8.41 mg/kg sòl. S’observa allunyament en totes les dosis utilitzades i la EC50 és de 2.57 mg/kg sòl. No s’han pogut determinar efectes a nivell neurològic a partir de la determinació de l’activitat de l’ AChE. A l’exposar D.magna als lixiviats del sòl contaminat no s’han observat efectes clars ni en la mortalitat ni en la reproducció. El mateix succeeix amb S.capricornutum. Les dosis d’aplicació del pesticida Confidor, representen una amenaça per a E.fetida pel que fa a efectes subletals però no podem dir el mateix per als organismes aquàtics.

O objetivo deste trabalho foi elucidar a atividade e a expressão isoenzimática das esterases, das peroxidases e das aspartato aminotransferases em função da infecção de plantas de trigo pelo Soil-borne wheat mosaic virus (SBWMV). Foram analisadas, aos 45 dias após a emergência, quatro cultivares e uma linhagem de trigo, com diferentes níveis de resistência ao SBWMV: BRS Guabiju, BRS 194, BRS 179, BR 23 e PF 980524. De modo geral, ocorreram diferenças qualitativas e quantitativas intra e interpopulacional, quando comparadas plantas assintomáticas e sintomáticas ao SBWMV. Para o sistema esterase, nove padrões de bandas foram determinados e para peroxidase e aspartato aminotransferase foram detectados três padrões de bandas, para ambas as condições. Padrões eletroforéticos foram observados para plantas infectadas, quando comparadas com as não infectadas, destacando-se a atividade da esterase, o que permitiu identificar com maior precisão o estado metabólico e diferenciado das células.

Congenital myasthenic syndromes (CMS) are clinically and genetically heterogeneous inherited disorders characterized by impaired neuromuscular transmission. Mutations in the acetylcholinesterase (AChE) collagen-like tail subunit gene (COlQ) cause recessive forms of synaptic CMS with end plate AChE deficiency. We present data on 15 COLQ -mutant CMS carrying 16 different mutations (9 novel ones identified) followed-up for an average period of 10 ears. The mean age at the first examination was 19 ears old (range from 3 to 48). We report relapses during short or long-term periods characterized by worsening of muscle weakness sometimes associated with respiratory crises. All the relapses ended spontaneously or with 3-4 DAP or ephedrine with no residual impairment. The triggering factors identified were esterase inhibitors, effort, puberty or pregnancy highlighting the importance of hormonal factors. There was no genotype-phenotype correlation. At the end of the follow-up, 80% of patients were ambulant and 87% of patients had no respiratory trouble in spite of severe relapses.

O objetivo deste trabalho foi avaliar os perfis proteicos e o desempenho fisiológico de sementes de café submetidas a diferentes métodos de processamento e secagem. Foram avaliados os processamentos por via seca e úmida, e as secagens natural, em terreiro, e artificial a 60ºC, ou a 60ºC até 30% de umidade e 40ºC até teor final de 11% (base úmida). Após serem processadas e secadas, as sementes foram avaliadas quanto ao desempenho fisiológico e submetidas a análises bioquímicas, por meio da eletroforese de proteínas resistentes ao calor LEA ("late embryogenesis abundant") e das enzimas superóxido dismutase, catalase, peroxidase, esterase, polifenoloxidase, isocitrato desidrogenase, álcool desidrogenase e malato desidrogenase. O perfil proteico de sementes de café é afetado pelo método de processamento e de secagem. Os cafés processados por via úmida apresentam maior tolerância à secagem - revelada pela maior atividade de enzimas antioxidativas e pelo melhor desempenho fisiológico - do que os processados por via seca. A atividade de proteínas resistentes ao calor e de enzimas antioxidantes é variável promissora para diferenciar a qualidade dos cafés submetidos a diferentes manejos pós-colheita.

Obesity is associated with a chronic low-grade inflammation, and specific antiinflammatory interventions may be beneficial for the treatment of type 2 diabetes and other obesity-related diseases. The lipid kinase PI3Kγ is a central proinflammatory signal transducer that plays a major role in leukocyte chemotaxis, mast cell degranulation, and endothelial cell activation. It was also reported that PI3Kγ activity within hematopoietic cells plays an important role in obesity-induced inflammation and insulin resistance. Here, we show that protection from insulin resistance, metabolic inflammation, and fatty liver in mice lacking functional PI3Kγ is largely consequent to their leaner phenotype. We also show that this phenotype is largely based on decreased fat gain, despite normal caloric intake, consequent to increased energy expenditure. Furthermore, our data show that PI3Kγ action on diet-induced obesity depends on PI3Kγ activity within a nonhematopoietic compartment, where it promotes energetic efficiency for fat mass gain. We also show that metabolic modulation by PI3Kγ depends on its lipid kinase activity and might involve kinase-independent signaling. Thus, PI3Kγ is an unexpected but promising drug target for the treatment of obesity and its complications.
Alterações isoenzimáticas em sementes de cultivares de soja em diferentes condições de armazenamento

O objetivo deste trabalho foi determinar alterações fisiológicas e isoenzimáticas em sementes de genótipos de soja, em diferentes condições de armazenamento. Utilizou-se o delineamento experimental inteiramente casualizado com quatro repetições, em arranjo fatorial com seis cultivares de soja (TMG 1176, TMG 1179, TMG 132, TMG 133, TMG 115 e GB 874) e cinco periodos de armazenamento (0, 2, 4, 6 e 8 meses), em dois ambientes de armazenamento (câmara fria e seca, a 10ºC e 50% de umidade relativa; e armazém convencional, em condições não controladas). A qualidade fisiológica foi avaliada por meio de testes de germinação, de envelhecimento acelerado e de frio. As expressões isoenzimáticas determinadas foram as de malato desidrogenase (MDH), álcool desidrogenase (ADH), esterase (EST), isocitrato liase (ICL), superóxido dismutase (SOD) e peroxidase (PO). Sementes de soja armazenadas em câmara fria e seca conservaram sua qualidade fisiológica. Após seis meses de armazenamento, em condições não controladas, a qualidade das sementes e as atividades isoenzimáticas de MDH, ADH, EST, ICL, SOD e PO diminuíram. No armazenamento em câmara fria e seca, não ocorreu alteração nas sementes. Os genótipos de soja apresentam diferentes níveis de tolerância ao armazenamento e expressões isoenzimáticas.

A cultura da aceroleira despertou um grande interesse do mercado consumidor, tendo em vista o alto teor de vitamina C (ácido ascórbico), que varia entre 1.325 a 2.250 mg por 100 mL de suco. Com a expansão da cultura, surgiram problemas fitossanitários, entre os quais a infecção das raízes da aceroleira por nematóides. Os produtos químicos utilizados no controle dos nematóides são agressivos ao meio ambiente, e a seleção de genótipos resistentes e tolerantes constitui-se na melhor alternativa para a solução do problema. O trabalho foi desenvolvido em casa de vegetação com 18 genótipos de aceroleira, com o objetivo de encontrar genótipos resistentes e tolerantes a Meloidogyne incognita raça 2 assistida por marcadores isoenzimáticos, para indicar plantas destinadas a porta-enxerto. A avaliação foi realizada 60 dias após a inoculação mediante análise das variáveis: número de ovos por planta e por grama de raiz, índice de galhas e massa de ovos, biomassa fresca relativa do sistema radicular e biomassa fresca relativa da parte aérea. Os resultados permitiram identificar o genótipo 023-CMF como menos suscetível, e os genótipos 027-CMF e 035-CMF, como mais suscetíveis. Estudos realizados através da eletroforese isoenzimática com os sistemas α esterase, fosfatase ácida e peroxidase, 20; 40 e 60 dias após a inoculação com ovos de M. incognita raça 2, possibilitaram relacionar a expressão de proteínas com a suscetibilidade. Os genótipos mais próximos geneticamente, com índice de similaridade 0,941, foram 027-CMF e 026-CMF, 046-CMF e 026-CMF, e 041-CMF e 026-CMF. O menor índice de similaridade genética (0,115) foi observado entre os genótipos 002-SPE e 036-CMF.

Nutritional factors, especially the protein and fat content of the diet, may change pancreatic morphology after ethanol induced injury. This study was performed to delineate the combined effects of a low fat diet and longterm ethanol ingestion on the rat pancreas. Male Sprague-Dawley rats were maintained with five different diets for 12 weeks and the pancreas removed on the day they were killed. Rats fed a very low fat diet without ethanol (5% of total calories as lipid) developed malnutrition, pancreatic steatosis, and reduction in zymogen granules content. Animals fed a 35% lipid diet with ethanol also developed pancreatic steatosis but changes in zymogen granules content were not detected. Both malnutrition and longterm ethanol consumption increased pancreatic cholesterol ester content, and their effects were additive. Pancreatic steatosis was accompanied with hypercholesterolaemia. Amylase, lipase, and cholesterol esterase content were reduced in malnourished rats; but longterm ethanol ingestion, regardless of the nutritional state, increased lipase content and decreased amylase. It is suggested that high serum cholesterol concentrations and increased pancreatic lipase activity could cause accumulation of cholesterol esters in acinar cells. Fat accumulation in the pancreas has been reported as the earliest histopathological feature in alcoholic patients and may be responsible for cytotoxic effects on the acinar cells at the level of the cell membrane. Although it is difficult to extrapolate results in this animal study to the human situation, the results presented in this work might explain the higher incidence of pancreatitis is malnourished populations as well as in alcoholic subjects that is reported in dietary surveys.

Este trabalho relata uma ocorrência de Meloidogyne enterolobii (sin. M. mayaguensis) no Estado do Mato Grosso do Sul, parasitando goiabeira (Psidium guajava), cv. Novo Milênio. O nematoide foi caracterizado e identificado pelo fenótipo da isoenzima esterase (Est) (Est M2, Rm: 0,7 e 0,9).

This work describes the formation of transformation products (TPs) by the enzymatic degradation at laboratory scale of two highly consumed antibiotics: tetracycline (Tc) and erythromycin (ERY). The analysis of the samples was carried out by a fast and simple method based on the novel configuration of the on-line turbulent flow system coupled to a hybrid linear ion trap – high resolution mass spectrometer. The method was optimized and validated for the complete analysis of ERY, Tc and their transformation products within 10 min without any other sample manipulation. Furthermore, the applicability of the on-line procedure was evaluated for 25 additional antibiotics, covering a wide range of chemical classes in different environmental waters with satisfactory quality parameters. Degradation rates obtained for Tc by laccase enzyme and ERY by EreB esterase enzyme without the presence of mediators were ∼78% and ∼50%, respectively. Concerning the identification of TPs, three suspected compounds for Tc and five of ERY have been proposed. In the case of Tc, the tentative molecular formulas with errors mass within 2 ppm have been based on the hypothesis of dehydroxylation, (bi)demethylation and oxidation of the rings A and C as major reactions. In contrast, the major TP detected for ERY has been identified as the “dehydration ERY-A”, with the same molecular formula of its parent compound. In addition, the evaluation of the antibiotic activity of the samples along the enzymatic treatments showed a decrease around 100% in both cases
