La caracterització funcional de dos gens en la peridermis, la ω hidroxilasa d'àcids grassos CYP86A33 -candidata per la funcionalització del carboni ω-terminal dels monòmers alifàtics de la suberina- i la ketoacyl-CoA sintasa StKCS6 -candidata per elongar àcids grassos o derivats llargs de suberina i ceres- es realitza per silenciament per RNA d'interferència en patata. La deficiència de CYP86A33 comporta una gran reducció dels monòmers principals de la suberina, l'àcid gras ω-hidroxilat i l'α,ω-diàcid C18:1, juntament amb una reducció total de la quantitat de suberina del 60%. Aquesta deficiència altera l'estructura lamel·lar típica de la suberina, així com també la funció barrera de la peridermis. La deficiència en StKCS6 comporta que els monòmers de la suberina de 28 carbonis o més llargs es redueixin i que els de 26 carbonis o més curts s'incrementin. Aquesta deficiència suggereix que la llargada dels compostos alifàtics pot contribuir a les propietats impermeabilitzants de la peridermis.

Os centros históricos são o resultado de um parque habitacional corrente, com uma grande uniformidade, coerência construtiva e espaços urbanos de modo a proporcionar uma identidade própria. Um dos grandes problemas urbanos são as vastas áreas degradadas do ponto de vista arquitetónico como também social, cultural e económico. Conhecidos os principais problemas dos edifícios de habitação e suas causas, para sua resolução existe necessidade de adaptar o processo de reabilitação tradicional ao conceito de sustentabilidade. Estes dois assuntos conjugados são atualmente emergentes, devido à necessidade de reabilitação do parque habitacional, nomeadamente centros históricos. A casa burguesa do Porto apresenta um elevado grau de degradação, porém apesar de existentes as ações de intervenção que sobre ela reincidem, não são proporcionais à necessidade atual. A cidade do Porto é marcada, maioritariamente, pelas Casas Burguesas, das quais foram executadas segundo padrões de conforto e de utilização da época. É, então, fundamental um estudo pormenorizado da casa burguesa do Porto, avaliando o contexto em que esta se encere (na cidade e na respetiva habitação), os subsistemas construtivos e as praticas de reabilitação.

Cells of the bacterial symbiont Xenorhabdus nematophila from the entomopathogenic nematode, Steinernema carpocapsae entered the pupae of Plutella xylostella after 15 minutes treatment with suspensions containing the bacterial cells. Secretions of Xenorhabdus nematophila, in either broth or water, were found lethal to the pupae of P. xylostella when applied in moist sand. The bacterial symbiont Xenorhabdus nematophila was found lethal to the pupae of greater wax moth (Galleria mellonella), beet armyworm (Spodoptera exigua), diamondback moth (Plutella xylostella) and black vine weevil (Otiorhynchus sulcatus) in the absence of the nematode vector and the cells of X. nematophila entered the haemocoele of the pupae.

Artificial diet studies were used to differentiate among physical and chemical mechanisms affecting the suitability to diamondback moth (Plutella xylostella L.), of 16 food substrates obtained by growing four different brassicas in the glasshouse or field and measuring the pest's performance on either leaf discs or a diet incorporating leaf powders. Leaves of Chinese cabbage and the cabbage cultivar 'Minicole' were, respectively, the most and least suitable leaves for the insect, but this ranking was reversed on artificial diet. Leaves of glasshouse-grown plants were more suitable than those of plants grown in the fields. Differences in the suitability of leaves to diamondback moth appeared to be largely determined by leaf toughness and surface wax load. Concentrations of individual glucosinolates in the brassicas probably acted as phagostimulants, so increasing their intrinsic susceptibility to diamondback moth, but the effect of the physical factors appeared more important.

Larvae of Galleria mellonella (Greater Wax Moth) have been shown to be susceptible to Campylobacter jejuni infection and our study characterizes this infection model. Following infection with C. jejuni human isolates, bacteria were visible in the haemocoel and gut of challenged larvae, and there was extensive damage to the gut. Bacteria were found in the extracellular and cell-associated fraction in the haemocoel, and it was shown that C. jejuni can survive in insect cells. Finally, we have used the model to screen a further 67 C. jejuni isolates belonging to different MLST types. Isolates belonging to ST257 were the most virulent in the Galleria model, whereas those belonging to ST21 were the least virulent.

Galleria mellonella (wax moth) larvae have elsewhere been shown to be susceptible to pathogens such as Francisella tularensis, Burkholderia mallei, and Pseudomonas aeruginosa. We report that the larvae are rapidly killed by Campylobacter jejuni at 37 degrees C. Three strains of C. jejuni tested, 11168H (human diarrheal isolate), G1 (human Guillain-Barre syndrome isolate), and 81-176 (human diarrheal isolate), were equally effective at killing G. mellonella larvae. A panel of defined mutants of C. jejuni 11168H, in known or putative virulence genes, showed different degrees of attenuation in G. mellonella larvae. A mutant lacking the O-methyl phosphoramidate (MeOPN) capsule side group was attenuated, clearly demonstrating that MeOPN has a role in virulence. This new model of C. jejuni infection should facilitate the identification of novel virulence genes.

Atmospheric CO2 concentration is hypothesized to influence vegetation distribution via tree–grass competition, with higher CO2 concentrations favouring trees. The stable carbon isotope (δ13C) signature of vegetation is influenced by the relative importance of C4 plants (including most tropical grasses) and C3 plants (including nearly all trees), and the degree of stomatal closure – a response to aridity – in C3 plants. Compound-specific δ13C analyses of leaf-wax biomarkers in sediment cores of an offshore South Atlantic transect are used here as a record of vegetation changes in subequatorial Africa. These data suggest a large increase in C3 relative to C4 plant dominance after the Last Glacial Maximum. Using a process-based biogeography model that explicitly simulates 13C discrimination, it is shown that precipitation and temperature changes cannot explain the observed shift in δ13C values. The physiological effect of increasing CO2 concentration is decisive, altering the C3/C4 balance and bringing the simulated and observed δ13C values into line. It is concluded that CO2 concentration itself was a key agent of vegetation change in tropical southern Africa during the last glacial–interglacial transition. Two additional inferences follow. First, long-term variations in terrestrial δ13Cvalues are not simply a proxy for regional rainfall, as has sometimes been assumed. Although precipitation and temperature changes have had major effects on vegetation in many regions of the world during the period between the Last Glacial Maximum and recent times, CO2 effects must also be taken into account, especially when reconstructing changes in climate between glacial and interglacial states. Second, rising CO2 concentration today is likely to be influencing tree–grass competition in a similar way, and thus contributing to the "woody thickening" observed in savannas worldwide. This second inference points to the importance of experiments to determine how vegetation composition in savannas is likely to be influenced by the continuing rise of CO2 concentration.

Modified fluorcanasite glasses were fabricated by either altering the molar ratios of Na(2)O and CaO or by adding P(2)O(5) to the parent stoichiometric glass compositions. Glasses were converted to glass-ceramics by a controlled two-stage heat treatment process. Rods (2 mm x 4 mm) were produced using the conventional lost-wax casting technique. Osteoconductive 45S5 bioglass was used as a reference material. Biocompatibility and osteoconductivity were investigated by implantation into healing defects (2 mm) in the midshaft of rabbit femora. Tissue response was investigated using conventional histology and scanning electron microscopy. Histological and histomorphometric evaluation of specimens after 12 weeks implantation showed significantly more bone contact with the surface of 45S5 bioglass implants when compared with other test materials. When the bone contact for each material was compared between experimental time points, the Glass-Ceramic 2 (CaO rich) group showed significant difference (p = 0.027) at 4 weeks, but no direct contact at 12 weeks. Histology and backscattered electron photomicrographs showed that modified fluorcanasite glass-ceramic implants had greater osteoconductivity than the parent stoichiometric composition. Of the new materials, fluorcanasite glass-ceramic implants modified by the addition of P(2)O(5) showed the greatest stimulation of new mineralized bone tissue formation adjacent to the implants after 4 and 12 weeks implantation. Supercritical carbon dioxide (SC-CO(2)) extractions of Brazilian cherry (Eugenia uniflora L.) were carried out under varied conditions of pressure and temperature, according to a central composite 2(2) experimental design, in order to produce flavour-rich extracts. The composition of the extracts was evaluated by gas chromatography coupled with mass spectrometry (GC/MS). The abundance of the extracted compounds was then related to sensory analysis results, assisted by principal component and factorial discriminant analysis (PCA and FDA, respectively). The identified sesquiterpenes and ketones were found to strongly contribute to the characteristic flavour of the Brazilian cherry. The extracts also contained a variety of other volatile compounds, and part of the fruit wax contained long-chain hydrocarbons that according to multivariate analysis, contributed to the yield of the extracts, but not the flavour. Volatile phenolic compounds, to which antioxidant properties are attributed, were also present in the extracts in high proportion, regardless of the extraction conditions. (C) 2010 Elsevier Ltd. Alcantarea (Bromeliaceae) has 26 species that are endemic to eastern Brazil, occurring mainly on gneiss-granitic rock outcrops (`inselbergs`). Alcantarea has great ornamental potential and several species are cultivated in gardens. Limited data is available in the literature regarding the leaf anatomical features of the genus, though it has been shown that it may provide valuable information for characterizing of Bromeliaceae taxa. In the present work, we employed leaf anatomy to better characterize the genus and understand its radiation into harsh environments, such as inselbergs. We also searched for characteristics potentially useful in phylogenetic analyses and in delimiting Alcantarea and Vriesea. The anatomical features of the leaves, observed for various Alcantarea species, are in accordance with the general pattern shown by other Bromeliaceae members. However, some features are notable for their importance for sustaining life on rock outcrops, such as: small epidermal thick-walled cells, uneven sinuous epidermal walls, hypodermis often differentiated into lignified layers with thick-walled cells, aquiferous hypodermis bearing collapsible cells, and the presence of well developed epicuticular stratum. Alcantarea leaves tend to show different shapes in the spongy parenchyma, and have chlorenchymatous palisade parenchyma arranged in more well-defined arches, when compared to Vriesea species from the same habitat.

Camarea is a South-American endemic genus comprising eight species. In the present work n-alkanes from foliar cuticular waxes of 23 specimens, representing seven species of Camarea were analyzed, aiming at establishing interspecific affinities and evaluating the usefulness of n-alkane distribution as species characteristic. The sampling included also specimens of Peixotoa rericulata and Janusia guaronitica (both Malpighiaceae). The results were used to obtain a phenogram indicating chemical affinities between species. The results are in agreement with morphological similarities among some Camarea species. Intraspecific variability was small, suggesting that n-alkane distribution may be useful for species characterization and establishment of links among Camarea species. The results support the recognition of Camarea triphylla as a synonym of Camarea axillaris and are not coherent with a hybrid condition of a population exhibiting morphological characteristics combining Camarea affinis and Camarea hirsuta, suggesting instead that the individuals analyzed belong either to Camarea hirsuta or a close species. Distribution of n-alkanes is inadequate to distinguish among Malpighiaceae genera: P reticulata has n-alkane distribution similar to several Cumarea species. (C) 2008 Elsevier Ltd. O jambo-vermelho (Syzygium malaccense (L.) Merryl et Perry), espécie pertencente à família Myrtaceae, possui frutos ricos em vitaminas, ferro e fósforo. Este trabalho teve por objetivos fornecer informações sobre aspectos morfológicos do fruto, semente e plântula e avaliar a influência do tamanho da semente na germinação dessa espécie. O fruto é uma baga piriforme, carnoso e indeiscente. O epicarpo é delgado, liso e avermelhado; o mesocarpo e o endocarpo são esbranquiçados e suculentos. Os frutos pesam, em média, 35,57g e medem 7,16cm de comprimento e 5,15cm de largura. As sementes são poliembriônicas e exalbuminosas, pesando, em média, 7,62g. Os cotilédones são maciços e esverdeados. A germinação é hipógea, e a emergência das plântulas inicia-se aos 36 dias após a semeadura. A raiz primária é longa e esbranquiçada. As raízes secundárias são curtas e filiformes. As sementes de maior tamanho são as mais vigorosas, não havendo diferença entre as de tamanho médio e pequeno. O estudo morfológico do jambo-vermelho pode ser utilizado para a identificação da espécie ou em relações ecológicas interespecíficas.

A presente pesquisa visa entender os fatores que explicam a manutenção dos aspectos artesanais no extrativismo da cera de carnaúba em Limoeiro do Norte. Justificamos a exploração desse tema pelo fascínio do autor pelo tema e por ser esse recurso ainda uma importante fonte de renda para produtores e trabalhadores rurais no respectivo município. Objetivamos nessa pesquisa identificar as razões pelas quais a atividade mantém seu padrão tecnológico intacto. Primeiro objetivamos uma descrição do processo produtivo da cera, tanto no passado, quanto no presente, comparando estes momentos e ressaltando o caráter artesanal da atividade, além de analisar a importância atual da atividade para aqueles que ainda lidam com esse tipo de produção, bem como procuramos revelar estrutura histórica e institucional que permeia a lógica e as escolhas destes produtores, a qual pode explicar a preservação destes aspectos artesanais. Utilizando o referencial teórico do institucionalismo, explicamos que a preservação destes aspectos artesanais, ocorre em virtude da prevalência de determinadas instituições, como a memória coletiva cultivada entre os rendeiros idosos e resistentes, a concentração fundiária, responsável pelo arrendamento e também as restritas possibilidades de investimento e poupança no processo produtivo, em face do baixo nível de renda de produtores, os quais com um produto pouco competitivo no mercado, pouco podem fazer para inovar tecnologicamente.

Este trabalho teve como objetivo avaliar diferentes temperaturas e coberturas na conservação de frutos de pimentão 'Magali-R'. No primeiro experimento, avaliou-se o uso de filme de PVC ou cera, e os frutos foram armazenados a 20ºC e 66%UR, e no segundo experimento associou-se o filme de PVC e cera, com armazenamento a 22°C (67% UR); 12ºC (90% UR) e 5ºC (87% UR). Acompanhou-se a evolução da qualidade dos frutos avaliando-se a massa fresca, a aparência, o aparecimento de podridões e os teores de sólidos solúveis, acidez titulável e ácido ascórbico. A proteção dos pimentões com cera ou filme de PVC foi favorável à manutenção da aparência e da massa fresca, e protegeu-os contra podridões, sem prejuízos aos teores de sólidos solúveis, acidez titulável e ácido ascórbico, com aumento significativo na vida útil para até 33 dias com o armazenamento a 5°C.
