Lutzomyia (Nyssomyia) whitmani s.l.is the main vector of cutaneous leishmaniasis in state of Mato Grosso, but little is known about environmental determinants of its spatial distribution on a regional scale. Entomologic surveys of this sand fly species, conducted between 1996 and 2001 in 41 state municipalities, were used to investigate the relationships between environmental factors and the presence of the species, and to develop a spatial model of habitat suitability. The relationship between averaged CDC light trap indexes and 15 environmental and socio-economic factors were tested by logistic regression (LR) analysis. Spatial layers of deforestation tax and the Brazilian index of gross net production (IGNP) were identified as significant explanatory variables for vector presence in the LR model, and these were then overlaid with habitat maps. The highest habitat suitability in 2001 was obtained for the heavily deforested areas in the Central-North, South, East, and Southwest of Mato Grosso, particularly in municipalities with lower IGNP values.

The incidence and clinical features of human coronaviruses (HCoVs) among Brazilian patients with respiratory illness are not well known. We investigated the prevalence of HCoVs among Brazilian outpatients and hospitalised patients with respiratory illnesses during 2009 and 2010. To identify the HCoVs, pancoronavirus and species-specific reverse-transcriptase polymerase chain reaction assays were performed. Five of 394 samples were positive for HCoVs (1.2%): 1/182 (0.5%) outpatients and 4/212 (1.8%) hospitalised patients. The OC43 and NL63 HCoVs were identified. Two patients were admitted to the intensive care unit. Underlying chronic disease was reported in cases and one diabetic adult died. HCoVs can cause lower respiratory infections and hospitalisation. Patients with pre-existing conditions and respiratory infections should be evaluated for HCoV infections.

The aim of this study was to characterize two metallo-β-lactamases (MBLs)-producing Pseudomonas aeruginosa clinical isolates showing meropenem susceptibility. Antimicrobial susceptibility was assessed by automated testing and Clinical and Laboratory Standards Institute agar dilution method. MBL production was investigated by phenotypic tests. Molecular typing was determined by pulsed field gel electrophoresis (PFGE). MBL-encoding genes, as well as their genetic context, were identified by polymerase chain reaction (PCR) and sequencing. The location of blaIMP-16 was determined by plasmid electrophoresis, Southern blot and hybridization. Transcriptional levels of blaIMP-16, mexB, mexD, mexF, mexY, ampC and oprD were determined by semi-quantitative real time PCR. The P. aeruginosa isolates studied, Pa30 and Pa43, showed imipenem and meropenem susceptibility by automated testing. Agar dilution assays confirmed meropenem susceptibility whereas both isolates showed low level of imipenem resistance. Pa30 and Pa43 were phenotypically detected as MBL producers. PFGE revealed their clonal relatedness. blaIMP-16 was identified in both isolates, carried as a single cassette in a class 1 integron that was embedded in a plasmid of about 60-Kb. Pa30 and Pa43 overexpressed MexAB-OprM, MexCD-OprJ and MexXY-OprM efflux systems and showed basal transcriptional levels of ampC and oprD. MBL-producing P. aeruginosa that are not resistant to meropenem may represent a risk for therapeutic failure and act as silent reservoirs of MBL-encoding genes.

Human respiratory syncytial virus (HRSV) causes severe infections among children and immunocompromised patients. We compared HRSV infections among Haematopoietic Stem Cell Transplant program (HSCT) patients and children using direct immunofluorescence (DFA), point-of-care RSV Bio Easy® and a polymerase chain reaction (PCR) assay. Overall, 102 samples from HSCT patients and 128 from children obtained positivity rate of 18.6% and 14.1% respectively. PCR sensitivity was highest mainly on samples collected after five days of symptoms onset. A combination of both DFA and reverse transcriptase-PCR methods for HSCT high-risk patients is the best diagnostic flow for HRSV diagnosis among these patients.

Tachia sp. are used as antimalarials in the Amazon Region and in vivo antimalarial activity of a Tachia sp. has been previously reported. Tachia grandiflora Maguire and Weaver is an Amazonian antimalarial plant and herein its cytotoxicity and antimalarial activity were investigated. Spectral analysis of the tetraoxygenated xanthone decussatin and the iridoid aglyone amplexine isolated, respectively, from the chloroform fractions of root methanol and leaf ethanol extracts was performed. In vitro inhibition of the growth of Plasmodium falciparum Welch was evaluated using optical microscopy on blood smears. Crude extracts of leaves and roots were inactive in vitro. However, chloroform fractions of the root and leaf extracts [half-maximal inhibitory concentration (IC50) = 10.5 and 35.8 µg/mL, respectively] and amplexine (IC50= 7.1 µg/mL) were active in vitro. Extracts and fractions were not toxic to type MRC-5 human fibroblasts (IC50> 50 µg/mL). Water extracts of the roots of T. grandiflora administered by mouth were the most active extracts in the Peters 4-day suppression test in Plasmodium berghei-infected mice. At 500 mg/kg/day, these extracts exhibited 45-59% inhibition five to seven days after infection. T. grandiflora infusions, fractions and isolated substance have potential as antimalarials.

Phospholipase is an important virulence factor for pathogenic fungi. In this study, we demonstrate the following: (i) the Paracoccidioides brasiliensis pld gene is preferentially expressed in mycelium cells, (ii) the plb1 gene is mostly up-regulated by infection after 6 h of co-infection of MH-S cells or during BALB/c mice lung infection, (iii) during lung infection, plb1, plc and pld gene expression are significantly increased 6-48 h post-infection compared to 56 days after infection, strongly suggesting that phospholipases play a role in the early events of infection, but not during the chronic stages of pulmonary infection by P. brasiliensis.

Lipophorin (Lp) is the main haemolymphatic lipoprotein in insects and transports lipids between different organs. In adult females, lipophorin delivers lipids to growing oocytes. In this study, the interaction of this lipoprotein with the ovaries of Rhodnius prolixus was characterised using an oocyte membrane preparation and purified radiolabelled Lp (125I-Lp). Lp-specific binding to the oocyte membrane reached equilibrium after 40-60 min and when 125I-Lp was incubated with increasing amounts of membrane protein, corresponding increases in Lp binding were observed. The specific binding of Lp to the membrane preparation was a saturable process, with a Kdof 7.1 ± 0.9 x 10-8M and a maximal binding capacity of 430 ± 40 ng 125I-Lp/µg of membrane protein. The binding was calcium independent and pH sensitive, reaching its maximum at pH 5.2-5.7. Suramin inhibited the binding interaction between Lp and the oocyte membranes, which was completely abolished at 0.5 mM suramin. The oocyte membrane preparation from R. prolixus also showed binding to Lp from Manduca sexta. When Lp was fluorescently labelled and injected into vitellogenic females, the level of Lp-oocyte binding was much higher in females that were fed whole blood than in those fed blood plasma.

O sistema brasileiro de avaliação de programas de pós-graduação vem evoluindo desde 1976, sendo considerado eficiente e moderno. No presente trabalho, o objetivo é propor um modelo de gestão de programas de pós-graduação stricto sensu em Administração do Brasil, com base nos sistemas de avaliação do Brasil e dos Estados Unidos. Para tanto, procedeu-se a uma pesquisa em oito programas: quatro no Brasil, recomendados pela Coordenação de Aperfeiçoamento de Pessoal de Nível Superior (CAPES) e quatro nos Estados Unidos, credenciados pela Association to Advance Collegiate School of Business (AACSB). O método de pesquisa foi o estudo de casos múltiplos de caráter exploratório com dois componentes básicos: pesquisa documental, por meio de um estudo em profundidade dos sistemas de avaliação; pesquisa de campo com os coordenadores de programas para identificar como os sistemas de avaliação são utilizados para desenvolver os programas. A partir desses estudos, foi proposto um modelo de gestão em que, além de atender aos requisitos do sistema de avaliação da CAPES, seja levada em consideração a realidade do programa e os elementos importantes para seu contínuo aprimoramento, dentre eles: missão e visão; plano estratégico; corpo docente; estrutura do programa; corpo discente; resultados que abrangem produção científico-tecnológica, egresso e inserção social.

Gene duplication leads to paralogy, which complicates the de novo assembly of genotyping-by-sequencing (GBS) data. The issue of paralogous genes is exacerbated in plants, because they are particularly prone to gene duplication events. Paralogs are normally filtered from GBS data before undertaking population genomics or phylogenetic analyses. However, gene duplication plays an important role in the functional diversification of genes and it can also lead to the formation of postzygotic barriers. Using populations and closely related species of a tropical mountain shrub, we examine 1) the genomic differentiation produced by putative orthologs, and 2) the distribution of recent gene duplication among lineages and geography. We find high differentiation among populations from isolated mountain peaks and species-level differentiation within what is morphologically described as a single species. The inferred distribution of paralogs among populations is congruent with taxonomy and shows that GBS could be used to examine recent gene duplication as a source of genomic differentiation of nonmodel species.

The position of a gene in the genome may have important consequences for its function. Therefore, when a new duplicate gene arises, its location may be critical in determining its fate. Our recent work in humans, mouse, and Drosophila provided a test by studying the patterns of duplication in sex chromosome evolution. We revealed a bias in the generation and recruitment of new gene copies involving the X chromosome that has been shaped largely by selection for male germline functions. The gene movement patterns we observed reflect an ongoing process as some of the new genes are very young while others were present before the divergence of humans and mouse. This suggests a continuing redistribution of male-related genes to achieve a more efficient allocation of male functions. This notion should be further tested in organisms employing other sex determination systems or in organisms differing in germline sex chromosome inactivation. It is likely that the selective forces that were detected in these studies are also acting on other types of duplicate genes. As a result, future work elucidating sex chromosome differentiation by other mutational mechanisms will shed light on this important process.

Este estudo foi realizado na bacia do rio Caí, no Rio Grande do Sul, em duas áreas com diferentes estados de conservação ambiental: uma fortemente alterada pela intensa avicultura e suinocultura e outra mais conservada. Para padronizar as coletas das larvas e pupas de simulídeos foram utilizados substratos artificiais, instalados nos arroios por períodos de 14 dias. Em todos os pontos de amostragens foram feitas coletas simultâneas de água para análise físico-química e microbiológica. Na área mais conservada foram coletadas onze espécies com as seguintes freqüências: Psaroniocompsa incrustata (42,95%), P. auripellita (17,66%), Chirostilbia pertinax (16,86%), Inaequalium subclavibranchium (8,43%), I. nogueirai (5,80%), P. anamariae (3,69%), I. clavibranchium (1,58%), C. riograndense (0,92%), Lutzsimulium hirticosta (0,92%), I. botulibranchium (0,79%), Thyrsopelma itaunense (0,40%). Na área de maior impacto foram coletadas apenas seis espécies: C. pertinax (84,31%), C. riograndense (7,52%), T. itaunense (4,68%), P. incrustata (2,94%), I. subclavibranchium (0,33%), Ectemnaspis dinellii (0,22%). As concentrações de Nitrato e Nitrito na água foram os fatores que melhor permitem distinguir os pontos amostrados. A concentração de nitrato esteve significativamente relacionada com C. pertinax (r²= 0,54; P < 0,03) e com T. itaunense (r²= 0,55; P < 0,03) e nitrito com C. riograndense (r²= 0,64; P < 0,02). Essas informações podem ser úteis aos programas de controle integrado dos simulídeos, atualmente em execução em pelo menos 170 municípios no Rio Grande do Sul.

O presente estudo realiza-se no âmbito do Mestrado em Direito Marítimo e Comércio Internacional e tem como título: Aspectos Jurídicos sobre a Repressão do Tráfico Ilícito de Drogas pela Via Marítima, Cabo Verde. Portanto pretende-se iniciar o mesmo esclarecendo possíveis dúvidas que possam surgir referente ao termo “droga” e o contexto histórico da ilegalidade das drogas. Recorde-se que há pouco mais de um século, precisamente no ano 1909, na China realizou-se a primeira conferência sobre o ópio que viria a entrar em vigor a nível mundial no ano de 1919, após ter sido incorporada no tratado de Versalhes. A cannabis entrou na lista de substâncias proibidas após a revisão da Convenção Internacional do Ópio realizada em Genebra no ano 1925. Perante esse cenário, conhecendo os avultados lucros do comércio da droga, os antigos comerciantes não pretendiam desistir dessa prática e encontraram no mar as vulnerabilidades necessárias para continuarem a realizar esse comércio, agora tipificado internacionalmente como crime. Logo, uma vez mais1, a visão antiga do Direito Internacional, principalmente, tratando-se de dois extremos, o Direito à Guerra (ius bellum) e o Direito à Paz (o ius pacis) tornou-se impotente para responder aos novos desafios do Direito Internacional. Assim sendo, o segundo capítulo, visa esclarecer factos relevantes do Direito Internacional que afectam directamente o tráfico ilícito de drogas pela via marítima, partindo da análise das Fontes e princípios do Direito Internacional, distinguindo o Direito Internacional Público do Direito Internacional Privado e evocando questões importantes no âmbito do Direito Penal Internacional que afetam o tráfico de estupefacientes e substâncias psicotrópicas. Embora o Direito do Mar e o Direito Marítimo sejam dois conceitos que aparentam ser semelhantes, o terceiro capítulo esclarece algumas diferenças 1 Tendo em conta que a pirataria no alto mar foi um dos primeiros crimes a explorar as fragilidades jurídicas existentes no âmbito do Direito Internacional publico, mais concretamente, sua subdivisão, Direito do Mar. Aspectos Jurídicos sobre a Repressão do Trafico de Drogas Ilícitas Pela Via Marítima –Cabo Verde entre eles, destacando os princípios que iluminam a relação entre os Estados, as delimitações e os regimes jurídicos aplicados nas áreas marítimas consagrados na Convenção das Nações Unidas Sobre o Direito do Mar, doravante designada de CNUDM. Dará ainda enfase aos principais aspectos sobre as fontes e os princípios do Direito do Mar e os principais fundamentos das competências dos juízes nas diversas áreas marítimas. O quarto e último capítulo visa abordar os instrumentos jurídicos mais importantes no âmbito da repressão ao tráfico ilícito de drogas pela via marítima, enaltecendo, desde já, o princípio da cooperação internacional estabelecido internacionalmente, expressando que, “Todos os Estados devem cooperar para a repressão do tráfico ilícito de estupefacientes e substâncias psicotrópicas praticado por navios no alto mar, com violação das convenções internacionais.”2 Esse pressuposto remete-nos, subitamente, para a análise das Convenções de maior importância criados pela ONU com essa finalidade, nomeadamente, a Convenção Única sobre Narcóticos de 1961, emendada pelo Protocolo de 1972, a Convenção sobre Substâncias Psicotrópicas de 1971 e a Convenção das Nações Unidas contra o Tráfico Ilícito de Narcóticos e Substâncias Psicotrópicas de 1988. O tema se desenvolve, sempre evocando os instrumentos jurídicos caboverdianos, tais como a lei da droga, 78/IV/93 de 12 Julho, o Código Marítimo de Cabo Verde, adiante referido pelas siglas CMCV, o Código Penal de Cabo Verde, doravante referido por CPCV e a lei mãe da nação cabo-verdiana, a Constituição da República de Cabo Verde, adiante designado por CRCV. Para facilitar a compreensão da matéria exposta, será tratado de forma sintetizada, o caso da abordagem a um veleiro de pavilhão norte-americano, realizado por autoridades de Cabo Verde, em conjunto com autoridades do Reino Unido e ocorrida em águas internacionais próximas de Cabo Verde.
