Generierung und Prozessierung oxidativer DNA Schäden --- Ziel dieser Arbeit war es, adaptive Antworten der Zellen auf einen DNA Schädigung zu untersuchen. Hierzu wurden Experimente zur Reparatur oxidierter Basen (Substrate der Basen Exzisions Reparatur (BER)) oder von Pyrimidindimeren (Substrate der Nukleotid Exzisions Reparatur (NER)) nach einer Vorbehandlung mit DNA-schädigender Agenzien durchgeführt. Die Ergebnisse zeigten, dass sowohl eine Vorbehandlung mit einer alkylierenden als auch mit einer oxidierenden Substanz zu einer adaptiven Erhöhung des zellulären Glutathionspiegels führte, die 16 h nach der Schädigung ihr Maximum erreichte. Jedoch waren die 8-oxoG Glykosylaseaktivitäten über einen Zeitraum von 18 h konstant. Diese Effekte waren unabhängig davon, ob Maus Embryofibroblasten, primäre oder p53 profiziente menschliche Zellen verwendet wurden. Die BER war ebenfalls in keiner der verschiedenen Zelllinien signifikant verbessert. Die adaptive Antwort bezüglich der Glutathionspiegel war also nicht mit einer entsprechenden Veränderung bei der DNA-Reparatur verbunden. Folglich ist die Reparatur von oxidativen DNA-Schäden durch eine vorausgehende Schädigung nicht induzierbar. Der zweite Teil der Untersuchungen zu der Reparatur beschäftigte sich mit der NER. Hierzu wurde die Reaktivierung eines mit UVB-Strahlung geschädigten Plasmids untersucht. Als Wirtszellen fungierten primäre menschliche Fibroblasten und Keratinozyten, die entweder mit UVB vorbehandelt oder ungeschädigt waren. Auch für die NER konnte keine signifikante Beschleunigung der Reparatur von Pyrimidindimeren durch eine Vorbehandlung festgestellt werden. Die Reaktivierung erfolgte ferner unabhängig vom p53-Status der Zellen, wie Versuche mit p53-siRNA zeigten. Neben der Prozessierung war die Generierung oxidativer DNA Schäden Gegenstand der Arbeit. Die verwendete Substanz Tirapazamin (TPZ) ist ein für hypoxische Zellen selektives, neues Zytostatikum und befindet sich momentan in Phase 2/3 der klinischen Prüfung. Ziel war es die von TPZ verursachten DNA Modifikationen zu charakterisieren, sowie die Toxizität und Genotoxizität zu untersuchen. Da es Hinweise auf eine Aktivierung von TPZ über eine Oxidoreduktase (OR) gab, wurden die Experimente in Wildtyp und hOR überexprimierenden Zellen durchgeführt. Die Quantifizierung der verursachten DNA-Modifikationen zeigte, dass der von TPZ verursachte Schaden in Zellen mit hOR erhöht war. Das erhaltene Schadensprofil der durch TPZ verursachten DNA-Modifikationen war dem Schadensprofil von durch Gamma-Strahlung intrazellulär verursachten Hydroxylradikalen sehr ähnlich. Da es nach der Aktivierung von TPZ durch eine OR zu einer Abspaltung von Hydroxylradikalen kommt, bestätigte dies den vermuteten Mechanismus. Weitere Untersuchungen mit t-Butanol, einem Hydroxylradikal Fänger, ergaben eine verminderte DNA-Schädigung, was ebenfalls für eine DNA-Schädigung durch Hydroxylradikale spricht. Untersuchungen zur Mutagenität zeigten das die Mutationsrate in Zellen mit hOR um das 4 fache erhöht ist. Erstaunlich war jedoch, dass der im gleichen Ausmaß von Gamma-Strahlung verursachte DNA-Schaden für die beobachtete Toxizität dieser verantwortlich war, während bei TPZ unter den gleichen Bedingungen keine Toxizität vorlag. Erklärt werden könnte die erhöhte Toxizität und Mutagenität durch so genannte geclusterte DNA-Schäden, die von Gamma-Strahlen, nicht jedoch von TPZ gebildet werden. Nach einer verlängerten Inkubation wurde sowohl für die Toxizität als auch für die Genotoxizität erneut ein verstärkender Effekt durch die OR bestätigt. Überraschend war weiterhin die von der OR unabhängige Generierung von Doppelstrangbrüchen, für die demnach ein grundsätzlich anderer Mechanismus, wie zum Beispiel eine direkte Interaktion mit der Topoisomerase II, angenommen werden muss.

The present thesis is focused on the study of Organic Semiconducting Single Crystals (OSSCs) and crystalline thin films. In particular solution-grown OSSC, e.g. 4-hdroxycyanobenzene (4HCB) have been characterized in view of their applications as novel sensors of X-rays, gamma-rays, alpha particles radiations and chemical sensors. In the field of ionizing radiation detection, organic semiconductors have been proposed so far mainly as indirect detectors, i.e. as scintillators or as photodiodes. I first study the performance of 4HCB single crystals as direct X-ray detector i.e. the direct photon conversion into an electrical signal, assessing that they can operate at room temperature and in atmosphere, showing a stable and linear response with increasing dose rate. A dedicated study of the collecting electrodes geometry, crystal thickness and interaction volume allowed us to maximize the charge collection efficiency and sensitivity, thus assessing how OSSCs perform at low operating voltages and offer a great potential in the development of novel ionizing radiation sensors. To better understand the processes generating the observed X-ray signal, a comparative study is presented on OSSCs based on several small-molecules: 1,5-dinitronaphthalene (DNN), 1,8-naphthaleneimide (NTI), Rubrene and TIPS-pentacene. In addition, the proof of principle of gamma-rays and alpha particles has been assessed for 4HCB single crystals. I have also carried out an investigation of the electrical response of OSSCs exposed to vapour of volatile molecules, polar and non-polar. The last chapter deals with rubrene, the highest performing molecular crystals for electronic applications. We present an investigation on high quality, millimeter-sized, crystalline thin films (10 – 100 nm thick) realized by exploiting organic molecular beam epitaxy on water-soluble substrates. Space-Charge-Limited Current (SCLC) and photocurrent spectroscopy measurements have been carried out. A thin film transistor was fabricated onto a Cytop® dielectric layer. The FET mobility exceeding 2 cm2/Vs, definitely assess the quality of RUB films.

Questo lavoro ha lo scopo di individuare una strada nella ricerca di un confronto tra canditati eventi di neutrino cosmico da parte dell'esperimento ANTARES ,nel Mar Mediterraneo a sud est della costa di Tolone, in Francia e gli Ultra High Energy Cosmic Rays dell'osservatorio Pierre Auger in Argentina. Si utilizzano dati raccolti dal telescopio di neutrini ANTARES tra il 2007 e il 2013, con energie dell'ordine del TeV, e quelli di Auger ottenuti tra il 2004 e il 2014, con energie dei raggi cosmici dell'ordine dell'EeV. Entrambi gli apparati sono basati sulla rivelazione di radiazione Cherenkov, ma l'osservatorio di Auger è dotato di un secondo rivelatore di luminescenza che lavora in sincrono per ridurre gli errori sistematici associati ai singoli rivelatori. Le distribuzioni degli eventi in base alle direzioni di arrivo e dell' energia osservata, sono i parametri necessari per la ricerca di una correlazione. Dalla sovrapposizione delle distribuzioni possiamo evidenziare le zone di visibilità caratteristiche di ciascun esperimento, e condurre un eventuale successiva analisi finalizzata ad una identificazione univoca di eventi coincidenti.

In recent years, several surveys have highlighted the presence of the rodent carcinogen furan in a variety of food items. Even though the evidence of carcinogenicity of furan is unequivocal, the underlying mechanism has not been fully elucidated. In particular, the role of genotoxicity in furan carcinogenicity is still not clear, even though this information is considered pivotal for the assessment of the risk posed by the presence of low doses of furan in food. In this work, the genotoxic potential of furan in vivo has been investigated in mice, under exposure conditions similar to those associated with cancer onset in the National Toxicology Program long-term bioassay. To this aim, male B6C3F1 mice were treated by gavage for 4 weeks with 2, 4, 8 and 15 mg furan/kg b.w./day. Spleen was selected as the target organ for genotoxicity assessment, in view of the capability of quiescent splenocytes to accumulate DNA damage induced by repeat dose exposure. The induction of primary DNA damage in splenocytes was evaluated by alkaline single-cell gel electrophoresis (comet assay) and by the immunofluorescence detection of foci of phosphorylated histone H2AX (gamma-H2AX). The presence of cross-links was probed in a modified comet assay, in which cells were irradiated in vitro with gamma-rays before electrophoresis. Chromosome damage was quantitated through the detection of micronuclei in mitogen-stimulated splenocytes using the cytokinesis-block method. Micronucleus induction was also assessed with a modified protocol, using the repair inhibitor 1-beta-arabinofuranosyl-cytosine to convert single-strand breaks in micronuclei. The results obtained show a significant (P < 0.01) increase of gamma-H2AX foci in mitogen-stimulated splenocytes of mice treated with 8 and 15 mg furan/kg b.w. and a statistically significant (P < 0.001) increases of micronuclei in binucleated splenocytes cultured in vitro. Conversely, no effect of in vivo exposure to furan was observed when freshly isolated quiescent splenocytes were analysed by immunofluorescence and in comet assays, both with standard and radiation-modified protocols. These results indicate that the in vivo exposure to furan gives rise to pre-mutagenic DNA damage in resting splenocytes, which remains undetectable until it is converted in frank lesions during the S-phase upon mitogen stimulation. The resulting DNA strand breaks are visualized by the increase in gamma-H2AX foci and may originate micronuclei at the subsequent mitosis.

Data collected with the surface detector array of the Pierre Auger Observatory during the period from January 1, 2004 to March 17, 2015 was examined for evidence of production of long-lived weakly interacting particles in interactions of ultra high energy cosmic rays in the atmosphere. The search was performed using extensive air showers with primary energies more than 10 EeV and arrival directions in the range of 57.5◦ to 77.5◦. There was no evidence of significant production of such particles. An upper limit on the fraction of extensive air showers in which such particles are produced was set.

The aim of this study was to evaluate the ability of dual energy X-rays absorptiometry (DXA) areal bone mineral density (aBMD) measured in different regions of the proximal part of the human femur for predicting the mechanical properties of matched proximal femora tested in two different loading configurations. 36 pairs of fresh frozen femora were DXA scanned and tested until failure in two loading configurations: a fall on the side or a one-legged standing. The ability of the DXA output from four different regions of the proximal femur in predicting the femoral mechanical properties was measured and compared for the two loading scenarios. The femoral neck DXA BMD was best correlated to the femoral ultimate force for both configurations and predicted significantly better femoral failure load (R2=0.80 vs. R2=0.66, P<0.05) when simulating a side than when simulating a standing configuration. Conversely, the work to failure was predicted similarly for both loading configurations (R2=0.54 vs. R2=0.53, P>0.05). Therefore, neck BMD should be considered as one of the key factors for discriminating femoral fracture risk in vivo. Moreover, the better predictive ability of neck BMD for femoral strength if tested in a fall compared to a one-legged stance configuration suggests that DXA's clinical relevance may not be as high for spontaneous femoral fractures than for fractures associated to a fall.

Supernova remnants are among the most spectacular examples of astrophysical pistons in our cosmic neighborhood. The gas expelled by the supernova explosion is launched with velocities ~1000 kilometers per second into the ambient, tenuous interstellar medium, producing shocks that excite hydrogen lines. We have used an optical integral-field spectrograph to obtain high-resolution spatial-spectral maps that allow us to study in detail the shocks in the northwestern rim of supernova 1006. The two-component Hα line is detected at 133 sky locations. Variations in the broad line widths and the broad-to-narrow line intensity ratios across tens of atomic mean free paths suggest the presence of suprathermal protons, the potential seed particles for generating high-energy cosmic rays.

The clinical advantage for protons over conventional high-energy x-rays stems from their unique depth-dose distribution, which delivers essentially no dose beyond the end of range. In order to achieve it, accurate localization of the tumor volume relative to the proton beam is necessary. For cases where the tumor moves with respiration, the resultant dose distribution is sensitive to such motion. One way to reduce uncertainty caused by respiratory motion is to use gated beam delivery. The main goal of this dissertation is to evaluate the respiratory gating technique in both passive scattering and scanning delivery mode. Our hypothesis for the study was that optimization of the parameters of synchrotron operation and respiratory gating can lead to greater efficiency and accuracy of respiratory gating for all modes of synchrotron-based proton treatment delivery. The hypothesis is tested in two specific aims. The specific aim #1 is to assess the efficiency of respiratory-gated proton beam delivery and optimize the synchrotron operations for the gated proton therapy. A simulation study was performed and introduced an efficient synchrotron operation pattern, called variable Tcyc. In addition, the simulation study estimated the efficiency in the respiratory gated scanning beam delivery mode as well. The specific aim #2 is to assess the accuracy of beam delivery in respiratory-gated proton therapy. The simulation study was extended to the passive scattering mode to estimate the quality of pulsed beam delivery to the residual motion for several synchrotron operation patterns with the gating technique. The results showed that variable Tcyc operation can offer good reproducible beam delivery to the residual motion at a certain phase of the motion. For respiratory gated scanning beam delivery, the impact of motion on the dose distributions by scanned beams was investigated by measurement. The results showed the threshold for motion for a variety of scan patterns and the proper number of paintings for normal and respiratory gated beam deliveries. The results of specific aims 1 and 2 provided supporting data for implementation of the respiratory gating beam delivery technique into both passive and scanning modes and the validation of the hypothesis.

The wet bulk density is one of the most important parameters of the physical and geological properties of marine sediments. The density is connected directly with sedimentation history and a few sedirnent properties. Knowledge of the fine scale density-depth structure is the base for many model calculations, for both sedimentological and palaeoclimatic research. A density measurement system was designed and built at the Alfred Wegener Institute in Bremerhaven for measuring the wet buk density of sediment cores with high resolution in a non-destructive way. The density is deterrnined by measuring the absorption of Gamma-rays in the sediment. This principle has been used since the 50's in materials research and in the geosciences. In the present case, Cs137 is used as the radioactive source and the intensity is measured by a detector system (scintillator and photomultiplier). Density values are obtainable in both longitudinal core sections and planar cross-sections (the latter are a function of the axial rotation angle). Special studies on inhomogenity can be applied with core rotation. Detection of ice rafted debris (IRD) is made possible with this option. The processes that run the density measurement system are computer controlled. Besides the absorption measurement the core diameter at every measurement point is determined with a potentiometric system. The data values taken are stored on a personal computer. Before starting routine measurements on the sediment cores, a few experiments conceming the statistical aspects of the gamma-ray signal and its accuracy were carried out. These experiments led to such things as the optimum operational parameters. A high spatial resolution in the mm-range is possible with the 4mm-thin gamma-ray measurements. Within five seconds the wet bulk density can be deterrnined with an absolute accuracy of 1%. A comparison between data measured with the new system and conventional measurements on core samples after core splitting shows an agreement within +I- 5% for most of the values. For this thesis, density determinations were carried out on ten sediment cores. A few sediment characteristics are obtainable from using just the standard measurement results without core rotation. In addition to differentes and steps in the absolute density range, variations in the "frequency" of the density-depth structure can be detected due to the close spatial measurement interval and high resolution. Examples from measurements with small (9°) and great (90°) angle increments show that abrupt and smooth transitional changes of sedirnent layers as well as ice rafted debris of several dimensions can be detected and distiflguished clearly. After the presentation of the wet bulk density results, a comparison with data from other investigations was made. Measurements of the electrical resistivity correlated very well with the density data because both parameters are closely related to the porosity of the sedirnent. Additionally, results from measurements of the magnetic susceptibility and from ultra-sonic wave velocity investigations were considered for a integrative interpretation. The correlation of these both parameters and wet bulk density data is strongly dependent on the local (environmental) conditions. Finally, the densities were compared with recordings from sediment-echographic soundings and an X-ray computer tomography analysis. The individual results of all investigations were then finally combined into an accurate picture of the core. Problems of ambiguity, which exist when just one Parameter is determined alone, can be reduced more or less according to the number of parameters and sedimentary characteristics measured. The important role of the density data among other parameters of such an integrated interpretation is evident. Evidence of this role include the high resolution of the measurement, the excellent accuracy and the key position within methods and parameters concerning marine sediments.

We have developed a new projector model specifically tailored for fast list-mode tomographic reconstructions in Positron emission tomography (PET) scanners with parallel planar detectors. The model provides an accurate estimation of the probability distribution of coincidence events defined by pairs of scintillating crystals. This distribution is parameterized with 2D elliptical Gaussian functions defined in planes perpendicular to the main axis of the tube of response (TOR). The parameters of these Gaussian functions have been obtained by fitting Monte Carlo simulations that include positron range, acolinearity of gamma rays, as well as detector attenuation and scatter effects. The proposed model has been applied efficiently to list-mode reconstruction algorithms. Evaluation with Monte Carlo simulations over a rotating high resolution PET scanner indicates that this model allows to obtain better recovery to noise ratio in OSEM (ordered-subsets, expectation-maximization) reconstruction, if compared to list-mode reconstruction with symmetric circular Gaussian TOR model, and histogram-based OSEM with precalculated system matrix using Monte Carlo simulated models and symmetries.

Recombination repair protein 1 (Rrp1) includes a C-terminal region homologous to several DNA repair proteins, including Escherichia coli exonuclease III and human APE, that repair oxidative and alkylation damage to DNA. The nuclease activities of Rrp1 include apurinic/apyrimidinic endonuclease, 3'-phosphodiesterase, 3'-phosphatase, and 3'-exonuclease. As shown previously, the C-terminal nuclease region of Rrp1 is sufficient to repair oxidative- and alkylation-induced DNA damage in repair-deficient E. coli mutants. DNA strand-transfer and single-stranded DNA renaturation activities are associated with the unique N-terminal region of Rrp1, which suggests possible additional functions that include recombinational repair or homologous recombination. By using the Drosophila w/w+ mosaic eye system, which detects loss of heterozygosity as changes in eye pigmentation, somatic mutation and recombination frequencies were determined in transgenic flies overexpressing wild-type Rrp1 protein from a heat-shock-inducible transgene. A large decrease in mosaic clone frequency is observed when Rrp1 overexpression precedes treatment with gamma-rays, bleomycin, or paraquat. In contrast, Rrp1 overexpression does not alter the spot frequency after treatment with the alkylating agents methyl methanesulfonate or methyl nitrosourea. A reduction in mosaic clone frequency depends on the expression of the Rrp1 transgene and on the nature of the induced DNA damage. These data suggest a lesion-specific involvement of Rrp1 in the repair of oxidative DNA damage.

The mutagen-sensitive CHO line irs1SF was previously isolated on the basis of hypersensitivity to ionizing radiation and was found to be chromosomally unstable as well as cross-sensitive to diverse kinds of DNA-damaging agents. The analysis of somatic cell hybrids formed between irs1SF and human lymphocytes implicated a human gene (defined as XRCC3; x-ray repair cross-complementing), which partially restored mitomycin C resistance to the mutant. A functional cDNA that confers mitomycin C resistance was transferred to irs1SF cells by transforming them with an expression cDNA library and obtaining primary and secondary transformants. Functional cDNA clones were recovered from a cosmid library prepared from a secondary transformant. Transformants also showed partial correction of sensitivity to cisplatin and gamma-rays, efficient correction of chromosomal instability, and substantially improved plating efficiency and growth rate. The XRCC3 cDNA insert is approximately 2.5 kb and detects an approximately 3.0-kb mRNA on Northern blots. The cDNA was mapped by fluorescence in situ hybridization to human chromosome 14q32.3, which was consistent with the chromosome concordance data of two independent hybrid clone panels.

Electromagnetic energy injected into the universe above a few hundred TeV is expected to pile up as γ radiation in a relatively narrow energy interval below 100 TeV due to its interaction with the 2.7^°K background radiation. We present an upper limit (90% C.L.) on the ratio of primary γ to charged cosmic rays in the energy interval 65–160 TeV (80–200 TeV) of 10.3 • 10^−3 (7.8 • 10^−3). Data from the HEGRA cosmic-ray detector complex consisting of a wide angle Čerenkov array (AIROBICC) measuring the lateral distribution of air Čerenkov light and a scintillator array, were used with a novel method to discriminate γ-ray and hadron induced air showers. If the presently unmeasured universal far infrared background radiation is not too intense, the result rules out a topological-defect origin of ultrahigh energy cosmic rays for masses of the X particle released by the defects equal to or larger than about 10^16 GeV.

We present optical, near-IR, and radio follow-up of 16 Swift bursts, including our discovery of nine afterglows and a redshift determination for three. These observations, supplemented by data from the literature, provide an afterglow recovery rate of 52% in the optical/near-IR, much higher than in previous missions (BeppoSAX, HETE-2, INTEGRAL, and IPN). The optical/near-IR afterglows of Swift events are on average 1.8 mag fainter at t = 12 hr than those of previous missions. The X-ray afterglows are similarly fainter than those of pre-Swift bursts. In the radio the limiting factor is the VLA threshold, and the detection rate for Swift bursts is similar to that for past missions. The redshift distribution of pre-Swift bursts peaked at z similar to 1, whereas the six Swift bursts with measured redshifts are distributed evenly between 0.7 and 3.2. From these results we conclude that ( 1) the pre-Swift distributions were biased in favor of bright events and low-redshift events, ( 2) the higher sensitivity and accurate positions of Swift result in a better representation of the true burst redshift and brightness distributions ( which are higher and dimmer, respectively), and (3) similar to 10% of the bursts are optically dark, as a result of a high redshift and/or dust extinction. We remark that the apparent lack of low-redshift, low-luminosity Swift bursts and the lower event rate than prelaunch estimates ( 90 vs. 150 per year) are the result of a threshold that is similar to that of BATSE. In view of these inferences, afterglow observers may find it advisable to make significant changes in follow-up strategies of Swift events. The faintness of the afterglows means that large telescopes should be employed as soon as the burst is localized. Sensitive observations in RIz and near-IR bands will be needed to discriminate between a typical z similar to 2 burst with modest extinction and a high-redshift event. Radio observations will be profitable for a small fraction (similar to 10%) of events. Finally, we suggest that a search for bright host galaxies in untriggered BAT localizations may increase the chance of finding nearby low-luminosity GRBs.
