Nos últimos anos, duas espécies de lagostas sapateiras, Scyllarides brasiliensis e S. deceptor, vêm se destacando nos desembarques pesqueiros de lagostas do Atlântico Sul Ocidental. Para espécies comercialmente importantes, o desenvolvimento de estudos que permitam conhecer a variabilidade e entender a dinâmica populacional é fundamental. Assim, o objetivo do primeiro capítulo desta tese foi avaliar a diversidade genética e a estrutura populacional dessas duas lagostas ao longo de aprox. 2.800 km da costa da América do Sul. Para as análises, foram empregados marcadores mitocondriais (citocromo oxidase I: COI; e a região controle: RC) e marcadores nucleares (13 loci de microssatélites desenvolvidos nesta tese). As duas espécies apresentaram altos níveis de variabilidade (S. deceptor: N = 200, mtDNA: h > 0,841, π > 0,005; microssatélites: He = 0,685; S. brasiliensis: N = 211, He = 0,554), distribuídos homogeneamente entre as localidades (S. deceptor: ΦST < -0,004, ΦCT < 0,016, FST global = 0,001, Dest global = 0,003, FCT < 0,002, P > 0,05, K = 1; S. brasiliensis: FST global = 0,004, Dest global = 0,001, FCT < 0,004, P > 0,05, K = 1). A ausência de estruturação nas duas espécies pode estar relacionada a características biológicas que promovem a conectividade entre localidades geograficamente distantes, como alta fecundidade e alto potencial de dispersão das larvas planctônicas. Além disso, os dados mitocondriais sugerem que a história demográfica de S. deceptor foi marcada por eventos de expansão populacionais e geográficos possivelmente relacionados às condições ambientais favoráveis dos episódios interglaciais do Pleistoceno Médio-Tardio. Diversos estudos têm mostrado que os fenômenos de inserção de regiões mitocondriais no DNA nuclear (NuMts) e heteroplasmia limitam a correta amplificação e identificação dos marcadores mitocondriais. Em estudos filogenéticos e de genética de populações, a presença inadvertida de sequências de diversas origens viola o principio de ortologia, o que pode resultar em inferências evolutivas erradas. Assim, o objetivo do segundo capítulo desta tese foi identificar e caracterizar os possíveis NuMts e sequências heteroplásmicas de três regiões mitocondriais (COI, RC e o gene da subunidade maior do RNA ribossomal: 16S) em quatro espécies do gênero Scyllarides (S. aequinoctialis, S. brasiliensis, S. deceptor e S. delfosi). A clonagem e sequenciamento de extratos de DNA genômico e DNA enriquecido com mtDNA revelaram que os genomas destas espécies podem exibir NuMts (que divergem entre 0,6 e 17,6% do mtDNA) e heteroplasmia (que divergem < 0,2% do mtDNA prevalente). Os NuMts surgiram possivelmente de vários eventos independentes de integração ao núcleo ao longo da história evolutiva do gênero Scyllarides. Dependendo do seu grau de similaridade com o mtDNA, a presença de NuMts nas análises filogenéticas no nível de gênero pode causar superestimativa do número de espécies e alterações nos comprimentos dos ramos e nas relações filogenéticas entre espécies.

An experiment was conducted with Labeo rohita fingerlings in an indoor static fish rearing water system of glass made aquaria. Five experimental diets A, B, C, D and E were formulated containing 33% dietary protein level in five treatments each having two replicates containing 12 fingerlings of mean total initial weight of 13.00±0.2g. Sixty days of feeding trial in this experiment showed that fish fed on diet 'A' containing fish meal and diet 'E' containing mixed plant sources protein had significantly highest and lowest growth respectively. However, no significant difference of growth was found in fish fed on diets C and D containing meat and bone meal, and mix of animal protein source diets respectively. The result showed that the apparent protein digestibility (APD) of diets 'A' and 'E' had significantly best and least values respectively. Food conversion ratio (FCR) and protein efficiency ratio (PER) ranged between 1.37 to 2.17 and 1.38 to 2.18 respectively. On the basis of observed FCR and PER diets 'A' and 'E' produced significantly highest and lowest growth respectively.

Tripura is a densely populated small state with meagre water resources. 47.51% of the population is constituted by socio-economically backward, illiterate, orthodox, tribal and scheduled castes. Some of them are nomad and a majority of the rest of the population is refugees from Bangladesh, but almost 100% is fish eater. Settlement of tribes in villages, provision of nutritious food and employment therefore calls for proper utilization of every resource they have. The State is poor in water resources but recently has created 21,636.23 ha of new water area. Tripura is rich in pig population, besides poultry birds. Paddy is the main crop cultivated in arable lands. An integration of livestock raising and land based agriculture with pisciculture practices around mini barrages will help in solving the problems to a great extent. The paper is an attempt to outline the prospects of integration of aquaculture with animal husbandry and land based crop culture in Tripura State.

Although shrimp head meal alone does not provide for good growth and survival, fish meal can provide high survival rate. The addition of shrimp head improves this diet. It is suggested that cholesterol present in shrimp could have caused this difference. Composition of the test diets is tabulated, as are proximate chemical analysis of the diets, and the mean initial weights, final weights, weight gains, survival rate, feed consumed, protein consumed, of Penaeus monodon postlarvae, feed conversion and protein efficiency ratio.

Identification of venomous species of Persian Gulf cone snails and characterization of venom composition and their features is so important from the point of medical importance. Marine cone snails from the genus Conus are estimated to consist of up to 700 species. The venom of cone snails has yielded a rich source of novel neuroactive peptides or conotoxins. The present study was aimed to study the analgesic effect of Persian Gulf Conus textile and its comparison with morphine in mouse model. The specimens of Conus textile were collected of Larak Island from depth of 7 m. The collected samples were transferred to laboratory alive and were stored at -700 c. he veno s ducts were separated and ho ogenized with deionized water he ixture centrifuged at rp for inutes upernatant was considered as extracted veno and stored at - C after lyophylization. The protein profile of venom determined by using SDS-PAGE and HPLC used to investigate the extracted venom and to evaluate the analgesic activity, formalin test was carried out. SDS-PAGE indicated several bands ranged between 6 and 250 kDa. Chromatogram of the venom demonstrated more than 44 large and small fractions. The amount of 10 ng of Conus crude venom and analgesic peptide showed the best anti-pain activity in formalin test. No death observed up to 100 mg/kg, which is 250,000 times higher than the effective dose.Venom characterization of Persian Gulf Conus textile may be of medical importance and potential for new pharmaceutical drugs as well.

In this study, we investigated the effects of animal-plant protein ratio in extruded and expanded diets on nutrient digestibility, nitrogen and energy budgets of juvenile soft-shelled turtle (Pelodiscus sinensis). Four extruded and expanded feeds (diets 1-4) were formulated with different animal-plant protein ratios (diet 1, 1.50:1; diet 2, 2.95:1; diet 3, 4.92:1; diet 4, 7.29:1). The apparent digestibility coefficients (ADCs) of dry matter and crude lipid for diet 1 were significantly lower than those for diets 2-4. There was no significant difference in crude protein digestibility among diets 1-4. The ADC of carbohydrate was significantly increased with the increase in animal-plant protein. Although nitrogen intake rate, faecal nitrogen loss rate and excretory nitrogen loss rate of turtles fed diet 1 were significantly higher than those fed diets 2-4, nitrogen retention rate, net protein utilization and biological value of protein in these turtles were significantly lower than those fed diets 2-4. In addition, energy intake rate, excretory energy loss rate and heat production rate of turtles fed diet 1 were also significantly higher than those fed diets 2-4. Faecal energy loss was significantly reduced with the increase in the animal-plant protein ratio. The ADC of energy and assimilation efficiency of energy significantly increased with a higher animal-plant protein ratio. The growth efficiency of energy in the group fed diet 1 was significantly lower than those in the groups fed diets 2-4. Together, our results suggest that the optimum animal-plant protein ratio in extruded and expanded diets is around 3:1.

In the present research, two Chinese rhesus monkeys were inoculated intravenously with 5000 TCID50 of SIVmac239. The changes in the numbers of CD4+ T lymphocyte in peripheral blood, plasma viral loads, proviral DNA and humoral antibodies against virus were periodically monitored during 121 days. At the early stage of infection, proviral DNA had been detected in PBMCs, and infectious SIVmac239 virus had been isolated from PBMCs. At the same period, the numbers of CD4+ T lymphocytes were significantly decreased, and maintained at low level during the 121-day period of infection. Plasma viral loads reached the peak at week 2 post-inoculation and kept at a steady state subsequently. Moreover, antibodies against viral proteins were detected from plasma. All the results showed that the two Chinese rhesus monkeys had been infected with SIVmac239 successfully. This animal model can be applied for further AIDS researches.
