Les sélénoprotéines sont des protéines auxquelles des sélénocystéines, soit le 21e acide aminé, sont incorporées durant leur traduction. Plus précisément, la sélénocystéine (Sec) est un dérivé métabolique de la sérine, mais structurellement équivalent à une cystéine dont on a remplacé l'atome de soufre par du sélénium. Elle se distingue des autres acides aminés puisqu’elle possède sa propre synthétase qui sert à convertir la sérine en Sec alors que le résidu est déjà fixé à l’ARNt. La position d’une Sec sur l’ARNm est indiquée par le codon UGA étant habituellement un signal STOP introduisant le concept de recoding. Grâce à une machinerie métabolique spécifique à l'ARNtSec et à la présence d’un SecIS (Selenocystein Insertion Sequence) sur l’ARNm, ce codon permet la présence d'une Sec dans la protéine. Il est connu que la synthèse débute avec l’acétylation de l’ARNt[Ser]Sec par la seryl-ARNt synthétase (SerRS) afin de donner la seryl-ARNt[Ser]Sec. Cette dernière est subséquemment phosphorylée par l’O-phosphoséryl-ARNt[Ser]Sec kinase (PSTK) qui donnera l’O-phosphoséryl-ARNt[Ser]Sec. Par la suite, un complexe de plusieurs protéines et cofacteurs, agissant comme machinerie pour l’incorporation des Sec durant la traduction, s’associe avec l’ARNt[Ser]Sec puis l’ARNm et, finalement, les composantes du ribosome. Parmi ces protéines, SepSecS catalyse l’étape finale de la synthèse des Sec en convertissant le O-phosphoseryl-ARNt[Ser]Sec en selenocysteinyl-ARNt[Ser]Sec utilisant le sélénophosphate comme source de sélénium. Des études récentes montrent que l’association avec SECp43 serait nécessaire pour que SepSecS joue son rôle et soit ségrégée au noyau pour s’associer à la machinerie de biosynthèse des sélénoprotéines, soit le complexe moléculaire qui reconnaît le codon UGA. Parmi les protéines de la machinerie de biosynthèse des sélénoprotéines que nous avons analysées, il y a eEFSec, RPL30, SPS2, SPS1, SBP2 et NSEP1. Nos résultats d’analyse de la dynamique de l’interaction entre les constituants de la machinerie de biosynthèse et d’incorporation des Sec, confirment plusieurs données de la littérature, mais remettent en question le modèle jusqu’à maintenant établi. Une meilleure compréhension de la dynamique des interactions entre ses constituants et la régulation de cette dynamique permet d’émettre des hypothèses quant au rôle de la machinerie de biosynthèse des sélénoprotéines et de l’importance de sa complexité. Nous avons analysé les interactions in vivo dans des cellules HEK293T au moyen de la technique de Protein-Fragment Complementation Assay (PCA) en couplant, par un clonage moléculaire, les gènes de chacune des protéines d’intérêt avec des fragments des gènes de la protéine luciférase (hRluc). Nous avons ainsi réalisé une fusion en N-terminal et en C-terminal des fragments de luciférase pour chacune des protéines d’intérêt. Puis, nous avons analysé la dynamique des interactions avec les composantes de la machinerie de biosynthèse des Sec. D’autres travaux seront essentiels afin de bâtir sur les résultats présentés dans cette recherche.

La purification est une métaphore désignant le moteur de la philosophie de la religion de Hegel. Elle est d’abord à l’œuvre dans la création de la Nature qui se consume pour produire la conscience de soi divine à travers l'esprit humain. En second lieu, elle s’opère dans l’objectivation des productions spirituelles de l’homme qui sont purifiées jusqu'à ce que l’Esprit soit auprès de soi dans le christianisme. La troisième purification est morale et trouve son fondement dans la Genèse, le judaïsme étant le premier à avoir identifié l'unité des natures humaine et divine. Le mythe témoignera également de la culpabilité en tant que l'homme n'exprime pas immédiatement sa divinité, mais sa finitude. La réalisation du divin impliquera donc la purification de la naturalité au profit de la substantialité. Le christianisme explicitera cette tâche par l’héroïsme de Jésus et cet héroïsme se perpétuera jusqu’à ce qu’émergent un individualisme moderne et une religion assurant la cohésion sociale : le protestantisme luthérien. Cet individualisme sera toutefois défectueux puisqu’il produira éventuellement davantage d’égoïsme que de réconciliation, ce qui donnera lieu à certaines critiques de l’analyse hégélienne du christianisme. En effet, Hegel croit toujours que la vitalité religieuse est nécessaire au fonctionnement de l’État, bien qu’elle soit dorénavant incapable de diffuser les sentiments de culpabilité et de responsabilité dans le corps social. Néanmoins, comme les valeurs du christianisme ont été épurées de leur contingence en passant dans les mœurs et dans l’État, il s’avérera que le corps social peut se passer d’une tradition religieuse vivante

Stock markets employ specialized traders, market-makers, designed to provide liquidity and volume to the market by constantly supplying both supply and demand. In this paper, we demonstrate a novel method for modeling the market as a dynamic system and a reinforcement learning algorithm that learns profitable market-making strategies when run on this model. The sequence of buys and sells for a particular stock, the order flow, we model as an Input-Output Hidden Markov Model fit to historical data. When combined with the dynamics of the order book, this creates a highly non-linear and difficult dynamic system. Our reinforcement learning algorithm, based on likelihood ratios, is run on this partially-observable environment. We demonstrate learning results for two separate real stocks.

The impact that “Romanization” and the development of urban centers had on the health of the Romano-British population is little understood. A re-examination of the skeletal remains of 364 nonadults from the civitas capital at Roman Dorchester (Durnovaria) in Dorset was carried out to measure the health of the children living in this small urban area. The cemetery population was divided into two groups; the first buried their dead organized within an east–west alignment with possible Christian-style graves, and the second with more varied “pagan” graves, aligned north–south. A higher prevalence of malnutrition and trauma was evident in the children from Dorchester than in any other published Romano-British group, with levels similar to those seen in postmedieval industrial communities. Cribra orbitalia was present in 38.5% of the children, with rickets and/or scurvy at 11.2%. Twelve children displayed fractures of the ribs, with 50% of cases associated with rickets and/or scurvy, suggesting that rib fractures should be considered during the diagnosis of these conditions. The high prevalence of anemia, rickets, and scurvy in the Poundbury children, and especially the infants, indicates that this community may have adopted child-rearing practices that involved fasting the newborn, a poor quality weaning diet, and swaddling, leading to general malnutrition and inadequate exposure to sunlight. The Pagan group showed no evidence of scurvy or rib fractures, indicating difference in religious and child-rearing practices but that both burial groups were equally susceptible to rickets and anemia suggests a shared poor standard of living in this urban environment.

The present study investigates the initiation of precipitating deep convection in an ensemble of convection-resolving mesoscale models. Results of eight different model runs from five non-hydrostatic models are compared for a case of the Convective and Orographically-induced Precipitation Study (COPS). An isolated convective cell initiated east of the Black Forest crest in southwest Germany, although convective available potential energy was only moderate and convective inhibition was high. Measurements revealed that, due to the absence of synoptic forcing, convection was initiated by local processes related to the orography. In particular, the lifting by low-level convergence in the planetary boundary layer is assumed to be the dominant process on that day. The models used different configurations as well as different initial and boundary conditions. By comparing the different model performance with each other and with measurements, the processes which need to be well represented to initiate convection at the right place and time are discussed. Besides an accurate specification of the thermodynamic and kinematic fields, the results highlight the role of boundary-layer convergence features for quantitative precipitation forecasts in mountainous terrain.

There has been an increasing interest in the impact of individual well-being on the attitudes and actions of people receiving services designed to offer support. If well-being factors are important in the uptake and success of service programmes it is important that the nature of the relationships involved is understood by service designers and implementers. As a contribution to understanding, this paper examines the impact of well-being on the uptake of intervention programmes for homeless people. From the literature on well-being a number of factors are identified that contribute towards overall well-being, which include personal efficacy and identity, but also more directly well-being can be viewed as personal or group/collective esteem. The impact of these factors on service use is assessed by means of two studies of homelessness service users, comparing the implementation of two research tools: a shortened and a fuller one. The conclusions are that the factors identified are related to service use. The higher the collective esteem – esteem drawn from identification with services and their users and providers – and the less that they feel isolated, the more benefits that homeless people will perceive with service use, and in turn the more likely they are to be motivated to use services. However, the most important factors in explaining service use are a real sense that it is appropriate to accept social support from others, a rejection of the social identity as homeless but a cultivation of being valued as part of a non-homeless community, and a positive perception of the impact of the service.

TGR5 is a G protein-coupled receptor that mediates bile acid (BA) effects on energy balance, inflammation, digestion and sensation. The mechanisms and spatiotemporal control of TGR5 signaling are poorly understood. We investigated TGR5 signaling and trafficking in transfected HEK293 cells and colonocytes (NCM460) that endogenously express TGR5. BAs (deoxycholic acid, DCA, taurolithocholic acid, TLCA) and the selective agonists oleanolic acid (OA) and 3-(2-chlorophenyl)-N-(4-chlorophenyl)-N, 5-dimethylisoxazole-4-carboxamide (CCDC) stimulated cAMP formation but did not induce TGR5 endocytosis or recruitment of β-arrestins, assessed by confocal microscopy. DCA, TLCA and OA did not stimulate TGR5 association with β-arrestin 1/2 or G protein-coupled receptor kinase (GRK) 2/5/6, determined by bioluminescence resonance energy transfer. CCDC stimulated a low level of TGR5 interaction with β-arrestin2 and GRK2. DCA induced cAMP formation at the plasma membrane and cytosol, determined using exchange factor directly regulated by cAMP (Epac2)-based reporters, but cAMP signals did not desensitize. AG1478, an inhibitor of epidermal growth factor receptor (EGFR) tyrosine kinase, the metalloprotease inhibitor batimastat, and methyl-β-cyclodextrin and filipin, which block lipid raft formation, prevented DCA stimulation of extracellular signal regulated kinase (ERK1/2). BRET analysis revealed TGR5 and EGFR interactions that were blocked by disruption of lipid rafts. DCA stimulated TGR5 redistribution to plasma membrane microdomains, localized by immunogold electron microscopy. Thus, TGR5 does not interact with β-arrestins, desensitize or traffic to endosomes. TGR5 signals from plasma membrane rafts that facilitate EGFR interaction and transactivation. An understanding of the spatiotemporal control of TGR5 signaling provides insights into the actions of BAs and therapeutic TGR5 agonists/antagonists.

Rationale:Metabolic Syndrome (MetS) is a high prevalence condition characterized by altered energy metabolism, insulin resistance and elevated cardiovascular risk.Objectives:Although many individual single nucleotide polymorphisms (SNPs) have been linked to certain MetS features, there are few studies analyzing the influence of SNPs on carbohydrate metabolism in MetS.Methods:904 SNPs (tag SNPs and functional SNPs) were tested for influence in eight fasting and dynamic markers of carbohydrate metabolism, performing an intravenous glucose tolerance test in 450 participants of the LIPGENE study.Findings:From 382 initial gene-phenotype associations between SNPs and any phenotypic variables, 61 (a 16 % of the pre-selected) remained significant after Bootstrapping. Top SNPs affecting glucose metabolism variables were as follows: fasting glucose: rs26125 (PPARGC1B); fasting insulin: rs4759277 (LRP1); C peptide: rs4759277 (LRP1); HOMA-IR: rs4759277 (LRP1); QUICKI: rs184003 (AGER); SI: rs7301876 (ABCC9), AIRg: rs290481 (TCF7L2) and DI: rs12691 (CEBPA).Conclusions:We describe here the top SNPs linked to phenotypic features in carbohydrate metabolism among aproximately 1000 candidate gene variations in fasting and postprandial samples of 450 patients with MetS from the LIPGENE study.

The new Max-Planck-Institute Earth System Model (MPI-ESM) is used in the Coupled Model Intercomparison Project phase 5 (CMIP5) in a series of climate change experiments for either idealized CO2-only forcing or forcings based on observations and the Representative Concentration Pathway (RCP) scenarios. The paper gives an overview of the model configurations, experiments related forcings, and initialization procedures and presents results for the simulated changes in climate and carbon cycle. It is found that the climate feedback depends on the global warming and possibly the forcing history. The global warming from climatological 1850 conditions to 2080–2100 ranges from 1.5°C under the RCP2.6 scenario to 4.4°C under the RCP8.5 scenario. Over this range, the patterns of temperature and precipitation change are nearly independent of the global warming. The model shows a tendency to reduce the ocean heat uptake efficiency toward a warmer climate, and hence acceleration in warming in the later years. The precipitation sensitivity can be as high as 2.5% K−1 if the CO2 concentration is constant, or as small as 1.6% K−1, if the CO2 concentration is increasing. The oceanic uptake of anthropogenic carbon increases over time in all scenarios, being smallest in the experiment forced by RCP2.6 and largest in that for RCP8.5. The land also serves as a net carbon sink in all scenarios, predominantly in boreal regions. The strong tropical carbon sources found in the RCP2.6 and RCP8.5 experiments are almost absent in the RCP4.5 experiment, which can be explained by reforestation in the RCP4.5 scenario.

Adult human neural crest-derived stem cells (NCSCs) are of extraordinary high plasticity and promising candidates for the use in regenerative medicine. Here we describe for the first time a novel neural crest-derived stem cell population within the respiratory epithelium of human adult inferior turbinate. In contrast to superior and middle turbinates, high amounts of source material could be isolated from human inferior turbinates. Using minimally-invasive surgery methods isolation is efficient even in older patients. Within their endogenous niche, inferior turbinate stem cells (ITSCs) expressed high levels of nestin, p75(NTR), and S100. Immunoelectron microscopy using anti-p75 antibodies displayed that ITSCs are of glial origin and closely related to nonmyelinating Schwann cells. Cultivated ITSCs were positive for nestin and S100 and the neural crest markers Slug and SOX10. Whole genome microarray analysis showed pronounced differences to human ES cells in respect to pluripotency markers OCT4, SOX2, LIN28, and NANOG, whereas expression of WDR5, KLF4, and c-MYC was nearly similar. ITSCs were able to differentiate into cells with neuro-ectodermal and mesodermal phenotype. Additionally ITSCs are able to survive and perform neural crest typical chain migration in vivo when transplanted into chicken embryos. However ITSCs do not form teratomas in severe combined immunodeficient mice. Finally, we developed a separation strategy based on magnetic cell sorting of p75(NTR) positive ITSCs that formed larger neurospheres and proliferated faster than p75(NTR) negative ITSCs. Taken together our study describes a novel, readily accessible source of multipotent human NCSCs for potential cell-replacement therapy.

Slavic and German colonization of the southern Baltic between the 8th and 15th centuries A.D. is well-documented archaeologically and historically. Despite the large number of pollen profiles from Poland, few palaeoecological studies have examined the ecological impact of a process that was central to the expansion of European, Christian, societies. This study aims to redress this balance through multiproxy analysis of lake sediments from Radzyń Chełminski, Northern Poland, using pollen, element geochemistry (Inductively Coupled-Optical Emission Spectroscopy [ICP-OES]), organic content, and magnetic susceptibility. The close association between lake and medieval settlements presents the ideal opportunity to reconstruct past vegetation and land-use dynamics within a well-documented archaeological, historical, and cultural context. Three broad phases of increasing landscape impact are visible in the pollen and geochemical data dating from the 8th/9th, 10th/11th, and 13th centuries, reflecting successive phases of Slavic and German colonization. This involved the progressive clearance of oak-hornbeam dominated woodland and the development of an increasingly open agricultural landscape. Although the castles and towns of the Teutonic Order remain the most visible signs of medieval colonization, the palynological and geochemical data demonstrate that the major phase of woodland impact occurred during the preceding phase of Slavic expansion; Germans colonists were entering a landscape already significantly altered.
