The water buffalo is vital to the lives of small farmers and to the economy of many countries worldwide. Not only are they draught animals, but they are also a source of meat, horns, skin and particularly the rich and precious milk that may be converted to creams, butter, yogurt and many cheeses. Genome analysis of water buffalo has advanced significantly in recent years. This review focuses on currently available genome resources in water buffalo in terms of cytogenetic characterization, whole genome mapping and next generation sequencing. No doubt, these resources indicate that genome science comes of age in the species and will provide knowledge and technologies to help optimize production potential, reproduction efficiency, product quality, nutritional value and resistance to diseases. As water buffalo and domestic cattle, both members of the Bovidae family, are closely related, the vast amount of cattle genetic/genomic resources might serve as shortcuts for the buffalo community to further advance genome science and biotechnologies in the species.

Nos últimos anos, os avanços nas Ciências Biológicas têm levado a sociedade a discutir diversas questões no campo da moral e da ética. Questões como engenharia genética, clonagem e pesquisas com células-tronco são questões chamadas de sociocientíficas por estarem na interface entre a ciência e a sociedade. Nesse trabalho buscamos entender como estudantes de Ensino Médio percebem e interpretam questões relacionadas à manipulação genética em seres humanos. Houve divisão de opiniões em relação à eugenia negativa, que se destina a remover características desfavoráveis das pessoas; mas a eugenia positiva, que busca melhoramento de características estéticas, foi rejeitada por todos os estudantes. As variações nas opiniões em relação ao assunto tratado podem ser, em grande medida, devidas às representações sociais dos estudantes.

As variantes gênicas da beta-lactoglobulina (β-LG) e da kappa-caseína (κ-CN) bovinas são associadas à produção, qualidade e características de processamento do leite. O objetivo deste trabalho foi analisar as frequências dos genótipos AA, AB e BB, por meio da técnica de PCR-RFLP, da β-LG e da κ-CN bovinas, e suas associações com a produção de leite (kg leite/dia) em bovinos das raças Girolanda, Holandesa e Jersey. Para a κ-CN, a frequência do genótipo AA foi maior nos animais das raças Holandesa (37%) e Girolanda (63%). Na raça Jersey, houve predomínio do genótipo BB (60%). Para a β-LG, o genótipo AB foi o mais encontrado nas raças Girolanda (54%) e Holandesa (58%), enquanto nos animais da raça Jersey houve predomínio do genótipo BB (45%). Houve associação do alelo B da κ-CN com maior produtividade leiteira nas raças Girolanda e Holandesa, e do alelo A da β-LG com maior produtividade de leite na raça Jersey. As variantes genéticas da κ-CN podem ser usadas como marcadores na seleção para a produtividade leiteira nas raças Girolanda e Holandesa. Para a raça Jersey, as variantes da β-LG seriam mais adequadas para essa seleção.

This paper deals with the effects of hair coat characteristics on the sweating rate of Brazilian Braford cows and estimation of heritabilities and genetic correlations of these traits. Data (n=1607) on hair length, coat thickness, hair diameter, number of hairs per unit area, coat reflectance and sweating rate were recorded from heifers and cows of a commercial herd managed on range under extensive system. The data were analyzed considering the following effects on the model for hair coat traits: classes of sires and contemporary groups; linear effects of month and genotype; linear and quadratic effects of age. The effect of sire was important (P<0.05) for all hair coat traits, except for number of hairs; contemporary groups affected (P<0.05) all hair coat traits; the effect of sampling month was important (P<0.05) for hair length and reflectance; genotype affected (P<0.05) hair length, diameter and coat reflectance; the quadratic effect of age was important (P<0.05) only for coat reflectance. Two models were used to analyze the sweating rate. The first model considered the following fixed effects: classes of contemporary groups and sires; linear effect of genotype, coat thickness, hair length, hair diameter, number of hairs, coat reflectance; linear and quadratic effects of time of day, age, air temperature, partial vapour pressure and radiant heat load. The second model used for the sweating rate considered the same fixed effects for the first model, except that the hair coat characteristics were adjusted for important effects used in the models to analyze hair coat traits. All meteorological factors and contemporary groups were important (P<0.05) on variation of sweating rate in both models. The Restricted Maximum. Likelihood (REML) method was used to estimate variance and covariance components under the sire model. Results included heritability estimates in narrow (h(2)) and broad (H) sense for single-trait analyzes: hair thickness (h(2)=0.16; H-2=0.26); hair length (h(2)=0.18; H-2=0.39); number of hairs (h(2)=0.08 +/- 0.07; H-2=0.08 +/- 0.07); hair diameter (h(2)=0.12 +/- 0.07; H-2=0.12 +/- 0.07); coat reflectance (h(2)=0.30; H-2=0.42); and sweating rate (h(2)=0.10 +/- 0.07; H-2=0.10 +/- 0.07). In general, the genetic correlations between the adaptive traits were favorable as for the direction to select for adaptation in tropical environment; however, they presented high standard errors. The results of this study imply that hair coat characteristics and sweating ability are important for the adaptability to heat stress and they must be better studied and further considered for selection for genetic progress of adaptation in tropical environment. The objective of this study was to evaluate the genotype x environment interaction for weaning and yearling weights, daily weight gain from weaning to 12 months of age and the growth performance in Canchim (5/8 Charolais + 3/8 Zebu) beef cattle estimated by a principal components analysis including those three traits. The environment was defined by season of birth (first and second semesters of the year). Genetic parameters were estimated by bayesian method with the Gibbs sampler using bivariate analyses (considering the trait in each of the two seasons as a different one) and models that included the fixed effects of year and month of birth, sex and age of cow (linear and quadratic) and the random effects of animal and residual. The results suggested that genetic evaluation and selection in Canchim beef cattle for the traits studied should consider the genotype and season of birth interaction.

The objectives of this study were to obtain heritability and genetic, phenotypic and environmental correlation estimates for birth (B), weaning (WW), yearling (YW) and 18 - (EW) and 24 - month (TW) weights, and to propose selection criteria for body weight in Canchim cattle. The data were analyzed by the least-squares method with models that included the fixed effects of herd, year and month of birth, sex of calf and age of cow at calving, and the random effects of sire within herd. The heritability estimates obtained were equal to 0.36 0.06 (BW), 0.47 0.06 (WW), 0.53 0.07 (YW), 0.54 0.08 (EW) and 0.27 0.06 (TW). The genetic correlations were equal to 0.51 (BW and WW), 0.36 (BW and YW), 0.14 (BW and EW), 0.00 (BW and TW), 0.92 (WW and YW), 0.77 (WW and EW), 0.75 (WW and TW), 0.94 (YW and EW), 0.86 (YW and TW) and 0.85 (EW and TW). The phenotypic correlations ranged from 0.19 to 0.72, and the environmental correlations from 0.11 to 0.61. The results showed that, in general, mass selection for weight will result in genetic progress, selection for weight at any age will result in correlated changes at other ages, and EW and TW are good selection criteria for increasing weight in the Canchim breed.

Seeds of Astronium fraxinifolium (Anarcadiaceae) were collected in two years, and submitted to tests of seed germination and vigour (first count, aging and electric conductivity), in order to verify the potencial of conservation and if variations in the seed quality presented on association with genetic parameters. Coefficient of heritability in average (ĥm 2) were 0.91 and 0.97 to germination; 0.90 and 0.96 to first count; 0.93 and 0.97 to aging and 0.95 and 0.99 to electric conductivity, for seeds collected in 1997 and 1998, respectively, evidencing a great heritability to all studied characteristics. Genetic variability was influenced more by environmental conditions of blooming and harvest of seeds than storage time.

Levels of genetic variability for in situ and ex situ genetic conservation were estimated in a population of Myracrodruon urundeuva using the PCR (polymerase chain reaction) technique with the AFLP (Amplified fragment-length polymorphism) genetic marker. Seeds for progeny tests were collected from 30 open-pollination trees (matrices) at Paulo de Faria Ecological Station - SP. From this genetic material, three progeny tests were installed on the Teaching and Research Farm of Ilha Solteira Faculty of Engineering - University of São Paulo State (UNESP), which is located in Selvlria - MS, Brazil. The analysis by genetic marker was conducted with three combinations of different starters EcoRl-Msel, resulting in a total number of 137 polymorphic bands, thus forming a table of binary data. These data were used for the analysis of genetic divergence and distance between progenies. High levels of genetic divergence were observed among families. Based on the Analysis of Molecular Variance (AMOVA), it was shown that 16.2% of genetic diversity is found among progenies and 83.8% within progenies, which suggests deviances of random matings. The grouping of progenies, based on genetic distances, suggests that progenies deriving from trees which are close to each other tend to be more similar. This, in turn, indicates that the population originating the seeds may be genetically structured.

The objective of this work was to study the effect of selective thinning on the estimates of genetic parameters in progenies of Pinus caribaea var. hondurensis. The progeny test was installed in June 1986, following a 10 × 10, triple square lattice design, containing ten trees in linear plots in 3.0 × 3.0m spacing. Twelve years after the planting, a selective thinning based on Multi-effect index (selection for DBH) was performed, leaving six trees per plot. The assessments were done in four situations: A (before thinning); B (among thinned trees); C (among remnant trees at 13 years of age) and D (two years after the thinning). The following traits were analyzed: total height (H), diameter at breast height (DBH), volume (VOL), stem form (FOR), foxtail (FT), wood density at 1.3 m (WD1) and wood density at the half height (WD2). The thinning led to increase in the estimates of variability among progenies for VOL and decrease for ALT and DBH. There was a high and positive genetic correlation among the growth traits, mainly between DBH and VOL. The latter remained unchanged after thinning. The narrow-sense heritabilities at the individual level, showed a slight increase while at the family mean level had a reduction with the thinning. The estimates of breeding values increased with the thinning. This may lead to an easier identification of the best progenies in further selection.

Data from purebred Simmental, Nellore and Canchim cattle breeds obtained from the respective Brazilian Associations of Breeders were used to estimate variance components and to predict genetic values for 365 days weight. The results obtained by Bayesian inference were compared to those from Restricted Maximum Likelihood (REML) and Best Linear Unbiased Prediction (BLUP), which are the most commonly used methods of estimation and prediction in animal breeding. The two methods presented similar point estimates but the study of the marginal posterior distributions in the Bayesian approach yields more detailed information about the parameters and other unknowns in the model.

The aim of this study was to investigate the genetic variation, the genotype × soil interaction and the selection among and within Corymbia citriodora progenies in three different kinds of soils (Red Latossol, Quartz Sand and Purple Latossol), which occur in the Luiz Antônio Experimental Station, São Paulo State, Brazil. The progeny test was established 1983, using 56 open-pollinated families of C. citriodora. Twenty fve years after planting the following traits were measured: height, diameter at breast height (Dbh), stem form and survival. Best growth occurred in Purple Latossol. Significant differences among progenies were detected for most traits in all sites, suggesting the possibility of improvement by selection. In the analysis with sites, Significant differences among locals, progenies and genotypes × soil interaction were detected, confiming that the tested material has sufficient genetic variation to be explored by selection in all sites. The analysis of genotype × soil interaction indicates that growth traits present single interaction and the same progenies can be selected in each site. On the order hand, genotype × soil interaction for stem form and survival was complex; and specifc progenies need to be selected for each kind of soil. The population, due to its high genetic variation and strong genetic control of traits, permits to obtain considerable genetic gains by selection among and within progenies.
