Stomach samples from three rockfish species, yellowtail (Sebastes f lavidus), widow (S. entomelas), and canary (S. pinniger) rockfish, seasonally collected off the Pacific Northwest in 1998 and 1999, provided quantitative information on the food habits of these species during and after the 1997–98 El Niño event. Although euphausiids were the most common major prey of all three predators, gelatinous zooplankton and fishes were the most commonly consumed prey items during some seasonal quarters. The influence of the El Niño event was evident in the diets. Anomalous prey items, including the southern euphausiid species Nyctiphanes simplex and juveniles of Pacific whiting (Merluccius productus) frequently appeared in the diets in the spring and summer of 1998. The results of stomach contents analyses, based on 905 stomach samples from 49 trawl hauls during seven commercial fishing trips and from 56 stations during research surveys, were consistent with the timing of occurrence and the magnitude of change in biomass of some zooplankton species reported from zooplankton studies in the northern California Current during the 1997–98 El Niño. Our findings indicate that the observed variations of prey groups in some rockfish diets may be a function of prey variability related to climate and environment changes.

The use of parasites as indicators of the stock structure of Pacific halibut (Hippoglossus stenolepis) in the northeast Pacific was investigated by using 328 adult (>55 cm fork length) halibut from 15 composite localities ranging from northern California to the northern Bering Sea and 96 juvenile (10–55 cm) halibut from five localities ranging from the northern Queen Charlotte Islands to the Bering Sea. Counts of eight selected parasite species (the juvenile acanthocephalans Corynosoma strumosum and C. villosum, the metacestode Nybelinia surmenicola, the digenean metacercaria Otodistomum sp., and the larval nematodes Anisakis simplex, Pseudoterranova decipiens, Contracaecum sp., and Spirurid gen. sp.) that produce infections of long duration, do not multiply in the host, and that have a relatively high abundance in at least one geographic locality were subjected to discriminant function analysis. Juvenile Pacific halibut showed no separation and, even though they were not heavily infected with parasites, the analysis suggested that juveniles could be a mixed stock. Three groups of adults were identified: fish from California to the southern Queen Charlotte Islands, those from the northern Queen Charlotte Islands to the central Bering Sea, and those from the central and north-ern Bering Sea. These groups suggest that the single stock concept be more thoroughly evaluated.

Qualitative estimation of phytoplankton and zooplankton of the northern Red Sea and Gulf of Aqaba were carried out from four sites: Sharm El-Sheikh, Taba, Hurghada and Safaga. A total of 106 species and varieties of phytoplankton were identified including 41 diatoms, 53 dinoflagellates, 10 cyanophytes and 2 chlorophytes. The highest number of species was recorded at Sharm El-Sheikh (46 spp), followed by Safaga (40 spp), Taba (30 spp), and Hurghada (23 spp). About 95 of the recorded species were previously mentioned by different authors in the Red Sea and Gulf of Suez. Eleven species are considered new to the Red Sea. About 115 species of zooplankton were recorded from the different sites. They were dominated by four main phyla namely: Arthropoda, Protozoa, Mollusca, and Urochordata. Sharm El-Sheikh contributed the highest number of species (91) followed by Safaga (47) and Taba (34). Hurghada contributed the least (26). Copepoda dominated the other groups at the four sites. The appearances of Spirulina platensis, Pediastrum simplex, and Oscillatoria spp. of phytoplankton in addition to the rotifer species and the protozoan Difflugia oblongata of zooplankton impart a characteristic feature of inland freshwater discharge due to wastewater dumping at sea in these regions resulting from the expansion of cities and hotels along the coast.

Sodium rutin sulfate (SRS) is a sulfated rutin modified from the natural flavonol glycoside rutin. Here, we investigated its in vitro anti-HIV and -HSV activities and its cytotoxic profile. Fifty percent inhibitory concentration (IC50) values of SRS against HIV-1 X4 virus IIIB, HIV-1 R5 isolates Ada-M and Ba-L were 2.3 +/- 0.2, 4.5 +/- 2.0 and 8.5 +/- 3.8 mu M with a selectivity index (SI) of 563, 575 and 329, respectively. Its IC50 against primary R5 HIV-1 isolate from Yunnan province in China was 13.1 +/- 5.5 mu M, with a Sl of 197. In contrast, unsulfated rutin had no activity against any of the HIV-1 isolates tested. Further study indicated that SRS blocked viral entry and virus-cell fusion likely through interacting with the HIV- I envelope glycoprotein. SRS also demonstrated some activity against human herpes simplex virus (HSV) with an IC50 of 88.3 +/- 0.1 mu M and a Sl of 30. The 50% cytotoxicity concentration (CC50) of SRS was >3.0 mM, as determined in human genital ME 180, HeLa and primary human foreskin fibroblast cells. Minimum inhibitory concentration of SRS for vaginal lactobacilli was >3.0 mM. These results collectively indicate that SRS represents a novel candidate for anti-HIV-1/HSV microbicide development. The gene targeting technique is a powerful tool for analyzing functions of cloned genes and for generating transgenic animals with site-directed integration of foreign genes. In order to develop this technique in fish, positive-negative selection (PNS) and homologous recombination vectors were constructed, and their expression was examined in fish cells. A vector (pNK) for PNS consists of the neomycin resistance gene (neo) as a positive selectable marker gene and the herpes simplex virus (HSV) thymidine kinase (tk) gene as a negative selectable marker gene. Positive selection with geneticin (G418) of epithelioma papulosum of carp (EPC) cells transfected with linearized pNK vector yielded 350 colonies, while double selection of transfected EPC cells with G418 and gancyclovir (Gc) resulted in nearly complete cell death, demonstrating that the PNS procedure is effective in fish cells. Homologous recombination vectors consist of the Xiphophorus melanoma receptor kinase (X mrk(Y)) gene as homologous sequence in addition to the neo and tk genes. Conditions for homologous recombination vector transfection and drug selection were established. After verification of the feasibility of expression of homologous recombination vectors in EPC cells, the first gene targeting experiments were attempted in the Xiphophorus melanoma cell line, PSM. Positive-negative selection of the targeting vector-transfectants led to a low enrichment in this particular cell line. The reasons for the low enrichment in PSM cells were discussed. High dimensional biomimetic informatics (HDBI) is a novel theory of informatics developed in recent years. Its primary object of research is points in high dimensional Euclidean space, and its exploratory and resolving procedures are based on simple geometric computations. However, the mathematical descriptions and computing of geometric objects are inconvenient because of the characters of geometry. With the increase of the dimension and the multiformity of geometric objects, these descriptions are more complicated and prolix especially in high dimensional space. In this paper, we give some definitions and mathematical symbols, and discuss some symbolic computing methods in high dimensional space systematically from the viewpoint of HDBI. With these methods, some multi-variables problems in high dimensional space can be solved easily. Three detailed algorithms are presented as examples to show the efficiency of our symbolic computing methods: the algorithm for judging the center of a circle given three points on this circle, the algorithm for judging whether two points are on the same side of a hyperplane, and the algorithm for judging whether a point is in a simplex constructed by points in high dimensional space. Two experiments in blurred image restoration and uneven lighting image correction are presented for all these algorithms to show their good behaviors.

An optimization method based on uniform design in conjunction with genetic algorithm is described. According to the proposed method, the uniform design technique was applied to the design of starting experiments, which can reduce the number of experiments compared with traditional simultaneous methods, such as simplex. And genetic algorithm was used in optimization procedure, which can improve the rapidity of optimal procedure. The hierarchical chromatographic response function was modified to evaluate the separation equality of a chromatogram. An iterative procedure was adopted to search for the optimal condition to improve the accuracy of predicted retention and the quality of the chromatogram. The optimization procedure was tested in optimization of the chromatographic separation of 11 alkaloids in reversed-phase ion pair chromatography and satisfactory optimal result was obtained. Local main chain dynamics of dissolved phenolphthalein polyethersulfone (PES-C) in solution with chloroform-d(1) were examined through C-13 NMR relaxation measurements. Spin-lattice relaxation times and NOE (nuclear Overhauser effects) factors were measured as a function of temperature. The relaxation data were interpreted in terms of main chain segmental motion by using the damped orientational diffusion model (DAMP) and the conformation jump model (VJGM) derived by Valeur, Jarry, Geny, and Monnerie. The simulation method used is N-SIMPLEX, which gives, in this study, a result of the object function less than 10(-4). Correlation times were obtained for the main chain motion of PES-C with these models and the results indicate that the main chain of PES-C are flexible. The comparison between PES-C and 1,2-polybutadiene is proposed. The distribution of the correlation time for the main chain motion by using VJGM model is discussed. The temperature dependence of correlation times for PES-C indicating the dynamical rigidity of its chains is obtained.

A detailed petrologic and mineralogic study was carried out on serpentinized peridotites dredged from the southern landward slopes of the Mariana Trench, in order to reveal the serpentinization process of these unusual rocks and to identify the sole presence of the mineral lizardite. The constituent minerals of these southern Mariana forearc peridotites are olivine, amphibole and spinel, as well as serpentine, chlorite and talc. Compared with serpentinite seamounts, the serpentinized peridotites from the southern Mariana forearc are characterized by the absence of magnetite and brucite, and the common presence of talc; besides, the serpentine mineral variety is simplex, only lizardite. Combining mineral chemistry and mineral phase relationships, we conclude that (1) the absence of magnetite in the serpentinized peridotites is due to incomplete serpentinization, other than magnetite, the iron end-member in olivine forms Fe-rich brucite and Fe-rich serpentine; (2) brucite is not stable with high silica activity, reacting with later SiO2-rich fluid and then forming lizardite, leading to a lack of brucite in these serpentinized peridotites; (3) the occurrence of talc is the result of later SiO2-rich fluid reactions with lizardite; and (4) the reason for the sole occurrence of lizardite is that the temperature condition of our study area was not high enough for the formation of antigorite (which is stable at > 500 degrees C). Despite the broad overlap of lizardite and chrysotile in growth temperature, differences in the modes of occurrence of lizardite and chrysotile, such as the scarcity of H2O, low porosity and permeability, as well as the actual situation of initial serpentinization in the study area, result in the absolute prevalence of lizardite over chrysotile in the area.
