Ethnopharmacological relevance
The two plants investigated here (Fagonia cretica L. and Hedera nepalensis K. Koch) have been previously reported as natural folk medicines for the treatment of diabetes but until now no scientific investigation of potential anti-diabetic effects has been reported.
Materials and methods
In vitro inhibitory effect of the two tested plants and their five isolated compounds on the dipeptidyl peptidase 4 (DPP-4) was studied for the assessment of anti-diabetic activity.
Results
A crude extract of Fagonia cretica possessed good inhibitory activity (IC50value: 38.1 μg/ml) which was also present in its n-hexane (FCN), ethyl acetate (FCE) or aqueous (FCA) fractions. A crude extract of Hedera nepalensis (HNC) possessed even higher inhibitory activity (IC50value: 17.2 μg/ml) and this activity was largely retained when further fractionated in either ethyl acetate (HNE; IC50: 34.4 μg/ml) or n-hexane (HNN; 34.2 μg/ml). Bioactivity guided isolation led to the identification of four known compounds (isolated for the first time) from Fagonia cretica: quinovic acid (1), quinovic acid-3β-O-β-d-glycopyranoside (2), quinovic acid-3β-O-β-d-glucopyranosyl-(28→1)-β-d-glucopyranosyl ester (3), and stigmasterol (4) all of which inhibited DPP-4 activity (IC50: 30.7, 57.9, 23.5 and >100 μM, respectively). The fifth DPP-4 inhibitor, the triterpenoid lupeol (5) was identified in Hedera nepalensis (IC50: 31.6 μM).
Conclusion
The experimental study revealed that Fagonia cretica and Hedera nepalensis contain compounds with significant DPP-4 inhibitory activity which should be further investigated for their anti-diabetic potential.

Durch asymmetrische Doppelbindungsisomerisierung mittels Me-DuPHOS-modifizierter Dihalogen-Nickel-Komplexe als Katalysatorvorstufen lassen sich aus 2-Alkyl-4,7-dihydro-1,3-dioxepinen hochenantiomerenreine 2-Alkyl-4,5-dihydro-1,3-dioxepine erhalten. Ein Ziel dieser Arbeit war es, die bisher noch unbekannte Absolutkonfiguration dieses Verbindungstyps zu bestimmen und darüber hinaus ihre Einsatzfähigkeit in der enantioselektiven organischen Synthese zu untersuchen. Zu diesem Zweck wurden enantiomerenangereichertes 2-Isopropyl- und 2-tert-Butyl-4,5-dihydro-1,3-dioxepin mit m-Chlorperbenzoesäure epoxidiert. Dabei bildeten sich die entsprechenden 3-Chlorbenzoesäure-(2-alkyl-5-hydroxy-1,3-dioxepan-4yl)-ester in hohen Ausbeuten und Diastereoselektivitäten. Von den vier zu erwartenden Diastereomeren wurden jeweils nur zwei mit einer Selektivität von mehr als 95:5 gebildet. Im Fall des 3-Chlorbenzoesäure-(2-isopropyl-5-hydroxy-1,3-dioxepan-4yl)-esters konnte das Haupt-diastereomer kristallin erhalten werden. Durch röntgenspektroskopische Untersuchung war es möglich, die Relativ-Konfiguration dieser Verbindung zu bestimmen. Die Ester lassen sich unter Ringverengung in 2-Alkyl-1,3-dioxan-4-carbaldehyde umlagern. Ausgehend von diesen Carbaldehyden stehen zwei Synthesewege zur Verfügung, welche zu Verbindungen führen deren Absolutkonfiguration bereits bekannt ist. So erhält man durch Reduktion 2-Alkyl-1,3-dioxan-4-yl-methanole, welche sich in 1,2,4-Butantriol überführen lassen. Oxidation ergibt die 2-Alkyl-1,3-dioxan-4-carbonsäuren, aus denen 3-Hydroxytetrahydrofuran-2-on gewonnen werden kann. Messung des Drehwertes dieser beiden literaturbekannten Verbindungen liefert nicht nur Information über deren Enantiomerenreinheit sondern ebenfalls über die Konfiguration ihres Stereozentrums. In Kombination mit der Relativ-Konfiguration des Esters ist somit ein Rückschluss auf die Absolutkonfiguration der eingesetzten 4,5-Dihydro-1,3-dioxepine möglich. Die auf den beschriebenen Wegen gewonnenen Substanzen finden Anwendung in der stereoselektiven organischen Synthese. Löst man die Chlorbenzoesäureester in Dichlormethan und behandelt sie mit wässriger Salzsäure, so entstehen die bicyclischen 2-Alkyltetrahydrofuro[2,3-d][1,3]dioxole. Auch bei diesen Verbindungen konnten hohe Enantio- und Diastereoselektivitäten erzielt werden. Der intermolekular verlaufende Reaktionsmechanismus der Bicyclus-Bildung, welcher unter Abspaltung eines den Alkylrest tragenden Aldehyds und dessen Neuanlagerung unter Ausbildung eines Acetals verläuft, konnte in dieser Arbeit durch ein Kreuzungsexperiment bestätigt werden. Umacetalisierung der Bicyclen liefert 2-Methoxytetrahydrofuran-3-ol, aus dem durch Acetalspaltung Tetrahydrofuran-2,3-diol erhalten wird, das die Halbacetalform der entsprechenden Desoxytetrose darstellt, die auf diese Weise in einer de novo-Synthese hergestellt werden kann.

In der vorliegenden Arbeit wurde die Synthese von optisch aktivem 9,14-Methylen-Lipoxin A4 untersucht. Lipoxin A4 und seine Derivate leiten sich von der Arachidonsäurekaskade ab wie die Prostaglandine, Prostacycline, Thromboxana und Leucotriene. Alle diese Verbindungen sind biologisch aktive Eicosanoide, die aus 20 C-Atomen bestehen. Lipoxine können im Organismus auf verschiedenen Biosynthesewegen entstehen: über die Lipoxygenase sowie über die Zell-Zell-Wechselwirkungen. Untersuchungen ergaben, dass die Lipoxine selektive biologische Wirkungen zeigen, die eine wichtige Rolle vor allem in Entzündungsprozessen und Infektabwehr zeigen. Da aber diese Moleküle rasch isomerisieren, sind diese hohen Aktivitäten mit einer sehr geringen Stabilität gekoppelt. Aufgrund dieser Instabilität konnte die biologisch aktive Konformation des Lipoxin A4 am Rezeptor nicht eindeutig bestimmt werden. Zur besseren Untersuchung der Lipoxinrezeptor-Anordnung wurden deshalb stabile Analoga synthetisiert. Die biologische Aktivität des nach Nokami et al hergestellten Lipoxin A4-Analogons weicht sehr stark von der des Lipoxins ab. Angeregt durch diese Arbeiten wurde in der Arbeitsgruppe Nubbemeyer die Idee entwickelt, das konjugierte Tetraensystem des Lipoxin A4-Moleküls durch das Cycloheptatrien nachzuahmen. Die CH2-Gruppe bildet eine Brücke, die die Isomerisierung vom aktiven cis-Isomer zu den inaktiven trans-Isomeren verhindern soll. Mit diesem Cycloheptatriengerüst als Lipoxin A4-Analogon hoffen wir das Lipoxingerüst unwesentlich zu verändern und die damit verbundene biologische Aktivität zu erhalten. Die Synthese des 9,14-Methylen-LXA4 soll möglichst konvergent erfolgen, so dass gegebenenfalls auf Bausteine zurückgegriffen werden kann, deren Aufbau bereits optimiert wurde. Eine derartige Strategie ermöglicht darüber hinaus die Herstellung einer großen Zahl von weiteren potentiell interessanten Verbindungen ohne komplettes Umstellen der Synthese. Wichtige Reaktionen im Verlauf dieser Synthese sind: bei der Synthese des C8-C20-Bausteins: Friedel-Crafts-Acylierungen, Haloform-Reaktion, Veresterung mittels Standardmethoden, enantioselektive Reduktion mit dem chiralen CBS-Katalysator und Schutzgruppenoperation. bei der Synthese des C1-C7-Bausteins: ex-chiral-pool-Synthese aus 2-Desoxy-D-ribose, Wittig-Reaktion, Hydrierung mit Pd/C, Schutzgruppenoperation, Abspaltung von Schutzgruppen und Swern-Oxidation zum Aldehyd. Die Schlüsselreaktion der ganzen Synthese ist die Eintopf-Variante der Julia-Olefinierung nach Kocienski: selektiver Aufbau des trans-Olefins durch Verknüpfung der beiden Bausteine. Nach weiteren Schutzgruppenoperationen (und Öffnung des Valerolactons) wird der Methylester des 9,14-Lipoxin A4 erhalten, dessen biologische Aktivität zweifelsfrei bewiesen wurde.

New coordination polymers [M(Pht)(4-MeIm)2(H2O)]n (M=Co (1), Cu (2); Pht2−=dianion of o-phthalic acid; 4-MeIm=4-methylimidazole) have been synthesized and characterized by IR spectroscopy, X-ray crystallography, thermogravimetric analysis and magnetic measurements. The crystal structures of 1 and 2 are isostructural and consist of [M(4-MeIm)2(H2O)] building units linked in infinite 1D helical chains by 1,6-bridging phthalate ions which also act as chelating ligands through two O atoms from one carboxylate group in the case of 1. In complex 1, each Co(II) atom adopts a distorted octahedral N2O4 geometry being coordinated by two N atoms from two 4-MeIm, three O atoms of two phthalate residues and one O atom of a water molecule, whereas the square-pyramidal N2O3 coordination of the Cu(II) atom in 2 includes two N atoms of N-containing ligands, two O atoms of two carboxylate groups from different Pht, and a water molecule. An additional strong O–H⋯O hydrogen bond between a carboxylate group of the phthalate ligand and a coordinated water molecule join the 1D helical chains to form a 2D network in both compounds. The thermal dependences of the magnetic susceptibilities of the polymeric helical Co(II) chain compound 1 were simulated within the temperature range 20–300 K as a single ion case, whereas for the Cu(II) compound 2, the simulations between 25 and 300 K, were made for a linear chain using the Bonner–Fisher approximation. Modelling the experimental data of compound 1 with MAGPACK resulted in: g=2.6, |D|=62 cm−1. Calculations using the Bonner–Fisher approximation gave the following result for compound 2: g=2.18, J=–0.4 cm−1.

The transport of a group of quinolone antibiotics across the human intestinal model, Caco-2 cells, was investigated. It was found that the transport of the quinolones generally correlated with the lipophilicity of the compounds, indicating the passive diffusional transcellular processes were involved. However, it was observed that the transport in both directions apical-to-basolateral and basolateral-to-apical was not equivalent, and polarised transport occurred. For all the quinolones studied except, BMS-284756-01, it was found that the basolateral-to-apical transport was significantly greater than the apical-to-basolateral transport. This finding suggested that the quinolones underwent a process of active secretion. The pKas and logPs for the quinolones were determined using potentiometric titrations. The measured logP values were compared with those determined using theoretical methods. The theoretical methods for calculating logP including the Moriguchi method correlated poorly with the measured logP values. Further investigations revealed that there may be an active transporter involved in the apical-to-basolateral transport of quinolones as well. This mechanism was sensitive to competing quinolones, but, it was unaffected by the metabolic inhibitor combination of sodium azide (15mM) with 2-deoxy-D-glucose (50mM). The basolateral-to-apical transport of quinolones was found to be sensitive to inhibition by a number of different inhibitors. The metabolic inhibitors, sodium azide (15mM) with 2-deoxy-D-glucose (50mM) and 2,4-dinitrophenol (1mM), were able to reduce the basolateral-to-apical transport of quinolones. A reduction in temperature from 37°C to 2°C caused an 80-fold decrease in the transport of gatifloxacin in both directions, however, this effect was not sufficient to abolish the greater basolateral-to-apical secretion. As with apical-to-basolateral transport, it was found that quinolones competed with gatifloxacin for basolateral-to-apical transport, both ofloxacin (100μM) and norfloxacin (100μM) significantly (P<0.003) decreased the basolateral-to-apical transport of gatifloxacin; however, ciprofloxacin (100μM and 300μM) had no effect. A number of inhibitors of various transport systems were also investigated. It was found that the anion transport inhibitor, probenecid (100 μM) had a significant inhibitory effect on the basolateral-to-apical transport of ciprofloxacin (P=0.039), while the cation transport inhibitor cimetidine (100μM and 500μM) had no effect. The organic anion exchange inhibitor 4,4'diisothiocyanostilbene-2-2' -disulphonic acid DIDS (400μM) also had a significant inhibitory effect (P=O.O 13). The PgP inhibitor and anion exchange inhibitor verapamil (400Mμ) was able to completely abolish the basolateral-to-apical secretion of gatifloxacin and bring it into line with the apical-to-basolateral flux. In conclusion, the apical-to-basolateral and basolateral-toapical transport of quinolones involved an active component. The basolateral-to-apical secretion was abolished by a verapamil (400μM), a bisubstrate for PgP and the anion transporter.

Purpose: To evaluate the potential of Lonicera macranthoids Hand. -Mazz. Yulei1 suspension culture system for enhanced production of the main secondary metabolite, chlorogenic acid. Methods: The callus of L. macranthoides Hand.-Mazz. “Yulei1” was suspension cultured in B5 liquid medium supplemented with different plant growth regulators. Biomass accumulation was calculated by weight method and chlorogenic acid production was measured using high performance liquid chromatography (HPLC). HPLC was carried out on C18 analytical column at 35 °C and the detection wavelength was set at 324 nm. Results: The results showed that maximum accumulation of biomass and chlorogenic acid were achieved 15 days after culture growth. The optimized conditions for biomass accumulation and chlorogenic acid production were 50 g/L of inoculum on fresh weight basis, B5 medium supplemented with plant growth regulators, 30 - 40 g/L sucrose and initial medium pH of 5.5. Maximum accumulation of chlorogenic acid and biomass were observed when the culture medium was supplemented with 2.0 mg/L6-BA. Optimal accumulation of chlorogenic acid was observed using combination of hormones 2.0 mg/L 6-Benzyladenine (BA) + 0.5 mg/L2, 4-Dichlorophenoxyacetic acid (2,4-D), while optimal accumulation of biomass was observed with 2.0 mg/L 6-BA + 2.0 mg/L2, 4-D. In addition, phenylalanine also contributed to the synthesis of chlorogenic acid at a concentration > 50 mg/L. Conclusion: Cell suspension cultures of L. macranthoides Hand.-Mazz. “Yulei1” have successfully been established. The findings provide a potential basis for large scale production of chlorogenic acid using cell suspension cultures of L. macranthoides.

Callus was initiated in three different ‘‘esculenta’’ taro cultivars by culturing corm slices in the dark on half-strength MS medium supplemented with 2.0 mg/l 2,4- dichlorophenoxyacetic acid (2,4-D) for 20 days followed by subculture of all corm slices to half-strength MS medium containing 1.0 mg/l thidiazuron (TDZ). Depending on the cultivar, 20–30% of corm slices produced compact, yellow, nodular callus on media containing TDZ. Histological studies revealed the presence of typical embryogenic cells which were small, isodiametric with dense cytoplasms. Somatic embryos formed when callus was transferred to hormone-free medium and *72% of the embryos germinated into plantlets on this medium. Simultaneous formation of roots and shoots during germination, and the presence of shoot and root poles revealed by histology, confirmed that these structures were true somatic embryos. Plants derived from somatic embryos appeared phenotypically normal following 2 months growth in a glasshouse. This method is a significant advance on those previously reported for the esculenta cultivars of taro due to its efficiency and reproducibility.

Embryogenic callus was initiated by culturing in vitro taro corm slices on agar-solidified half-strength MS medium containing 2.0 mg/L 2,4-dichlorophenoxyacetic acid (2,4-D) for 20 days followed by transfer to 1.0 mg/L thidiazuron (TDZ). Callus was subsequently proliferated on solid medium containing 1.0 mg/L TDZ, 0.5 mg/L 2,4- D and 800 mg/L glutamine before transfer to liquid medium containing the same components but with reduced glutamine (100 mg/L). After 3 months in liquid culture on an orbital shaker, cytoplasmically dense cell aggregates began to form. Somatic embryogenesis was induced by plating suspension cells onto solid media containing reduced levels of hormones (0.1 mg/L TDZ, 0.05 mg/L 2,4-D), high concentrations of sucrose (40–50 g/L) and biotin (1.0 mg/L). Embryo maturation and germination was then induced on media containing 0.05 mg/L benzyladenine (BA) and 0.1 mg/L indole-3-acetic acid (IAA). Histological studies of the developing embryos revealed the presence of typical shoot and root poles suggesting that these structures were true somatic embryos. The rate of somatic embryos formation was 500–3,000 per mL settledcell volume while approximately 60% of the embryos regenerated into plants.

A copolymer comprising 1,4-diketopyrrolo[3,4-c]pyrrole (DPP) and thieno[3,2-b]thiophene moieties, PDBT-co-TT, shows high hole mobility of up to 0.94 cm2 V-1 s-1 in organic thin-film transistors. The strong intermolecular interactions originated from π-π stacking and donor-acceptor interaction lead to the formation of interconnected polymer networks having an ordered lamellar structure, which have established highly efficient pathways for charge carrier transport.
