Background Non-alcoholic fatty liver disease (NAFLD) is the most common chronic liver disorder in industrialized countries, yet its pathophysiology is incompletely understood. Small-molecule metabolite screens may offer new insights into disease mechanisms and reveal new treatment targets. Methods Discovery (N = 33) and replication (N = 66) of liver biopsies spanning the range from normal liver histology to non-alcoholic steatohepatitis (NASH) were ascertained ensuring rapid freezing under 30 s in patients. 252 metabolites were assessed using GC/MS. Replicated metabolites were evaluated in a murine high-fat diet model of NAFLD. Results In a two-stage metabolic screening, hydroquinone (HQ, pcombined = 3.0 × 10−4) and nicotinic acid (NA, pcombined = 3.9 × 10−9) were inversely correlated with histological NAFLD severity. A murine high-fat diet model of NAFLD demonstrated a protective effect of these two substances against NAFLD: Supplementation with 1% HQ reduced only liver steatosis, whereas 0.6% NA reduced both liver fat content and serum transaminase levels and induced a complex regulatory network of genes linked to NALFD pathogenesis in a global expression pathway analysis. Human nutritional intake of NA equivalent was also consistent with a protective effect of NA against NASH progression. Conclusion This first small-molecular screen of human liver tissue identified two replicated protective metabolites. Either the use of NA or targeting its regulatory pathways might be explored to treat or prevent human NAFLD.

The gerbil model of ischemia was used to determine the effect of carotid occlusion on energy metabolites in cellular layers of discrete regions of the hippocampus and dentate gyrus. Levels of glucose, glycogen, ATP and phosphocreatine (PCr) were unchanged after 1 minute of ischemia. However, 3 minutes of ischemia produced a dramatic decrease in net levels of all metabolites. No additional decrease was observed after 15 minutes of ischemia. Re-establishment of the blood flow for 5 minutes after a 15 minute ischemic episode returned all metabolites to pre-ischemia levels. Concentrations of glucose and glycogen were elevated in sham-operated animals as a function of the pentobarbital anesthetic employed. In other studies, elevated GABA levels (produced by inhibiting GABA-transaminase with (gamma)-vinyl-GABA (GVG)) were found to decrease the rate of utilization of the high-energy phosphate metabolites ATP and PCr in the mouse cortex. In addition, glucose and glycogen levels were increased. Thus, tonic inhibition by GABA produced decreased cellular activity. Additional experiments demonstrated the attenuation of ischemia-induced metabolite depletion in cellular layers of regions of the hippocampus, dentate gyrus and cortex after GVG administration. Under ether, 1 minute of bilateral carotid occlusion produced a dramatic decrease in metabolite levels. After GVG treatment, the decrease was blocked completely for glucose, glycogen and ATP, and partially for PCr. Therefore, GABA-transaminase inhibition produced increased levels of GABA which subsequently decreased cellular activity. The protection against ischemia may have been due to (a)decreased metabolic rate; the available energy stores were utilized at a slower rate, and (b)increased levels of energy substrates; additional supplies available to maintain viability. These data suggest that the functional state of neural tissue can determine the response to metabolic stress. ^

The joint modeling of longitudinal and survival data is a new approach to many applications such as HIV, cancer vaccine trials and quality of life studies. There are recent developments of the methodologies with respect to each of the components of the joint model as well as statistical processes that link them together. Among these, second order polynomial random effect models and linear mixed effects models are the most commonly used for the longitudinal trajectory function. In this study, we first relax the parametric constraints for polynomial random effect models by using Dirichlet process priors, then three longitudinal markers rather than only one marker are considered in one joint model. Second, we use a linear mixed effect model for the longitudinal process in a joint model analyzing the three markers. In this research these methods were applied to the Primary Biliary Cirrhosis sequential data, which were collected from a clinical trial of primary biliary cirrhosis (PBC) of the liver. This trial was conducted between 1974 and 1984 at the Mayo Clinic. The effects of three longitudinal markers (1) Total Serum Bilirubin, (2) Serum Albumin and (3) Serum Glutamic-Oxaloacetic transaminase (SGOT) on patients' survival were investigated. Proportion of treatment effect will also be studied using the proposed joint modeling approaches. ^ Based on the results, we conclude that the proposed modeling approaches yield better fit to the data and give less biased parameter estimates for these trajectory functions than previous methods. Model fit is also improved after considering three longitudinal markers instead of one marker only. The results from analysis of proportion of treatment effects from these joint models indicate same conclusion as that from the final model of Fleming and Harrington (1991), which is Bilirubin and Albumin together has stronger impact in predicting patients' survival and as a surrogate endpoints for treatment. ^

Although more than 100 genes associated with inherited retinal disease have been mapped to chromosomal locations, less than half of these genes have been cloned. This text includes identification and evaluation of candidate genes for three autosomal dominant forms of inherited retinal degeneration: atypical vitelliform macular dystrophy (VMD1), cone-rod dystrophy (CORD), and retinitis pigmentosa (RP). ^ VMD1 is a disorder characterized by complete penetrance but extremely variable expressivity, and includes macular or peripheral retinal lesions and peripappilary abnormalitites. In 1984, linkage was reported between VMD1 and soluble glutamate-pyruvate transaminase GPT); however, placement of GPT to 8q24 on linkage maps had been debated, and VMD1 did not show linkage to microsatellite markers in that region. This study excluded linkage between the loci by cloning GPT, identifying the nucleotide substitution associated with the GPT sozymes, and by assaying VMD1 family samples with an RFLP designed to detect the substitution. In addition, linkage of VMD1 to the known dominant macular degeneration loci was excluded. ^ CORD is characterized by early onset of color-vision deficiency, and decreased visual acuity, However, this retinal degeneration progresses to no light perception, severe macular lesion, and “bone-spicule” accumulations in the peripheral retina. In this study, the disorder in a large Texan family was mapped to the CORD2 locus of 19q13, and a mutation in the retina/pineal-specific cone-rod homeobox gene (CRX) was identified as the disease cause. In addition, mutations in CRX were associated with significantly different retinal disease phenotypes, including retinitis pigmentosa and Leber congenital amaurosis. ^ Many of the mutations leading to inherited retinal disorders have been identified in genes like CRX, which are expressed predominantly in the retina and pineal gland. Therefore, a combination of database analysis and laboratory investigation was used to identify 26 novel retina/pineal-specific expressed sequence tag (EST) clusters as candidate genes for inherited retinal disorders. Eight of these genes were mapped into the candidate regions of inherited retinal degeneration loci. ^ Two of the eight clusters mapped into the retinitis pigmentosa RP13 candidate region of 17p13, and were both determined to represent a single gene that is highly expressed in photoreceptors. This gene, the Ah receptor-interacting like protein-1 (AIPL1), was cloned, characterized, and screened for mutations in RP13 patient DNA samples. ^

Por se tratar de um elemento essencial às plantas e um metal pesado ao mesmo tempo, o níquel requer atenção quanto aos aspectos da fisiologia de plantas e ambiental. Além disso, existe um intervalo estreito entre as exigências nutricionais e os teores tóxicos às plantas. Neste contexto, objetivou-se avaliar o efeito do Ni no sistema solo-planta, com foco no ciclo do N e a disponibilidade do elemento no solo, por meio de experimento em condições controladas, utilizando vasos distribuídos inteiramente ao acaso, utilizando-se esquema fatorial 2 x 5, com sete repetições cada tratamento. O primeiro fator foi constituído de duas saturações por base (50 e 70%) e o segundo de cinco doses de Ni (0; 0,1; 0,5; 1,0 e 10,0 mg dm-3 de solo). Os vasos foram preenchidos com 8 dm3 de terra e cultivados com soja [Glycine max (L.) Merrill] sucedida por girassol (Helianthus annuus L.). Os parâmetros qualitativos e quantitativos: altura de plantas (AP), diâmetro do caule (DC), número de nós (NN), estádio fenológico (EF), índice SPAD e, diâmetro do capítulo (DCap) (para girassol) foram avaliadas aos 30 e 60 dias após a emergência (d.a.e.) de cada cultivo. Plantas inteiras de soja, amostradas em quatro vasos de cada tratamento, foram coletadas no estádio R1. Na mesma ocasião foram coletadas amostras de solo da rizosfera. Em seguida, as plantas coletadas foram divididas em: folhas; raízes (nódulos na soja) e parte aérea. Foram determinados nas folhas utilizadas para diagnose em soja e girassol: os teores de macro e micronutrientes, as atividades da redutase do nitrato e da urease e as concentrações dos ácidos orgânicos: oxálico, malônico, succínico, málico, tartárico, fumárico, oxaloacético, cítrico e lático. Os mesmos ácidos orgânicos foram determinados em raízes secundárias de girassol e nódulos de soja. Foram realizadas avaliações ultraestruturais por meio de microscopia eletrônica de transmissão (MET) em raízes de girassol, e estruturais e de tonalidade em nódulos de soja, por meio de microscopia de luz. No solo, foram determinadas: atividade urease, desidrogenase, Ni total e fitodisponível pelos métodos: Mehlich-1, Mehlich-3 e DTPA. No período de maturidade fisiológica de cada cultura foi realizada a colheita das plantas dos vasos restantes para determinação de produção de grãos, teores de Ni na planta inteira e Ni e N nos grãos. Ao final dos dois experimentos foi realizada nova coleta de solo para extração sequencial de Ni. O índice SPAD em soja aos 60 d.a.e., a produção de massa seca da parte aérea da soja e da raiz de girassol foram influenciados pela saturação por bases, doses de níquel e pela a interação destes. Foram influenciados pelas saturações por base e doses de níquel (fatores isolados): para soja: AP aos 60 d.a.e., NN aos 30 e 60 d.a.e., SPAD aos 30 d.a.e.; para girassol: AP e NN aos 30 e 60 d.a.e., DC e SPAD aos 30 d.a.e. As demais variáveis avaliadas aos 30 e 60 d.a.e. foram influenciadas apenas pela saturação por bases, ou doses de Ni separadamente. As plantas de soja e girassol apresentaram maiores teores de Ni nos diferentes tecidos avaliados (exceto grãos) quando cultivadas sob V50%. A produção de grãos de soja e girassol não foi influenciada pelos tratamentos, porém o teor de N dos grãos de soja influenciado pelas doses de Ni na V70%. A atividade da enzima urease nas folhas de soja e girassol foi responsiva positivamente ao aumento das doses de Ni. Quatro dos ácidos orgânicos avaliados e o teor de N nas folhas e nos grãos foram maiores nas plantas cultivadas sob V70% com a dose de 0,5 mg dm-3 de Ni. As doses de Ni bem com as saturações por bases influenciaram diretamente o balanço de nutrientes das plantas. Os extratores Mehlich-1, Mehlich-3 e DTPA apresentaram elevado coefienciente de correlação entre a fração de Ni disponível no solo e a concentração do elemento nas plantas de soja e girassol, sendo o extrator DTPA o que apresentou maior coeficiente de correlação. O Ni apresentou distribuição variável entre as diferentes frações do solo em função dos tratamentos. Os solos dos tratamentos com saturação por bases de 70% apresentaram maior concentração de Ni ligado a carbonato, comparado aos tratamentos sob saturação por bases de 50%. A distribuição do Ni entre as frações do solo seguiu a seguinte orgem: ligado a carbonato < trocável < ligado a óxidos < matéria orgânica < residual. A saturação por bases exerceu efeito diferenciado para a atividade da urease no solo em função da cultura avaliada. Por sua vez, o Ni exerceu efeito diferenciado sobre a atividade de desidrogenase em função da cultura estudada

Several immunomodulatory factors are involved in malaria pathogenesis. Among them, heme has been shown to play a role in the pathophysiology of severe malaria in rodents, but its role in human severe malaria remains unclear. Circulating levels of total heme and its main scavenger, hemopexin, along with cytokine/chemokine levels and biological parameters, including hemoglobin and creatinine levels, as well as transaminase activities, were measured in the plasma of 237 Plasmodium falciparum-infected patients living in the state of Odisha, India, where malaria is endemic. All patients were categorized into well-defined groups of mild malaria, cerebral malaria (CM), or severe noncerebral malaria, which included acute renal failure (ARF) and hepatopathy. Our results show a significant increase in total plasma heme levels with malaria severity, especially for CM and malarial ARF. Spearman rank correlation and canonical correlation analyses have shown a correlation between total heme, hemopexin, interleukin-10, tumor necrosis factor alpha, gamma interferon-induced protein 10 (IP-10), and monocyte chemotactic protein 1 (MCP-1) levels. In addition, canonical correlations revealed that heme, along with IP-10, was associated with the CM pathophysiology, whereas both IP-10 and MCP-1 together with heme discriminated ARF. Altogether, our data indicate that heme, in association with cytokines and chemokines, is involved in the pathophysiology of both CM and ARF but through different mechanisms.

Bacillus thuringiensis (Bt) transgenic cotton has shown changes of vegetative and reproductive growth characteristics. The objective of this study was to investigate the physiological change of nitrogen metabolism that related closely to the growth in Bt cotton cultivars. The study Was undertaken on two 131 transgenic cotton cultivars and their parents, one conventional (Xingyang822) and recurrent parent (Sumian No. 9), the other a hybrid (Kumian No. 1) and female parent (Yumian No. 1), during the 2001 and 2002 growing seasons at the Yangzhou University Farm, Yangzhou, China. In the 2001 study, The results indicated that the Bt cotton cultivars were higher than their parents in leaf total nitrogen, free amino acid and soluble protein content, greater in NR and GPT activity, and lower in protease activity, during peak square and boll developing period. The biggest increase of total nitrogen was at peak boll period, which increased by 36.01 and 18.96% for Kumian No. I and Xingyang822, respectively. There were similar results for free amino acid and soluble protein content. The results showed further in 2002 study that NR activity increased dramatically at peak square and early boll open period, the biggest increase at early boll open period, with Kumian No. I and Xingyan,822 being 87.5 and 61.4% higher than their parent, respectively, the biggest increase of GPT activity was at peak boll period, with Kumian No. I and Xingyang822 being 39.1 and 29.1% higher than their parent, respectively. However, protease activity of Bt cultivars reduced significantly before flowering and early boll open period, the biggest decrease was before flowering period, with Kumian No. I being more than 30%, Xingyang822 being 26.5% at peak square period. Moreover, the boll total nitrogen content reduced sharply. The results suggest that the Bt cotton cultivars have higher intensity of leaf nitrogen metabolism than their parent, especially during square and boll development period. It is disadvantage for square development and earlier boll maturity under high nitrogen condition. The cultural practice should aim at reducing leaf nitrogen metabolic strength and keep the balance of vegetative and reproductive growth. (C) 2003 Elsevier B.V. All rights reserved.

Bt transgenic cotton has not shown the same level of resistance to bollworm in China, as in other major Bt cotton growing areas of the world. The objective of this study was to investigate the effects of high temperature on the CryIA insecticidal protein content and nitrogen metabolism, in the leaf of Bt transgenic cotton. The study was undertaken on two transgenic cotton cultivars, one conventional (Xinyang 822) and the other a hybrid (Kumian No. 1), during the 2001 and 2002 growing seasons at the Yangzhou University Farm, Yangzhou, China. In the 2001 study, potted cotton plants were exposed to 37 C for 24 h under glasshouse conditions at three growth stages peak square, peak flowering and peak boll developing periods. Based on the 2001 results, in 2002 the same two cultivars were exposed to the same temperature for 48 h at two growth stages-peak flowering and boll developing periods. The results of the study indicated that the insecticidal protein content of the leaf was not significantly affected by the stress during the square and flowering periods. However, exposure to high temperature for 24h during the boll period reduced the CryIA protein content by approximately 51% in the cultivar Kumian No 1, and 30% in Xinyang 822 in the 2001 study, and by approximately 73 and 63% for 48 h with the same cultivars, respectively, in the 2002 study. Glutamic-pyruvic transaminase (GPT) activity, total free amino acid and soluble protein content, and the activity of protease in the leaf, showed relatively little change in response to high temperature in the flowering period. However, exposure to high temperature in the boll period resulted in the following changes - a reduction of GPT activity, a sharp increase in free amino acid content, a significant decrease in soluble protein content, and significant increases in the activity of protease. The results suggest that high temperature may result in the degradation of soluble protein in the leaf, with a resulting decline in the level of the toxin CryIA. It is believed that this may be the cause of the reduced efficacy of Bt cotton in growing conditions in China, where temperatures during the boll period often reach 36-40&DEG; C. © 2004 Elsevier B.V All rights reserved.

The principal aim of this work was to examine the effects of antiepileptic drugs (AEDs) on vision. Vigabatrin acts by increasing GABA at brain inhibitory synapses by irreversibly binding to GABA-transaminase. Remacemide is a novel non-competitive NMDA receptor antagonist and fast sodium channel inhibitor that results in the inhibition of the NMDA receptors located in the neuronal membrane calcium channels increasing glutamate in the brain. Vigabatrin has been shown to cause a specific pattern of visual field loss, as one in three adults taking vigabatrin have shown a bilateral concentric constriction. Remacemide has unknown effects on vision. The majority of studies of the effects of AEDs on vision have not included the paediatric population due to difficulties assessing visual field function using standard perimetry testing. Evidently an alternative test is required to establish and monitor visual field problems associated with AEDs both in children and in adults who cannot comply with perimetry. In order to test paediatric patients exposed to vigabatrin, a field-specific visual evoked potential was developed. Other tests performed on patients taking either vigabatrin or remacemide were electroretinograms, electro-oculograms, multifocal VEPs and perimetry. Comparing these tests to perimetry results from vigabatrin patients the field specific VEP was found to have a high sensitivity and specificity, as did the 30Hz flicker amplitude. The modified VEP was also found to provide useful results in vigabatrin patients. Remacemide did not produce a similar visual field loss to vigabatrin although macular vision was affected. The field specific VEP is a useful method for detecting vigabatrin associated visual field loss that is well tolerated by young children. This technique combined with the ERG under light adapted (30Hz flicker) condition is presently the superior method for detecting vigabatrin-attributed peripheral field defects present in children below the developmental age of 9. The effects of AEDs on vision should be monitored carefully and the use of multifocal stimulation allows for specific areas of the retina and visual pathway to be monitored.

Vigabatrin (VGB) is a transaminase inhibitor that elicits its anitepileptic effect by increasing GABA concentrations in the brain and retina. - Assess whether certain factors predispose patients to develop severe visual field loss. - Develop a sensitive algorithm for investigating the progression of visual field loss. - Determine the most sensitive clinical regimen for diagnosing VGB-attributed visual field loss. - Investigate whether the reports of central retinal sparing are accurate. The investigations have resulted in a number of significant findings: - The anatomical evidence in combination with the pattern of visual field loss suggests that the damage induced by VGB therapy occurs at retinal level, and is most likely a toxic effect. - The quantitative algorithm, designed within the course of this investigation, provided increased sensitivity in determining the severity of visual field loss. - Maximum VGB dose predisposes patients to develop severe visual field loss. - The SITA Standard algorithm was found to be as sensitive and significantly faster, in diagnosing visual field defects attributed to VGB, when compared to the Full Threshold algorithm. The Full Threshold was found to be the most repeatable between visits. - The normal SWAP 10-2 database provided an effective method of differentiating SWAP defects. - SWAP, FDT and the mfERG have increased sensitivity in detecting visual field loss attributed to VGB. The pattern of visual field loss from these investigations suggests that VGB produces a diffuse effect across the retina including subtle central abnormalities and more severe peripheral defects. - Abnormalities detected using the mfERG have suggested that VGB adversely affects the photoreceptors Müller, amacrine and ganglion cells in the retina. An urgent review of the manufacturers recommended maximum dose for VGB is required.

Se realizó un estudio descriptivo; en un universo de 96 pacientes pertenecientes a la clínica de SIDA del hospital. Los datos demográficos se obtuvieron por entrevista directa y los resultados de los exámenes de laboratorio fueron copiados de la historia clínica. Estos se analizaron con la ayuda del Software Epi-Info. Resultados: la mayoría fueron hombres, con edad menor a 45 años, casados, solteros y con ocupación de obreros y quehaceres domésticos. Los fármacos más utilizados fueron zidovudina, lamivudina y efavirenz: zidovudina produjo anemia en el 15.5de los casos y depresión medular en el 2.4; lamivudina provocó miositis en el 31, neuropatía periférica en el 4y efavirenz ocasionó elevación de las transaminasas, glutámico pirúvico y glutámico oxalacético, en el 51.2. En la evaluación de las transaminasas, creatinfosfoquinasa, creatinina, glucosa, colesterol total, triglicéridos, hemoglobina, carga viral y los linfocitos TCD4 no hubo diferencia significativa entre la valoración inicial y en la de control. Conclusión: efavirenz produjo la más alta frecuencia de efectos colaterales se relaciono con la elevación de transaminasas, seguido por zidovudina que produjo anemia y depresión medular; lamivudina provoco miositis y neuropatía periférica

Cancer cells have been noted to have an altered metabolic phenotype for over ninety years. In the presence of oxygen, differentiated cells predominately utilise the tricarboxylic acid (TCA) cycle and oxidative phosphorylation to efficiently produce energy and the metabolites necessary for protein and lipid synthesis. However, in hypoxia, this process is altered and cells switch to a higher rate of glycolysis and lactate production to maintain their energy and metabolic needs. In cancer cells, glycolysis is maintained at a high rate, even in the presence of oxygen; a term described as “aerobic glycolysis”. Tumour cells are rapidly dividing and have a much greater need for anabolism compared to normal differentiated cells. Rapid glucose metabolism enables faster ATP production as well as a greater redistribution of carbons to nucleotide, protein, and fatty acid synthesis, thus maximising cell growth. Recently, other metabolic changes, driven by mutations in genes related to the TCA cycle, indicate an alternative role for metabolism in cancer, the “oncometabolite”. This is where a particular metabolite builds up within the cell and contributes to the tumorigenic process. One of these genes is isocitrate dehydrogenase (IDH) IDH is an enzyme that forms part of the tricarboxylic acid (TCA) cycle and converts isocitrate to α-ketoglutarate (α-KG). It exists in three isoforms; IDH1, IDH2 and IDH3 with the former present in the cytoplasm and the latter two in the mitochondria. Point mutations have been identified in the IDH1 and IDH2 genes in glioma which result in a gain of function by converting α-KG to 2-hydroxyglutarate (2HG), an oncometabolite. 2HG acts as a competitive inhibitor of the α-KG dependent dioxygenases, a superfamily of enzymes that are involved in numerous cellular processes such as DNA and histone demethylation. It was hypothesised that the IDH1 mutation would result in other metabolic changes in the cell other than 2HG production, and could potentially identify pathways which could be targeted for therapeutic treatment. In addition, 2HG can act as a potential competitive inhibitor of α-KG dependent dioxygenases, so it was hypothesised that there would be an effect on histone methylation. This may alter gene expression and provide a mechanism for tumourogenesis and potentially identify further therapeutic targets. Metabolic analysis of clinical tumour samples identified changes associated with the IDH1 mutation, which included a reduction in α-KG and an increase in GABA, in addition to the increase in 2HG. This was replicated in several cell models, where 13C labelled metabolomics was also used to identify a possible increase in metabolic flux from glutamate to GABA, as well as from α-KG to 2HG. This may provide a mechanism whereby the cell can bypass the IDH1 mutation as GABA can be metabolised to succinate in the mitochondria by GABA transaminase via the GABA shunt. JMJ histone demethylases are a subset of the α-KG dependent dioxygenases, and are involved in removing methyl groups from histone tails. Changes in histone methylation are associated with changes in gene expression depending on the site and extent of chemical modification. To identify whether the increase in 2HG and fall in α-KG was associated with inhibition of histone demethylases a histone methylation screen was used. The IDH1 mutation was associated with an increase in methylation of H3K4, which is associated with gene activation. ChiP and RNA sequencing identified an increase in H3K4me3 at the transcription start site of the GABRB3 subunit, resulting in an increase in gene expression. The GABRB3 subunit forms part of the GABA-A receptor, a chloride channel, which on activation can reduce cell proliferation. The IDH1 mutation was associated with an increase in GABA and GABRB3 subunit of the GABA-A receptor. This raises the possibility of GABA transaminase as a potential therapeutic target. Inhibition of this enzyme could reduce GABA metabolism, potentially reducing any beneficial effect of the GABA shunt in IDH1 mutant tumours, and increasing activation of the GABA-A receptor by increasing the concentration of GABA in the brain. This in turn may reduce cell proliferation, and could be achieved by using Vigabatrin, a GABA transaminase inhibitor licensed for use in epilepsy.

O trabalho teve por objetivo avaliar a dinâmica de nitrogênio, em cultivo heterotrófico, a partir da cianobactéria Aphanothece microscopica Nägeli, sob o escopo de uma biorrefinaria. Neste sentido, foi avaliada a contribuição dos compostos nitrogenados não proteicos, na dinâmica de distribuição do nitrogênio, na biomassa gerada pelo micro-organismo em estudo, quando cultivado em sistema autotrófico e heterotrófico. Para o cultivo em condições autotróficas, foi utilizado o meio padrão BG-11, enquanto que, para o cultivo em condições heterotróficas, foi empregado o efluente da indústria de laticínios. Inicialmente, foi avaliada a contribuição dos pigmentos na fração nitrogenada não proteica tendo como base dois experimentos. No primeiro experimento foi selecionada a melhor condição para a produção de pigmentos, expressos pela clorofila-a em sistema heterotrófico, tendo como base os parâmetros C/N (20, 40 e 60), N/P (5, 10 e 15) e concentração de inóculo (100, 200 e 300 mg.L-1), mediante um planejamento fatorial 23 . Os experimentos foram conduzidos em biorreator heterotrófico a 20°C, pH 7,6 e aeração contínua de 1VVM. A melhor condição de produção de pigmento foi indicada como sendo a 200 mg.L-1 de concentração celular, razões C/N 20 e N/P 10. Com base nestes resultados, um segundo experimento foi delineado, visando avaliar a contribuição de pigmentos na fração de nitrogênio não proteico, bem como avaliar a produção de clorofila-a e ficobiliproteínas (ficocianina, aloficocianina e ficoeritrina), sob influência da luz e do meio de cultivo. Foi possível destacar teores superiores de ficobiliproteínas na biomassa gerada no cultivo heterotrófico. No entanto, com notada diferença (p≤0,05) nos teores de clorofila-a, quando são comparadas as concentrações na biomassa de meios autotróficos (10,7 mg.g-1) e heterotróficos (1,0 mg.g-1). Fato este compensado pelo menor tempo de cultivo registrado para atingir o final do experimento, quando o micro-organismo é cultivado em condições heterotróficas. Fica demonstrado assim, ainda, a importante contribuição dos pigmentos na fração de nitrogênio não proteico. Na sequência, um terceiro e quarto experimentos foram delineados, visando avaliar a influência do nitrogênio inorgânico intracelular na fração não proteica e na produção de proteína, assim como a caracterização da fração proteica quanto ao seu perfil aminoacídico. O estudo da dinâmica do nitrogênio intracelular demonstrou que o N-NH4 + foi a forma nitrogenada predominante, perfazendo importante fração de N-NP, sendo, portanto, os teores de N-NP significativamente dependente dos teores de pigmentos e nitrogênio intracelular. Os aminogramas das biomassas geradas pelos cultivos autotróficos e heterotróficos indicaram como aminoácidos majoritários o ácido glutâmico e aspártico, seguidos por valina, leucina e isoleucina, e como minoritários, lisina, glicina e metionina. O perfil aminoacídico caracterizou-se por apresentar aminoácidos essenciais como isoleucina, metionina + cisteína, fenilalanina + tirosina, valina e treonina em concentrações superiores ao preconizado pela FAO/WHO. A caracterização da fração proteica quanto ao perfil aminoacídico qualificou esta biomassa como fonte potencial de proteína. Os resultados obtidos neste trabalho demonstram a influência e dinâmica de distribuição dos compostos nitrogenados em Aphanothece microscopica Nägeli. Fica demonstrado, ainda, que a implementação do conceito de biorrefino, no tipo de agroindústria estudado, poderá representar importantes possibilidades de aproveitamento sustentável do efluente gerado.
