In this qualitative user evaluation participants in the project refugee guide, Falun, was interviewed about their experience of the project and how the project contributed to their integration into society and establishment on the labor market. The collection of empirical data has been made by six interviews. The results of the evaluation indicate that participants feel that they through the project had the opportunity to meet new people, practice the language and learn about each other's culture. The results also show that participants are satisfied with the project. Some participants expressed a desire to meet with several people at once while others don´t have that need. Furthermore, the interviews shows that establishment on labor market occurred through job placement and not through participation in the project. Integration proved to be a difficult concept to define, but the interviews show that the participants have a perception that it is about learning the Swedish language and culture.

Länsstyrelsen Dalarna och Borlänge kommun startade höstterminen 2013 pilotprojektet ANDT på schemat – En satsning för ökad kunskap och minskad ANDT-användning i Dalarna. Förutom att syftet var att eleverna skulle få möjlighet att förbättra sina kunskaper om temat ANDT skulle undervisningen även bidra till att utveckla elevernas kritiska tänkande och förmåga att reflektera över sina värderingar och attityder. Eleverna skulle också ges möjligheter att utveckla sina förmågor att söka information via internet, kritiskt granska källor, träna att arbeta i projektform samt skriva en projektrapport. Syftet med utvärderingen är att beskriva och tolka på vilka sätt ett nyutvecklat kursmaterial för att undervisa om ANDT-frågor uppfattas fungera i praktiken. Följande frågeställningar formuleras med utgångspunkt i detta: Hur uppfattar de medverkande lärarna och eleverna att pilotprojektet ANDT på schemat fungerade avseende att: utveckla elevernas kunskaper om ANDT-frågor påverka elevernas motivation och intresse för ökad kunskap inom ANDT-området påverka elevernas värderingar och attityder inom ANDT-området utveckla elevernas förmågor att söka information på internet och att kritiskt granska källor utveckla elevernas förmågor att arbeta projektinriktat i grupper samt att skriva en projektrapport Data har samlats in genom enkätundersökning till de elever som deltog i pilotprojektet, genom intervjuer med lärarna, observationer vid klassrumsbesök och de rapporter som eleverna producerade under projekttiden. Resultatet redovisas tematiserat och följer de tre faser som undervisningen i ANDT på schemat byggdes upp runt, d.v.s. startblock, resa och målgång. Erfarenheterna från projektet visar bl.a. att en majoritet av eleverna svarade att de fick tillräcklig information både om ämnesinnehåll och att de förstod hur arbetet skulle genomföras. Alla elever var dock inte med ”på tåget”. Det var cirka 15 procent av eleverna som ansåg att de inte hade fått en bra introduktion till temat, men framför allt var det drygt 20 procent av flickorna och nästan 30 procent av pojkarna som uppgav att de inte fått tydliga instruktioner till hur arbetet skulle genomföras. Under arbetets gång var det sedan cirka 15 procent av eleverna som uppgav att arbetet i deras grupp inte fungerade och drygt 15 procent av flickorna och drygt 20 procent av pojkarna som svarade att alla i deras grupp inte medverkade i arbetet. En relativt liten andel (knappt 10 %) ansåg att de inte fick det stöd och hjälp i arbetet som de behövde. Det var även relativt många som tyckte att det inte var lika givande att arbeta projektinriktat i grupper som det är med ”vanlig” undervisning, 17 procent av flickorna och 24 procent av pojkarna. Dessa elever tycker inte heller att det var vare sig ett roligt, intressant eller meningsfullt sätt att arbeta på. En överväldigande majoritet av eleverna upplever ändå att deras kunskaper om ANDT-frågor hade förbättrats. I det avseendet framstår det sätt som att undervisningen genomförts på som att det har fungerat väl, liksom att eleverna har utvecklat sin förmåga att söka information och värdera källor på internet. Många av eleverna säger också att de utvecklat sin förmåga att skriva en projektrapport. Däremot förefaller det vanskligt att veta något mer bestämt om hur elevernas attityder och värderingar påverkats. En positiv tolkning kan dock vara att många elever fått upp ögonen för det riskfyllda med att nyttja alkohol, narkotika, dopning och tobak och därför också i en del avseenden omprövat sina tidigare ståndpunkter. Resultatet av projektet visar att det verkar finnas en potential för utveckling av startblocket, framför allt genom att lägga mer tid på introduktionen av ämnesinnehållet och att ge mer utrymme åt att beskriva hur arbetet är tänkt att genomföras. Det verkar också finnas en förbättringspotential genom att erbjuda eleverna hjälp och stöd under hela processen och att lärarna är uppmärksamma på hur samarbetet i grupperna fungerar. Det framstår som att lärarens roll som ledare för arbetsprocessen än mer behöver sättas i centrum. Resultatet kan sammanfattas som att en överväldigande majoritet av eleverna upplever att deras kunskaper om ANDT-frågor förbättrats. I det avseendet framstår det sätt som att undervisningen genomförts på som att det har fungerat väl. När det däremot gäller elevernas intresse för att arbeta med ämnesövergripande projektarbete i gruppform är bilden mera splittrad. Det är visserligen en klar majoritet av eleverna som anger att detta sätt att arbeta både är roligt, intressant och meningsfullt och att de kan rekommendera andra elever att arbeta på detta sätt. Mot detta står att runt 20 procent av eleverna säger att de inte alls uppskattar denna arbetsform. Detta skapar svårigheter om målet är att alla elever ska vara delaktiga i undervisningen. Å andra sidan förefaller det sannolikt att även när andra undervisningsformer används så attraheras inte heller då alla elever. En slutsats i det avseendet är att det, trots att alla elever inte uppskattar detta sätt att arbeta, mycket som talar för att detta sätt att undervisa framstår som värt att utveckla vidare.

A Wolbachiapertence a um grupo de bactérias intracelulares, transmitidas maternalmente, que se encontram amplamente distribuídas nos artrópodes. Estes endossimbiontes encontram-se normalmente nos tecidos do sistema reprodutor dos artrópodes e têm a capacidade manipular a sua reproduçãode modo a garantir a sua transmissão à descendência e rápida dispersão na população.A capacidade de manipulação reprodutiva da Wolbachia tornou-a o alvo de diversos estudos para uma maior e melhor perceção da sua implicação em processos biológicos e evolutivos e por acreditar-se que esta bactéria é uma promissora ferramenta no controlo de populações de insetos que são pragas agrícolas. Os afídeos são um grupo de insetos associados às plantas que podem ter um efeito devastador nas culturas agrícolas e hortícolas pois não só retiram nutrientes às plantas como podem ser vetores de doenças. Embora durante muito tempo se pensasse que estes insetos não albergavam a Wolbachia estudos recentes mostram que são várias as espécies de afídeos infetados com esta bactéria. O principal objetivo deste trabalho é estudar a prevalência de infeção por Wolbachia assim como a caracterização das suas estirpes em amostras de afídeos dos Arquipélagos da Madeira e dos Açores. Neste estudo foram analisadas 545 amostras de afídeos, 361 provenientes do Arquipélago da Madeira e 184 dos Açores. Utilizando a técnica da “Polymerase Chain Reaction” (reação em cadeia da polimerase) amplificou-se o gene 16S rRNA (RNA ribossomal) e verificou-se que 32 destas amostras encontravam-se infetadas com Wolbachia sendo a maior parte das amostras infetadas provenientes dos Açores. Para determinar a estirpe que infeta estes afídeos utilizou-se a tipagem sequencial multilocus (MLST) com os genesglutamil-tRNA amidotransferase, subunidade B (gatB), citocromo c oxidase, subunidade I (coxA), proteína hipotética conservativa (hcpA) e proteína da divisão celular (ftsZ). A análise filogenética realizada para os diferentes genes mostrou que grande parte das amostras analisadas estão incluídas em dois dos novos supergrupos descobertos para Wolbachia, supergrupo M e N.Foi detetada a presença da mesma estirpe de Wolbachia, supergrupo N, em duas espécies diferentes de afídeos, Neophyllaphis podocarpi e Aphis spiraecola da mesma planta hospedeira, Podocarpus macrophyllus. Esta infeção reforça a ideia de que a Wolbachia não recorre só a transmissão vertical para se difundir na população mas utiliza também a transmissão horizontal. A deteção e caracterização das estirpes de Wolbachia é essencial para um maior entendimento sobre a sua origem e forma de disseminação. Esta informação é importante para desenvolvimento de estratégias de controlo de pestes recorrendo a estes endossimbiontes.

Carbon fiber ultramicroelectrodes are shown to be suitable for adsorptive stripping potentiometric measurements of trace DNA and RNA. The origin of the carbon fiber has a profound effect upon its suitability for trace analysis of nucleic acids, with the 'Aesar' materials performing most favorably. The resulting ultramicroelectrodes offer effective adsorptive accumulation of DNA and RNA from unstirred microliter-volume solutions, and are shown to be useful in adsorptive stripping transfer experiments. The influence of the surface pretreatment and accumulation conditions is described, along with the analytical-performance characteristics. The detection limits are 6, 15 and 40 mu g/l tRNA, ssDNA and dsDNA, respectively (5 min accumulation). (C) 1998 Elsevier B.V. S.A.

The family Callichthyidae comprises eight genera of fishes widely distributed across the Neotropical region. In the present study, sequences of the mitochondrial genes 12S rRNA, 16S rRNA, ND4, tRNA(His), and tRNA(Scr) were obtained from 28 callichthyid specimens. The sample included 12 species of Corydoras, three species of Aspidoras, two species of Brochis, Dianema, Lepthoplosternum, and Megalechis, and two local populations of Callichthys and Hoplosternum. Sequences of Nematogenys inermis (Nematogenyidae), Trichomycterus areolatus, and Henonemus punctatus (Trichomycteridae), Astroblepus sp. (Astroblepidae), and Neopleeostomus paranensis, Delturus parahybae, and Hemipsilichthys nimius (Loricariidae) were included as the outgroup. Phylogenetic analyses were performed by using the methods of maximum parsimony and maximum likelihood. The results of almost all analyses were very similar. The family Callichthyidae is monophyletic and comprises two natural groups: the subfamilies Corydoradinae (Aspidoras, Brochis, and Corydoras) and Callichthyinae (Callichthys, Dianema, Hoplosternum, Lepthoplosternum, and Megalechis), as previously demonstrated by morphological studies. The relationships observed within these subfamilies are in several ways different from those previously proposed on the basis of morphological data. The previous uncertain placement of Lysapsus and Pseudis within the neobatrachians was recently resolved by molecular and morphological studies, which supported them as members of the Hylinae subfamily. Their inter- and intrageneric relationships, however, have long been under debate and no studies shed light on these questions. Aiming to elucidate such questions, this paper used 3.2 kb comprising the mitochondrial genes 12S, tRNA valine, 16S and cytochrome b, and the nuclear exon 1 coding for rhodopsin, to all representatives of both genera (except to two subspecies of Pseudis paradoxa). The results identified three major clades: the clade 1 was composed by Lysapsus species and subspecies; clade 2 included the subspecies of the Pseudis paradoxa (Pseudis paradoxa paradoxa, P. paradoxa platensis and P. paradoxa occidentalis), P. fusca, P. bolbodactyla and P. tocantins, and clade 3 was composed by Pseudis southern Brazil species (Pseudis cardosoi and P. minuta). As closely related taxa we found Pseudis minuta + P. cardosoi; P. tocantins + P. fusca, and the subspecies within each genus. Evidence that Pseudis is not monophyletic with respect to Lysapsus was found and we suggest Lysapsus to be a junior synonym of Pseudis. Based on pair-wise comparison among gene sequences, we also suggest that the subspecies of Pseudis paradoxa and Lysapsus limellum must be considered as full species. The application of adsorptive stripping potentiometry to the reductive detection of nucleic acids at mercury electrodes is reported. Compared to analogous voltammetric stripping modes, constant current potentiometric stripping analysis (PSA) effectively addresses the hydrogen discharge background problem, and hence greatly improves the characteristics of the superimposed cytosine/adenine (CA) reduction peak. Compared to earlier schemes for trace measurements of nucleic acids at mercury or carbon electrodes that rely on anodic signals arising from the guanine residue, convenient quantitation can now be carried out in connection with the cytosine and adenine residues. Variables influencing the adsorptive PSA response are explored and optimized. With five minute accumulation, the detection limits for tRNA, ssDNA and dsDNA are 30 mu g l(-1), 60 mu g l(-1) and 2 mg l(-1), respectively. Such different values reflect the strong dependence of the PSA CA signal upon the nucleic-acid structure. This allows the quantitation of ssDNA or tRNA in the presence of dsDNA, and offers new possibilities for electrochemical studies of DNA structure and interactions.

Restriction fragment length polymorphism (RFLP) and sequence analyses of the PCR-amplified 16S-23S rDNA intergenic spacer (ITS) were used for differentiating Acidithiobacillus thiooxidans strains from other related acidithiobacilli, including A. ferrooxidans and A. caldus. RFLP fingerprints obtained with AluI, DdeI, HaeIII, HinfI and MspI enabled the differentiation of all Acidithiobacillus reference strains into species groups. The A. thiooxidans strains investigated (metal mine isolates) yielded identical RFLP patterns to the A. thiooxidans type strain (ATCC 19377(T)), except for strain DAMS, which had a distinct pattern for all enzymes tested. Fourteen A. ferrooxidans mine strains were assigned to 3 RFLP groups, the majority of which were grouped with A. ferrooxidans ATCC 23270(T). The spacer region of one representative strain from each of the RFLP groups obtained was subjected to sequence analysis, in addition to eleven additional A. thiooxidans strains isolated from sediment and water samples, and A. caldus DSM 8584(T). The tRNA(IIe) and tRNA(Ala) genes, present in all strains analyzed, showed high sequence similarity. Phylogenetic analysis of the ITS sequences differentiated all three Acidithiobacillus species. Inter- and infraspecific genetic variations detected were mainly due to the size and sequence polymorphism of the ITS3 region. Mantel tests showed no significant correlation between ITS sequence similarity and the geographical origin of strains. The results showed that the 16S-23S rDNA spacer region is a useful target for the development of molecular-based methods aimed at the detection, rapid differentiation and identification of acidithiobacilli. The evidentiary basis of the currently accepted classification of living amphibians is discussed and shown not to warrant the degree of authority conferred on it by use and tradition. A new taxonomy of living amphibians is proposed to correct the deficiencies of the old one. This new taxonomy is based on the largest phylogenetic analysis of living Amphibia so far accomplished. We combined the comparative anatomical character evidence of Haas (2003) with DNA sequences from the mitochondrial transcription unit HI (12S and 16S ribosomal RNA and tRNA(Valine) genes, 2,400 bp of mitochondrial sequences) and the nuclear genes histone H3, rhodopsin, tyrosinase, and seven in absentia, and the large ribosomal subunit 28S (approximate to 2,300 bp of nuclear sequences; ca. 1.8 million base pairs; x ($) over bar = 3.7 kb/terminal). The dataset includes 532 terminals sampled from 522 species representative of the global diversity of amphibians as well as seven of the closest living relatives of amphibians for outgroup comparisons.

A molecular phylogenetic analysis of the Hyla pulchella species group was performed to test its monophyly, explore the interrelationships of its species, and evaluate the validity of the taxa that were considered subspecies of H. pulchella. Approximately 2.8 kb from the mitochondrial genes 12s, tRNA valine, 16s, and Cytochrome b were sequenced. The analysis included 50 terminals representing 10 of the 14-15 species currently recognized in the H. pulchella group, including samples from several localities for some taxa, several outgroups, as well as two species previously suspected to be related with the group (Hyla guentheri and Hyla hischoffi). The results show that the H. pulchella and Hyla circumdata groups are distantly related, and, therefore, should be recognized as separate groups. As currently defined, the H. pulchella group is paraphyletic with respect to the Hyla polytaenia group; therefore, we recognize the Hyla polytaenia clade in the H. pulchella group. Two subspecies of H. pulchella recognized by some authors are considered full species including Hyla pulchella riojana because it is only distantly related to H. pulchella, and Hyla pulchella cordobae because molecular and non-molecular evidence suggests that it is specifically distinct. With the inclusion of the H. polytaenia clade, H. guentheri, and H. bischoffi, and the recognition of the two former subspecies of H. pulchella as distinct species, the H. pulchella group now comprises 25 described species. All representatives of the H. pulchella group with an Andean distribution are monophyletic and nested within a clade from the Atlantic forest from south-southeastern Brazil/northeastern Argentina, and Cerrado gallery forest from central Brazil. The eukaryotic translation initiation factor 2 (eIF2) binds the methionyl-initiator tRNA in a GTP-dependent mode. This complex associates with the 40 S ribosomal particle, which then, with the aid of other factors, binds to the 5' end of the mRNA and migrates to the first AUG codon, where eIF5 promotes GTP hydrolysis, followed by the formation of the 80 S ribosome. Here we provide a comparative sequence analysis of the β subunit of eIF2 and its archaeal counterpart (aIF2β). aIF2β differs from eIF2β in not possessing an N-terminal extension implicated in binding RNA, eIF5 and eIF2B. The remaining sequences are highly conserved, and are shared with eIF5. Previously isolated mutations in the yeast eIF2β, which allow initiation of translation at UUG codons due to the uncovering of an intrinsic GTPase activity in eIF2, involve residues that are conserved in aIF2β, but not in eIF5. We show that the sequence of eIF2B homologous to aIF2β is sufficient for binding eIF2γ, the only subunit with which it interacts, and comprises, at the most, 78 residues, eIF5 does not interact with eIF2γ, despite its similarity with eIF2β, probably because of a gap in homology in this region. These observations have implications for the evolution of the mechanism of translation initiation.

In the present study, we describe the cloning and characterization of a new SINE-like element from O. niloticus (ROn-2) and show the distribution of this SINE and a previously isolated SINE, ROn-1, in the chromosomes of O. niloticus. The ROn-2 element is 359 base pairs (bp) in length, contains short direct terminal repeats, a tRNA-related region similar to tRNA Val and tRNA Arg, a tRNA-unrelated region, and a poly-A tail. Analysis of the chromosomal distribution of ROn-1 and ROn-2 by fluorescent in situ hybridization showed that both SINE sequences are present in all chromosomes of tilapia, and organized in small clusters. The only exception was a large cluster of ROn-1 repeats found in the middle of the long arm of chromosome 1. In view of our data we discuss the hypothesis that the absence of large clusters of SINE sequences and the structural composition of these sequences may explain the absence of base-specific fluorochrome bands in the chromosomes of tilapia.

Hylidae is a large family of American, Australopapuan, and temperate Eurasian treefrogs of approximately 870 known species, divided among four subfamilies. Although some groups of Hylidae have been addressed phylogenetically, a comprehensive phylogenetic analysis has never been presented. The first goal of this paper is to review the current state of hylid systematics. We focus on the very large subfamily Hylinae (590 species), evaluate the monophyly of named taxa, and examine the evidential basis of the existing taxonomy. The second objective is to perform a phylogenetic analysis using mostly DNA sequence data in order to (1) test the monophyly of the Hylidae; (2) determine its constituent taxa, with special attention to the genera and species groups which form the subfamily Hylinae, and c) propose a new, monophyletic taxonomy consistent with the hypothesized relationships. We present a phylogenetic analysis of hylid frogs based on 276 terminals, including 228 hylids and 48 outgroup taxa. Included are exemplars of all but 1 of the 41 genera of Hylidae (of all four nominal subfamilies) and 39 of the 41 currently recognized species groups of the species-rich genus Hyla. The included taxa allowed us to test the monophyly of 24 of the 35 nonmonotypic genera and 25 species groups of Hyla. The phylogenetic analysis includes approximately 5100 base pairs from four mitochondrial (12S, tRNA valine, 16S, and cytochrome b) and five nuclear genes (rhodopsin, tyrosinase, RAG-1, seventh in absentia, and 28S), and a small data set from foot musculature. Concurring with previous studies, the present analysis indicates that Hemiphractinae are not related to the other three hylid subfamilies. It is therefore removed from the family and tentatively considered a subfamily of the paraphyletic Leptodactylidae. Hylidae is now restricted to Hylinae, Pelodryadinae, and Phyllomedusinae. Our results support a sister-group relationship between Pelodryadinae and Phyllomedusinae, which together form the sister taxon of Hylinae. Agalychnis, Phyllomedusa, Litoria, Hyla, Osteocephalus, Phrynohyas, Ptychohyla, Scinax, Smilisca, and Trachycephalus are not monophyletic. Within Hyla, the H. albomarginata, H. albopunctata, H. arborea, H. boons, H. cinerea, H. eximia, H. geographica, H. granosa, H. microcephala, H. miotympanum, H. tuberculosa, and H. versicolor groups are also demonstrably nonmonophyletic. Hylinae is composed of four major clades. The first of these includes the Andean stream-breeding Hyla, Aplastodiscus, all Gladiator Frogs, and a Tepuian clade. The second clade is composed of the 30-chromosome Hyla, Lysapsus, Pseudis, Scarthyla, Scinax (including the H. uruguaya group), Sphaenorhynchus, and Xenohyla. The third major clade is composed of Nyctimantis, Phrynohyas, Phyllodytes, and all South American/West Indian casque-headed frogs: Aparasphenodon, Argenteohyla, Corythomantis, Osteocephalus, Osteopilus, Tepuihyla, and Trachycephalus. The fourth major clade is composed of most of the Middle American/Holarctic species groups of Hyla and the genera Acris, Anotheca, Duellmanohyla, Plectrohyla, Pseudacris, Ptychohyla, Pternohyla, Smilisca, and Triprion. A new monophyletic taxonomy mirroring these results is presented where Hylinae is divided into four tribes. Of the species currently in Hyla, 297 of the 353 species are placed in 15 genera; of these, 4 are currently recognized, 4 are resurrected names, and 7 are new. Hyla is restricted to H. femoralis and the H. arborea, H. cinerea, H. eximia, and H. versicolor groups, whose contents are redefined. Phrynohyas is placed in the synonymy of Trachycephalus, and Pternohyla is placed in the synonymy of Smilisca. The genus Dendropsophus is resurrected to include all former species of Hyla known or suspected to have 30 chromosomes. Exerodonta is resurrected to include the former Hyla sumichrasti group and some members of the former H. miotympanum group. Hyloscirtus is resurrected for the former Hyla armata, H. bogotensis, and H. larinopygion groups. Hypsiboas is resurrected to include several species groups - many of them redefined here - of Gladiator Frogs. The former Hyla albofrenata and H. albosignata complexes of the H. albomarginata group are included in Aplastodiscus. New generic names are erected for (1) Agalychnis calcarifer and A. craspedopus; (2) Osteocephalus langsdorffii; the (3) Hyla aromatica, (4) H. bromeliacia, (5) H. godmani, (6) H. mixomaculata, (7) H. taeniopus, (8) and H. tuberculosa groups; (9) the clade composed of the H. pictipes and H. pseudopuma groups; and (10) a clade composed of the H. circumdata, H. claresignata, H. martinsi, and H. pseudopseudis groups.
