Background Helichrysum species are used extensively for stress-related ailments and as dressings for wounds normally encountered in circumcision rites, bruises, cuts and sores. It has been reported that Helichysum species are used to relief abdominal pain, heart burn, cough, cold, wounds, female sterility, menstrual pain. Results From the extracts of Helichrysum foetidum (L.) Moench, six known compounds were isolated and identified. They were 7, 4′-dihydroxy-5-methoxy-flavanone (1), 6′-methoxy-2′,4, 4′-trihydroxychalcone (2), 6′-methoxy-2′,4-dihydroxychalcone -4′-O-β-D-glucoside (3), apigenin (4), apigenin-7-O-β-D-glucoside (5), kaur-16-en-18-oic acid (6) while two known compounds 3,5,7-trihydroxy-8-methoxyflavone (12), 4,5-dicaffeoyl quinic acid (13) together with a mixture of phytosterol were isolated from the methanol extract of Helichrysum mechowianum Klatt. All the compounds were characterized by spectroscopic and mass spectrometric methods, and by comparison with literature data. Both extracts and all the isolates were screened for the protease inhibition, antibacterial and antifungal activities. In addition, the phytochemical profiles of both species were investigated by ESI-MS experiments. Conclusions These results showed that the protease inhibition assay of H. foetidum could be mainly attributed to the constituents of flavonoids glycosides (3, 5) while the compound (13) from H. mechowianum contributes to the stomach protecting effects. In addition, among the antibacterial and antifungal activities of all the isolates, compound (6) was found to possess a potent inhibitor effect against the tested microorganisms. The heterogeneity of the genus is also reflected in its phytochemical diversity. The differential bioactivities and determined constituents support the traditional use of the species. Molecular modelling was carried out by computing selected descriptors related to drug absorption, distribution, metabolism, excretion and toxicity (ADMET).

This study reports on the influence of heat and hydrogen peroxide combination on the inactivation kinetics of two heat resistant molds: Neosartorya fischeri and Paecilomyces variotii. Spores of different ages (1 and 4 months) of these molds were prepared and D-values (the time required at certain temperature/hydrogen peroxide combination to inactivate 90% of the mold ascospores) were determined using thermal death tubes. D-values found for P. variotii ranged from 1.2 to 25.1 s after exposure to different combinations of heat (40 or 60 degrees C) and hydrogen peroxide (35 or 40% w/w) while for N. fischeri they varied from 2.7 to 14.3 s after exposure to the same hydrogen peroxide concentrations and higher temperatures (60 or 70 degrees C). The influence of temperature and hydrogen peroxide concentration on the d-values varied with the genus of mold and their ages. A synergistic effect of heat and hydrogen peroxide in reducing D-values of Paecilomyces variotti and N. fischeri has been observed. In addition to strict control of temperature, time and hydrogen concentration, hygienic storage and handling of laminated paperboard material must be considered to reduce the probability of package's contamination. All these measures together will ensure package's sterility that is imperative for the effectiveness of aseptic processing and consequently to ensure the microbiological stability of processed foods during shelf-life. Objective: To compare the polymerization status of mouse oocyte spindles exposed to various temperatures at various stages of meiosis. Design: Experimental animal study. Setting: University animal laboratory. Animal(s): CF1 mice. Intervention(s): Immature oocytes matured to metaphase I (MI), telophase I (TI), and metaphase II (MII) were incubated at 37 degrees C (control), room temperature (RT), or 4 degrees C for 0, 10, 30, and 60 minutes. Spindle analysis subsequently was performed using polarized field microscopy and immunocytochemistry. Spindles of TI and MII oocytes that underwent vitrification and warming were analyzed also by immunocytochemistry. Main Outcome Measure(s): Detection of polymerized meiotic spindles. Result(s): At RT, and after 60 minutes at 4 degrees C, a significant time-dependent decrease in the percentage of polymerized meiotic spindles was observed in MI and MII oocytes, but not in TI oocytes. The polymerization of TI spindles at 4 degrees C was similar to that of TI spindles at 4 degrees C that underwent vitrification and warming. Conclusion(s): Significant differences in the microtubule dynamics of MI, TI, and MII oocytes incubated at different temperatures were observed. This study aimed at enumerating molds (heat-labile and heat-resistant) on the surface of paperboard material to be filled with tomato pulps through an aseptic system and at determining the most heat-and hydrogen peroxide-resistant strains. A total of 118 samples of laminated paperboard before filling were collected, being 68 before and 50 after the hydrogen peroxide bath. Seven molds, including heat-resistant strains (Penicillium variotii and Talaromyces flavus) with counts ranging between 0.71 and 1.02 CFU/cm(2) were isolated. P. variotii was more resistant to hydrogen peroxide than T. flavus and was inactivated after heating at 85 degrees C/15 min. When exposed to 35 % hydrogen peroxide at 25 degrees C, T. flavus (F5E2) and N. fischeri (control) were less resistant than P. variotti (F1A1). P. citrinum (F7E2) was shown to be as resistant as P. variotti. The D values (the time to cause one logarithmic cycle reduction in a microbial population at a determined temperature) for spores of P. variotii (F1A1) and N. fischeri (control) with 4 months of age at 85 and 90 degrees C were 3.9 and 4.5 min, respectively. Although the contamination of packages was low, the presence of heat-and chemical-resistant molds may be of concern for package sterility and product stability during shelf-life. To our knowledge, this is the first report that focuses on the isolation of molds, including heat-resistant ones, contaminating paperboard packaging material and on estimating their resistance to the chemical and physical processes used for packaging sterilization.

A produção em escala comercial de sementes híbridas de cebola (Allium cepa) tem sido conduzida com o emprego de dois sistemas de macho-esterilidade do tipo genética-citoplasmática (CMS-S e CMS-T) em associação ao citoplasma normal (macho-fértil). No entanto, a análise molecular desses diferentes tipos citoplasmáticos ainda não está disponível para um grande número de acessos de cebola adaptados para cultivo em regiões tropicais. Além de adaptação às condições edafoclimáticas do Brasil, muitos desses acessos apresentam tolerância a doenças, sendo de potencial valor como genitores de híbridos. O presente trabalho visou identificar os tipos citoplasmáticos de acessos de cebola de diferentes grupos morfoagronômicos de interesse para o melhoramento genético no Brasil, usando a reação da polimerase em cadeia (PCR) com 'primers' específicos para regiões polimórficas do genoma mitocondrial de cebola. Foi observada, nos 66 acessos amostrados, a presença dos três principais tipos de citoplasma descritos para cebola (S, N e T). Foi constatada maior frequência do citoplasma S (56%) seguido do citoplasma T (25,8%). Em 18,2% das amostras, foi encontrado exclusivamente o citoplasma N. Essa caracterização pode ser útil para guiar a escolha de materiais genéticos dentro dos programas de melhoramento com objetivo de desenvolver cultivares híbridas adaptadas às condições tropicais.

[EN] The atmospheric CO2 level is rising. Its greenhouse effect is partially mitigated by terrestrial (plants) and marine photosynthetic organisms (algae, phytoplankton), and also by the less-known chemosynthetic bacteria. Within this group of bacteria, nitrifiers have a direct and indirect impact on carbon fixation because, on one hand, they are autotrophs and, on the other, they release inorganic nitrogenous nutrients that feed other photoautotrophs. A new assay which simplifies the measurement of nitrification would improve our knowledge about the ocean’s capacity to fix CO2. Knowing how to cultivate these microbes from marine water samples is a first step towards developing new nitrification detection techniques. During the last six months, we have isolated and cultured a natural assembledge of marine nitrifiers. Our larger objective is to develop a way to enzymatically detect nitrification. However, to do this, we need large quantities of nitrifiers. Consequently, at this point, culturing this marine nitrifier community is our priority. We have learned that pH, nutrient levels, air flow, temperature, low light and sterility are critical for growing healthy nitrifiers. With this knowledge we will now be able to conduct experiments with the nitrifiers and develop the methodology that we seek.

Identifizierung, Sequenzierung und Charakterisierung des Dmxl1-Gen in Mus musculus sowie die funktionelle Analyse durch Knock-OutrnrnBei Dmxl1 handelt es sich um ein neuartiges Gen aus Mus musculus. Das ebenfalls in der vorliegenden Arbeit bioinformatisch untersuchte Gen DMXL1 ist das zu Dmxl1 homologe Gen des Menschen. Beide Gene bestehen aus 43 Exons, das murine Dmxl1 codiert für eine mRNA von 10992 bp bzw. 12210 bp, das humane DMXL1 kodiert für eine cDNA von 11082 bp, der offene Leserahmen umfasst bei der Maus 9042 bp. In der Maus konnte ein mögliches alternatives Polyadenylierungssignal identifiziert werden. Zwischen beiden Spezies sind die Exonpositionen und ihre Längen hoch konserviert. Dmxl1 liegt auf dem Crick-Strang von Chromosom 18 Bande C, der translatierte Bereich erstreckt sich auf genomischer Ebene über 129558 bp und die Orientierung verläuft in Richtung Centromer. Dmxl1 und DMXL1 gehören damit zu den größten bekannten Genen in Maus und Mensch. Bei beiden Spezies liegen die DmX-Homologen genomisch innerhalb eines Bereichs der Isochoren-Klasse L1 in einer Gen-armen Region. Die Anzahl der repetitiven Elemente innerhalb der Genregion von Dmxl1 liegt 6% unter dem erwarteten Wert eines L1 Isochors, die Anzahl beim Menschen liegt 4% über dem erwarteten Wert. Um die mögliche Promotorstruktur von Dmxl1 darzustellen, wurden umfangreiche in silico-Analysen der Region um den putativen Transkriptionsstart vorgenommen. Mit Hilfe der gewonnenen Daten konnte ein Transkriptionstartpunkt identifiziert werden. Zudem wurde eine Promotorstruktur erarbeitet, bei der angenommen werden kann, dass sie eine gute Näherung an die tatsächlich vorhandenen Bindungsstellen von Transkriptionsfaktoren darstellt. Die mit bioinformatischen Werkzeugen erzeugte virtuelle Promotor- und Enhancerstruktur zeigt das Potenzial, Dmxl1 basal und ubiquitär zu exprimieren. Gleichzeitig zeigen diese Daten, dass Dmxl1 vermutlich in einigen Geweben der Keimbahn, im Fettgewebe, dem blutbildenen System und während der Embryogenese hochkomplex reguliert werden kann. Eine regulierte Expression zur Steuerung des Energiestoffwechsels ist ebenfalls wahrscheinlich. Diese Ergebnisse passen sehr gut zu den experimentell ermittelten Daten und den beobachteten Phänotypen Dmxl1-chimärer Mäuse.rnDie abgeleitete Aminosäuresequenz umfasst in der Maus 3013 AS, im Menschen 3027 AS, der Vergleich der abgeleiteten Aminosäuresequenzen zeigt eine Identität von 89,3 % und eine Similarität von 94,7 % zwischen beiden Spezies. Im Dmxl1/DMXL1-Protein von Maus und Mensch konnten mindestens 24 und maximal 36 WD-Wiederholungseinheiten identifiziert werden, zudem wurden eine Reihe weiterer konservierter Proteinmotive gefunden. Die in silico-Strukturanalysen beider abgeleiteter Aminosäuresequenzen lässt vermuten, dass sich C- und N-terminal WD-Propellerstrukturen befinden. In dieser Arbeit gelang eine C-terminale Rekonstruktion einer 10-blättrigen Propellerstruktur, denkbar ist jedoch auch eine Struktur mit mindestens drei WD-Propellern, wenn eine prädominante Struktur mit Propellern aus jeweils sieben Propellerblättern angenommen wird.rnDas primäre Ziel dieser Arbeit, die Etablierung einer stabilen Mauslinie mit diruptiertem Dmxl1-Gen konnte aufgrund einer beobachteten Haploinsuffizienz nicht erreicht werden. Trotz zahlreicher Transformationen von Maus-Stammzelllinien konnte letztlich nur eine stabil transformierte Linie mit einem Dmxl1-Null-Allel identifiziert werden, was auch zu den theoretischen Daten und den angenommenen Aufgaben von Dmxl1 als komplex und diffizil reguliertes Multifunktions-Protein passt. Aus der transformierten Mauszelllinie konnten chimäre Mäuse entwickelt werden, die in Abhängigkeit von dem Ausmaß des Chimärismus phänotypisch massive Schädigungen aufwiesen. Neben einer Teilsterilität wurden massive Fettleibigkeit und ein ausgeprägter Hypogonadismus beobachtet. Keines der Tiere war in der Lage das Dmxl1-Null-Allel zu transduzieren. Die Tiere waren nur sehr eingeschränkt fertil, die wenigen Nachkommen entsprachen genotypisch und phänotypisch ausschließlich den verwendeten Blastocysten.rn

La presente Tesi di Dottorato intende affrontare una lettura critica della Casa in Belvederestraße 60, realizzata dall’architetto Oswald Mathias Ungers (Kaisersesch, 12 luglio 1926 – Köln, 30 settembre 2007), nel 1958-’59 a Köln-Müngersdorf, come studio per sé ed abitazione per la propria famiglia. Questo primo oggetto della ricerca viene considerato evidente espressione delle convinzioni formali e compositive dell’architetto, negli anni Cinquanta e Sessanta. A differenza di altri progetti residenziali coevi ed antecedenti, frutto di un’elaborazione autonoma, la prima casa che costruisce per sé riflette una maggiore libertà di pensiero, dettata dalla coincidenza delle figure di progettista e committente; a ciò si aggiunge anche una precisa volontà dichiarativa ed ideologica. Proprio quest’ultimo aspetto permette di introdurre il secondo oggetto della Tesi: il manifesto “ideologico”, Zu einer neuen Architektur, scritto dallo stesso Oswald Mathias Ungers e da Reinhard Gieselmann, alla fine del 1960; un breve testo che espone, con toni perentori ed inappellabili, il punto di vista dei due architetti nei confronti di un panorama architettonico e critico, caratterizzato da una sterilità di pensiero dilagante, a causa dell’egemonia costruttiva funzionalista. La ricerca indaga quindi le forti reciprocità delle due opere: casa e testo, viste in chiave di “manifesto scritto e manifesto costruito”. Il primo legame tra i due soggetti è senza dubbio la concomitanza temporale, (tra il 1958 ed il 1960) associata ad un rapporto causa-effetto, tale per cui il manifesto viene redatto a difesa delle aspre critiche scaturite dalla pubblicazione della casa sulla rivista Bauwelt. Il secondo nesso è la possibilità di comprendere le accezioni effettive dei termini impiegati nella redazione del testo, attraverso le forme di una delle opere maggiormente personali dell’architetto, estraendone il senso e conferendogli un’immagine architettonica. Si vuole creare così un rapporto biunivoco di traducibilità, dell’architettura nello scritto e della semantica ungersiana in azioni compositive.

Endometriosis is a significant gynecologic condition that can cause both pain and infertility and affects up to 15% of women during their reproductive years. In peritoneal endometriotic lesions, the expression of peroxisome proliferation-activated receptor gamma, a nuclear receptor with antiinflammatory and neuroprotective roles, is positively correlated with the pain reported by patients.

This pilot study evaluated whether combination of partial removal of ovarian tissue for cryobanking followed by ovarian stimulation and cryopreservation of oocytes can improve the efficacy of fertility preservation without further delaying cancer treatment. Initial partial removal of ovarian tissue did not substantially affect the average number and quality of retrieved oocytes after ovarian stimulation in this study.
