Arabinogalactan proteins (AGPs) are proteoglycans of higher plants, which are implicated in growth and development. We recently have shown that two AGPs, NaAGP1 (from Nicotiana alata styles) and PcAGP1 (from Pyrus communis cell suspension culture), are modified by the addition of a glycosylphosphatidylinositol (GPI) anchor. However, paradoxically, both AGPs were buffer soluble rather than membrane associated. We now show that pear suspension cultured cells also contain membrane-bound GPI-anchored AGPs. This GPI anchor has the minimal core oligosaccharide structure, d-Manα(1–2)-d-Manα(1–6)-d-Manα(1–4)-d-GlcN-inositol, which is consistent with those found in animals, protozoa, and yeast, but with a partial β(1–4)-galactosyl substitution of the 6-linked Man residue, and has a phosphoceramide lipid composed primarily of phytosphingosine and tetracosanoic acid. The secreted form of PcAGP1 contains a truncated GPI lacking the phosphoceramide moiety, suggesting that it is released from the membrane by the action of a phospholipase D. The implications of these findings are discussed in relation to the potential mechanisms by which GPI-anchored AGPs may be involved in signal transduction pathways.

In spite of much effort, no one has succeeded in isolating and characterizing the enzyme(s) responsible for synthesis of cellulose, the major cell wall polymer of plants. We have characterized two cotton (Gossypium hirsutum) cDNA clones and identified one rice (Oryza sativa) cDNA that are homologs of the bacterial celA genes that encode the catalytic subunit of cellulose synthase. Three regions in the deduced amino acid sequences of the plant celA gene products are conserved with respect to the proteins encoded by bacterial celA genes. Within these conserved regions, there are four highly conserved subdomains previously suggested to be critical for catalysis and/or binding of the substrate UDP-glucose (UDP-Glc). An overexpressed DNA segment of the cotton celA1 gene encodes a polypeptide fragment that spans these domains and binds UDP-Glc, while a similar fragment having one of these domains deleted does not. The plant celA genes show little homology at the N- and C-terminal regions and also contain two internal insertions of sequence, one conserved and one hypervariable, that are not found in the bacterial gene sequences. Cotton celA1 and celA2 genes are expressed at high levels during active secondary wall cellulose synthesis in developing cotton fibers. Genomic Southern blot analyses in cotton demonstrate that celA forms a small gene family.

Five microsatellite loci are presented for prickly acacia, Acacia nilotica ssp. indica (Benth.) Brenan, an introduced weed of national significance in Australia. These microsatellite loci were obtained through the construction of an enriched library and their use will enable us to determine the genetic origin and extent of genetic diversity of this weed in Australia.

To examine the effects of recent habitat fragmentation, we assayed genetic diversity in a rain forest endemic lizard, the prickly forest skink (Gnypetoscincus queenslandiae), from seven forest fragments and five sites in continuous forest on the Atherton tableland of northeastern Queensland, Australia. The rain forest in this region was fragmented by logging and clearing for dairy farms in the early 1900s and most forest fragments studied have been isolated for 50-80 years or nine to 12 skink generations. We genotyped 411 individuals at nine microsatellite DNA loci and found fewer alleles per locus in prickly forest skinks from small rain forest fragments and a lower ratio of allele number to allele size range in forest fragments than in continuous forest, indicative of a decrease in effective population size. In contrast, and as expected for populations with small neighbourhood sizes, neither heterozygosity nor variance in allele size differed between fragments and sites in continuous forests. Considering measures of among population differentiation, there was no increase in F-ST among fragments and a significant isolation by distance pattern was identified across all 12 sites. However, the relationship between genetic (F-ST) and geographical distance was significantly stronger for continuous forest sites than for fragments, consistent with disruption of gene flow among the latter. The observed changes in genetic diversity within and among populations are small, but in the direction predicted by the theory of genetic erosion in recently fragmented populations. The results also illustrate the inherent difficulty in detecting genetic consequences of recent habitat fragmentation, even in genetically variable species, and especially when effective population size and dispersal rates are low.

Erwinia amylovora is a necrogenic bacterium that causes fire blight of the Maloideae subfamily of Roseacae, such as apple and pear. It provokes necrosis in aerial parts of susceptible host plants and the typical hypersensitive reaction in non-host plants. The secreted hatpin, HrpN(ea), is able by itself to induce an active cell death in non-host plants. Ion flux modulations were shown to be involved early in such processes but very few data are available on the plasma membrane ion channel activities responsible for the pathogen-induced ion fluxes. We show here that HrpNea induces cell death in non-host Arabidopsis thaliana suspension cells. We further show that two cystic fibrosis transmembrane conductance regulator modulators, glibenclamide and bromotetramisole, can regulate anion channel activities and HrpN(ea)-induced cell death. O presente estudo foi desenvolvido com o objetivo de compreender a construção da identidade profissional de enfermeiros docentes do curso de graduação em Enfermagem de uma instituição privada localizada na cidade de São Paulo. Para tanto, discute os processos de constituição identitária pautado nos estudos de Vera Placco de Souza, no âmbito nacional, e de Maurice Tardif e Claude Dubar, no âmbito internacional. Em uma etapa exploratória, foi aplicado um questionário semi-estruturado a cinco professoras da pós-graduação em Enfermagem e realizada uma entrevista semi-diretiva com dois desses participantes com o objetivo de refinar os instrumentos de coleta. A pesquisa principal fez uso de entrevista realizada com sete professores do curso de graduação em Enfermagem da mesma instituição. Os dados, tanto do estudo exploratório como da pesquisa principal, foram analisados tendo como referência os pressupostos da análise de conteúdo sistematizados por Laurence Bardin e Maria Laura P. B. Franco. Os resultados indicaram que os enfermeiros se tornam professores impelidos pelo desejo de ensinar e de compartilhar conhecimentos, oportunidade de trabalho, flexibilidade de horário e complementação da renda profissional, e são influenciados por professores de sua trajetória formativa e por familiares. As características valorizadas por eles em um professor foram: o domínio do conteúdo ensinado, sobretudo o saber técnico; a busca contínua do conhecimento; ética; profissionalismo; humanização. Nota-se um predomínio das características que envolvem o conhecimento dos conteúdos em detrimento das questões pedagógicas. A reflexão e preocupação com a Docência estão presentes em suas falas, assim como a busca constante para a superação das dificuldades existentes na profissão. Eles consideram a Docência uma profissão com um papel social de destaque do mesmo modo que a Enfermagem. Outro fator destacado nas respostas foi a forte representação do enfermeiro na carreira profissional dos pesquisados. Em seus relatos, foi possível perceber que sua construção como docentes ocorre de forma contínua ao longo da carreira, o que fica evidenciado em suas falas quando afirmam serem melhores professores agora do que no início de sua trajetória profissional e que, apesar de ser uma caminhada espinhosa inicialmente, tornou-se prazerosa e significativa com o passar do tempo. Ressalta-se aqui a importância de se valorizar os saberes do professor, partindo deles e trabalhando-os teórica e conceitualmente para que o docente amplie a compreensão do processo de ensino em saúde que, no mundo contemporâneo, tornou-se de alta complexidade.

The cactus pear has become over the years an important forage alternative for brazilian semiarid region, especially during long periods of drought. Despite its importance for agriculture, its cultivation has dispensed basic crop practices and fundamentals technical-scientific basis about its climatic requirements. Thus, the main objective of this study was to elaborate the agroclimatic zoning of cactus pear (Opuntia sp.) for the state of Paraíba. The agroclimatic zoning of cactus pear was based on climatic indicators outlined in the literature and climatological data of precipitation and temperature (mean, maximum, and minimum) from 97 locations in the state of Paraíba. According to the results, the region of ‘Borborema’ is the most favorable for the cultivation of cactus pear. The regions of ‘Agreste’, ‘Sertão’, and coastal part of Litoral may be used but with restrictions. However, the cultivation of cactus pear is recommended throughout the state of Paraíba, except the coastal part of the Litoral and the region around Areia. In both cases, the inability is due to excessive precipitation.
