O presente trabalho parte da caracterização da Biblioteca UNL para desenvolver um estudo de avaliação dos principais Sistemas Integrados de Gestão de Bibliotecas (SIGB) em open source. Os SIGB são utilizados na automatização dos processos de trabalho das bibliotecas, através de diversos módulos funcionais e opções de configuração, e permitem a interoperabilidade através de normas e protocolos. O open source define-se como abordagem para o desenvolvimento e distribuição de software baseada no livre acesso ao código fonte, estando assim em oposição ao conceito de software proprietário. A Biblioteca UNL é caracterizada através da sua organização, dos seus espaços e recursos, dos seus processos de gestão documental e da infraestrutura tecnológica subjacente. O open source é abordado a partir da sua contextualização histórica, da organização dos seus projectos, das vantagens e inconvenientes para as instituições e do desenvolvimento e adopção de SIGB open source.

Existent computer programming training environments help users to learn programming by solving problems from scratch. Nevertheless, initiating the resolution of a program can be frustrating and demotivating if the student does not know where and how to start. Skeleton programming facilitates a top-down design approach, where a partially functional system with complete high level structures is available, so the student needs only to progressively complete or update the code to meet the requirements of the problem. This paper presents CodeSkelGen - a program skeleton generator. CodeSkelGen generates skeleton or buggy Java programs from a complete annotated program solution provided by the teacher. The annotations are formally described within an annotation type and processed by an annotation processor. This processor is responsible for a set of actions ranging from the creation of dummy methods to the exchange of operator types included in the source code. The generator tool will be included in a learning environment that aims to assist teachers in the creation of programming exercises and to help students in their resolution.

The methanol extract of Leptospira interrogans serovar canicola was purified by precipitation with acetone or acetone and chloroform. The antigenicity of the antigen was not altered by heating or treatment with pepsin and pronase. However the antigenicity was lost when the antigen was treated with periodic acid. Chemical analysis revealed the presence of 40% carbohydrate (22% methylpentose, 28%; hexoses),4% protein, 20% lipid and 2,7% phosphate. The complement fixation test with sera from patients with leptospirosis agreed with the microscopic agglutination reaction.

The mineral content (phosphorous (P), potassium (K), sodium (Na), calcium (Ca), magnesium (Mg), iron (Fe), manganese (Mn), zinc (Zn) and copper (Cu)) of eight ready-to-eat baby leaf vegetables was determined. The samples were subjected to microwave-assisted digestion and the minerals were quantified by High-Resolution Continuum Source Atomic Absorption Spectrometry (HR-CS-AAS) with flame and electrothermal atomisation. The methods were optimised and validated producing low LOQs, good repeatability and linearity, and recoveries, ranging from 91% to 110% for the minerals analysed. Phosphorous was determined by a standard colorimetric method. The accuracy of the method was checked by analysing a certified reference material; results were in agreement with the quantified value. The samples had a high content of potassium and calcium, but the principal mineral was iron. The mineral content was stable during storage and baby leaf vegetables could represent a good source of minerals in a balanced diet. A linear discriminant analysis was performed to compare the mineral profile obtained and showed, as expected, that the mineral content was similar between samples from the same family. The Linear Discriminant Analysis was able to discriminate different samples based on their mineral profile.

The yeast Saccharomyces cerevisiae is a useful model organism for studying lead (Pb) toxicity. Yeast cells of a laboratory S. cerevisiae strain (WT strain) were incubated with Pb concentrations up to 1,000 μmol/l for 3 h. Cells exposed to Pb lost proliferation capacity without damage to the cell membrane, and they accumulated intracellular superoxide anion (O2 .−) and hydrogen peroxide (H2O2). The involvement of the mitochondrial electron transport chain (ETC) in the generation of reactive oxygen species (ROS) induced by Pb was evaluated. For this purpose, an isogenic derivative ρ0 strain, lacking mitochondrial DNA, was used. The ρ0 strain, without respiratory competence, displayed a lower intracellular ROS accumulation and a higher resistance to Pb compared to the WT strain. The kinetic study of ROS generation in yeast cells exposed to Pb showed that the production of O2 .− precedes the accumulation of H2O2, which is compatible with the leakage of electrons from the mitochondrial ETC. Yeast cells exposed to Pb displayed mutations at the mitochondrial DNA level. This is most likely a consequence of oxidative stress. In conclusion, mitochondria are an important source of Pb-induced ROS and, simultaneously, one of the targets of its toxicity.

In this paper, we present a deterministic approach to tsunami hazard assessment for the city and harbour of Sines, Portugal, one of the test sites of project ASTARTE (Assessment, STrategy And Risk Reduction for Tsunamis in Europe). Sines has one of the most important deep-water ports, which has oil-bearing, petrochemical, liquid-bulk, coal, and container terminals. The port and its industrial infrastructures face the ocean southwest towards the main seismogenic sources. This work considers two different seismic zones: the Southwest Iberian Margin and the Gloria Fault. Within these two regions, we selected a total of six scenarios to assess the tsunami impact at the test site. The tsunami simulations are computed using NSWING, a Non-linear Shallow Water model wIth Nested Grids. In this study, the static effect of tides is analysed for three different tidal stages: MLLW (mean lower low water), MSL (mean sea level), and MHHW (mean higher high water). For each scenario, the tsunami hazard is described by maximum values of wave height, flow depth, drawback, maximum inundation area and run-up. Synthetic waveforms are computed at virtual tide gauges at specific locations outside and inside the harbour. The final results describe the impact at the Sines test site considering the single scenarios at mean sea level, the aggregate scenario, and the influence of the tide on the aggregate scenario. The results confirm the composite source of Horseshoe and Marques de Pombal faults as the worst-case scenario, with wave heights of over 10 m, which reach the coast approximately 22 min after the rupture. It dominates the aggregate scenario by about 60 % of the impact area at the test site, considering maximum wave height and maximum flow depth. The HSMPF scenario inundates a total area of 3.5 km2. © Author(s) 2015.

In order to know the importance of chicken as natural reservoir of Campylobacter lari in Iquitos, Peru; samples were obtained by cloacal swabs from 200 chickens and immediately placed into a semisolid enrichment medium; these were streaked on modified Skirrow Agar. The organism was isolated from 21 (10.5%) samples, corresponding 58.8% to biovar I and 41.2% to biovar II (Lior scheme). The results provide evidence that chicken appear to be prominent reservoirs of Campylobacter lari in Iquitos.

Tendo por referência a diretiva 2006/95/CE, o trabalho desenvolvido no contexto da disciplina de Dissertação/Projeto/Estágio do Mestrado de Engenharia de Instrumentação e Metrologia, decorreu nas instalações do IEP (Instituto Electrotécnico Português) e teve como objetivo principal o desenvolvimento de um procedimento de avaliação dos efeitos fotobiológicos no olho e pele provocados por fontes de emissão contínua (LED), doravante designado método alternativo ao de referência. Os dois métodos, alternativo e de referência, utilizam respectivamente um foto-radiómetro multicanal e um espetro-radiómetro. O procedimento desenvolvido (método alternativo) de acordo com a norma EN/IEC62471) consiste na aquisição dos valores de irradiância com recurso a um foto-radiómetro e posterior determinação dos valores da radiância, com os quais se faz a avaliação dos efeitos fotobiológicos, para fontes de luz LED (Light Emitting Diode) ou GLS (General Lighting Service). A consulta detalhada da norma EN/IEC62471 e a pesquisa sobre os conceitos, definições, equipamentos e metodologias relacionadas com o tema em causa, constituiu o primeiro passo deste projecto. Com recurso aos dois equipamentos, uma fonte de luz LED (módulo de 12 lâmpadas LED) é avaliada em relação aos perigos (ou riscos) actínico UV e UV-A, ao perigo da luz azul e ainda o perigo térmico na retina e térmico na pele, permitindo fazer uma análise comparativa dos resultados. O método alternativo revelou-se bastante flexível e eficaz, proporcionando bons resultados em termos da irradiância e radiância dos referidos efeitos fotobiológicos. A comparação destes resultados com os valores limites de exposição mencionados na norma EN/IEC6247 permitiu afirmar que a fonte de luz LED avaliada não representa perigo fotobiológico para a saúde humana e classifica-se no grupo de risco “isento”. Uma vez cumpridos os objectivos, entendeu-se que seria uma mais-valia para o trabalho já realizado, estudar outro caso prático. Sendo assim, fez-se a avaliação da radiação de apenas um dos LED´s que constituíam a fonte usada nos ensaios anteriores, com o espetro-radiómetro (método de referência) e com uma distância de 200 mm entre a fonte e o medidor. Neste caso verificaram-se diferenças significativas nas quantidades obtidas quando comparadas com os valores normativos. Concluiu-se que o efeito fotobiológico da luz azul insere-se no grupo de “isento”, sem perigo para a saúde. Contudo, o efeito térmico da retina apresenta um aumento considerável da quantidade de radiância, embora dentro do grupo de risco “isento”. Esta classificação de grupos de risco. Face aos resultados obtidos, pode confirmar-se que as lâmpadas LED apresentam segurança fotobiológica, atendendo aos baixos valores de irradiância e radiância dos efeitos fotobiológicos estudados. Pode ainda afirmar-se que a utilização do foto-radiómetro em alternativa ao espetro-radiómetro se revela mais eficaz do ponto de vista de metodologia prática. Este trabalho demonstra a robustez desses dois equipamentos de avaliação dos efeitos fotobiológicos, e procura estabelecer uma linha de orientação para a prevenção dos efeitos adversos na pele e olhos de todos os seres humanos sujeitos à radiação ótica artificial. Quanto às incertezas de medições, em relação ao processo de medição com foto-radiómetro, a sua estimação não se realizou, devido a não rastreabilidade entre as medições indicadas pelo fabricante, no certificado de calibração e as medidas realizadas por outras entidades. Contudo, é propõe-se a sua realização em trabalhos futuros dentro desse âmbito. As incertezas dos resultados de medições com espetro-radiómetro foram parcialmente estimadas. Atendendo às potencialidades do sistema de medição, propõe-se como trabalho futuro, a aplicação da norma IEC62478, que faz parte da aplicação da norma EN/IEC62471 na avaliação do efeito da luz azul, com base na determinação da temperatura de cor correlacionada (CCT) de lâmpadas ou sistemas de lâmpadas incluindo luminárias. Os valores de irradiância e radiância adquiridos nos processos de avaliação, tanto com foto-radiómetro como espectro-radiómetro foram gravados em ficheiro Excel para um CD e anexados a este trabalho.

Apart from cryptococcosis and histoplasmosis, which are mycoses contained by T cell-mediated mechanisms of host defense, fungemia is rarely found in AIDS patients. The frequency of fungemia due to Candida spp. has been reported to be as low as 1 %. We report a non-neutropenic AIDS patient who presented a candidemia which probably arose from her gastrointestinal tract.

We present a case of prenatal diagnosis of congenital rubella. After birth, in addition to traditional serologic and clinical examinations to confirm the infection, we could identify the virus in the "first fluid aspirated from the oropharynx of the newborn", using polimerase chain reaction (PCR). We propose that this first oropharynx fluid (collected routinely immediately after birth) could be used as a source for identification of various congenital infection agents, which may not always be easily identified by current methods

Among the Cyanoprokaryota, the genera Synechocystis and Synechococcus have rarely been studied with respect to potential toxicity. This is particularly true with marine environments where studies about the toxicity of cyanobacteria are restricted to filamentous forms at the warmer temperate and tropical regions and also to filamentous forms at cold seas such as the Baltic Sea. In this study, we describe the effects of cyanobacterial strains of the Synechocystis and Synechococcus genera isolated from the marine coast of Portugal, on marine invertebrates. Crude and partially purified extracts at a concentration of 100 mg/ml of freeze-dried material of the marine strains were tested for acute toxicity in nauplii of the brine shrimp Artemia salina, in the rotifer Brachionus plicatillis and in embryos of the sea urchin Paracentrotus lividus and the mussel Mytilus galloprovincialis. The cyanobacterial extracts, especially the crude extract, had an impact on A. salina nauplii. No significant toxic effects were registered against the rotifer. A negative impact of all strains was recorded on the embryonic development of the sea urchin, with toxic effects resulting in an inhibition of embryogenesis or development of smaller larvae. To the mussel embryos, the effects of cyanobacterial extracts resulted in a complete inhibition of embryogenesis. The results of all assays indicate that Synechocystis and Synechococcus marine strains contained toxic compounds to marine invertebrates.

We report the isolation of Fonsecaea pedrosoi from thorns of the plant Mimosa pudica L. at the place of infection identified by one of our patients. Clinical diagnosis of chromoblastomycosis was established by direct microscopic examination and cultures from the patient's lesion. The same species was isolated from the patient and from the plant. Scanning electron microscopy of the surface of the thorns showed the characteristic conidial arrangement of F. pedrosoi. These data indicate that M. pudica could be a natural source of infection for the fungus F. pedrosoi.
