Circadian growth rhythm of the juvenile sporophyte of the brown alga Undaria pirznatifida was measured with the computer-aided image analysis system in constant florescent white light under constant temperature (10 degrees C). The growth rhythm persisted for 4 d in constant light with a free-running period of 25. 6 h. Egg release from filamentous gametophytes pre-cultured in the light - dark regime was evaluated for six consecutive days at fixed time intervals in constant white light and 12 h light per day. Egg release rhythm persisted for 3 d in both regimes, indicating the endogenous nature. Temporal scale of egg release and gametogenesis in 18, 16, 12 and 8 h light per day were evaluated respectively using vegetatively propagated filamentous gametophytes. Egg release occurred 2 h after the onset of dark phase and peaked at midnight. Evaluation of the rates of oogonium formation, egg release or fertilization revealed no significant differences in four light-dark regimes, indicating; the great plasticity of sexual reproduction. No photoperiodic effect in gametogenesis in terms of oogonium formation and egg release was found, but fertilization in short days was significantly higher than in long days. Results of this investigation further confirmed the general occurrence of circadian rhythms in inter-tidal seaweed species.

Field-collected tetrasporophytes of Palmaria palmata were tumbled in 300-L outdoor tanks from January to August at ambient daylength or in a constant short-day (SD) regime (8 h light per day), both at 10 or 15 degrees C. Tetrasporangia were massively induced after 2.5 months under SD conditions at 10 degrees C and completely lacking at 15 degrees C, both under SD or ambient daylength conditions, with a few tetrasporangia present at 10 degrees C and ambient daylength. Elongation rates of tagged tetrasporophytic thalli peaked from March to April in all four conditions, when the biomass densities in the outdoor tanks were close to 2.5 kg fresh weight m(-2). Under all four conditions, juvenile proliferations started to appear in June from the margins of the old fronds, and attained approximately 1 cm in length by the end of July. Approximately 80% of the tetraspores were released during the first three dark phases in a light/dark regime, and the remaining 20% during the light phases. A minimum of 10 min darkness was observed to trigger spore release. White light inhibited tetraspore release, while a similar number of spores were released in continuous red light or in the light/dark regime, although with no significant differences of spore release during subjective days and nights. Sporelings were successfully derived from the released tetraspores for mass propagation of the male gametophyte in 2000-L outdoor tanks in a greenhouse. Mass production of male gametophytic sporelings of P. palmata was completed two times by SD induction of tetrasporangia at 10 degrees C, release of spores in darkness and culturing the sporelings until they were ready to be propagated vegetatively in greenhouse tanks. One experiment lasted from January to October 2001, with spore release in June, and the second from September to April 2003, with spore release in January. 以红藻中的角叉菜(Chondrus ocellatus)、龙须菜(Gracilaria lemaneiformis)、真江蓠(Gracilaria asiatica)和海膜(Halymenia sinensis)为材料,观察了红藻幼苗的早期发育,通过组织培养构建无性繁殖系,并初步探讨了红藻无性繁殖系的发育及分化机理,为海藻无性繁殖系的获得提供理论基础和依据。 在室内进行了角叉菜的四分孢子和果孢子的细胞培养,并观察其早期发育过程。结果表明,四分孢子和果孢子的早期发育是相似的,可主要分为三个阶段:早期分裂阶段、盘状体阶段和直立体形成阶段。 切段组织培养表明,红藻无性繁殖系的获得主要通过两种途径:一种为切段组织经过培养可直接形成再生植株;另一种为切段组织经培养脱分化,诱导出愈伤组织或类愈伤组织,愈伤组织或类愈伤组织再分化,形成盘状体,盘状体继续发育即可形成再生植株。 通过切段组织培养,获得了龙须菜和真江蓠再生植株。在PES培养液中,添加6-BA(0.25 mg/L)对龙须菜切段组织再生新枝的诱导有促进作用,诱导率可高达84.4%,高浓度的IAA或6-BA(4 mg/L)对切段组织有损伤作用。实验证明伤口大小对切段组织再生新枝有影响,且伤口太大对再生新枝的形成不利。龙须菜的切段组织培养过程中表现出极性。 通过组织培养,诱导出海膜类愈伤组织—丝状体。添加植物生长调节类物质(IAA 和 6-BA)对海膜丝状体的诱导无促进作用。海膜切片在消毒海水中培养即可获得丝状体。海膜丝状体的外植体来源主要有两个:一是健康的藻体; 另一是盘状体,其丝状体的诱导率分别为80% 和90%。筛选出了丝状体适宜的生长条件:13-23℃,10-15 μE•m−2• s−1,12h:12h(Light: Dark),其中18℃为最佳生长温度。高的光照强度(50 μE•m−2•s−1)对丝状体产生损伤作用。 红藻无性繁殖系的发育途径,根据其获得途径也有两种情况。经过切段组织培养直接获得的无性繁殖系,是切段切口处已分化的细胞表现其全能性,恢复分裂能力,再生新枝,切段表现出极性。极性的上端产生新枝,下端形成固着器,成为一个完整的植株。经过愈伤组织和类愈伤组织获得的无性繁殖系,是切口处已分化的细胞表现其全能性,经脱分化形成愈伤组织和类愈伤组织,附着在基质上后,再分化形成盘状体,按照孢子的早期发育途径,形成直立体幼苗。 通过SDS-聚丙烯酰胺凝胶电泳分析,对龙须菜正常藻体及无性繁殖系发育过程中蛋白的表达进行检测,检测出表达差异的蛋白质,推测这两个蛋白可能与龙须菜切段组织再生有关,其中30 KD的蛋白仅在再生新枝中表达,150 KD和125 KD的蛋白仅在正常生长的龙须菜和龙须菜切段的极性下端中表达。 根据高等植物中与萌芽、分生组织以及愈伤组织发育相关的基因序列保守区设计了五对特异引物,通过PCR和RT-PCR扩增检测及DNA测序分析,对红藻无性繁殖系早期发育相关的基因序列,进行了初步分析。

Angelman syndrome (AS) is a neurobehavioral disorder associated with mental retardation, absence of language development, characteristic electroencephalography (EEG) abnormalities and epilepsy, happy disposition, movement or balance disorders, and autistic behaviors. The molecular defects underlying AS are heterogeneous, including large maternal deletions of chromosome 15q11-q13 (70%), paternal uniparental disomy (UPD) of chromosome 15 (5%), imprinting mutations (rare), and mutations in the E6-AP ubiquitin ligase gene UBE3A (15%). Although patients with UBE3A mutations have a wide spectrum of neurological phenotypes, their features are usually milder than AS patients with deletions of 15q11-q13. Using a chromosomal engineering strategy, we generated mutant mice with a 1.6-Mb chromosomal deletion from Ube3a to Gabrb3, which inactivated the Ube3a and Gabrb3 genes and deleted the Atp10a gene. Homozygous deletion mutant mice died in the perinatal period due to a cleft palate resulting from the null mutation in Gabrb3 gene. Mice with a maternal deletion (m-/p+) were viable and did not have any obvious developmental defects. Expression analysis of the maternal and paternal deletion mice confirmed that the Ube3a gene is maternally expressed in brain, and showed that the Atp10a and Gabrb3 genes are biallelically expressed in all brain sub-regions studied. Maternal (m-/p+), but not paternal (m+/p-), deletion mice had increased spontaneous seizure activity and abnormal EEG. Extensive behavioral analyses revealed significant impairment in motor function, learning and memory tasks, and anxiety-related measures assayed in the light-dark box in maternal deletion but not paternal deletion mice. Ultrasonic vocalization (USV) recording in newborns revealed that maternal deletion pups emitted significantly more USVs than wild-type littermates. The increased USV in maternal deletion mice suggests abnormal signaling behavior between mothers and pups that may reflect abnormal communication behaviors in human AS patients. Thus, mutant mice with a maternal deletion from Ube3a to Gabrb3 provide an AS mouse model that is molecularly more similar to the contiguous gene deletion form of AS in humans than mice with Ube3a mutation alone. These mice will be valuable for future comparative studies to mice with maternal deficiency of Ube3a alone.

Inland waters are of global biogeochemical importance. They receive carbon inputs of ~ 4.8 Pg C/ y of which, 12 % is buried, 18 % transported to the oceans, and 70 % supports aquatic secondary production. However, the mechanisms that determine the fate of organic matter (OM) in these systems are poorly defined. One aspect of this is the formation of organo-mineral complexes in aquatic systems and their potential as a route for OM transport and burial vs. their use as carbon (C) and nitrogen (N) sources within aquatic systems. Organo-mineral particles form by sorption of dissolved OM to freshly eroded mineral surfaces and may contribute to ecosystem-scale particulate OM fluxes. We experimentally tested the availability of mineral-sorbed OM as a C & N source for streamwater microbial assemblages and streambed biofilms. Organo-mineral particles were constructed in vitro by sorption of 13C:15N-labelled amino acids to hydrated kaolin particles, and microbial degradation of these particles compared with equivalent doses of 13C:15N-labelled free amino acids. Experiments were conducted in 120 ml mesocosms over 7 days using biofilms and water sampled from the Oberer Seebach stream (Austria). Each incubation experienced a 16:8 light:dark regime, with metabolism monitored via changes in oxygen concentrations between photoperiods. The relative fate of the organo-mineral particles was quantified by tracing the mineralization of the 13C and 15N labels and their incorporation into microbial biomass. Here we present the initial results of 13C-label mineralization, incorporation and retention within dissolved organic carbon pool. The results indicate that 514 (± 219) μmol/ mmol of the 13:15N labeled free amino acids were mineralized over the 7-day incubations. By contrast, 186 (± 97) μmol/ mmol of the mineral-sorbed amino acids were mineralized over a similar period. Thus, organo-mineral complexation reduced amino acid mineralization by ~ 60 %, with no differences observed between the streamwater and biofilm assemblages. Throughout the incubations, biofilms were observed to leach dissolved organic carbon (DOC). However, within the streamwater assemblage the presence of both organo-mineral particles and kaolin particles was associated with significant DOC removal (-1.7 % and -7.5 % respectively). Consequently, the study demonstrates that mineral and organo-mineral particles can limit the availability of DOC in aquatic systems, providing nucleation sites for flocculation and fresh mineral surfaces, which facilitate OM-sorption. The formation of these organo-mineral particles subsequently restricts microbial OM degradation, potentially altering the transport and facilitating the burial of OM within streams.

La présente thèse avait pour mandat d’examiner la question suivante : quels sont les indices visuels utilisés pour catégoriser le sexe d’un visage et comment sont-ils traités par le cerveau humain? La plupart des études examinant l’importance de certaines régions faciales pour la catégorisation du sexe des visages présentaient des limites quant à leur validité externe. L’article 1 visait à investiguer l’utilisation des indices achromatiques et chromatiques (sur l’axe xy) dans un contexte de plus grande validité externe. Pour ce faire, nous avons utilisé la technique Bubbles afin d’échantillonner l’espace xy de visages en couleurs n’ayant subi aucune transformation. Afin d’éviter les problèmes liés à la grande répétition des mêmes visages, nous avons utilisé un grand nombre de visages (c.-à-d. 300 visages caucasiens d’hommes et de femmes) et chaque visage n’a été présenté qu’une seule fois à chacun des 30 participants. Les résultats indiquent que la région des yeux et des sourcils—probablement dans le canal blanc-noir—est l’indice le plus important pour discriminer correctement le genre des visages; et que la région de la bouche—probablement dans le canal rouge-vert—est l’indice le plus important pour discriminer rapidement et correctement le genre des visages. Plusieurs études suggèrent qu’un indice facial que nous n’avons pas étudié dans l’article 1—les distances interattributs—est crucial à la catégorisation du sexe. L’étude de Taschereau et al. (2010) présente toutefois des données allant à l’encontre de cette hypothèse : les performances d’identification des visages étaient beaucoup plus faibles lorsque seules les distances interattributs réalistes étaient disponibles que lorsque toutes les autres informations faciales à l’exception des distances interattributs réalistes étaient disponibles. Quoi qu’il en soit, il est possible que la faible performance observée dans la condition où seules les distances interattributs étaient disponibles soit explicable non par une incapacité d’utiliser ces indices efficacement, mais plutôt par le peu d’information contenue dans ces indices. L’article 2 avait donc comme objectif principal d’évaluer l’efficacité—une mesure de performance qui compense pour la faiblesse de l’information disponible—des distances interattributs réalistes pour la catégorisation du sexe des visages chez 60 participants. Afin de maximiser la validité externe, les distances interattributs manipulées respectaient la distribution et la matrice de covariance observées dans un large échantillon de visages (N=515). Les résultats indiquent que les efficacités associées aux visages ne possédant que de l’information au niveau des distances interattributs sont un ordre de magnitude plus faibles que celles associées aux visages possédant toute l’information que possèdent normalement les visages sauf les distances interattributs et donnent le coup de grâce à l’hypothèse selon laquelle les distances interattributs seraient cuciale à la discrimination du sexe des visages. L’article 3 avait pour objectif principal de tester l’hypothèse formulée à la fin de l’article 1 suivant laquelle l’information chromatique dans la région de la bouche serait extraite très rapidement par le système visuel lors de la discrimination du sexe. Cent douze participants ont chacun complété 900 essais d’une tâche de discrimination du genre pendant laquelle l’information achromatique et chromatique des visages était échantillonnée spatiotemporellement avec la technique Bubbles. Les résultats d’une analyse présentée en Discussion seulement confirme l’utilisation rapide de l’information chromatique dans la région de la bouche. De plus, l’utilisation d’un échantillonnage spatiotemporel nous a permis de faire des analyses temps-fréquences desquelles a découlé une découverte intéressante quant aux mécanismes d’encodage des informations spatiales dans le temps. Il semblerait que l’information achromatique et chromatique à l’intérieur d’une même région faciale est échantillonnée à la même fréquence par le cerveau alors que les différentes parties du visage sont échantillonnées à des fréquences différentes (entre 6 et 10 Hz). Ce code fréquentiel est compatible avec certaines évidences électrophysiologiques récentes qui suggèrent que les parties de visages sont « multiplexées » par la fréquence d’oscillations transitoires synchronisées dans le cerveau.

Rationale: Anticipatory nausea (AN) is a poorly controlled side-effect experienced by chemotherapy patients. Currently, pharmacotherapy is restricted to benzodiazepine anxiolytics, which have limited efficacy, significant sedative effects, and induce dependency. The non-psychoactive phytocannabinoid, cannabidiolic acid (CBDA), has shown considerable efficacy in pre-clinical AN models, however determination of its neuromotor tolerability profile is crucial to justify clinical investigation. Provisional evidence for appetite-stimulating properties also requires detailed investigation. Objectives: To assess the tolerability of CBDA in locomotor activity, motor coordination and muscular strength tests, and additionally for ability to modulate feeding behaviours. Methods: Male Lister hooded rats administered CBDA (0.05-5 mg/kg; p.o.) were assessed in habituated open field (for locomotor activity), static beam and grip strength tests. A further study investigated whether these CBDA doses modulated normal feeding behaviour. Finally, evidence of anxiolytic-like effects in the habituated open field prompted testing of 5 mg/kg CBDA for anxiolytic-like activity in unhabituated open field, light/dark box and novelty-supressed feeding (NSF) tests. Results: CBDA had no adverse effects upon performance in any neuromotor tolerability test, however anxiolytic-like behaviour was observed in the habituated open field. Normal feeding behaviours were unaffected by any dose. CBDA (5 mg/kg) abolished the increased feeding latency in the NSF test induced by the 5-HT1AR antagonist, WAY-100,635, indicative of anxiolytic-like effects, but had no effect on anxiety-like behaviour in the novel open field or light/dark box. Conclusions: CBDA is very well tolerated and devoid of the sedative side-effect profile of benzodiazepines, justifying its clinical investigation as a novel AN treatment.

Thyroid hormone levels are implicated in mood disorders in the adult human but the mechanisms remain unclear partly because, in rodent models, more attention has been paid to the consequences of perinatal hypo and hyperthyroidism. Thyroid hormones act via the thyroid hormone receptor (TR) alpha and beta isoforms, both of which are expressed in the limbic system. TR's modulate gene expression via both unliganded and liganded actions. Though the thyroid hormone receptor (TR) knockouts and a transgenic TRalpha1 knock-in mouse have provided us valuable insight into behavioral phenotypes such as anxiety and depression, it is not clear if this is because of the loss of unliganded actions or liganded actions of the receptor or due to locomotor deficits. We used a hypothyroid mouse model and supplementation with tri-iodothyronine (T3) or thyroxine (T4) to investigate the consequences of dysthyroid hormone levels on behaviors that denote anxiety. Our data from the open field and the light-dark transition tests suggest that adult onset hypothyroidism in male mice produces a mild anxiogenic effect that is possibly due to unliganded receptor actions. T3 or T4 supplementation reverses this phenotype and euthyroid animals show anxiety that is intermediate between the hypothyroid and thyroid hormone supplemented groups. In addition, T3 but not T4 supplemented animals have lower spine density in the CA1 region of the hippocampus and in the central amygdala suggesting that T3-mediated rescue of the hypothyroid state might be due to lower neuronal excitability in the limbic circuit.

A genomic region neighboring the alpha-synuclein gene, on rat chromosome 4, has been associated with anxiety- and alcohol-related behaviors in different rat strains. In this study, we have investigated potential molecular and physiological links between alpha-synuclein and the behavioral differences observed between Lewis (LEW) and Spontaneously Hypertensive (SHR) inbred rats, a genetic model of anxiety. As expected, LEW rats appeared more fearful than SHR rats in three anxiety models: open field, elevated plus maze and light/dark box. Moreover, LEW rats displayed a higher preference for alcohol and consumed higher quantities of alcohol than SHR rats. alpha-Synuclein mRNA and protein concentrations were higher in the hippocampus, but not the hypothalamus of LEW rats. This result inversely correlated with differences in dopamine turnover in the hippocampus of LEW and SHR rats, supporting the hypothesis that alpha-synuclein is important in the downregulation of dopamine neurotransmission. A novel single nucleotide polymorphism was identified in the 30-untranslated region (3`-UTR) of the alpha-synuclein cDNA between these two rat strains. Plasmid constructs based on the LEW 3`-UTR sequence displayed increased expression of a reporter gene in transiently transfected PC12 cells, in accordance with in-vivo findings, suggesting that this nucleotide exchange might participate in the differential expression of alpha-synuclein between LEW and SHR rats. These results are consistent with a novel role for alpha-synuclein in modulating rat anxiety- like behaviors, possibly through dopaminergic mechanisms. Since the behavioral and genetic differences between these two strains are the product of independent evolutionary histories, the possibility that polymorphisms in the alpha-synuclein gene may be associated with vulnerability to anxiety- related disorders in humans requires further investigation. Aromatherapy uses essential oils (EOs) for several medical purposes, including relaxation. The association between the use of aromas and a decrease in anxiety could be a valuable instrument in managing anxiety in an ever increasing anxiogenic daily life style. Linalool is a monoterpene commonly found as the major volatile component of EOs in several aromatic plant species. Adding to previously reported sedative effects of inhaled linalool, the aim of this study was to investigate the effects of inhaled linalool on anxiety, aggressiveness and social interaction in mice. Additionally, we investigated the effects of inhaled linalool on the acquisition phase of a step-down memory task in mice. Inhaled linalool showed anxiolytic properties in the light/dark test, increased social interaction and decreased aggressive behavior; impaired memory was only seen the higher dose of linalool. These results strengthen the suggestion that inhaling linalool rich essential oils can be useful as a mean to attain relaxation and counteract anxiety. (C) 2009 Elsevier GmbH. All rights reserved.

We conducted a longitudinal study about daily variation of Wistar male rats' behavior in the elevated plus-maze (EPM) evaluated in the 1st, 2nd, 3rd, 6th, 12th, and 18th months of life. Animals were submitted to the plus-maze in 12 sessions at 2-h intervals (n=72, 6 per time point). Spontaneous rest-activity rhythm of four animals was assessed by observation of 24-h videotape records. Time series were analyzed by Cosinor method. Behavioral rates on the six occasions and in light and dark phases were compared by means of two-way ANOVA with repeated measures. Exploratory behavior in EPM was smaller in the light phase and in older animals. Higher values of open and closed arms exploration were observed in the first and third months of the dark phase, and in the first month of the light phase. Adjustment to the 24-h period was significant at all stages for rest-activity data, number of entries in closed arms, and time on center, and for three to five stages for open-arm exploration. In general, 24 h variability was more pronounced in younger animals compared with older ones. The present study showed that: (1) a significant amount of total variability of the behavioral indexes analyzed could be attributed to 24 h variation, (2) light/dark phases differences in EPM exploration were present at all developmental stages, (3) older Wistar rats explored less the EPM and were less active in their home cage compared with younger ones, and (4) behavioral indexes (EPM) decrease was phase related and partially related to a reorganization of rest-activity rhythm. It has been proposed that the ascending dorsal raphe (DR)-serotonergic (5-HT) pathway facilitates conditioned avoidance responses to potential or distal threat, while the DR-periventricular 5-HT pathway inhibits unconditioned flight reactions to proximal danger. Dysfunction on these pathways would be, respectively, related to generalized anxiety (GAD) and panic disorder (PD). To investigate this hypothesis, we microinjected into the rat DR the benzodiazepine inverse receptor agonist FG 7142, the 5-HT1A receptor agonist 8-OH-DPAT or the GABA(A) receptor agonist muscimol. Animals were evaluated in the elevated T-maze (ETM) and light/dark transition test. These models generate defensive responses that have been related to GAD and PD. Experiments were also conducted in the ETM 14 days after the selective lesion of DR serotonergic neurons by 5,7-dihydroxytriptamine (DHT). In all cases, rats were pre-exposed to one of the open arms of the ETM 1 day before testing. The results showed that FG 7142 facilitated inhibitory avoidance, an anxiogenic effect, while impairing one-way escape, an anxiolytic effect. 8-OH-DPAT, muscimol, and 5,7-DHT-induced lesions acted in the opposite direction, impairing inhibitory avoidance while facilitating one-way escape from the open arm. In the light/dark transition, 8-OH-DPAT and muscimol increased the time spent in the lighted compartment, an anxiolytic effect. The data supports the view that distinct DR-5-HT pathways regulate neural mechanisms underlying GAD and PD. (C) 2002 Elsevier B.V. B.V. All rights reserved.
