A via das quinureninas é a principal rota de degradação do aminoácido triptofano. Os metabólitos dessa via, comumente chamados de quinureninas, estão envolvidos em vários processos fisiológicos e patológicos e, recentemente, algumas quinureninas foram relacionadas à fisiopatologia de várias doenças neurodegenerativas. Tendo em vista que dados da literatura são contraditórios no que se refere à geração de espécies reativas a partir de algumas quinureninas e considerando que as concentrações de alguns desses metabólitos estão elevadas em várias doenças neurodegenerativas, este trabalho teve por objetivo investigar os efeitos in vitro de alguns intermediários da via das quinureninas, particularmente a 3-hidroxiquinurenina (3HKyn), a quinurenina (Kyn), o ácido 3-hidroxiantranílico (3HAA), o ácido antranílico (AA) e o ácido quinolínico (QA) sobre alguns parâmetros de estresse oxidativo em córtex cerebral de ratos de 30 dias de idade. Verificamos que a 3HKyn e o 3HAA diminuíram as substâncias reativas ao ácido tiobarbitúrico (TBA-RS) e a quimiluminescência em córtex cerebral de ratos jovens, o que indica um efeito antioxidante desses compostos, ao passo que a Kyn e o AA não alteraram os parâmetros de lipoperoxidação. Por outro lado, o QA aumentou a peroxidação lipídica nesta estrutura cerebral por aumentar significativamente as medidas de TBA-RS e quimiluminescência. Além disso, se pode verificar uma prevenção significativa da oxidação da GSH causada pelo 3HAA, enquanto o QA, na presença de íon ferroso e ácido ascórbico, diminuiu significativamente as concentrações de glutationa reduzida, o que indica que esse efeito seja mediado por radicais hidroxila gerados através da reação de Fenton. Já a 3HKyn, a Kyn e o AA não alteraram significativamente as concentrações de GSH. Também se verificou que a 3HKyn diminuiu a oxidação do diacetato de 2, 7-diclorofluoresceína, além de mostrar a propriedade de seqüestrar radicais peroxila e hidroxila. Por outro lado, o 3HAA somente seqüestrou radicais peroxila, sugerindo que a estrutura orto-aminofenólica é essencial para o composto possuir propriedades antioxidantes. Com o objetivo de verificar se o tempo de exposição à 3HKyn alterava a sua atividade antioxidante, determinamos a reatividade antioxidante total (TAR) e os valores de TBA-RS em células C6 cultivadas de glioma de ratos ao longo de 48 h na ausência (controle) ou presença de 3HKyn. Os resultados demonstraram um aumento da TAR e uma diminuição das TBA-RS pela 3HKyn em tempos curtos de exposição (1-6 horas), sendo que o metabólito não alterou significativamente esses parâmetros em tempos maiores de exposição, sugerindo uma diminuição da capacidade antioxidante da 3HKyn ao longo do tempo. Também verificamos uma diminuição do potencial antioxidante total (TRAP) e da TAR pelo QA em homogeneizado de córtex cerebral Finalmente, foi evidenciado qua a 3HKyn, na concentração de 100 µM, foi capaz de prevenir os efeitos tóxicos causados pelo QA e pelo ácido glutárico (GA). O GA é a principal neurotoxina que se encontra acumulada na acidemia glutárica tipo I. Em resumo, nossos resultados sugerem um efeito antioxidante da 3HKyn e do 3HAA e um efeito pró-oxidante do QA in vitro em córtex cerebral de ratos jovens. As alterações provocadas pelas várias quinureninas foram obtidas nas concentrações de 10, 100 e 500 µM. Embora não saibamos as concentrações dessas substâncias em doenças neurodegenerativas em que eles se acumulam, é possível que, em uma situação in vivo, a produção de substâncias antioxidantes através da rota das quinureninas poderia contrabalançar os efeitos tóxicos causados por outros metabólitos como o QA.

Já está bem estabelecido na literatura que as doenças pulmonares podem causar diretamente alterações na mecânica cardíaca. Estas alterações acarretam prejuízos que invariavelmente induzem o estabelecimento da insuficiência cardíaca. Aproximadamente 30% dos casos de insuficiência cardíaca estão relacionados à falência do ventrículo direito, sendo que esta situação é a principal causa de morbidade e mortalidade pós-transplante cardíaco. As alterações patológicas do ventrículo direito resultantes da disfunção do pulmão são denominadas de Cor pulmonale. Estas manifestações, quando crônicas, se caracterizam por hipertrofia e dilatação do ventrículo direito secundária à hipertensão pulmonar, evoluindo progressivamente para o desenvolvimento de insuficiência cardíaca congestiva. Embora os mecanismos envolvidos no desenvolvimento da insuficiência cardíaca ainda não tenham sido completamente elucidados, vários estudos apontam para a participação das espécies reativas de oxigênio (ERO) nestes processos. A utilização da droga monocrotalina (MCT) para desenvolvimento de Cor pulmonale, é o modelo experimental mais amplamente aplicado para o estudo de hipertensão pulmonar, hipertrofia seletiva de ventrículo direito e insuficiência cardíaca. Apesar da constante evolução no desenvolvimento de fármacos que melhorem tanto a morbidade quanto a mortalidade de portadores de doenças cardiorrespiratórias, a utilização de terapêuticas não farmacológicas como o exercício físico, principalmente em programas de reabilitação, tem demonstrado resultados positivos no tratamento destas doenças. Um dos mecanismos responsáveis por estes benefícios é a modulação do estresse oxidativo O objetivo deste estudo foi avaliar as alterações induzidas por um protocolo de treinamento físico, em parâmetros morfométricos (evolução do peso corporal, congestão hepática e pulmonar, hipertrofia cardíaca), hemodinâmicos (pressão sistólica e diastólica final do ventrículo direito (PSVD e PDFVD) e índices de contratilidade e relaxamento (±dP/dt)) e no estresse oxidativo cardíaco avaliado pela atividade das enzimas antioxidantes superóxido dismutase (SOD), catalase (CAT), glutationa peroxidase (GPx) e glutationa S-transferase (GST), pela quimiluminescência (QL) e pelas espécies reativas ao ácido tiobarbitúrico (TBARS) em ratos tratados com a droga monocrotalina. Foram utilizados ratos Wistar machos pesando ≅ 180g subdivididos em quatro grupos: controle sedentário (CS), monocrotalina sedentário (MS), controle treinado (CT), e monocrotalina treinado (MT). O treinamento físico constituiu-se de corrida em esteira rolante adaptada (5 vezes por semana, 0.9/0.6 Km/h), durante três, quatro ou cinco semanas. A síndrome Cor pulmonale foi induzida por dose única da droga monocrotalina (MCT, 60mg/kg i.p.). Os animais tratados com MCT apresentaram hipertrofia de ventrículo direito, sendo que na terceira semana de tratamento, o exercício físico foi capaz de evitar o desenvolvimento da mesma. O tratamento com MCT promoveu um aumento significativo na PSVD e PDFVD, em todos os tempos de tratamento, sendo que o protocolo de treinamento físico foi capaz de atenuar a PDFVD, na quarta semana de tratamento. Ainda com relação aos resultados hemodinâmicos, os animais tratados com MCT apresentaram aumento significativo na +dP/dt e diminuição significativa -dP/dt do ventrículo direito, nos três tempos de tratamento. Os animais exercitados apresentaram uma diminuição na +dP/dt e um aumento na -dP/dt nestes mesmos períodos experimentais Em conjunto, estes dados indicam que os animais tratados com MCT se apresentavam em um estágio transitório de insuficiência cardíaca compensada. Na quarta e quinta semanas, nos animais tratados com MCT, houve um aumento significativo do dano causado aos lipídios de membrana, medido através da QL em eritrócitos e do TBARS em homogeneizado de tecido cardíaco, sendo este efeito reduzido pelo exercício físico. Com relação aos dados de TBARS, os animais tratados com MCT e exercitados mostraram uma diminuição significativa nos valores quando comparados aos demais grupos, em todos os tempos experimentais. A atividade das enzimas antioxidantes SOD, CAT GPx e GST avaliadas tanto em eritrócitos quanto em homogeneizado de tecido cardíaco, apresentaram respostas tempo e tecido dependentes. Apesar de que tanto o exercício quanto a MCT tenham sido capaz de modular a ação enzimática antioxidante durante o desenvolvimento do protocolo experimental, as alterações mais significativas foram relacionadas a GPx e GST. Em conjunto, nossos resultados indicam que o exercício foi capaz de modular as alterações fisiológicas tradicionalmente induzidas pela administração de MCT e que estas alterações foram dependentes do tempo de tratamento e do tecido avaliado. Possivelmente, os benefícios gerados pelo exercício físico tenham sido induzidos por uma melhora nos danos no leito vascular pulmonar com uma conseqüente diminuição da pós-carga imposta ao ventrículo direito, além de uma adaptação positiva do sistema de defesa antioxidante. Estes resultados corroboram os benefícios da indicação de um programa regular de exercícios físicos tanto para terapêutica de reabilitação quanto para manutenção da qualidade de vida de portadores de hipertensão pulmonar e insuficiência cardíaca direita.

Os polímeros do tipo poli-hidroxialcanoatos (PHAs) são poliésteres bacterianos que apresentam as propriedades de termoplásticos e elastômeros biodegradáveis. A síntese deste polímero em plantas de interesse agroindustrial tem sido vista como uma área promissora dentro da biotecnologia de polímeros para a produção em grande escala com baixos custos. Contudo, esta tarefa requer o aprimoramento de diferentes metodologias bioquímicas e moleculares, além de maximizar os processos de extração destes polímeros biológicos. A produção de PHAs em peroxissomos ou no citoplasma de Saccharomyces cerevisiae, por meio da expressão de uma PHA-sintase bacteriana, pode servir como indicador e modulador do fluxo de carbonos que percorre vias biossintéticas como a síntese de novo de ácidos graxos e a β-oxidação. Esta levedura tem sido usada como eucarioto modelo para manipular as rotas envolvidas na síntese de PHAs do tipo MCL (PHA com um número médio de carbonos), um polímero menos cristalino e com menor ponto de fusão quando comparado ao PHA-SCL (PHA com um número pequeno de carbonos). A enzima PhaG (3-hidroxidecanoil-ACP-CoA transacilase) é responsável pela conexão entre a síntese de ácidos graxos e a produção de PHA-MCL em bactérias do gênero Pseudomonas, em meios de cultura contendo uma fonte de carbono nãorelacionada como gliconato, etanol ou acetato. Para tentar estabelecer esta rota metabólica em S. cerevisiae, o presente trabalho avaliou a coexpressão de PhaGPa e PhaC1Pa (PHA-sintase) de P. aeruginosa para a síntese de PHA-MCL a partir de uma fonte de carbono não-relacionada em leveduras. Contudo, a presença de PhaGPa não alterou a composição ou a quantidade de PHA-MCL em relação à cepa controle contendo apenas PhaC1Pa citoplasmática ou direcionada ao peroxissomo, independentemente da fonte de carbono utilizada (rafinose ou ácido oléico). Este resultado permite sugerir que a ligação entre a síntese de ácidos graxos e a produção de PHA-MCL em S. cerevisiae não foi estabelecida, provavelmente devido à ausência de algum passo enzimático que limita o desvio de substratos da síntese de ácidos graxos para a produção de PHA-MCL em organismos que não são capazes de acumular naturalmente este polímero quando cultivados em fontes de carbono não-relacionadas.A levedura S. cerevisiae tem sido usada como um sistema modelo para estudar a β-oxidação de ácidos graxos insaturados em peroxissomos. A produção de PHA-MCL pela expressão de PhaC1Pa em peroxissomos de cepas selvagens e mutantes nulos de S. cerevisiae para as enzimas auxiliares da β-oxidação (Eci1p, Sps19p e Dci1p), multiplicadas em meio de cultivo contendo um ácido graxo insaturado como fonte de carbono, permitiu monitorar o fluxo de carbonos que percorre as vias dependente de isomerase, redutase e di-isomerase. Desta forma, o presente estudo permitiu avaliar a β- oxidação in vivo dos ácidos graxos linoléico conjugado, 9-cis,11-trans-CLA (ácido rumênico) ou 10-cis,13-cis-nonadecadienóico, para determinar a contribuição das vias alternativas na degradação destes substratos pela utilização de cepas selvagens, mutantes nulos e linhagens contendo um plasmídio multicópia para os genes ECI1 (Δ3- Δ2-enoil-CoA isomerase), SPS19 (2,4-dienoil-CoA redutase) e DCI1 (Δ3,5-Δ2,4-dienoil- CoA isomerase). As linhagens selvagens foram capazes de sintetizar PHA-MCL quando cultivadas em ácido rumênico, mas a atividade da enzima Eci1p foi essencial para a degradação deste CLA, indicando que a via dependente de isomerase é a única rota in vivo necessária para a β-oxidação do ácido rumênico em peroxissomos de S. cerevisiae. A contribuição da enzima di-isomerase (Dci1p) para a degradação do ácido 10- cis,13-cis-nonadecadienóico foi avaliada em cepas selvagens, mutantes nulos dci1Δ e linhagens de S. cerevisiae contendo os plasmídeos multicópia. De acordo com o conteúdo e a quantidade de PHA formado, a β-oxidação de ácidos graxos cisinsaturados em um carbono ímpar é, in vivo, independente da di-isomerase. Embora este resultado possa indicar o mesmo padrão de envolvimento de Dci1p na degradação de ácidos graxos cis-insaturados em um carbono ímpar em mitocôndrias de mamíferos, esta via alternativa deve ser mais bem investigada em eucariotos superiores.

A redução do Cr(VI) para Cr(III) diminui a toxidade deste metal no ambiente uma vez que, o Cr(III) é insolúvel às membranas biológicas. Assim a redução microbiana do Cr(VI) é uma alternativa para reduzir os impactos ambientais causados por este metal, utilizado em diversos processos industriais. O objetivo deste trabalho foi selecionar microrganismos a partir de solo contaminado com cromo, caracterizar sua capacidade de redução do Cr(VI) durante o crescimento celular e purificar parcialmente a enzima cromo redutase do Bacillus sp. ES29, através da precipitação com sulfato de amônio (45-75%), cromatografia de gel filtração (Sephadex G-25) e cromatografia de interação hidrofóbica (Octyl Sepharose). A atividade de redução do Cr(VI) pelos isolados foi quantificada com o reagente de s-difenilcarbazida. No isolamento, foram obtidas 20 bactérias resistentes a cromo(VI). Seis destas foram capazes de reduzir 100 mg L-1 Cr(VI) em 24 horas. Um dos isolados foi identificado, através de testes bioquímicos, como pertencete ao gênero Bacillus, sendo tolerante a 750 mg L-1 Cr(VI) e reduzindo mais de 40% do Cr(VI) durante o crescimento celular. Na purificação parcial da enzima foi obtido um fator de purificação de 11,2, aumentando a atividade específica da enzima acima de 11 vezes, porém se faz necessário mais passos de purificações para obtenção desta enzima pura. As bactérias selecionadas e a enzima parcialmente purificada, foram eficientes na redução do Cr(VI) e apresentam potencial para outros estudos, visando a aplicação em processos de biorremediação.

The relationship between lipid peroxidation, antioxidant defense and diabetic osteopenia remains unclear. This study evaluated the relationship between lipid peroxidation index, antioxidant defense parameter and bone metabolism in a premenopausal diabetic model by measurements such as thiobarbituric acid-reactive substances concentration (TBARS) and reduced glutathione (GSH) content in brain homogenates, histomorphometric analysis, biomechanical testing and bone mineral density (BMD). Female Wistar rats with regular estrous cycle were divided into two groups: Group 1: control rats (n = 15) and Group 2: diabetic rats (n = 15). Diabetes mellitus was induced by alloxan and confirmed by glycemia 250 mg/dL. The experimental period understood 1 and 5 after days induction and 45, 75 and 120 days after the installation of diabetes mellitus.The lipid peroxidation index, measured by TBARS concentration, showed a significant increase (p<0.05) in diabetic animals in comparison to animals control. However, the antioxidant parameter, measured by GSH content, was significantly decrease (p<0.05) in diabetic animals. Histomorphometric analysis showed a significant increase (p<0.05) in femoral trabecular separation together with a significant decrease (p<0.05) in trabecular thickness and reduced trabecular bone volume in diabetic rats. Moreover, biomechanical testing and BMD values were significant decrease (p<0.05) in diabetic group. Thus, our results demonstrated that increased lipid peroxidation and altered antioxidant defense could be related to the development of oxidative stress and diabetic osteopenia in premenopausal rats

Schistosomiasis is an ancient disease caused by helminth Schistosoma mansoni and is a public health problem in Brazil. The granulomatous lesion, typical of the disease, associates itself with increase in the oxidative damage through the generation of free radicals. The aim of this work was to evaluate the occurrence of changes in parameters oxidant / antioxidant that are part of the human defense system, and observe whether they would cause oxidative stress in subjects with schistosomiasis. Moreover, correlating with some biochemical and hematological parameters. Two groups were selected for study, consisting of individuals of both sexes, aged between 16 and 30 years. A control group, formed by individuals without schistosomiasis (n = 30) and a test group, formed by individuals with schistosomiasis (n = 30). The evaluation of lipid peroxidation in plasma was performed by determination of malondialdehyde and antioxidant defense by the quantification of reduced glutathione and catalase activity. For the parameters that assess oxidative stress, the results showed a decrease in the content of reduced glutathione and no change in the activity of catalase, with an increase in the value of malondialdehyde. Therefore, the data found suggest the occurrence of oxidative stress in subjects with schistosomiasis. Of the parameters that assess hepatic function, only levels of aspartate aminotransferase have been high, while there was a decrease of bilirubine. There was a significant change in the lipid profile (p <0.5), however with regard to the renal function of patients, there was a decrease in creatinine. The assessment hematological, made through hemogram and the quantification of hemoglobin, shows increase of eosinophils individuals in the group test, which can be related to the presence of the parasite. The amendments suggest the involvement of oxidative stress in the pathophysiology of this disease

OBJECTIVE: The aim of this work was to analyse some oxidative stress parameters in patients of Systemic Lúpus Erythematosus. PATIENTS AND METHODS: Determinations of reduced glutathione content in whole blood were carried out. The activity of superoxide dismutase, gluthatione peroxidase and catalase in erythrocytes and the concentration of reactive substances of acid thiobarbituric in plasma of patients female (n =19) with SLE no activity of disease (Mex-SLEDAI < 2), with average ages of 32 ± 11 years, through the spectrophotometrical methods and from healthy individuals (n =30). Statistical data were analyzed by student t-test, p<0,05. RESULTS: Our data indicated a significant decrease on the activity of catalase and significant increase on the concentration of reactive substances of acid thiobarbituric in patients with SLE comparing with healthy individuals. There was no significant difference in other parameters. CONCLUSION: The results showed that oxidative stress has a role in the pathogenesis of the disease in SLE, even in patients without active disease.

Recently, it has been a increasing interest in the antioxidative role of natural products to aid the endogenous protective biological systems against the deleterious effects of oxygen (ROS) and nitrogen (RNS) reactive species. Many antioxidant compounds, naturally occurring from plant sources. Natural antioxidants can protect and prevent the human body from oxidative stress and retard the progress of many diseases in which free radical are involved. Several plants used in the folk medicine to treat certain disorders that are accompanied by inflammation and other pharmacological properties have been proved their attributed properties, such antioxidant activity. Turnera ulmifolia Linn. var. elegans (Turneraceae), frequently employed by population as a medicinal plant, demonstrated antioxidant activity by in vitro and in vivo assays, using its leaf hydroethanolic extract (10%) he in vitro DPPH radical-scanvenging activity showed a strong antioxidant activity (86.57% ± 0.14), similar to Carduus marianus and catequine effects. For the in vivo assays, adult female Wistar rats (n=48) with carbon tetrachloride hepatic injury induced (2,5mL/kg i.p.) were used, Six groups or rats were uses (n=8) [G1 = control (1,25 mL/kg i.p. vehicle); G2 = CCl4 (2,5 mL/kg i.p.); G3 = CCl4 + extract 7 days (500 mg/kg p.o.); G4 = CCl4 + Legalon® 7 days (50 mg/kg p.o.), G5 = CCl4 + extract 21 days (500 mg/kg p.o.) e G6 = CCl4 + Legalon® 21 days (50 mg/kg p.o.)]. The hepatic oxidative injury was evaluated through biochemical parameters [alanine amino transferase (ALT), aspartate amino transferase (AST)] histopathological study, while thiobarbituric acid reactive products (TBAR), glutathione (GSH), catalase (CAT), superoxide dismutase (SOD), and glutathione peroxidase (GPx) levels were used to evaluate proantioxidant parameters. The plant extract tested was found effective as hepatoprotective as evidenced by a decreasing in the ALT and AST activities (p<0.001) and TBAR (plasma, p<0.001 and liver, p<0.001). Levels of GSH (blood, p<0.001 and liver, p<0.001) and antioxidant enzymes [CAT erythrocyte (p<0.05) and hepatic (p<0.01); SOD erythrocyte (p<0.001) and hepatic (p<0.001); GPx erythrocyte (p<0.001) and hepatic (p<0.001)] were also significantly increased. Histopathological changes induced by CCl4 were significantly reduced by the extract treatment. The data obtained were comparable to that of Legalon®, a reference hepatoprotective drug. The results showed that T. ulmifolia leaf extract protects against CCl4 induced oxidative damage. Therefore, this effect must be associated to its antioxidant activity, attributed to the phenolic compounds, present in these extract, which can act as free radical scavengers

Bone is a dynamic tissue that is in constant process of remodeling in response to mechanical stress and hormonal changes. This study aimed to understand the relationship between the biochemical changes, which women in the menopausal transition are subject to, and how the use of an alternative therapy with lipoic acid (LA) could influence these changes. The study of double-blind, was carried out in perimenopausal women that underwent a three month treatment with 600 mg of AL compared with another group that received placebo during the same period. This study showed that women had a waist circunference and body mass index above the values recommended by WHO (WC ≥ 80 cm; BMI > 25kg/m2). Associated with this, these women had increased concentrations of total cholesterol and triglycerides, and borderline LDL (Total Cholesterol > 200mg/dL; Triglycerides > 150mg/dL; LDL >130mg/dL). These changes were not affected by treatment with AL. There were no shifts in liver profile (ALT, AST and GGT), kidney profile (urea, creatinine, total protein and albumin), mineral profile (Total Calcium, Ionized Calcium, Phosphorus and Magnesium) as well in bone markers (osteocalcin, Total Alkaline Phosphatase and Tartrate Resistant Acid Phosphatase) after treatment with LA. The results of the oxidative profile showed that treatment with LA decreased GPx activity (p < 0,01), while for the TBARS, GSH and SOD activity there were no differences. With regard to SOD, this enzyme will submit to be high in the placebo group after 3 months of study (p<0,05). The expression of RANKL mRNA was reduced (p < 0,05) and of RANK increased (p <0.001), after treatment with LA, while the expression of IL-6 and TNF-ɑ genes were no changed. We conclude that women already in the perimenopause stage have changes in lipid profile and body composition that could induce shifts in oxidative and bone metabolism. However, LA treatment has provided an effective effect in the oxidative and bone profile since the earliest markers such as GPx activity and mRNA expression of RANKL, respectively, were reduced associated with no change in SOD activity. These results suggest a beneficial and protective effect of LA, indicating it potential as an alternative treatment to help the to prevent the complications associated with estrogen deficiency

The congenital facial clefts are characterized by incomplete formation of the structures that separate the oral and nasal cavity. It is known that several environmental and genetic factors are involved in its development, among these, polymorphisms associated with folic acid metabolism have been investigated. In this sense, the objective was to observe the frequency of polymorphisms C677T and A1298C methylenetetrahydrofolate reductase gene (MTHFR), methionine synthase A2756G of (MTR), A66G of methionine synthase reductase (MTRR) A80G and the reduced folate carrier (RFC1) in patients with non-syndromic oral clefts, trying to match them with their development. Methods: We studied 140 patients with non-syndromic oral clefts and their mothers and 175 control subjects with their mothers, who underwent a questionnaire to obtain family information. Were collecting blood for DNA extraction from patients and their mothers to identify the genotypes of both by PCRRFLP, in addition to carrying out the determination of glucose, AST, ALT and serum creatinine, folic acid and vitamin B12 Serum and plasma homocysteine, and the hemogram. Results: Most patients have cleft lip and palate (55.8%), followed by isolated cleft palate (24.2%) and cleft lip (20%). Regarding gender, 62% of patients were male and 48% female and, after subdivision of the type of screwdriver according to sex was found a prevalence of males in the cracks of the type lip and palate (69 %) and lip (69.2%) and in the case of cleft palate was a female predominance (59%). The average concentration of serum folate in the group of mothers of cleft patients was significantly lower (13.8 ± 2.4 ng / mL) compared with the group of mothers of control subjects (18.8 ± 3.4 ng / mL) This was also observed for the group of cleft children as compared to controls, the dosage of folic acid had a significant difference with values of 15.6 ± 0.6 (ng / mL) and 17.9 ± 0.6 (ng / mL), respectively. For the biochemical measurements of glucose, AST, ALT and creatinine were not statistically different, nor was observed for haematological parameters performed. In assessing the frequency of polymorphisms C677T and A1298C MTHFR, A2756G MTR, MTRR A66G and A80G of the RFC1 there was no statistically significant difference in genotype distribution between cases and controls both for mothers and in the cleft. Conclusion: Although not observed association of polymorphisms with the development of cracks, the decrease in serum folate in the group of cleft patients and their mothers may reflect a disturbance in the metabolism of this metabolite, necessitating further studies such as studies methylation and expression to further elucidate the involvement of folate in the development of oral clefts

The aim of this study was to determine the effects of the use of rosuvastatin in patients with atherosclerosis, in relation to blood parameters of selenium and selenoproteins, and also observe possible changes in gene expression of selenoproteins in these patients. The sample consisted of 27 adult and elderly patients with a clinical diagnosis of coronary artery disease undergoing angioplasty, treated at Natal Hospital Center hospital, Natal, RN. Patients were treated with rosuvastatin 10 mg/day during four months. Anthropometric variables such as body mass index (BMI) and Waist circumference (WC) were measured before and after treatment, as well as lipid profile, blood glucose and liver enzymes (AST and ALT). The diet of the patients was also analyzed using 24-hour diet recall. We analyzed the concentrations of selenium in plasma and erythrocytes, and also the activity of Glutathione Peroxidase and gene expression by Real Time PCR of selenoproteins GPx1, SelP1 and SelN1. Patients had mean age of 61.0 ± 9.4 years, 59.3% were men and 40.7% were women. After four months of treatment there was significant reduction of CA and, according to BMI, most were overweight. The intake of macronutrients, cholesterol, polyunsaturated fatty acids, monounsaturated and saturated was adequate, but the energy and fiber intake was below the recommendations. Regarding the selenium intake was observed a high prevalence of inadequacy. As expected, after treatment with rosuvastatin, a significant reduction in total cholesterol, LDL and glucose, which was not observed for HDL. Selenium concentrations in plasma and erythrocytes showed no changes, keeping within the established cutoffs. We observed a significant increase in GPx enzyme activity and mRNA expression of GPX1 and SEPN1, but not for gene SEPP1. Thus, it was found that treatment with rosuvastatin did not reduce the expression of selenoproteins. More studies are needed to clarify the effects of rosuvastatin on gene expression of selenoproteins in patients with atherosclerosis

Spondias sp. (Anacardiaceae), popularly known as cajá-umbu, is an endemic plant from Northeastern Brazil, where their leaves are widely used in folk medicine to treat inflammatory processes, while their fruits have a great agro industrial potential. This study was designed to evaluate hepatoprotective, antinociceptive, antioxidant, antimicrobial and anti-inflammatory properties, as well as the acute toxicity and repeated dose 28, using a methanolic extract (MES), a fraction rich in flavonoids (FRF) and a precipitate from Spondias sp.leaves. The antioxidant activity of them was valued to evaluate their free radical scavenger capacity by DPPH test, whereas MES and FRF were used to evaluate while the preventive action on carbon tetrachloride (CCl4)-induced hepatotoxicity. Seven groups (n=5) of female Wistar rats were used as follows: control group, CCl4-intoxicated group treated with EMS (500 mg/kg) for 7 days, three CCl4-intoxicated groups treated with FRF (25, 50 and 75 mg/kg) for 7 days and the CCl4-intoxicated group treated with Legalon ® (silimarina; (phytotherapeutic reference) (50 mg/kg; 7 days). MES and FRF showed a protective action against liver injury induced by CCl4, being observed a significant reduction of serum enzyme activity marker of liver damage (alanine transaminase and aspartate transaminase). On the other hand, the lipid peroxidation (SRAT) decrease, as well as the increase of glutathione content and enzyme activity of antioxidant defense system (SOD, CAT, GPx) toward near normal values indicated the ability of EMS to restore the oxidative imbalance induced by CCl4. The histological analysis confirmed the hepatoprotection, compared to degenerative changes in CCl4-treated group. This hepatoprotetor effect was similar to that shown by Legalon®. The in vitro high antioxidant capacity of extract (93.16 ± 1.00%) showed analogous results to those obtained by Carduus marianus BHT (reference standard). This fact explains the obtained results in vivo. Although no antimicrobial activity was detected, EMS and FRF promoted the antinociceptive effect induced in the second phase by the intraplantar formalin test, evidencing the anti-inflammatory action; confirmed by the carrageenan-induced peritonitis model. The evaluation of the mechanical allodynia (CFA a 80%) demonstrated the involvement of the Spondias sp. chemical composition in the anti-inflammatory activity toward the acute processes. The acute exposure and repeated dose during 28 days did not produce significant changes in the parameters that evaluate toxicity. Together the experimental results reveal, that Spondias sp. leaf extracts have a promising potential in pharmaceutical area, and due to its non-toxic condition present efficiency and security

Post-menopause is a period of women s life cycle that is characterized by estrogen depletion and therefore increasing cardiovascular diseases, neurodegenerative disorders, urogenital atrophy, osteoporosis, hot flushes and sexual discomfort incidences. Estrogen is a hormone with comfirmed antioxidant action and its depletion is related to oxidative stress instalation and damaging various important biomolecules. Regular physical activity has been identified as a factor involved in reducing women s post-menopausal complications in addition to improving antioxidant defense by reducing the oxidative damage and consequently improving life s quality in this part of the population. This study aims to evaluate the influence of hypoestrogenism in antioxidant adaptation due to regular exercise, by determining reduced glutathione (GSH) and Thiobarbituric Acid Reactive Substances (SRAT) concentrations and antioxidant enzymes glutathione peroxidase (GPx), Superoxide Dismutase (SOD) and Catalase (CAT) activities in blood, brain and liver of rats. To achieve this goal we used 50 Wistar rats, weighing 180-250g which were divided into two groups, control - GC (25) and ooforectomized - GO (25). Each group was subdivided into five subgroups: Not-trained - S (5), Not-trained Acute Exercise - SEA (5), regular exercise 30 days - E30 (5), regular exercise 60 days - E60 (5) and regular exercise 90 days - E90 (5). Each of the three subgroups exercised regularly was subjected to acute exercise on the eve and the day of sacrifice to collect biological samples of blood, liver and brain and subsequent determination of SRAT concentration, GSH content and antioxidant enzymes GPx, SOD and CAT activities. The results indicated that the sedentary animals acutely exercised presented oxidative stress and regular physical activity led to antioxidant adaptation. In ooforectomized group the antioxidant adaptation seen in control animals showed to be impaired. Unlike the results from blood and liver, in brain there was a shield against oxidative damage originated by the exercise and that hypoestrogenism led to a loss of this natural antioxidant potential. Therefore, hypoestrogenism interferes negatively in antioxidant adaptation due to regular exercise

Studies report that the pathophysiological mechanism of diabetes complications is associated with increased production of Reactive Oxygen Species (ROS)-induced by hyperglycemia and changes in the capacity the antioxidant defense system. In this sense, the aim of this study was to evaluate changes in the capacity of antioxidant defense system, by evaluating antioxidant status, gene expression and polymorphisms in the genes of GPx1, SOD1 and SOD2 in children, adolescents and young adults with type 1 diabetes. We studied 101 individuals with type 1 diabetes (T1D) and 106 normoglycemic individuals (NG) aged between 6 and 20 years. Individuals with type 1 diabetes were evaluated as a whole group and subdivided according to glycemic control in DM1G good glycemic control and DM1P poor glycemic control. Glycemic and metabolic control was evaluate by serum glucose, glycated hemoglobin, triglycerides, total cholesterol and fractions (HDL and LDL). Renal function was assessed by measurement of serum urea and creatinine and albumin-to-creatinine ratio (ACR) in spot urine. Antioxidant status was evaluate by content of reduced glutathione (GSH) in whole blood and the activity of erythrocyte enzymes glutathione peroxidase (GPx) and superoxide dismutase (SOD). We also analyzed gene expression and gene polymorphisms of GPx1 (rs1050450), SOD1 (rs17881135) and SOD2 (rs4880) by the technique of real-time PCR (Taqman®). Most individuals with DM1 (70.3%) had poor glycemic control (glycated hemoglobin> 8%). Regarding the lipid profile, individuals with type 1 diabetes had significantly elevated total cholesterol (p <0.001) and LDL (p <0.000) compared to NG; for triglycerides only DM1NC group showed significant increase compared to NG. There was an increase in serum urea and RAC of individuals with DM1 compared to NG. Nine individuals with type 1 diabetes showed microalbuminuria (ACR> 30 mg / mg). There was a decrease in GSH content (p = 0.006) and increased erythrocyte GPx activity (p <0.001) and SOD (p <0.001) in DM1 group compared to NG. There was no significant difference in the expression of GPx1 (p = 0.305), SOD1 (.365) and SOD2 (0.385) between NG and DM1. The allele and genotype frequencies of the polymorphisms studied showed no statistically significant difference between the groups DM1 and NG. However, the GPx1 polymorphism showed the influence of erythrocyte enzyme activity. There was a decrease in GPx activity in individuals with type 1 diabetes who had a polymorphic variant T (p = 0.012). DM1 patients with the polymorphic variant G (AG + GG) for polymorphism of SOD2 (rs4880) showed an increase in the RAC (p <0.05). The combined data suggest that glucose control seems to be the predominant factor for the emergence of changes in lipid profile, renal function and antioxidant system, but the presence of the polymorphisms studied may partly contribute to the onset of complications
