A espécie Tomato severe rugose virus (ToSRV) é a predominante em áreas de cultivo de pimentão no Estado de São Paulo. Sua ocorrência na cultura é relativamente recente de modo que não existem informações sobre os danos causados nesta cultura. Os objetivos do presente trabalho foram avaliar a produtividade e qualidade dos frutos de pimentão de três cultivares (Magda, Amanda e Rubia R) quando infectadas com o ToSRV. Verificou-se acentuada redução no número de frutos e menor crescimento das plantas, porém, o ToSRV não influenciou significativamente na massa, diâmetro e comprimento dos frutos. Os resultados obtidos até o momento permitem concluir que o ToSRV causa danos em pimentão e que há necessidade de estudos visando resistência ao ToSRV.

Four citrus fruit varieties cultivated in Brazil (two kinds of sweet orange and two kinds of tangerine) were analyzed for physicochemical characteristics contents of total phenolics, total carotenoids and ascorbic acid, and antioxidant activities. The antioxidant activities of aqueous, methanolic, and acetone extracts of the citrus fruit juices were assessed on the basis of their ability to scavenge 1,1-diphenyl-2-picrylhydrazyl (DPPH(center dot)). The cravo tangerine has the highest content of citric acid, while the pera orange is richest in ascorbic acid. The lima orange has the highest total phenolic contents, and the ponkan the highest total carotenoids. The antioxidant activities, expressed as the concentration of antioxidant able to scavenge 50% of the initial DPPH(center dot) (EC(50)), ranged from 139.1+/-27.3 to 182.2+/-28.8 g extract/l for juice of orange varieties and 186.3+/-29.6 to 275.5+/-3.3g extract/l for juice of tangerine citrus varieties. In methanolic extracts the EC(50) ranged from 192.5+/-43.1 to 267.4+/-41.4 g extract/l for orange varieties and from 225.2+/-69.8 to 336.3+/-27.2 g extract/l for tangerine varieties. For EC(50) values of acetone fractions, there were no statistically significant differences between the different varieties. For every citrus fruit in the present study, the radical scavenging capacity was higher in the aqueous than in the methanolic or acetone fractions.

Doxorubicin (DOX) is an efficient chemotherapeutic agent used against several types of tumors; however, its use is limited due to severe cardiotoxicity. Since it is accepted that reactive oxygen species are involved in DOX-induced cardiotoxicity, antioxidant agents have been used to attenuate its side effects. To determine tomato-oleoresin protection against cardiac oxidative DNA damage induced by DOX, we distributed Wistar male rats in control (C), lycopene (L), DOX (D) and DOX+lycopene (DL) groups. They received corn oil (C, D) or tomato-oleoresin (5 mg/kg body wt. day) (L, DL) by gavage for a 7-week period. They also received saline (C, L) or DOX (4 ma/kg body wt.) (D, DL) intraperitoneally at the 3rd, 4th, 5th, and at 6th week. Lycopene absorption was checked by HPLC. Cardiac oxidative DNA damage was evaluated by the alkaline Comet assay using formamidopyrimidine-DNA glycosylase (FPG) and endonuclease III (endo 111). Cardiomyocyte levels of SBs, SBs FPG and SBs Endo III were higher in rats from D when compared to other groups. DNA damage levels in cardiomyocytes from DL were not different when compared to C and L groups. The viability of cardiomyocytes from D or DL was lower than C or L groups (p < 0.01). Lycopene levels (mean +/- S.D. nmol/kg) in saponified hearts were similar between L (47.43 +/- 11.78) and DL (49.85 +/- 16.24) groups. Our results showed: (1) lycopene absorption was confirmed by its cardiac levels; (2) DOX-induced oxidative DNA damage in cardiomyocyte; (3) tomato-oleoresin supplementation protected against cardiomyocyte oxidative DNA damage. Various studies have shown that lycopene, a non-provitamin A carotenoid, exerts antioxidant, antimutagenic and anticarcinogenic activities in different in vitro and in vivo systems. However, the results concerning its chemopreventive potential on rat hepatocarcinogenesis are ambiguous. The aim of the present study was to investigate the antigenotoxic and anticarcinogenic effects of dietary tomato oleoresin adjusted to lycopene concentration at 30, 100 or 300ppm (administered 2 weeks before and during or 8 weeks after carcinogen exposure) on liver of male Wistar rats treated with a single intraperitoneal dose of 20 or 100 mg/kg of diethylnitrosamine (DEN), respectively. The level of DNA damage in liver cells and the development of putative preneoplastic single hepatocytes, minifoci and foci of altered hepatocytes (FHA) positive for glutathione S-transferase (GST-P) were used as endpoints. Significant reduction of DNA damage was detected when the highest lycopene concentration was administered before and during the DEN exposure (20 mg/kg). However, the results also showed that lycopene consumption did not reduce cell proliferation in normal hepatocytes or the growth of initiated hepatocytes into minifoci positive for GST-P during early regenerative response after 70% partial hepatectomy, or the number and area of GST-P positive FHA induced by DEN (100 mg/ kg) at the end of week 10. Taken together, the data suggest a chemopreventive effect of tomato oleoresin against DNA damage induced by DEN but no clear effectiveness in initiating or promoting phases of rat hepatocarcinogenesis. Background: the genus Arachis includes Arachis hypogaea (cultivated peanut) and wild species that are used in peanut breeding or as forage. Molecular markers have been employed in several studies of this genus, but microsatellite markers have only been used in few investigations. Microsatellites are very informative and are useful to assess genetic variability, analyze mating systems and in genetic mapping. The objectives of this study were to develop A. hypogaea microsatellite loci and to evaluate the transferability of these markers to other Arachis species.Results: Thirteen loci were isolated and characterized using 16 accessions of A. hypogaea. The level of variation found in A. hypogaea using microsatellites was higher than with other markers. Cross-transferability of the markers was also high. Sequencing of the fragments amplified using the primer pair AhII from 17 wild Arachis species showed that almost all wild species had similar repeated sequence to the one observed in A. hypogaea. Sequence data suggested that there is no correlation between taxonomic relationship of a wild species to A. hypogaea and the number of repeats found in its microsatellite loci.Conclusion: These results show that microsatellite primer pairs from A. hypogaea have multiple uses. A higher level of variation among A. hypogaea accessions can be detected using microsatellite markers in comparison to other markers, such as RFLP, RAPD and AFLP. The microsatellite primers of A. hypogaea showed a very high rate of transferability to other species of the genus. These primer pairs provide important tools to evaluate the genetic variability and to assess the mating system in Arachis species.

Plant responses against pathogens cause up-and downward shifts in gene expression. To identify differentially expressed genes in a plant-virus interaction, susceptible tomato plants were inoculated with the potyvirus Pepper yellow mosaic virus (PepYMV) and a subtractive library was constructed from inoculated leaves at 72 h after inoculation. Several genes were identified as upregulated, including genes involved in plant defense responses (e. g., pathogenesis-related protein 5), regulation of the cell cycle (e. g., cytokinin-repressed proteins), signal transduction (e. g., CAX-interacting protein 4, SNF1 kinase), transcriptional regulators (e. g., WRKY and SCARECROW transcription factors), stress response proteins (e. g., Hsp90, DNA-J, 20S proteasome alpha subunit B, translationally controlled tumor protein), ubiquitins (e. g., polyubiquitin, ubiquitin activating enzyme 2), among others. Downregulated genes were also identified, which likewise display identity with genes involved in several metabolic pathways. Differential expression of selected genes was validated by macroarray analysis and quantitative real-time polymerase chain reaction. The possible roles played by some of these genes in the viral infection cycle are discussed.

Total and individual carotenoid concentrations were determined by spectro photometry and HPLC, in raw tubers of a sample of 23 accessions of Solanum phureja potatoes taken at random from the world germplasm collection following its stratification on tuber flesh color. Lutein, zeaxanthin, violaxanthin, antheraxanthin and beta-carotene were detected in all accessions and three distinct patterns of carotenoid accumulation were evidenced by cluster analysis. Accessions in group 1 showed the highest concentrations of total carotenoids (1258-1840 mu g 100 g(-1) FW) comprised largely of zeaxanthin (658-1290 mu g 100 g(-1) FW) with very low or no presence of beta-carotene (below 5.4 mu g 100 g(-1) FW). Accessions in group 2 presented moderate total carotenoid concentrations with violaxanthin, antheraxanthin, lutein and zeaxanthin as the major carotenoids. Accessions in group 3 showed low concentrations of total carotenoids (97-262 mu g 100 g(-1) FW) and very low or no zeaxanthin, with lutein and violaxanthin as the predominant carotenoids and relatively high concentrations of beta-carotene(up to 27 mu g 100 g(-1) FW). Five accessions with significant concentrations of zeaxanthin were identified with the accession 703566 showing the highest concentration (1290 p g 100 g(-1) FW). This value is to our knowledge higher than any value previously reported for potatoes, including those achieved through genetic modification. For the 23 S. phureja accessions, total carotenoid concentration was positively and significantly correlated with antheraxanthin and zeaxanthin concentrations, and negatively and significantly correlated with beta-carotene concentration. Anacanthorus penilabiatus n. sp. is described from the serrasalmid fish, Piaractus mesopotamicus (Holmberg, 1887), cultivated in the Centro de Aqüicultura, Universidade Estadual Paulista. The new species is characterized by having a relatively straight copulatory organ with a long lip on the distal margin and a median longitudinal flap, and a copulatory ligament. The large size of the infrapopulations of this species of parasite indicates that it should be considered a potential agent causing losses in aquaculture of the fish host.

Desenvolveu-se um trabalho de pesquisa na fazenda Jordão (município de Araguari, MG), na época de inverno, com o objetivo de verificar o desempenho agronômico de genótipos de tomateiro tipo Santa Cruz. Utilizou-se o delineamento experimental de blocos casualizados, com 14 tratamentos (genótipos) e quatro repetições. A parcela experimental foi constituída por duas fileiras com 12 plantas cada, no espaçamento de 1,00 m entre linhas e 0,60 m entre plantas (duas plantas por cova). As colheitas iniciaram-se em 25/8/96 e encerraram em 19/10/96, perfazendo um total de 17. de forma geral, todos os genótipos apresentaram produtividades elevadas, comparativamente ao rendimento médio nacional, principalmente os genótipos Santa Clara I-5300, Débora Plus, Santa Clara, Santa Clara Importada e Santa Clara III, todos com produtividade superior a 125 t/ha, podendo ser recomendados para plantio na região, no período de inverno. Apenas o genótipo Tom-556 e a cultivar Ângela Gigante I-5100 não tiveram peso médio de frutos superior a 100 g. As cultivares Saladinha, Santa Clara Importada, Concord e Jumbo, tiveram peso médio superior a 130 g, os quais se destacaram, também, em porcentagem de frutos tipo extra AA, juntamente com Santa Clara, Santa Clara I-5300, IAC Santa Clara, enquanto os genótipos Tom-556, Tex-015 e Ângela Gigante I-5100, se destacaram em frutos tipo extra A.

O trabalho foi desenvolvido na fazenda Jordão (município de Araguari, MG), na época do verão (período das águas), com o objetivo de verificar o desempenho agronômico de genótipos de tomateiro tipo Santa Cruz. Utilizou-se o delineamento experimental blocos casualizados, com 16 tratamentos (genótipos) e quatro repetições. A parcela experimental foi constituída por duas fileiras com 12 plantas cada, no espaçamento de 1,00 m entre linhas e 0,55 m entre plantas (1 planta/cova). Efetuaram-se 17 colheitas, sendo a primeira aos 69 dias após o transplante. Vários genótipos apresentaram um bom desempenho agronômico, principalmente Saladinha, Débora Plus, SM-16 e Atlas, podendo ser cultivados no período de verão. Apenas Saladinha e Atlas ultrapassaram 140 g de peso médio, destacando-se também em frutos do tipo extra AA. Observou-se uma correlação significativa e negativa com r = -0,52 e -0,54 na primeira avaliação, e r = -0,55 e -0,45 na segunda avaliação para a produção total e produção comercial, respectivamente, em relação à incidência de geminivírus nos diferentes genótipos. Os híbridos Saladinha e SM-16 apresentaram o menor número de plantas viróticas, enquanto Santa Clara Importada, Santa Clara, Jumbo AG-592 e IAC Santa Clara, apresentaram o maior número.

O trabalho avaliou a eficiência dos isolados (141/3, 233, 233/1, 245, 245/1, 251, 251/2, 251/5 e 257) de Fusarium oxysporum não patogênico ao tomateiro (Lycopersicon esculentum), no controle da murcha vascular causada por Fusarium oxysporum f. sp. lycopersici, raça 2 em plântulas de tomateiro cv. Viradoro. Para verificar o efeito dos isolados de F. oxysporum não patogênicos, o sistema radicular de plântulas de tomateiro, com 30 dias de idade, foi imerso na suspensão de conídios (10(6) ml-1) e as mudas transplantadas para substrato de cultivo. Após 35 dias do transplante foi verificado que esses isolados não foram patogênicos às plantas de tomateiro, nem afetaram o desenvolvimento das mudas. A eficiência dos isolados de Fusarium oxysporum não patogênicos no controle da murcha foi determinada imergindo-se as raízes de mudas de tomateiro em suspensão de conídios (10(6) conídios ml-1) e transplantando-as em substratos previamente infestados com os isolados de F. oxysporum f.sp. lycopersici, raça 2 (10(5) conídios ml-1 de substrato). Transcorridos 35 dias do transplante, foram realizadas as avaliações da severidade na escala de 1=planta sadia a 6=planta morta ou com vasos coloridos e folhas murchas até o ponteiro e altura das mudas. Os isolados de F. oxysporum não patogênicos foram eficientes em reduzir a severidade da doença e em manter normal o seu desenvolvimento. Esses resultados evidenciam a atividade antagônica dos isolados de F. oxysporum não patogênico no controle da murcha vascular do tomateiro, causada por Fusarium oxysporum f. sp. lycopersici raça 2.
