Both skeletal and cardiac muscles daily burn tremendous amounts of ATP to meet the energy requirements for contraction. So, it is not surprising that the maintenance of mitochondrial morphology, number, distribution and functionality in striated muscle are important for muscle homeostasis. In these tissues mitochondria present the added dimension of two populations, the intermyofibrillar (IMF) and the subsarcolemmal (SS) mitochondria, being IMF the most abundant one. In the present thesis, the molecular mechanisms harboured in mitochondria of striated muscles were studied using animal models, to better comprehend the role of mitochondrial plasticity in several pathophysiological conditions such as aging, diabetes mellitus and bladder cancer. The comparative analysis of IMF and SS populations isolated from heart evidenced a higher respiratory chain activity of mitochondria interspersed in the contractile apparatus. The higher susceptible of SS respiratory chain complexes subunits to carbonylation, but not to nitration, seems to justify the lower respiratory chain activity observed in this mitochondrial population. Our results showed that in heart from aged mice there is an accumulation of dysfunctional mitochondria. The age-related decrease of oxidative phosphorylation activity seems to be justified, at least partially, by the increased proneness of mitochondrial proteins as OXPHOS subunits and MnSOD to oxidative modifications. Moreover, a sedentary lifestyle seems to worsen the functional consequences of aging in heart by increasing mitochondrial proteins susceptibility to nitration. In skeletal muscle from rats with type 1 diabetes mellitus induced by streptozotocin administration, we verified the accumulation of dysfunctional mitochondria due, at least in part, to the impairment of PQC system. Indeed, the decreased activity of AAA proteases was accompanied by the accumulation of oxidatively modified mitochondrial proteins with impact in respiratory chain activity. The diminishing of mitochondria activity also underlies cancer-induced muscle wasting. Indeed, using a rat model of chemically induced urothelial carcinoma we verified that the loss of gastrocnemius mass was related to mitochondrial dysfunction due to, at least partially, the down-regulation of PQC system involving the mitochondrial proteases paraplegin and Lon. PQC impairment resulted in the accumulation of oxidatively modified mitochondrial proteins. In overall, regardless the pathophysiological stimuli that promote mitochondrial alterations, there are similarities in the pattern of disease-related mitochondrial plasticity. The diminished capacity for ATP production in striated muscle seems to be due to increased oxidative damage of mitochondrial proteins, namely subunits of respiratory chain complexes, metabolic proteins and MnSOD. Our data highlighted, for the first time, the impact of mitochondrial PQC system impairment in the accumulation of oxidized proteins, exacerbating the dysfunction of this organelle in striated muscle in several pathophysiological conditions.

Cachexia is a complex syndrome characterized by severe weight loss frequently observed in cancer patients and associated with poor prognosis. Cancer cachexia is also related to modifications in cardiac muscle structure and metabolism leading to cardiac dysfunction. In order to better understand the cardiac remodeling induced by bladder cancer and the impact of exercise training after diagnosis on its regulation, we used an animal model of bladder cancer induced by exposition to N-butyl-N-(4-hydroxybutyl)-nitrosamine (BBN) in the drinking water. Healthy animals and previously BBN exposed animals were submitted to a training program in a treadmill at a speed of 20m/min, 60 min/day, 5 days/week during 13 weeks. At the end of the protocol, animals exposed to BBN presented a significant decrease of body weight, in comparison with control groups, supporting the presence of cancer cachexia. Morphological analysis of the cardiac muscle sections revealed the presence of fibrosis and a significant decrease of cardiomyocyte’s cross-sectional area, suggesting the occurrence of cardiac dysfunction associated with bladder cancer. These modifications were accompanied by heart metabolic remodeling characterized by a decreased fatty acid oxidation given by diminished levels of ETFDH and of complex II subunit  from the respiratory chain. Exercise training promoted an increment of connexin 43, a protein involved in cardioprotection, and of c-kit, a protein present in cardiac stem cells. These results suggest an improved heart regenerative capacity induced by exercise training. In conclusion, endurance training seems an attractive non-pharmacological therapeutic option for the management of cardiac dysfunction in cancer cachexia.

Understanding heart development on a molecular level is a requirement for uncovering the causes of congenital heart diseases. Several genes have been implicated as critical for heart development. However, the inducers of these genes as well as their targets and pathways, remain largely unknown. We have identified a promoter element of chick cCer able to drive EGFP expression in a population of cells that consistently exit from the anterior primitive streak region, from as early as stage HH3+, and that later will populate the heart. Using this promoter element as a tool allowed us to identify novel genes previously not known to potentially play a role in heart development. In order to identify and study genes expressed and involved in the correct development and differentiation of the vertebrate heart precursor cell (HPC) lineages, a differential screening using Affymetrix GeneChip® system technologies was performed. Remarkably, this screening led to the identification of more than 700 transcripts differentially expressed in the heart forming regions (HFR). Bioinformatic tools allowed us to filter the large amount of data generated from this approach and to select a few transcripts for in vivo validation. Five genes were selected for further characterization by whole mount in situ hybridization leading to the validation of their expression in the HPC. From those, Adtk1 and Ccbe1 were selected for functional analysis. Regarding to ccbe1, a more detailed WISH analysis was performed and showed that Ccbe1 is expressed specifically on the cardiac progenitors regions at HH4, more specifically in primary heart field and at later stages is present in the second heart field. Further functional analyses by knockdown and overexpression revealed an important role for Ccbe1 in early heart tube formation. Moreover, the results presented in this thesis suggested that Ccbe1 is a key gene during heart development and might be limited to multipotent and highly proliferative progenitors and downregulated upon cellular commitment into more specific cardiac phenotypes. Other of the genes identified, Adtk1 was also subjected to further functional studies. Knockdown of Adtk1 using morpholino oligonucleotides suggested that it might be necessary for the migration and fusion of the heart tube as well as for neural tube closure.

O processamento de sinais de fluxo sanguíneo tem sido objecto de interesse de muitos investigadores, no entanto até este momento, tanto quanto se sabe, não existe caracterização de sinais de ultra-som Doppler de fluxo sanguíneo em vasos cardíacos com inserção de bypass. A presente dissertação, inserida nos objectivos de um projecto de investigação internacional, apresenta as metodologias desenvolvidas com vista à tipificação de sinais ultra-sónicos de fluxo sanguíneo recolhidos em bypass cardíacos e à verificação da variabilidade da composição do sangue artificial com a temperatura ambiente e com a utilização de diferentes marcas dos seus compostos. No que concerne à composição do sangue artificial verifica-se que a utilização de detergentes de marcas diferentes não influencia o espectrograma dos sinais ultra-sónicos; contudo, o glicerol brasileiro possibilita melhor dispersão das partículas de PVC que o glicerol português. Verifica-se também que o aumento da temperatura a que o sangue artificial e o ambiente envolvente se encontram potencia claramente a identificação das envolventes do espectrograma correspondente. Os sinais ultra-sónicos de fluxo sanguíneo foram distinguidos em siglas FL, FR e FT e foram pré-processados removendo-se o ruído de baixas frequências (até 430Hz) e as baixas intensidades de potência (inferiores a um limiar de 35). Após a determinação da envolvente dos ciclos cardíacos, foi calculado o ciclo cardíaco médio de cada sigla. Analisando as características dos ciclos cardíacos médios: tempos de subida e descida do pico máximo, tempo acima da média do ciclo cardíaco médio, índice de pulsatilidade, frequências máxima e média e ainda a duração temporal, conseguiu identificar-se que os sinais FT apresentam durações temporais maiores e um espectrograma maioritariamente acima da frequência média do ciclo cardíaco, os sinais FR apresentam maior índice de pulsatilidade e maiores valores de frequência máxima. Os sinais FL apresentam características intermédias.

The fact that the adult brain is able to produce new neurons or glial cells from neural stem cells (NSC) became one of the most interesting and challenging fields of research in neuroscience. Endogenous adult neurogenesis occurs in two main regions of the brain: the subventricular zone (SVZ) of the lateral ventricles and the subgranular zone (SGZ) in the dentate gyrus. Brain injury may be accompanied by increased neurogenesis, although neuroinflammation promotes the activation of microglial cells that can be detrimental to the neurogenic process. Nitric oxide (NO) is one of the factors released by microglia that can be proneurogenic. The mechanism by which NO promotes the proliferation of NSCs has been intensively studied. However, little is known about the role of NO in migration, survival and differentiation of the newborn cells. The aim of this work was to investigate the role of NO from inflammatory origin in proliferation, migration, differentiation and survival of NSCs from the dentate gyrus in a mouse model of status epilepticus. We also assessed neuroinflammation in the same injury model. Our work showed that NO increased proliferation of the early-born cells after seizures, but is detrimental for their survival. NO also increased migration of neuroblasts. Moreover, NO was important to maintain long-term neuroinflammation. Taken together, these results show that NO may be a good target to promote proliferation and migration of NSCs following seizures, but compromises survival of early-born cells.

A síndrome de Churg-Strauss (SCS) ou angeíte granulomatosa alérgica é uma doença rara caracterizada pela presença de asma, eosinofilia e vasculite dos pequenos e, por vezes, dos médios vasos. O pulmão, coração, pele e nervos periféricos são frequentemente atingidos. Os autores descrevem o caso clínico de uma doente do sexo feminino de 47 anos de idade, internada por lesões purpúricas dolorosas com uma semana de evolução, localizadas nas superfícies extensoras dos membros inferiores. Nos antecedentes pessoais destacava-se asma brônquica com 7 anos de evolução, rinite alérgica e sinusite. A avaliação laboratorial revelou leucocitose com eosinofilia e elevação dos parâmetros de inflamação. Os anticorpos citoplasmáticos anti-neutrófilos eram negativos. A avaliação neurológica e o estudo electroneuromiográfico mostraram uma polineuropatia periférica assimétrica. A biopsia cutânea revelou uma vasculite necrotizante com infiltrado perivascular rico em eosinófilos. O diagnóstico de SCS foi apoiado pelos achados clínicos e histopatológicos, pelo que se iniciou corticoterapia sistémica que resultou numa melhoria clínica marcada. As manifestações cutâneas podem ser uma forma de apresentação clínica do SCS, sendo o seu reconhecimento essencial para a instituição precoce de terapêutica e para a prevenção de lesões irreversíveis em órgãos vitais.
