The study site was located in the Disko Bay off Qeqertarsuaq, western Greenland. Due to land-connected sea ice coverage during winter, 2 sampling sites were combined. At the first site in winter (21 February to 23 March 2008), sampling was conducted through a hole in the ice at ca. 65 to 160 m depth approximately 0.5 nautical mile (n mile) south of Qeqertarsuaq (69° 14' N, 53° 29' W). In spring and summer (9 April to 18 July), sampling was done at a monitoring station 1 n mile south from Qeqertarsuaq (69° 14' N, 53° 23' W) at 300 m depth. Sampling was carried out between 10:00 and 17:00 h. During sampling from the ice, mesozooplankton was collected using a modified WP-2 net (45 µm) equipped with a closing mechanism (Hydrobios). Samples were collected in 3 depth strata (0-50, 50-100, and 100-150 m). During ship-based sampling, mesozooplankton was collected with a multinet (50 µm) equipped with a flow meter (Multinet, Hydrobios type midi), and 2 additional depth strata (150-200m and 200-250 m) were included. In addition to the seasonal study one diurnal investigation with sampling every 6 h was conducted from 29 April at 12:00 h to 30 April 30 at 12:00 h. Samples were immediately preserved in buffered formalin (5% final concentration) for later analyses. Biomass values of the different copepod species were calculated based on measurements of prosome length, and length/weight relationships. Two regressions for Calanus spp. were established for biomass calculations: one applicable prior to and during the phytoplankton bloom until 4 May, and another from 9 May onwards.

High resolution studies from the Propeller Mound, a cold-water coral carbonate mound in the NE Atlantic, show that this mound consists of >50% carbonate justifying the name "carbonate mound". Through the last ~300,000 years approximately one third of the carbonate has been contributed by cold-water corals, namely Lophelia pertusa and Madrepora oculata. This coral bound contribution to the carbonate budget of Propeller Mound is probably accompanied by an unknown portion of sediments buffered from suspension by the corals. However, extended hiatuses in Propeller Mound sequences only allow the calculation of a net carbonate accumulation. Thus, net carbonate accumulation for the last 175 kyr accounts for only <0.3 g/cm2/kyr, which is even less than for the off-mound sediments. These data imply that Propeller Mound faces burial by hemipelagic sediments as has happened to numerous buried carbonate mounds found slightly to the north of the investigated area.

ODP Leg 131 recovered nannofossil-bearing sediments from Site 808 in the Nankai Trough, western Pacific Ocean. Three holes were examined for nannofossils, 808A, 808B, and 808C. A total of 22 nannofossil events were recognized, of which 10 are used as zonal markers. The sediments recovered from Hole 808A (0-111.4 mbsf) contain Pleistocene nannofossil assemblages that are mostly well preserved. All samples from this hole were assigned to nannofossil Zone NN21. The nannofossil assemblages observed in Hole 808B (111.0-358.8 mbsf) are poorly to well preserved and were all assigned to the Pleistocene. The NN21/NN20 Boundary is placed at 230.7 ± 4.4 mbsf. Hole 808C was cored from 298.5 to 1327 mbsf and basalt was reached at 1289.9 mbsf. The sediments recovered range in age from the upper part of Zone NN20 of the Pleistocene to Zone NN5 of the middle Miocene and contain poorly to well-preserved nannofossil assemblages. The Pliocene/Pleistocene Boundary, marked by the FO Gephyrocapsa caribbeanica, was placed at 776.3 ±1.6 mbsf, and the Miocene/Pliocene Boundary is tentatively placed at 955.9 ±1.5 mbsf. The lowermost sediments above basement as well as a sediment sample intercalated between basalt flows are assigned to Zone NN5, with an age of approximately 15 Ma. Age estimates provided by nannofossils show that the sedimentation rate in the trench-fill deposits of the Nankai Trough was very high, 800-1350 m/m.y (0-0.46 Ma), whereas in the Shikoku Basin deposits (> 0.46 Ma), the sedimentation rate was much lower (24-200 m/m.y). These age estimates also provide an extrapolated age of approximately 15 Ma for the basaltic basement at Site 808.

Alkenone unsaturation ratios and planktonic delta18O records from sediment cores of the Alboran, Ionian and Levantine basins in the Mediterranean Sea show pronounced variations in paleo-temperatures and -salinities of surface waters over the last 16,000 years. Average sea surface temperatures (SSTs) are low during the last glacial (averages prior to 13,000 years: 11-15°C), vary rapidly at the beginning of the Holocene, and increase to 17-18°C at all sites during S1 formation (dated between 9500 and 6600 calendar years). The modern temperature gradient (2-3°C) between the Mediterranean sub-basins is maintained during formation of sapropel S1 in the Eastern Mediterranean Sea. After S1, SSTs have remained uniform in the Alboran Sea at 18°C and have fluctuated around 20°C in the Ionian and Levantine Basin sites. The delta18O of planktonic foraminifer calcite decreases by 2 per mil from the late glacial to S1 sediments in the Ionian Basin and by 2.8 per mil in the Levantine Basin. In the Alboran Sea, the decrease is 1.7 per mil. Of the 2.8 per mil decrease in the Levantine Basin, the effect of global ice volume accounts for a maximum of 1.05 per mil and the temperature increase explains only a maximum of 1.3 per mil. The remainder is attributed to salinity changes. We use the temperature and salinity estimates to calculate seawater density changes. They indicate that a reversal of water mass circulation is not a likely explanation for increased carbon burial during S1 time. Instead, it appears that intermediate and deep water formation may have shifted to the Ionian Sea approximately 2000 years before onset of S1 deposition, because surface waters were as cold, but saltier than surface water in the Levantine Basin during the Younger Dryas. Sapropel S1 began to form at the same time, when a significant density decrease also occurred in the Ionian Sea.

Increasing pCO2 (partial pressure of CO2 ) in an "acidified" ocean will affect phytoplankton community structure, but manipulation experiments with assemblages briefly acclimated to simulated future conditions may not accurately predict the long-term evolutionary shifts that could affect inter-specific competitive success. We assessed community structure changes in a natural mixed dinoflagellate bloom incubated at three pCO2 levels (230, 433, and 765 ppm) in a short-term experiment (2 weeks). The four dominant species were then isolated from each treatment into clonal cultures, and maintained at all three pCO2 levels for approximately 1 year. Periodically (4, 8, and 12 months), these pCO2 -conditioned clones were recombined into artificial communities, and allowed to compete at their conditioning pCO2 level or at higher and lower levels. The dominant species in these artificial communities of CO2 -conditioned clones differed from those in the original short-term experiment, but individual species relative abundance trends across pCO2 treatments were often similar. Specific growth rates showed no strong evidence for fitness increases attributable to conditioning pCO2 level. Although pCO2 significantly structured our experimental communities, conditioning time and biotic interactions like mixotrophy also had major roles in determining competitive outcomes. New methods of carrying out extended mixed species experiments are needed to accurately predict future long-term phytoplankton community responses to changing pCO2 .

The ROV operations had three objectives: (1) to check, whether the "Cherokee" system is suited for advanced benthological work in the high latitude Antarctic shelf areas; (2) to support the disturbance experiment, providing immediate visual Information; (3) to continue ecological work that started in 1989 at the hilltop situated at the northern margin of the Norsel Bank off the 4-Seasons Inlet (Weddell Sea). The "Cherokee" is was equipped with 3 video cameras, 2 of which support the operation. A high resolution Tritech Typhoon camera is used for scientific observations to be recorded. In addition, the ROV has a manipulator, a still camera, lights and strobe, compass, 2 lasers, a Posidonia transponder and an obstacle avoidance Sonar. The size of the vehicle is 160 X 90 X 90cm. In the present configuration without TMS (tether management system) the deployment has to start with paying out the full cable length, lay it in loops on deck and connect the glass fibres at the tether's spool winch. After a final technical check the vehicle is deployed into the water, actively driven perpendicular to the ship's axis and floatings are fixed to the tether. At a cable length of approx. 50 m, the tether is tightened to the depressor by several cable ties and both components are lowered towards the sea floor, the vehicle by the thruster's propulsion and the depressor by the ship's winch. At 5 m intervals the tether has to be tied to the single conductor cable. In good weather conditions the instruments supporting the navigation of the ROV, especially the Posidonia system, allow an operation mode to follow the ship's course if the ship's speed is slow. Together with the lasers which act as a scale in the images they also allow a reproducible scientific analysis since the transect can be plotted in a GIS system. Consequently, the area observed can be easily calculated. An operation as a predominantly drifting system, especially in areas with bottom near currents, is also possible, however, the connection of the tether at the rear of the vehicle is unsuitable for such conditions. The recovery of the system corresponds to that of the deployment. Most important is to reach the surface of the sea at a safe distance perpendicular to the ship's axis in order not to interfere with the ship's propellers. During this phase the Posidonia transponder system is of high relevance although it has to be switched off at a water depth of approx. 40 m. The minimum personal needed is 4 persons to handle the tether on deck, one person to operate the ship's winch, one pilot and one additional technician for the ROV's operation itself, one scientist, and one person on the ship's bridge in addition to one on deck for whale watching when the Posidonia system is in use. The time for the deployment of the ROV until it reaches the sea floor depends on the water depth and consequently on the length of the cable to be paid out beforehand and to be tightened to the single conductor cable. Deployment and recovery at intermediate water depths can last up to 2 hours each. A reasonable time for benthological observations close to the sea floor is 1 to 3 hours but can be extended if scientifically justified. Preliminary results: after a first test station, the ROV was deployed 3 times for observations related to the disturbance experiment. A first attempt to Cross the hilltop at the northern margin of the Norsel Bank close to the 4- Seasons Inlet was successful only for the first hundreds of metres transect length. The benthic community was dominated in biomass by the demosponge Cinachyra barbata. Due to the strong current of approx. 1 nm/h, the design of the system, and an expected more difficult current regime between grounded icebergs and the top of the hilltop the operation was stopped before the hilltop was reached. In a second attempt the hilltop was successfully crossed because the current and wind situation was much more suitable. In contrast to earlier expeditions with the "sprint" ROV it was the first time that both slopes, the smoother in the northeast and the steeper in the southwest were continuously observed during one cast. A coarse classification of the hilltop fauna shows patches dominated by single taxa: cnidarians, hydrozoans, holothurians, sea urchins and stalked sponges. Approximately 20 % of the north-eastern slope was devastated by grounding icebergs. Here the sediments consisted of large boulders, gravel or blocks of finer sediment looking like an irregularly ploughed field. On the Norsel Bank the Cinachyra concentrations were locally associated with high abundances of sea anemones. Total observation time amounted to 11.5 hours corresponding to almost 6-9 km transect length.

Coastal upwelling systems account for approximately half of global ocean primary production and contribute disproportionately to biologically driven carbon sequestration. Diatoms, silica-precipitating microalgae, constitute the dominant phytoplankton in these productive regions, and their abundance and assemblage composition in the sedimentary record is considered one of the best proxies for primary production. The study of the sedimentary diatom abundance (SDA) and total organic carbon content (TOC) in the five most important coastal upwelling systems of the modern ocean (Iberia-Canary, Benguela, Peru-Humboldt, California and Somalia-Oman) reveals a global-scale positive relationship between diatom production and organic carbon burial. The analysis of SDA in conjunction with environmental variables of coastal upwelling systems such as upwelling strength, satellite-derived net primary production and surface water nutrient concentrations shows different relations between SDA and primary production on the regional scale. At the global-scale, SDA appears modulated by the capacity of diatoms to take up silicic acid, which ultimately sets an upper limit to global export production in these ocean regions.

O objetivo desta investigação foi verificar se, diante da autonomia e vocação que possuem, bem como da possibilidade de receber amparo e incentivos governamentais, as universidades pertencentes à região do Grande ABC atenderam as recomendações feitas pelo PNPG vigente. Para tanto, foram analisadas cinquenta e sete dissertações e duas teses da área de ciências sociais aplicadas, publicadas no período entre 2011 e 2014, pelas seguintes instituições: Universidade Metodista de São Paulo (UMESP); Universidade Municipal de São Caetano do Sul (USCS); Universidade Federal do ABC (UFABC) e Faculdade de Engenharia Industrial (FEI). Essa averiguação se deu ao redor de dois eixos organizadores do PNPG 2011-2014: o terceiro eixo – o aperfeiçoamento da avaliação e sua expansão para outros segmentos do sistema de CT&I (formação de pós-graduados voltados para atividades extra-acadêmicas/setor empresarial) e o quarto eixo – a multi e a interdisciplinaridade entre as principais características da pós-graduação e importantes temas da pesquisa (promover, por meio de programas, áreas de concentração e linhas de pesquisa, a convergência de temas e compartilhamento de problemas em oposição à sua mera associação ou sobreposição). Este estudo – qualitativo, bibliográfico, documental, exploratório, descritivo, tipo estado do conhecimento – se desenvolveu por meio de pesquisa documental e de análise de conteúdo temático categorial. A coleta de dados foi feita por meio dos repositórios digitais de teses e dissertações mantidos na internet pelas Universidades, cuja produção científica foi investigada. Após a análise dos dados ficou demonstrado que aproximadamente 68,96% da produção científica da área de ciências sociais aplicadas, publicada pelas universidades do Grande ABC, no período entre 2011 e 2014, corresponde às expectativas do PNPG atual no que diz respeito às recomendações constantes no terceiro eixo. Em relação às recomendações feitas no texto do quarto eixo, vemos que aproximadamente 31,03% dos trabalhos selecionados atendem às expectativas do Plano, apresentando em sua estrutura, segundo a fundamentação teórica utilizada neste trabalho, características de interdisciplinaridade.

We previously demonstrated that α1B-adrenergic receptor (AR) gene transcription, mRNA, and functionally coupled receptors increase during 3% O2 exposure in aorta, but not in vena cava smooth muscle cells (SMC). We report here that α1BAR mRNA also increases during hypoxia in liver and lung, but not heart and kidney. A single 2.7-kb α1BAR mRNA was detected in aorta and vena cava during normoxia and hypoxia. The α1BAR 5′ flanking region was sequenced to −2,460 (relative to ATG +1). Transient transfection experiments identify the minimal promoter region between −270 and −143 and sequence between −270 and −248 that are required for transcription of the α1BAR gene in aorta and vena cava SMC during normoxia and hypoxia. An ATTAAA motif within this sequence specifically binds aorta, vena cava, and DDT1MF-2 nuclear proteins, and transcription primarily initiates downstream of this motif at approximately −160 in aorta SMC. Sequence between −837 and −273 conferred strong hypoxic induction of transcription in aorta, but not in vena cava SMC, whereas the cis-element for the transcription factor, hypoxia-inducible factor 1, conferred hypoxia-induced transcription in both aorta and vena cava SMC. These data identify sequence required for transcription of the α1BAR gene in vascular SMC and suggest the atypical TATA-box, ATTAAA, may mediate this transcription. Hypoxia-sensitive regions of the α1BAR gene also were identified that may confer the differential hypoxic increase in α1BAR gene transcription in aorta, but not in vena cava SMC.

Methyl chloride transferase catalyzes the synthesis of methyl chloride from S-adenosine-l-methionine and chloride ion. This enzyme has been purified 2,700-fold to homogeneity from Batis maritima, a halophytic plant that grows abundantly in salt marshes. The purification of the enzyme was accomplished by a combination of ammonium sulfate fractionation, column chromatography on Sephadex G100 and adenosine-agarose, and TSK-250 size-exclusion HPLC. The purified enzyme exhibits a single band on SDS/PAGE with a molecular mass of approximately 22.5 kDa. The molecular mass of the purified enzyme was 22,474 Da as determined by matrix-associated laser desorption ionization mass spectrometry. The methylase can function in either a monomeric or oligomeric form. A 32-aa sequence of an internal fragment of the methylase was determined (GLVPGCGGGYDVVAMANPER FMVGLDIXENAL, where X represents unknown residue) by Edman degradation, and a full-length cDNA of the enzyme was obtained by rapid amplification of cDNA ends–PCR amplification of cDNA oligonucleotides. The cDNA gene contains an ORF of 690 bp encoding an enzyme of 230 aa residues having a predicted molecular mass of 25,761 Da. The disparity between the observed and calculated molecular mass suggests that the methylase undergoes posttranslational cleavage, possibly during purification. Sequence homologies suggest that the B. maritima methylase defines a new family of plant methyl transferases. A possible function for this novel methylase in halophytic plants is discussed.

There is strong converging evidence that the intermediate and medial part of the hyperstriatum ventrale of the chick brain is a memory store for information acquired through the learning process of imprinting. Neurons in this memory system come, through imprinting, to respond selectively to the imprinting stimulus (IS) neurons and so possess the properties of a memory trace. Therefore, the responses of the intermediate and medial part of the hyperstriatum ventrale neurons to a visual imprinting stimulus were determined before, during, and after training. Of the total recorded population, the proportions of IS neurons shortly after each of two 1-h training sessions were significantly higher (approximately 2 times) than the pretraining proportion. However, ≈4.5 h later this proportion had fallen significantly and did not differ significantly from the pretraining proportion. Nevertheless, ≈21.5 h after the end of training, the proportion of IS neurons was at its highest (approximately 3 times the pretraining level). No significant fluctuations occurred in the proportions of neurons responding to the alternative stimulus. In addition, nonmonotonic changes were found commonly in the activity of 230 of the neurons tracked individually from before training to shortly after the end of training. Thus the pattern of change in responsiveness both at the population level and at the level of individual neurons was highly nonmonotonic. Such a pattern of change is not consistent with simple models of memory based on synaptic strengthening to asymptote. A model is proposed that accounts for the changes in the population responses to the imprinting stimulus in terms of changes in the responses of individual neurons.
