In this research reared white western shrimp (Litopenaeus vannamei ,Boone, 1931) with five diet with five different protein level contain 20%, 25%, 30%, 35% and 40% and three salinity level contain 15-17 ppt, 27-30 ppt,and 40-45 ppt researched protein percent effect and water salinity on growth, survival, feed conversion ratio, hemolymph osmolatity, hemolymph protein and corpse protein contain. In this research was 15 sorrow with 3 repeat and used from 45 tanks with 300 liters capacity. Shrimps first weight average was about 2 grams and after 60 days culture cropped down results: Shrimps biomass growth in 15-17 ppt salinity was higher than anther salinities who had meaning different with growth in 40-45 ppt salinity ( p< 0.05). But hadn’t meaning different with growth 27-30 ppt salinity. survival rate in 15-17 ppt salinity was 97.03 who was lower than another salinities. survival percent in 24-30 ppt salinity and 40-45 ppt salinity was 99.33% Highest biomass growth in different diets was in diet number 5 with 40 percent protein that it had meaning different with another diets (p<0.05) . although with informed to product expense in different diets. One kilogram shrimp product expense in different diets hadn’t meaning different (P<0.05) Survival rate in different diets hadn’t meaning different lowest feed conversion ratio was 1.67 in 15-14 salinity that hadn’t meaning different with another salinities also corpse protein quantity in different salinities and different diets hadn’t meaning different. Hemolymph Osmolality in 15-17 ppt salinity was 573.88 mOsm/kg had meaning different with hemolymph osmolality in 27-30 ppt salinity that was 650. 380 mOsm/kg and in 40-45 ppt salinity was 630.38 mOsm/kg. Hemolymph protein in 15-17 ppt salinity was 124.72 mg/ml had meaning different with hemlymph protein in 27-30 ppt salinity that was 136.52 mg/ml but hadn’t meaning different with hemolymph protein in 40-45 ppt salinity that was 128.84 mg/ml. Hemolymph protein in different diets hadn’t meaning different (p<0.05). The aim of the seawater irrigation system (SIS) is to clean up shrimp pond effluent and provide high quality seawater for shrimp farming. The system has 3 components: water intake; treatment reservoir and discharge system. There are criteria for site selection because shrimp farmers are required to form associations so they can work closely together. The construction site must be on the coastal area outside a mangrove forest and located away from a production agricultural area. All construction sites must have undergone an environmental impact assessment, and should be located on the area listed in Thailand's Coastal Zone Management Plan. Five SIS projects, which cover a culture area of 6,500 ha with 1,300 farmers (families), were completed and operated. The Department of Fisheries has planned for another 28 projects, that will cover almost 44,000 ha of culture area.

Seabass (Lates calcarifer) are raised in ponds and cages in Southeast Asia. Details are given of the 2 phases involved in the pond and culture culture systems -- nursery and grow-out. Both monoculture and polyculture systems are used for pond production of seabass. Polyculture may involve milkfish and tilapia, groupers, seaweed, tilapia or snapper, grouper and shrimp.

Live 'agiis' have been proven to be a good feed for shrimp for the past ten years by polyculture fish farmers in the province of Capiz, in the island of Panay, west central Philippines. A brief account is given of culture and feeding operations. Its cultivation period is short and seeds are readily available. It can reduce dependence on trash fish which is now getting to be scarce; it also seems much cheaper. Perhaps this fast-growing tiny bivalve can be scientifically investigated by students of aquaculture as feed for other commercial aquaculture species. Its scientific identification can be a good start.

Penaeus monodon postlarvae were fed with different percentages (0%, 25%, 50%, 75% and 100%) of the herbal appetizer Zingiber officinalis enriched Artemia. After 30 days of culture (i.e. PL-1-30), a very positive result was found in Z. officinalis-enriched Artemia-fed postlarvae. The unenriched Artemia-fed postlarvae consumed 91.0 mg/animal/30 days of feed, whereas the Z. officinalis-enriched Artemia increased their consumption to 127.9 mg/animal/30 days. A similar pattern was noticed in feed absorbed (110.2 mg), dry weight growth (26.7 mg) and feed catabolized (83.2 mg) in Z. officinalis-enriched Artemia because of enzymatic activities. The conversion efficiency of unenriched postlarva was 17.19%, whereas in 100% Z. officinalis-enriched Artemia, the maximum conversion efficiency was 20.85%. The net production efficiency increased significantly (P < 0.05) to 22% from that of the unenriched Artemia-fed postlarvae. The administration of Z. officinalis in all levels produced significantly (P < 0.05) higher weight gain and specific growth rate. The utilization efficiency of feed increased proportionately to the percentages of Z. officinalis. Digestive enzyme activity (amylase, protease and lipase) increased significantly (P < 0.05) in the 50%, 75% and 100% enrichment. Among the different percentages of enrichment, the 100% Z. officinalis-enriched Artemia-fed postlarvae performed better in the overall status.

This study aimed at evaluating the ploidy effects on growth performances of Chinese shrimp (Fenneropenaeus chinensis Osbeck, 1765) reared in different salinities under laboratory conditions. In the acute salinity experiment, there was no difference (P > 0.05) in tolerance observed in triploid and diploid shrimp due to abrupt salinity changes. The lethal salinity for 50% of the individuals in 96 h at 23-25 degrees C was about 2 g L-1 in both triploids and diploids. While for the chronic salinity experiment, statistical analyses confirmed that the differences in growth performances including the specific growth rate (SGR), the feeding rate (FR), feed conversion efficiency (FCE) and intermoult period (IP) between triploid and diploid were related to salinity. Diploid shrimp reared in 20 g L-1 exhibited highest SGR (P < 0.05), while triploids performed well in 20 and 30 g L-1 salinities (P < 0.05). Based on the survival and growth data, the optimal salinity for the culture of diploid F. chinensis should be 20 g L-1 and for triploids it should be between 20 and 30 g L-1.

Lectin is regarded as a potential molecule involved in immune recognition and phagocytosis through opsonization in crustacean. Knowledge on lectin at molecular level would help us to understand its regulation mechanism in crustacean immune system. A novel C-type lectin gene (Fclectin) was cloned from hemocytes of Chinese shrimp Fenneropenaeus chinensis by 3' and 5' rapid amplification of cDNA ends (RACE) PCR. The full-length cDNA consists of 1482 bp with an 861 bp open reading frame, encoding 287 amino acids. The deduced amino acid sequence contains a putative signal peptide of 19 amino acids. It also contains two carbohydrate recognition domains/C-type lectin-like domains (CRD1 and CRD2), which share 78% identity with each other. CRD1 and CRD2 showed 34% and 30% identity with that of mannose-binding lectin from Japanese lamprey (Lethenteron japonicum), respectively. Both CRD1 and CRD2 of Fclectin have I I amino acids residues, which are relatively invariant in animals' C-type lectin CRDs. Five residues at Ca2+ binding site I are conserved in Fclectin. The potential Ca2+/carbohydrate-binding (site 2) motif QPD, E, NP (Gln-Pro-Asp, Glu, Asn-Pro) presented in the two CRDs of Fclectin may support its ability to bind galactose-type sugars. It could be deduced that Fclectin is a member of C-type lectin superfamily. Transcripts of Fclectin were found only in hemocytes by Northern blotting and RNA in situ hybridization. The variation of mRNA transcription level in hemocytes during artificial infection with bacteria and white spot syndrome virus (WSSV) was quantitated by capillary electrophoresis after RT-PCR. An exploration of mRNA expression variation after LPS stimulation was carried out in primarily cultured hemocytes in vitro. Expression profiles of Fclectin gene were greatly modified after bacteria, LPS or WSSV challenge. The above-stated data can provide us clues to understand the probable role of C-type lectin in innate immunity of shrimp and would be helpful to shrimp disease control. (c) 2006 Elsevier Ltd. Triploid Penaeus (Fenneropenaeus) chinensis was successfully produced by heat shock. Their metamorphosis, the relationship between body weight and length and difference in appearance between triploids and their diploid siblings under laboratory culture were studied. Hematological studies showed a smaller number of haemocytes, but larger cell volume, in triploids than in diploids. Triploid shrimp did not show higher growth during the immature stage, but exhibited superior growth during the maturation stage. Characteristics of reproductive organs indicated that triploid shrimp may be sterile and sex ratio can be changed through triploidization of shrimp. This paper summarizes the progress made in triploid shrimp research which would be helpful in understanding more about triploids of crustaceans. (c) 2006 Elsevier B.V. All rights reserved.

The present study revealed the importance of marine actinomycetes as a potent source of bio active secondary metabolites. The selected isolates were capable of protecting Peaneus monodon against WSSV infection. They also proved to be inhibitory to vibrios and is a rich pool of hydrolytic enzymes. Their capacity to proliferate in saline environments and their property of non-pathogenicity to prawns makes them good candidates to be applied as probionts in penaeid shrimp aquaculture. They also enhanced the immune status of shrimps challenged with WSSV and act as a good source of antioxidants. Exploitation of the potential for the prophylactic and therapeutic measures in aquatic animal health management would be highly rewarding. This work is a preliminary study targeting marine actinomycetes as a source of antiviral compounds and as probionts in Penaeus monodon culture systems. More work is needed to understand the nature and mode of action of the bioactive compound, the various aspects of immune and antioxidant responses under challenge and when exposed to pro active treatments, and the dose and frequency of application of such compounds under rearing conditions.

Unveiling the molecular and regulatory mechanisms that prevent in vitro transformation in shrimp remains elusive in the development of continuous cell lines, with an arduous history of over 25 years (Jayesh et al., 2012). Despite presenting challenges to researchers in developing a cell line, the billion dollar aquaculture industry is under viral threat. In addition, the regulatory mechanisms that prevent in vitro transformation and carcinoma in shrimps might provide new leads for the development of anti-ageing and anti-cancer interventions in human (Vogt, 2011) and in higher vertebrates. This highlights the importance of developing shrimp cell lines, to bring out effective prophylactics against shrimp viruses and for understanding the mechanism that induce cancer and ageing in human.. Advances in molecular biology and various gene transfer technologies for immortalization of cells have resulted in the development of hundreds of cell lines from insects and mammals, but yet not a single cell line has been developed from shrimp and other marine invertebrates. With this backdrop, the research described in this thesis attempted to develop molecular tools for induced in vitro transformation in lymphoid cells from Penaeus monodon and for the development of continuous cell lines using conventional and novel technologies to address the problems at cellular and molecular level.

Penaeid shrimps are, perhaps, the most important fishery resource of the coastal waters of our country. Their exceptionally tasty. protein-rich flesh tops any seafood in foreign exchange earnings. No wonder, the demand of shrimp, the "Pinkish Gold of the Sea" (MPEDA. 1992). is increasing in the world market. The study of the growth of an organism is important in understanding the conditions under which optimum growth occurs. It is also important in getting an insight into the various factors that influence growth. Studies on the growth pattern of commercially important species of shrimp and of the factors that influence their growth rate are essential for the successful cultivation of shrimps.

Immortal cell lines have not yet been reported from Penaeus monodon, which delimits the prospects of investigating the associated viral pathogens especially white spot syndrome virus (WSSV). In this context, a method of developing primary hemocyte culture from this crustacean has been standardized by employing modified double strength Leibovitz-15 (L-15) growth medium supplemented with 2% glucose, MEM vitamins (1 ), tryptose phosphate broth (2.95 g l 1), 20% FBS, N-phenylthiourea (0.2 mM), 0.06 lgml 1 chloramphenicol, 100 lgml 1 streptomycin and 100 IU ml 1 penicillin and hemolymph drawn from shrimp grown under a bio-secured recirculating aquaculture system (RAS). In this medium the hemocytes remained viable up to 8 days. 5-Bromo-20-deoxyuridine (BrdU) labeling assay revealed its incorporation in 22 ± 7% of cells at 24 h. Susceptibility of the cells to WSSV was confirmed by immunofluoresence assay using a monoclonal antibody against 28 kDa envelope protein of WSSV. A convenient method for determining virus titer as MTT50/ml was standardized employing the primary hemocyte culture. Expression of viral genes and cellular immune genes were also investigated. The cell culture could be demonstrated for determining toxicity of a management chemical (benzalkonium chloride) by determining its IC50. The primary hemocyte culture could serve as a model for WSSV titration and viral and cellular immune related gene expression and also for investigations on cytotoxicity of aquaculture drugs and chemicals

Lack of shrimp cell lines has hindered the study of pollutants which adversely affects shrimp health and its export value. In this context a primary haemocyte culture developed from Penaeus monodon was employed for assessing the cytotoxicity and genotoxicity of two heavy metal compounds, cadmium chloride and mercuric chloride and two organophosphate insecticides, malathion and monocrotophos. Using MTT assay 12 h IC50 values calculated were 31.09 16.27 mM and 5.52 1.16 mM for cadmium chloride and mercuric chloride and 59.94 52.30 mg l 1 and 186.76 77.00 mg l 1 for malathion and monocrotophos respectively. Employing Comet assay, DNA damage inflicted by these pollutants on haemocytes were evaluated and the pollutants induced DNA damage in >60% of the cells. The study suggested that haemocyte culture could be used as a tool for quantifying cytotoxicity and genotoxicity of aquaculture drugs, management chemicals and pollutants
