Histone deacetylase 3 (HDAC3) plays a critical role in the maintenance of endothelial integrity and other physiological processes. In this study, we demonstrated that HDAC3 undergoes unconventional splicing during stem cell differentiation. Four different splicing variants have been identified, designated as HD3α, -β, -γ, and -Δ, respectively. HD3α was confirmed in stem cell differentiation by specific antibody against the sequences from intron 12. Immunofluorescence staining indicated that the HD3α isoform co-localized with CD31-positive or α-smooth muscle actin-positive cells at different developmental stages of mouse embryos. Overexpression of HD3α reprogrammed human aortic endothelial cells into mesenchymal cells featuring an endothelial-to-mesenchymal transition (EndMT) phenotype. HD3α directly interacts with HDAC3 and Akt1 and selectively activates transforming growth factor β2 (TGFβ2) secretion and cleavage. TGFβ2 functioned as an autocrine and/or paracrine EndMT factor. The HD3α-induced EndMT was both PI3K/Akt- and TGFβ2-dependent. This study provides the first evidence of the role of HDAC3 splicing in the maintenance of endothelial integrity.

Epithelial to mesenchymal transition (EMT) is a process whereby epithelial cells undergo transition to a mesenchymal phenotype and contribute directly to fibrotic disease. Recent studies support a role for EMT in cutaneous fibrotic diseases including scleroderma and hypertrophic scarring, though there is limited data on the cytokines and signalling mechanisms regulating cutaneous EMT. We investigated the ability of TGF-β and TNF-α, both over-expressed in cutaneous scleroderma and central mediators of EMT in other epithelial cell types, to induce EMT in primary keratinocytes and studied the signalling mechanisms regulating this process. TGF-β induced EMT in normal human epidermal keratinocytes (NHEK cells) and this process was enhanced by TNF-α. EMT was characterised by changes in morphology, proteome (down-regulation of E-cadherin and Zo-1, and up-regulation of vimentin and fibronectin), MMP secretion and COL1α1 mRNA expression. TGF-β and TNF-α in combination activated SMAD and p38 signalling in NHEK cells. P38 inhibition with SB203580 partially attenuated EMT, whereas SMAD inhibition using SB431542 significantly inhibited EMT and also reversed established EMT. These data highlight the retained plasticity of adult keratinocytes and support further studies of EMT in clinically relevant in vivo models of cutaneous fibrosis, and investigation of SMAD inhibition as a potential therapeutic intervention. This article describes the evolution of the labour market position of youth in European advanced economies in the last 3 decades, its main institutional determinants and policy instruments. In this period of time, we have witnessed the rise of the phenomenon of youth unemployment and, more recently, of joblessness. Still, youth fares quite differently across European countries. These differentials seem to be linked to institutional features of educational systems (school-to-work transitions) and labour markets (insiders-outsiders divide), and their interactions. Remedies have concentrated on the supply side of this equation (ALMP, minimum wages), leaving on the side the demand one (job creation). We conclude drawing some implications for the Chilean youth labour market concerning the necessity to develop active labour market measures to improve the transition to work processes, and to deal with the strong segmentation which still characterizes it.

Reversible work extraction from identical quantum systems via collective operations was shown to be possible even without producing entanglement among the sub-parts. Here, we show that implementing such global operations necessarily imply the creation of quantum correlations, as measured by quantum discord. We also reanalyze the conditions under which global transformations outperform local gates as far as maximal work extraction is considered by deriving a necessary and sufficient condition that is based on classical correlations.

To what extent is the therapist-client relationship damaged following client perpetuated violence and what steps can we take to diminish its impact? Much of the information we have on client violence comes from multiple mental health disciplines in the US and the UK over the last 20-30 years and has formed a useful, though sometimes dated and sporadic, quantitative baseline to delineate a range of issues. However, there is limited systematic research on how practitioner psychologists process the violence in the course of their everyday practice and how this impacts the therapist-client dynamic. Using Interpretative Phenomenological Analysis (IPA), we explored seven therapists’ experiences of client violence across a range of work sites – acute psychiatric hospital wards, forensic hospital wards and community mental health teams. Three main themes were documented: processing the moment-to-moment experience of client violence; professional vulnerabilities and needs as a result of client violence; and the ruptured therapeutic relationship. Strategies for supporting practicing psychologists and providing continuing professional care for clients include challenging self-doubt and re-energizing professional competencies as well as repairing ourselves and repairing the therapeutic relationship. Recommendations for credentialing and regulatory bodies in relation to client violence are also highlighted.

The growing number of robotic solutions geared to interact socially with humans, social robots, urge the study of the factors that will facilitate or hinder future human robot collaboration. Hence the research question: what are the factors that predict intention to work with a social robot in the near future. To answer this question the following socio-cognitive models were studied, the theory of reasoned action, the theory of planned behavior and the model of goal directed behavior. These models purport that all the other variables will only have an indirect effect on behavior. That is, through the variables of the model. Based on the research on robotics and social perception/ cognition, social robot appearance, belief in human nature uniqueness, perceived warmth, perceived competence, anthropomorphism, negative attitude towards robots with human traits and negative attitudes towards interactions with robots were studied for their effects on attitude towards working with a social robot, perceived behavioral control, positive anticipated emotions and negative anticipated emotions. Study 1 identified the social representation of robot. Studies 2 to 5 investigated the psychometric properties of the Portuguese version of the negative attitude towards robots scale. Study 6 investigated the psychometric properties of the belief in human nature uniqueness scale. Study 7 tested the theory of reasoned action and the theory of planned behavior. Study 8 tested the model of goal directed behavior. Studies 7 and 8 also tested the role of the external variables. Study 9 tested and compared the predictive power of the three socio-cognitive models. Finally conclusion are drawn from the research results, and future research suggestions are offered.

OBJECTIVE: Ability to work and live independently is of particular concern for patients with Parkinson's disease (PD). We studied a series of PD patients able to work or live independently at baseline, and evaluated potential risk factors for two separate outcomes: loss of ability to work and loss of ability to live independently. METHODS: The series comprised 495 PD patients followed prospectively. Ability to work and ability to live independently were based on clinical interview and examination. Cox regression models adjusted for age and disease duration were used to evaluate associations of baseline characteristics with loss of ability to work and loss of ability to live independently. RESULTS: Higher UPDRS dyskinesia score, UPDRS instability score, UPDRS total score, Hoehn and Yahr stage, and presence of intellectual impairment at baseline were all associated with increased risk of future loss of ability to work and loss of ability to live independently (P ≤ 0.0033). Five years after initial visit, for patients ≤70 years of age with a disease duration ≤4 years at initial visit, 88% were still able to work and 90% to live independently. These estimates worsened as age and disease duration at initial visit increased; for patients >70 years of age with a disease duration >4 years, estimates at 5 years were 43% able to work and 57% able to live independently. CONCLUSIONS: The information provided in this study can offer useful information for PD patients in preparing for future ability to perform activities of daily living.

Letter (2 page typed form letter with a handwritten note from Don Loker) regarding a special service held by DeVeaux School to re-inter the body of Mrs. Maria Woodruff, the first wife of Judge Samuel DeVeaux. Her body was originally buried at DeVeaux School in Niagara Falls, New York. The location of the ceremony was the cemetery in St. Davids, Ontario. Maria died on April 23, 1815 at the age of 19. This notice was sent by Rev. Alec Pudwell, chaplain of DeVeaux School, Niagara Falls, New York, May 6, 1963.

This paper examines the workaholism phenomenon in different work situations in Colombian company. Workaholism was defined as the individual’s steady and considerable allocation of time to work, which is not derived from external necessities (1). The research studies about workaholics and workaholism have been increasing a lot in the last years (2). Workaholism is an addiction that actually is affecting a lot of people around the world and has serious consequences in personal life, in the community and also in economy. Some of these researches are directed to explore ways to diagnose when a person is workaholic and when this situation may affect the performanceof the individual in work, daily life activities and especially in psychosocial area. Objective: this pilot study contributes to identify if Colombian workers present the main characteristicsof workaholism and if the job they perform is related to the presence of the characteristics of this addiction. Materials and method: for this pilot study used the Dutch Work Addiction Scale(DUWAS), this test suggests when a person has work addiction, trough the evaluation of two main components working excessively and working compulsively. Results: the study find differences for the two groups: the 67% of the AE group are over the average while only the 33% of the members of the O group are over it. Conclusions: these percentages show that the combinations of the components of workaholism are more evident in the population belonging to the administrative/executive jobs group, giving evidence that workaholism is presented in greater proportion in the population performance management positions.
