Legume plants carefully control the extent of nodulation in response to rhizobial infection. To examine the mechanism underlying this process we conducted a detailed analysis of the Lotus japonicus hypernodulating mutants, har1-1, 2 and 3 that define a new locus, HYPERNODULATION ABERRANT ROOT FORMATION (Har1), involved in root and symbiotic development. Mutations in the Har1 locus alter root architecture by inhibiting root elongation, diminishing root diameter and stimulating lateral root initiation. At the cellular level these developmental alterations are associated with changes in the position and duration of root cell growth and result in a premature differentiation of har1-1 mutant root. No significant differences between har1-1 mutant and wild-type plants were detected with respect to root growth responses to 1-aminocyclopropane1-carboxylic acid, the immediate precursor of ethylene, and auxin; however, cytokinin in the presence of AVG (aminoetoxyvinylglycine) was found to stimulate root elongation of the har1-1 mutant but not the wild-type. After inoculation with Mesorhizobium loti, the har1 mutant lines display an unusual hypernodulation (HNR) response, characterized by unrestricted nodulation (hypernodulation), and a concomitant drastic inhibition of root and shoot growth. These observations implicate a role for the Har1 locus in both symbiotic and non-symbiotic development of L. japonicus, and suggest that regulatory processes controlling nodule organogenesis and nodule number are integrated in an overall mechanism governing root growth and development.

This paper marks the first in a series of studies into the potential use of pyrolysis products in the development of more sustainable practices within the agricultural industry. In this study, the immediate benefits of the application of biochar to crop yields of Raphanus sativus (radishes) are assessed. Furthermore, the study reports on the preliminary findings into the potential application of pyroligneous acid (wood vinegar) as a biocidal agent against crop disease. Although germination tests undertaken on biochar/compost blends of up to 1: 2, by weight, showed no significant adverse effect from the addition of the nutrient rich carbonaceous solid, evidence of substantial increases in crop yield through the addition of biochar were not observed. In sharp contrast, zones of inhibition were observed at 3-10 vol. % upon application of pyroligneous acid to two causal agents responsible for certain diseases in vegetable and fruit crops, i.e. Rhizobium radiobacter (agrobacterium tumefaciens) and Xanthomonas campestris, highlighting the versatility in the application of pyrolysis products and avenues for exploration in the development of this biomass conversion technology.

Legumes have the unique ability to host nitrogen-fixing Rhizobium bacteria as symbiosomes inside root nodule cells. To get insight into this key process, which forms the heart of the endosymbiosis, we isolated specific cells/tissues at different stages of symbiosome formation from nodules of the model legume Medicago truncatula using laser-capture microdissection. Next, we determined their associated expression profiles using Affymetrix Medicago GeneChips. Cells were collected from the nodule infection zone divided into a distal (where symbiosome formation and division occur) and proximal region (where symbiosomes are mainly differentiating), as well as infected cells from the fixation zone containing mature nitrogen fixing symbiosomes. As non-infected cells/tissue we included nodule meristem cells and uninfected cells from the fixation zone. Here, we present a comprehensive gene expression map of an indeterminate Medicago nodule and selected genes that show specific enriched expression in the different cells or tissues. Validation of the obtained expression profiles, by comparison to published gene expression profiles and experimental verification, indicates that the data can be used as digital "in situ''. This digital "in situ'' offers a genome-wide insight into genes specifically associated with subsequent stages of symbiosome and nodule cell development, and can serve to guide future functional studies.

Este trabalho teve como objetivos avaliar a capacidade de utilização de fontes de C, tolerância à salinidade e temperatura, caracterizar genotipicamente e determinar a eficiência agronômica de novos isolados de rizóbio em condições de campo.

Although since 1984 there are Rhizobium strains available for common bean inoculation in Brazil, a survey in 2014 showed that only 15% of the Brazilian farmers adopt inoculation as a routine practice. Part of this disbelief in relation to common beans inoculation can be attributed to the fact that supposedly, bean inoculation would not be able to support increased yield levels in high-tech farms, being appropriate only for small farms. A series of demonstration units carried out since 2012, under farm conditions in the Cerrado region, have showed that a 10.8% mean increase in yield were obtained only with inoculation in relation to the treatment without inoculation and without nitrogen fertilization (whose average yield was 3163 kg ha-1). It was also found that inoculation with supplementation of 60 kg ha-1 of N promoted yield levels (4010 kg ha-1) higher than those with 120 kg ha-1 N as urea (3411 kg ha-1).

A inoculação de sementes de feijão com rizóbio tem apresentado resultados inconsistentes quanto à sua eficiência em suprir nitrogênio para as plantas. Objetivou-se com esta pesquisa determinar o Índice de Suficiência de Nitrogênio (ISN), utilizando o clorofilômetro portátil em plantas de feijão-comum, submetidas à inoculação das sementes com rizóbio no plantio e à adubação nitrogenada em cobertura e, também, definir entre os dois primeiros trifólios do feijoeiro, qual é o adequado para ser usado na tomada das leituras com o clorofilômetro. O delineamento experimental usado foi em blocos ao acaso, com quatro repetições. No primeiro estudo, avaliaram-se os tratamentos: sem adubação nitrogenada em cobertura e inoculação das sementes com rizóbio, com aplicação de 160 kg ha-1 de N aos oito dias após a emergência das plântulas e as doses de 20, 40, 60 e 80 kg ha-1 de N aplicadas em cobertura. No estudo de definição do trifólio, avaliaram-se as doses 0, 30, 60 e 120 kg ha-1 de N. O ISN determinado com o clorofilômetro permitiu determinar se a planta de feijão estava suprida de nitrogênio, quer seja proveniente da fixação biológica ou da adubação nitrogenada. Não se observou diferença entre os valores das leituras com o clorofilômetro no primeiro e segundo trifólios do feijão-comum. Pela maior facilidade de identificação em campo e a certeza de que se encontra totalmente formado, recomenda-se a utilização do primeiro trifólio na tomada das leituras com o clorofilômetro.

Soil salinity affects rhizobia both as free-living bacteria and in symbiosis with the host. The aim of this study was to examine the transcriptional response of the Lotus microsymbiont Mesorhizobium loti MAFF303099 to salt shock. Changes in the transcriptome of bacterial cells subjected to a salt shock of 10% NaCl for 30 min were analyzed. From a total of 7231 protein-coding genes, 385 were found to be differentially expressed upon salt shock, among which 272 were overexpressed. Although a large number of overexpressed genes encode hypothetical proteins, the two most frequently represented COG categories are "defense mechanisms" and "nucleotide transport and metabolism". A significant number of transcriptional regulators and ABC transporters genes were upregulated. Chemotaxis and motility genes were not differentially expressed. Moreover, most genes previously reported to be involved in salt tolerance were not differentially expressed. The transcriptional response to salt shock of a rhizobium with low ability to grow under salinity conditions, but enduring a salinity shock, may enlighten us concerning salinity stress response mechanisms.

Beans fromcowpea cultivars fertilized with mineral N or inoculated with various rhizobium strainsmay contain different nitrogen concentrations and nitrogen metabolite composition, which affects the beans? defense mechanisms against pests. In this study, the population growth of Callosobruchus maculatus reared on beans from four cowpea cultivars fertilized with different nitrogen sources was evaluated. The factors tested were beans from four cowpea cultivars and seven different nitrogen sources: mineral N fertilization, inoculation with five strains of symbiotic diazotrophic bacteria, and soil nitrogen (absolute control).

Sclerolobium panicudatum Vogel (taxi-branco-da-terra-firme) uma espécie nativa da Região Amazônica, de rápido crescimento, elevada produção de biomassa, capaz de associar-se com bactérias do gênero Rhizobium e com fungos micorrízicos. A madeira desta espécie possui características comparáveis àquelas tradicionalmente utilizadas no Sul do Brasil como fonte de energia. É de suma importância que sejam realizados estudos sobre as exigências nutricionais desta essência florestal tão promissora. Visando estudar o crescimento, foi desenvolvida a presente pesquisa, com o objetivo de avaliar os efeitos da aplicação do Fosfato natural trauíra superior na fase dc muda. O experimento foi instalado em casa de vegetação da Embrapa Amazônia Oriental, em vasos contendo Latossolo Amarelo álico. O delineamento experimental foi de blocos ao acaso, em esquema fatorial 4 x 5, sendo quatro doses: 0, 30, 60 e 90 ppm de fósforo (P); e cinco idades: 30. 60. 90. 120 c 150 dias. Foram avaliadas as variáveis: altura, número de folhas, área foliar, produção de matéria seca das folhas, caule, pane aérea, raiz e total. Constatou-se efeito significativo em todas as variáveis em função das idades, o número de folhas apresentou um ponto máximo de 9,59 folhas/planta, aos 110.62 dias. Quanto à aplicação das doses de fósforo, observou-se efeito significativo apenas nas variáveis área foliar, obtendo-se ponto máximo de 515.91 cm²/planta com aplicação da dose estimada de 52,86 ppm de P e na variável produção de matéria seca foliar com ponto máximo de 2,44 g/planta na dose estimada de 44,25 ppm de P.

Este trabalho teve por objetivo verificar a influência do biocarvão e da utilização de inoculante sobre o desenvolvimento e a nodulação de mudas de gliricídia. O experimento foi desenvolvido em viveiro da Embrapa Semiárido. Foram utilizadas quatro doses de biocarvão (0%, 10%, 20% e 30% em volume), na presença e ausência de inoculante formado pelas estirpes de rizóbio BR8801 e BR8802. A inoculação reduziu a altura das mudas, mas aumentou a matéria seca de raiz e de parte aérea das mudas. A adição de biocarvão ao substrato aumentou a matéria fresca de parte aérea das mudas. Assim, o inoculante testado e o biocarvão permitiram a produção de mudas de gliricídia de melhor qualidade.
