Interactions of viral proteins play an important role in the virus life cycle, especially in capsid assembly. Andean potato mottle comovirus (APMoV) is a plant RNA virus with a virion formed by two coat proteins (CP42 and CP22). Both APMoV coat protein open reading frames were cloned into pGBT9 and pGAD10, two-hybrid system vectors. HF7c yeast cells transformed with the p9CP42 construct grew on yeast dropout selection media lacking tryptophan and histidine. Clones also exhibited ß-galactosidase activity in both qualitative and quantitative assays. These results suggest that CP42 protein contains an amino acid motif able to activate transcription of His3 and lacZ reporter genes in Saccharomyces cerevisiae. Several deletions of the CP42 gene were cloned into the pGBT9 vector to locate the region involved in this activation. CP42 constructions lacking 12 residues from the C-terminal region and another one with 267 residues deleted from the N-terminus are still able to activate transcription of reporter genes. However, transcription activation was not observed with construction p9CP42deltaC57, which does not contain the last 57 amino acid residues. These results demonstrate that a transcription activation domain is present at the C-terminus of CP42 between residues 267 and 374.

Ipomoea imperati (Convolvulaceae) lives on the sandy shores of the Brazilian coast and in other areas of the world. The anti-inflammatory activity of a methanol-water extract of the leaves of I. imperati was investigated in experimental models of acute and subchronic inflammation. Topical application of the extract (10 mg/ear) inhibited mouse ear edema induced by croton oil (89.0 ± 1.3% by the lipid fraction with an IC50 of 3.97 mg/ear and 57.0 ± 1.3% by the aqueous fraction with an IC50 of 3.5 mg/ear) and arachidonic acid (42.0 ± 2.0% with an IC50 of 4.98 mg/ear and 31.0 ± 2.0% with an IC50 of 4.72 mg/ear). Phospholipase A2, purified from Apis mellifera bee venom, was also inhibited by the extract (5.0 mg/ml lipid and aqueous fraction) in vitro in a dose-dependent manner (85% by the lipid fraction with an IC50 of 3.22 mg/ml and 25% by the aqueous fraction with an IC50 of 3.43 mg/ml). The methanol-water extract of I. imperati (1000 mg/kg) administered by the oral route also inhibited the formation of cotton pellet-induced granulomas (73.2 ± 1.2% by the lipid fraction and 56.14 ± 2.7% by the aqueous fraction) and did not cause gastric mucosal lesions. I. imperati extracts (10 mg/ml) also inhibited in a dose-dependent manner the muscle contractions of guinea pig ileum induced by acetylcholine and histamine (IC50 of 1.60 mg/ml for the lipid fraction and 4.12 mg/ml for the aqueous fraction). These results suggest the use of I. imperati as an anti-inflammatory and antispasmodic agent in traditional medicine.

A batata é uma das principais hortaliças cultivadas no Brasil, constituindo-se em um dos mais importantes alimentos na dieta humana, em decorrência de sua disponibilidade e características nutricionais. Entretanto, membros da família Solanaceae produzem, durante crescimento e após colheita, compostos potencialmente tóxicos denominados glicoalcalóides, dos quais alfa-solanina e alfa-chaconina predominam. A concentração máxima desses compostos em batata in natura considerada segura para consumo humano é estimada em 200mg·kg-1, expressa como glicoalcalóides totais (GAT). No presente estudo, foram extraídos e quantificados os GAT em amostras de tubérculos de batata in natura comercializados na cidade de Campinas, SP. A técnica utilizada foi a cromatografia líquida de alta eficiência (CLAE) com coluna C18 e detetor de arranjo de diodos. O valor médio de recuperação foi 102,6% para alfa-solanina e 100,2% para alfa-chaconina. Os limites de quantificação foram 2,5µg·mL-1 para alfa-solanina e 1,4µg·mL-1 para alfa-chaconina. As concentrações de GAT em amostras individuais de tubérculos inteiros das diferentes variedades e tipos estudados (Bintje, Monaliza, Asterix e Bolinha) variaram de 22,4 a 246,9mg·kg-1. Das amostras analisadas, 82% apresentaram níveis de GAT inferiores a 100mg·kg-1, o que indica que as variedades de batatas estudadas podem ser consideradas seguras para consumo humano.

O objetivo deste trabalho foi caracterizar química e fisicamente batatas minimamente processadas durante o armazenamento refrigerado. Batatas (Solanum tuberosum, L.) ágata e monalisa foram minimamente processadas como minibatatas. Após o processamento, as batatas foram acondicionadas sob vácuo parcial e, posteriormente, armazenadas em câmaras frias a 5 e 15ºC, por nove dias. A cada três dias, foram avaliadas as seguintes variáveis: firmeza, atividade da polifenoloxidase e peroxidase, açúcares solúveis totais, amido e vitamina C total. Nas batatas armazenadas a 15ºC, constatou-se que, após nove dias de armazenamento, sua firmeza era 3,3 vezes menor em batatas monalisa e 4,3 vezes menor para a cultivar ágata, quando comparadas com o produto recém-processado. A atividade da polifenoloxidase mostrou-se praticamente estável em batatas monalisa armazenadas a 5ºC. Batatas monalisa minimamente processadas apresentaram maior atividade da peroxidase a 5ºC, sendo 86% maior do que a atividade desta enzima em batatas ágata ao final do período experimental. O teor inicial de açúcares solúveis totais nas batatas minimamente processadas era 28% maior na cultivar monalisa, quando comparada com tubérculos de ágata. As duas cultivares apresentaram tendência de elevação do teor de amido nos primeiros três dias, para as duas temperaturas estudadas. Em ambas as cultivares, armazenamento a 5ºC possibilitou maior manutenção dos teores de vitamina C.

Na fritura, os óleos e gorduras sofrem degradação formando os compostos polares, como os triacilgliceróis polimerizados e os monômeros de triacilgliceróis oxidados, estes últimos têm sido relacionados a patologias e se constituem de grupos como os monoepoxiácidos, monocetoácidos e monohidroxiácidos. Foram determinados os teores destes três grupos de substâncias, através da análise cromatográfica a gás dos ésteres metílicos de ácidos graxos após derivação a frio das amostras e concentração posterior das substâncias de interesse mediante extração em fase sólida, e os teores dos ácidos graxos polares polimerizados, utilizando a combinação das técnicas cromatográficas de adsorção em coluna e por exclusão, em gordura de soja parcialmente hidrogenada e óleos de soja e de palma, aquecidos a 180 °C por 25 horas, provenientes de experimentos de fritura. Foram encontrados teores de 3,30 a 8,24; 1,51 a 4,32 e 3,42 a 8,71 mg.g-1 de monoepoxiácidos, monocetoácidos e monohidroxiácidos, respectivamente, nas amostras com níveis de compostos polares de 20,8 a 44,1% (m/m), e predominância de formação de polímeros. Os resultados sugeriram que óleos e gorduras monoinsaturados apresentam maior tendência à formação de monômeros oxidados e menor tendência à formação de polímeros, ao contrário dos óleos poliinsaturados.

Óleos de palma e de soja foram utilizados em experimentos de fritura de batatas em fritadeiras elétricas domésticas de 1 L com relação inicial, superfície sobre volume, de 0,3 cm-1. Em cinco dias consecutivos cada óleo foi submetido por um período de 5 horas por dia à temperatura de (183,1 ± 1,7) °C. Após 25 horas, a perda de ácidos graxos foi de 15,0 e 62,2% (m/m), respectivamente, para os ácidos oléico e linoléico, no óleo de palma; e de 6,5; 24,9 e 39,7% (m/m), respectivamente, para os ácidos oléico, linoléico e linolênico, no óleo de soja. Os teores inicial e final de polímeros foram de 0,4 e 23,7% (m/m) no óleo de palma, e de 0,5 e 30,7% (m/m) no óleo de soja. A relação entre a perda dos ácidos graxos e a formação de polímeros mostrou uma correlação forte com coeficientes de correlação de 0,9951 e 0,9740 para os óleos de palma e de soja, respectivamente. Concluiu-se que maiores graus de alteração ocorreram em ácidos graxos mais insaturados; e que a alteração pode ser eficazmente verificada através da determinação quantitativa de polímeros.

Potato pulp waste (PPW) drying was investigated under different experimental conditions (temperatures from 50 to 70 °C and air flow from 0.06 to 0.092 m³ m- 2 s- 1) as a possible way to recover the waste generated by potato chip industries and to select the best-fit model to the experimental results of PPW drying. As a criterion to evaluate the fitting of mathematical models, a method based on the sum of the scores assigned to the four evaluated statistical parameters was used: regression coefficient (R²), relative mean error P (%), root mean square error (RMSE), and reduced chi-square (χ²). The results revealed that temperature and air velocity are important parameters to reduce PPW drying time. The models Midilli and Diffusion had the lowest sum values, i.e., with the best fit to the drying data, satisfactorily representing the drying kinetics of PPW.

Sweet potato is an important staple, and it is mainly known for its contribution of β-carotene in human diet. The effects of cultivar and habitat on this pigment and other nutritional characteristics of the crop still require investigation. In this study, three locally bred cultivars of sweet potato, two of which are orange-fleshed, were grown in three different agro-ecological areas to determine soluble sugar content, β-carotene, and total antioxidants of roots. In addition antioxidant activity, total carotenoids, and chlorophyll content were determined in edible leaves. Reducing sugars, β-carotene, total antioxidants capacity, total carotenoids, and chlorophyll content were significantly affected by environmental conditions. The location at lower altitude and closer to the coastline showed high evapotranspiration, thus reducing sugar content, antioxidant activity, and phytonutrients in both storage roots and leaves. Absence of water stress in agro-ecological locations further inland and at higher altitudes was associated with an increase in these compounds. Free radical scavenging activity of DPPH was higher in the storage roots (610.49 µmoles TE/100g) than in the leaves (426.06 µmoles TE/100g); nevertheless, opposite results were found for the ferric ion reducing activity (FRAP). The deep orange-fleshed cultivar A45 contained high β-carotene (15 mg/100g), which is enough to meet RDA for vitamin A. There is evidence of agro-ecological effect on sweet potato nutritional value.

Egg yolk was partially replaced (0, 25, 50, 75, and 100%) with octenyl succinic anhydride (OSA)-modified potato starch in a reduced-fat mayonnaise formulation to curtail the problems associated with high cholesterol and induced allergic reactions. The physicochemical properties included parameters such as: pH, fat content, and emulsion stability of the formulations analyzed. The samples with 75% and 100% egg yolk substitute showed the maximum emulsion stability (>95% after two of months storage), and they were selected according to cholesterol content, particle size distributions, dynamic rheological properties, microstructure, and sensory characteristic. A significant reduction (84-97%) in the cholesterol content was observed in the selected samples. Particle size analysis showed that by increasing the amount of OSA starch, the oil droplets with the peak size of 70 µm engulfed by this compound became larger. The rheological tests elucidated that in the absence of egg yolk, OSA starch may not result in a final product with consistent texture and that the best ratio of the two emulsifiers (OSA starch/egg yolk) to produce stable reduced-fat, low cholesterol mayonnaise is 75/25. The microscopic images confirmed the formation of a stable cohesive layer of starch surrounding the oil droplets emulsified in the samples selected.

AbstractPurple sweet potato (PSP) can provide products with attractive color besides nutritious benefits in food processing. So, the compositions and color stability of an aqueous anthocyanin-based PSP extract were investigated in order to promote its wide use in food industry. PSP anthocyanins were extracted with water, and nine individual anthocyanins (48.72 ug mL–1 in total, 24.36 mg/100 g fresh PSP in yield) were found by HPLC analysis. The PSP extract also contained 17.11 mg mL–1 of protein, 0.44 mg mL–1 of dietary fiber, 2.82 mg mL–1 of reducing sugars, 4.02 ug mL–1 of Se, 54.21 ug mL–1 of Ca and 60.83 ug mL–1 of Mg. Changes in color and stability of the PSP extract, as affected by pH, heat, light and extraction process, were further evaluated. Results indicated that PSP anthocyanins had good stability at pH 2.0-6.0, while the color of PSP extract kept stable during 30 days of storage at 20 °C in dark. Both UV and fluorescent exposure weakened the color stability of PSP extract and UV showed a more drastic effect in comparison. A steaming pretreatment of fresh PSP is beneficial to the color stability.

The aim of this work was to study techniques to extract and purify of anthocyanins from purple-blue potato. This topic was determined as a master’s thesis and it was done in collaboration with the Food Chemistry and Food Development Department of University of Turku and Department of Chemical and Process Engineering at Lappeenranta University of Technology. At first, purple-blue potatoes were pretreated in four types of boiled, raw, freeze-dried and dried boiled potato for extraction. They were mixed with aqueous acidified ethanol (ethanol:water:acetic acid 40%:53%:7% v/v) for conventional extraction. Boiled potato was selected as a best pretreated potato. Different ethanol concentration and extraction time were examined and the mixture of 80% in 24 h resulted in maximum anthocyanin content (132.23 mg/L). As conventional extraction method of anthocyanins was non-selective, some of impurities such as free sugars might accelerate anthocyanin degradation. Therefore, to obtain anthocyanins in purified form, adsorption as a promising selective method was used to recovery and isolate anthocyanins. It was carried out with six adsorbents. Among those, Amberlite XAD-7HP, a nonionic acrylic ester adsorbent, was found to have the best performance. In an adsorption column, flow rate of 3 mL/min was selected as the loading flow rate among four tested flow rates. Eluent volume and flow rate were 3 BV of aqueous acidified ethanol (75%, v/v) and 1 mL/min for desorption. The quantification of the total anthocyanin contents was performed by pH-differential method using UV-vis spectrophotometer. The resulting anthocyanin solution after purification was almost free from free sugars which were the major cause for degradation of anthocyanins. The average anthocyanin concentration in the purified and concentrated sample was obtained 1752.89 mg/L.

The presented thesis considered three different system approach topics to ensure yield and plant health in organically grown potatoes and tomatoes. The first topic describes interactions between late blight (Phytophthora infestans) incidence and soil nitrogen supply on yield in organic potato farming focussing in detail on the yield loss relationship of late blight based on results of several field trials. The interactive effects of soil N-supply, climatic conditions and late blight on the yield were studied in the presence and absence of copper fungicides from 2002-2004 for the potato cultivar Nicola. Under conditions of central Germany the use of copper significantly reduced late blight in almost all cases (15-30 %). However, the reductions in disease through copper application did not result in statistically significant yield increases (+0 – +10 %). Subsequently, only 30 % of the variation in yield could be attributed to disease reductions. A multiple regression model (R²Max), however, including disease reduction, growth duration and temperature sum from planting until 60 % disease severity was reached and soil mineral N contents 10 days after emergence could explain 75 % of the observed variations in yield. The second topic describes the effect of some selected organic fertilisers and biostimulant products on nitrogen-mineralization and efficiency, yield and diseases in organic potato and tomato trials. The organic fertilisers Biofeed Basis (BFB, plant derived, AgroBioProducts, Wageningen, Netherlands) and BioIlsa 12,5 Export (physically hydrolysed leather shavings, hair and skin of animals; ILSA, Arizignano, Italy) and two biostimulant products BioFeed Quality (BFQ, multi-compound seaweed extract, AgroBioProducts) and AUSMA (aqueous pine and spruce needle extract, A/S BIOLAT, Latvia), were tested. Both fertilisers supplied considerable amounts of nitrogen during the main uptake phases of the crops and reached yields as high or higher as compared to the control with horn meal fertilisation. The N-efficiency of the tested fertilisers in potatoes ranged from 90 to 159 kg yield*kg-1 N – input. Most effective with tomatoes were the combined treatments of fertiliser BFB and the biostimulants AUSMA and BFQ. Both biostimulants significantly increased the share of healthy fruit and/or the number of fruits. BFQ significantly increased potato yields (+6 %) in one out of two years and reduced R. solani-infestation in the potatoes. This suggests that the biostimulants had effects on plant metabolism and resistance properties. However, no effects of biostimulants on potato late blight could be observed in the fields. The third topic focused on the effect of suppressive composts and seed tuber health on the saprophytic pathogen Rhizoctonia solani in organic potato systems. In the present study 5t ha-1 DM of a yard and bio-waste (60/40) compost produced in a 5 month composting process and a 15 month old 100 % yard waste compost were used to assess the effects on potato infection with R. solani when applying composts within the limits allowed. Across the differences in initial seed tuber infestation and 12 cultivars 5t DM ha-1 of high quality composts, applied in the seed tuber area, reduced the infestation of harvested potatoes with black scurf, tuber malformations and dry core tubers by 20 to 84 %, 20 to 49 % and 38 to 54 %, respectively, while marketable yields were increased by 5 to 25 % due to lower rates of wastes after sorting (marketable yield is gross yield minus malformed tubers, tubers with dry core, tubers with black scurf > 15% infested skin). The rate of initial black scurf infection of the seed tubers also affected tuber number, health and quality significantly. Compared to healthy seed tubers initial black scurf sclerotia infestation of 2-5 and >10 % of tuber surface led in untreated plots to a decrease in marketable yields by 14-19 and 44-66 %, a increase of black scurf severity by 8-40 and 34-86 % and also increased the amount of malformed and dry core tubers by 32-57 and 109-214 %.

As we initiate entomological research on potato (Solanum tuberosum L.) in Uganda, there is need to understand farmers’ knowledge of existing insect pest problems and their management practices. Such information is important for designing a suitable intervention and successful integrated pest management (IPM) strategy. A farm household survey using a structured questionnaire was conducted among 204 potato farmers in six districts of Uganda (i.e., Kabale, Kisoro, Mbale, Kapchorwa, Mubende, and Kyegegwa) during August and September 2013. Diseases, insect pests, price fluctuations, and low market prices were the four highest ranked constraints in potato production, in order of decreasing importance. Cutworms (Agrotis spp.), aphids (Myzus persicae (Sulzer)), and potato tuber moth (Phthorimaea operculella (Zeller)) were the three most severe insect pests. Ants (Dorylis orantalis Westwood), whiteflies (Bemisia tabaci (Gennadius)), and leafminer flies (Liriomyza huidobrensis (Blanchard)) were pests of moderate importance. Major yield losses are predominantly due to late blight (Phytophthora infestans (Mont.) de Bary) and reached 100% without chemical control in the districts of Kabale, Kisoro, Mbale, and Kapchorwa. On average, farmers had little to moderate knowledge about pest characteristics. The predominant control methods were use of fungicides (72% of respondents) and insecticides (62% of respondents). On average, only 5% of the 204 farmers knew about insect pests and their natural enemies. This lack of knowledge calls for training of both farmers and extension workers in insect pest identification, their biology, and control. Empowering farmers with knowledge about insect pests is essential for the reduction of pesticide misuse and uptake of more environmentally friendly approaches like IPM. Field surveys would need follow-up in order to assess the actual field infestation rates and intensities of each insect pest and compare the results with the responses received from farmers.

La caracterització funcional de dos gens en la peridermis, la ω hidroxilasa d'àcids grassos CYP86A33 -candidata per la funcionalització del carboni ω-terminal dels monòmers alifàtics de la suberina- i la ketoacyl-CoA sintasa StKCS6 -candidata per elongar àcids grassos o derivats llargs de suberina i ceres- es realitza per silenciament per RNA d'interferència en patata. La deficiència de CYP86A33 comporta una gran reducció dels monòmers principals de la suberina, l'àcid gras ω-hidroxilat i l'α,ω-diàcid C18:1, juntament amb una reducció total de la quantitat de suberina del 60%. Aquesta deficiència altera l'estructura lamel·lar típica de la suberina, així com també la funció barrera de la peridermis. La deficiència en StKCS6 comporta que els monòmers de la suberina de 28 carbonis o més llargs es redueixin i que els de 26 carbonis o més curts s'incrementin. Aquesta deficiència suggereix que la llargada dels compostos alifàtics pot contribuir a les propietats impermeabilitzants de la peridermis.
