Compounds of plant origin and food components have attracted scientific attention for use as agents for cancer prevention and treatment. Wine contains polyphenols that were shown to have anti-cancer and other health benefits. The survival pathways of Akt and extracellular signal-regulated kinase (Erk), and the tumor suppressor p53 are key modulators of cancer cell growth and survival. In this study, we examined the effects of wine on proliferation and survival of human Non-small cell lung cancer (NSCLC) cells and its effects on signaling events.

Le diabète de type 2 et l'obésité sont des problèmes de santé majeurs et les peuples autochtones sont particulièrement à risque. Pour remédier à ce problème largement répandu dans les populations autochtones canadiennes pour qui la médication moderne n’est pas culturellement adaptée, notre équipe s’est donné comme objectif d’étudier les activités potentielles antidiabétique et anti-obésité de la pharmacopée traditionnelle des Cris de la Baie James. Le but de cette étude est de tester l’hypothèse selon laquelle certaines plantes médicinales pourraient inhiber l'absorption intestinale du glucose, une activité anti-hyperglycémique qui, par la même occasion, contribuerait à combattre l’obésité. Les extraits éthanoliques de dix-sept plantes médicinales de la forêt boréale ont été testés dans des cellules intestinales Caco-2 et comparés à l’effet d’inhibiteurs compétitifs connus, tels que la phlorizine et la phlorétine. Ces inhibiteurs sont des composés polyphénoliques qui partagent de nombreuses caractéristiques structurelles avec des constituants moléculaires de plusieurs plantes Cri. Les résultats démontrent que treize des dix-sept extraits de plantes ont inhibé de façon significative l'absorption intestinale du 3H-D-glucose. Pour valider ces effets in vivo, quatre extraits ont été administrés à des rats Wistar par gavage intragastrique (250 mg/kg) en même temps qu’un bolus de glucose (3 g/kg). Suite à ce gavage, deux de ces extraits ont restreint l’augmentation de la glycémie d'environ 40% par rapport à un contrôle sans extrait. Ces résultats indiquent qu’une inhibition compétitive de l'absorption intestinale du glucose peut être atteinte par des extraits bruts de plantes médicinales. La prise de ces plantes durant les repas aiderait à un meilleur contrôle post-prandial de la glycémie, particulièrement chez les personnes à risque.

Kurzfassung: Der Markt für ökologische Lebensmittel wächst stark. Verbraucher kaufen Produkte aus ökologischem Landbau aus einer Vielzahl von Gründen. Ein Teil dieser Gründe lässt sich nicht auf die Produktqualität zurückführen, sondern beruht auf der Annahme, dass sich der Produktionsprozess des Ökologischen Landbaus hinsichtlich der Schonung von Umweltressourcen, der Nachhaltigkeit der Produktion und sozialen Komponenten vom konventionellen Anbau unterscheidet. Daneben spielt der Wunsch nach einer gesunden Ernährung eine Rolle. Ökologische Lebensmittel können als Vertrauensgüter verstanden werden. Lebensmittelskandale machten in den vergangenen Jahren auch vor ökologischen Lebens¬mitteln nicht Halt. Folgerichtig erschütterte dies das Vertrauen der Verbraucher in ökologische Produkte. Mit steigender Produktion könnte die Gefahr, das weitere solche Ereignisse auftreten, steigen. Daher besteht Bedarf für Methoden, die die ökologische Produktqualität im Sinne einer Authentizitätsprüfung prüfen. Eine solche Prüfung könnte sich auf die Analyse sekundärer Pflanzenstoffe stützen. Diese Gruppe von Pflanzeninhaltsstoffen spielt bei der Diskussion um die besondere Qualität ökologischer Pflanzenprodukte eine große Rolle. Postuliert wird, dass ökologisch angebaute Pflanzen mangels mineralischer Düngung und mangels Schädlingsbekämpfung mit synthetischen Pestiziden einem erhöhten Stress ausgesetzt sind. Dies soll sich in einem höheren Niveau der mit den Selbstverteidigungsmechanismen der Pflanze eng verbundenen sekundären Pflanzenstoffe ausdrücken. Wichtige Untergruppen der sekundären Pflanzenstoffe sind Carotinoide und Polyphenole. An Weizen (Triticum aestivum L. und Triticum durum L.) und Möhre (Daucus carota L.) als für den ökologischen Landbau wichtigen Produkten wurden Messungen der Carotinoid- und Polyphenolkonzentration mit dem Ziel durchgeführt, die potentielle Eignung dieser Pflanzenstoffe als Biomarker zur Authentizitätsprüfung ökologischer Produkte zu evaluieren. Dazu wurden Proben aus ökologischem und konventionellem Anbau (Paarvergleich) untersucht. Diese stammten aus Langzeit-Feldversuchen (Weizen aus dem DOK- und dem MASCOT-Versuch), Feldversuchen und von Betriebspaaren untersucht. Ein generell höheres Niveau sekundärer Pflanzenstoffe in Möhren bzw. Weizen aus ökologischem Anbau gegenüber Proben aus konventionellem Anbau wurde nicht gefunden. Die Carotinoide waren weder bei der Möhre noch beim Weizen zur Authentizitätsprüfung geeignet. Die Konzentration der Carotinoide wurde stark durch die nicht dem Anbau¬verfahren zuzuordnenden Faktoren Klima, Sorte und Standort beeinflusst. Die Luteinkonzentration war das einzige durch das Anbauverfahren systematisch beeinflusste Carotenoid bei Weizen und Möhre. Die Unterschiede der Luteinkonzentration waren aber im Paarvergleich von Proben (ökologischer versus konventioneller Anbau) nicht durchgängig signifikant. Die Eignung von Polyphenolen als potentielles Authentizitätskriterium wurde nur an Möhren geprüft. Im Paarvergleich unterschieden sich die Konzentrationen einzelner Polyphenole signifikant und konsistent über Probenjahre und Standorte, nicht jedoch über Sorten hinweg. Wie bei den Carotinoiden konnte auch hier ein starker Einfluss von Probenjahr, Standort und Sorte gezeigt werden. Trotz der Variation durch diese nicht dem Anbau zuzuordnenden Faktoren war eine korrekte Klassifizierung der Proben nach Anbauverfahren möglich. Dies wurde mittels Diskriminanzanalyse getestet. Die Polyphenole sind daher potentiell als Authentizitätskriterium geeignet.

Nos últimos anos estudos efectuados com frutos vermelhos têm revelado a sua riqueza em compostos bioactivos,sobretudo devido a presença de compostos polifenólicos. Neste trabalho foram avaliados os sub-produtos da ginja (Prunus cerasus L.) resultantes da indústria do licor de Óbidos, através da determinação do seu conteúdo em compostos fenólicos totais e, em particular, em antocianinas. As amostras de folhagens (pedúnculos e folhas) e de bagaço (película e película+caroço) foram extraídas por maceração através de 2 métodos de extracção diferentes, utilizando o etanol e o metanol como solventes. Na generalidade observaram-se diferenças significativas entre os teores em fenóis totais e antocianinas, quer dos extractos obtidos a partir de diferentes fracções (folhagem ou bagaço), quer dos extractos obtidos a partir da mesma fracção mas utilizando diferentes solventes. Os extractos das folhagens apresentaram valores de fenóis totais mais elevados que os extractos de bagaço. O metanol foi o solvente mais eficaz na extracção dos fenóis totais e das antocianinas para todas as amostras em estudo. Para as amostras de bagaço as amostras sem caroço (amostras de película) foram as que apresentaram valores mais elevados de fenóis totais e de antocianinas. Os resultados obtidos apontam no sentido dos sub-produtos agro-industriais analisados poderem constituir uma promissora fonte de compostos polifenólicos de custo reduzido, com potencial para aplicação em formulações dermocosméticas. Estudos futuros serão efectuados para avaliar o potencial destes resíduos como ingrediente antioxidante.

Agricultural policy liberalisation, concern about unhealthy diets and growing recognition of the importance of sustainable land use have fostered interest in the development of competitive food chains based around products that are beneficial to the rural environment. We review the potential for foods with enhanced health attributes based on alternative varieties/breeds and production systems to traditional agriculture which has been predominantly motivated by yields. We concentrate on soft fruit, which is an important source of polyphenols, and grazing livestock systems that have the potential for improving fatty acid profiles in meat products and find there to be clear scientific potential, but limited research to date. Consumer research suggests considerable acceptance of such products and willingness to pay sufficient to cover additional production costs. Purchase of such foods could have major implications for agricultural land use and the rural environment. There is little research to date on specific healthier food products, but spatially explicit models are being developed to assess land use and environmental implications of changing demand and husbandry methods.

Platelets play a substantial role in cardiovascular disease, and for many years there has been a search for dietary components that are able to inhibit platelet function and therefore decrease the risk of cardiovascular disease. Platelets can be inhibited by alcohol, dietary fats and some antioxidants, including a group of compounds, the polyphenols, found in fruits and vegetables. A number of these compounds have been shown to inhibit platelet function both in vitro and in vivo. In the present study the effects of the hydroxycinnamates and the flavonoid quercetin on platelet activation and cell signalling in vitro were investigated. The hydroxycinnamates inhibited platelet function, although not at levels that can be achieved in human plasma by dietary intervention. However, quercetin inhibited platelet aggregation at levels lower than those previously reported. Quercetin was also found to inhibit intracellular Ca mobilisation and whole-cell tyrosine protein phosphorylation in platelets, which are both processes essential for platelet activation. The effect of polyphenols on platelet aggregation in vivo was also investigated. Twenty subjects followed a low-polyphenol diet for 3 d before and also during supplementation. All subjects were supplemented with a polyphenol-rich meal every lunchtime for 5 d. Platelet aggregation and plasma flavonols were measured at baseline and after 5 d of dietary supplementation. Total plasma flavonoids increased significantly after the dietary intervention period (P = 0.001). However, no significant changes in ex vivo platelet aggregation were observed. Further investigation of the effects of individual polyphenolic compounds on platelet function, both in vitro and in vivo, is required in order to elucidate their role in the relationship between diet and the risk of cardiovascular disease.

There is increasing evidence to suggest that neuroinflammatory processes contribute to the cascade of events that lead to the progressive neuronal damage observed in neurodegenerative disorders such as Parkinson’s disease and Alzheimer’s disease. Therefore, treatment regimes aimed at modulating neuroinflammatory processes may act to slow the progression of these debilitating brain disorders. Recently, a group of dietary polyphenols known as flavonoids have been shown to exert neuroprotective effects in vivo and in neuronal cell models. In this review we discuss the evidence relating to the modulation of neuroinflammation by flavonoids. We highlight the evidence which suggests their mechanism of action involves: 1) attenuation of the release of cytokines, such as interleukin-1β (IL-1β) and tumor necrosis factor-alpha (TNF-α); 2) an inhibitory action against inducible nitric oxide synthase (iNOS) induction and subsequent nitric oxide (NO•) production; 3) inhibition of the activation of NADPH oxidase and subsequent reactive oxygen species generation; 4) a capacity to down-regulate the activity of pro-inflammatory transcription factors such as nuclear factor-κB (NF-κB); and 5) the potential to modulate signalling pathways such as mitogen-activated protein kinase (MAPK) cascade. We also consider the potential of these dietary compounds to represent novel therapeutic agents by considering their metabolism in the body and their ability to access the brain via the blood brain barrier. Finally, we discuss future areas of study which are necessary before dietary flavonoids can be established as therapeutic agents against neuroinflammation.

There is increasing interest in the ability of diets rich in polyphenols to modulate age-related diseases and promote healthy ageing. We have conducted a pilot experiment with eight tomato varieties to correlate the total antioxidant capacity of the tomato variants with the specific constituent flavonoids present. A strong correlation was observed with the flavonol rhamnoglucoside rutin but not with other flavonoids, such as naringenin chalcone, or hydroxycinnamates, such as chlorogenic, which are also present in the tomato. To test the rigor of this correlation a second study was undertaken with a further 37 tomato varieties selected for low, medium and high rutin levels. We show that the flavonol rutin contributes to the greatest extent to the antioxidant capacity of tomatoes and suggest that this flavonoid may be a useful target for up-regulation in tomatoes in order to improve their antioxidant status.

Flavonoids have been proposed to act as beneficial agents in a multitude of disease states, including cancer, cardiovascular disease, and neurodegenerative disorders. The biological effect of these polyphenols and their in vivo circulating metabolites will ultimately depend on the extent to which they associate with cells, either by interactions at the membrane or more importantly their uptake. This review summarises the current knowledge on the cellular uptake of flavonoids and their metabolites with particular relevance to further intracellular metabolism and the generation of potential new bioactive forms. Uptake and metabolism of the circulating forms of flavanols, flavonols, and flavanones into cells of the skin, the brain, and cancer cells is reviewed and potential biological relevance to intracellular formed metabolites is discussed.

Emerging evidence suggests that a group of dietary-derived phytochemicals known as flavonoids are able to induce improvements in memory acquisition, consolidation, storage and retrieval. These low molecular weight polyphenols are widespread in the human diet, are absorbed to only a limited degree and localise in the brain at low concentration. However, they have been found to be highly effective in reversing age-related declines in memory via their ability to interact with the cellular and molecular architecture of the brain responsible for memory. These interactions include an ability to activate signalling pathways, critical in controlling synaptic plasticity, and a potential to induce vascular effects capable of causing new nerve cell growth in the hippocampus. Their ability to activate the extracellular signal-regulated kinase (ERK1/2) and the protein kinase B (PKB/Akt) signalling pathways, leading to the activation of the cAMP response element-binding protein (CREB), a transcription factor responsible for increasing the expression of a number of neurotrophins important in de. ning memory, will be discussed. How these effects lead to improvements in memory through induction of synapse growth and connectivity, increases in dendritic spine density and the functional integration of old and new neurons will be illustrated. The overall goal of this critical review is to emphasize future areas of investigation as well as to highlight these dietary agents as promising candidates for the design of memory-enhancing drugs with relevance to normal and pathological brain ageing (161 references).
