This thesis investigated well-ordered block copolymer (BCP) thin film characteristics and their use for nanoscale pattern formation using a series of polystyrene-block-polymethylmethacrylate (PS-b-PMMA), polystyrene-blockpolydimethylsiloxane (PS-b-PDMS) and polystyrene-block-poly(ethylene oxide) (PS-b-PEO) systems of various molecular weights. BCP thin films, which act as an ‘on-chip’ etch mask and material templates, are highly promising self-assembling process for future scalable nanolithography. Unlike conventional BCP processing methods, the work in this thesis demonstrates that well-ordered patterns can be achieved in a few seconds compared to several hours by use of a non-conventional microwave assisted technique. As a result, well-ordered BCP nanoscale structures can be developed in industry appropriate periods facilitating their incorporation into current technologies. An optimised and controlled plasma dry etch process was used for successful pattern transfer to the underlying silicon substrate. Long range ordered BCP templates were further modified by selective metal inclusion technique to form a hard mask template towards fabrication of high aspect ratio nanopillars and nanowires. The work described here is centred on how these templates might be used to generate function at substrate surfaces. Herein we describe a number of innovations which might allow their successful uptake in a number of applications.

This paper reports a new strategy, recursive directional ligation by plasmid reconstruction (PRe-RDL), to rapidly clone highly repetitive polypeptides of any sequence and specified length over a large range of molecular weights. In a single cycle of PRe-RDL, two halves of a parent plasmid, each containing a copy of an oligomer, are ligated together, thereby dimerizing the oligomer and reconstituting a functional plasmid. This process is carried out recursively to assemble an oligomeric gene with the desired number of repeats. PRe-RDL has several unique features that stem from the use of type IIs restriction endonucleases: first, PRe-RDL is a seamless cloning method that leaves no extraneous nucleotides at the ligation junction. Because it uses type IIs endonucleases to ligate the two halves of the plasmid, PRe-RDL also addresses the major limitation of RDL in that it abolishes any restriction on the gene sequence that can be oligomerized. The reconstitution of a functional plasmid only upon successful ligation in PRe-RDL also addresses two other limitations of RDL: the significant background from self-ligation of the vector observed in RDL, and the decreased efficiency of ligation due to nonproductive circularization of the insert. PRe-RDL can also be used to assemble genes that encode different sequences in a predetermined order to encode block copolymers or append leader and trailer peptide sequences to the oligomerized gene.

Free-radical polymerization of methyl methacrylate and styrene using conventional organic initiators in the room temperature ionic liquid, 1-butyl-3-methylimidazolium hexafluorophosphate ([ C(4)mim][PF6]) is rapid and produces polymers with molecular weights up to 10x higher than from benzene; both polymerization and isolation of products were achieved without using VOCs, offering economic as well as environmental advantages.

Photodynamic therapy (PDT) is a medical treatment in which a combination of a photosensitising drug and visible light causes destruction of selected cells. Due to the lack of true selectivity of preformed photosensitisers for neoplastic tissue and their high molecular weights, PDT of superficial skin lesions has traditionally been mediated by topical application of the porphyrin precursor 5-aminolevulinic acid (ALA). Objective: This article aims to review the traditional formulation-based approaches taken to topical delivery of ALA and discusses the more innovative strategies investigated for enhancement of PDT mediated by topical application of ALA and preformed photosensitisers. Methods: All of the available published print and online literature in this area was reviewed. As drug delivery of agents used in PDT is still something of an emerging field, it was not necessary to go beyond literature from the last 30 years. Results/conclusion: PDT of neoplastic skin lesions is currently based almost exclusively on topical application of simple semisolid dosage forms containing ALA or its methyl ester. Until expiry of patents on the current market-leading products, there is unlikely to be a great incentive to engage in design and evaluation of innovative formulations for topical PDT, especially those containing the more difficult-to-deliver preformed photosensitisers.

The palladium-catalyzed copolymerization of styrene and CO in an ionic liquid solvent, 1-hexylpyridinium bis(trifluoromethanesulfonyl) imide, gave improved yields and increased molecular weights compared to polymerizations run in methanol.

There is a gulf between the enormous information content of the various genome projects and the understanding of the life of the parasite in the host. In vitro studies with adult Schistosoma mansoni using several substrates suggest that the excretory system contains both P-glycoproteins and multiresistance proteins. If both these families of protein were active in vivo, they could regulate parasite metabolism and be responsible for the excretion of drugs. During skin penetration, membrane-impermeant molecules of a wide range of molecular weights can be taken into the cercaria and schistosomulum through the nephridiopore, through the surface membrane or through both. We speculate that this uptake process might stimulate novel signalling pathways involved in growth and development.

Predicable and controlled degradation is not only central to the accurate delivery of bioactive agents and drugs, it also plays a vital role in key aspects of bone tissue engineering. The work addressed in this paper investigates the utilisation of e-beam irradiation in order to achieve a controlled (surface) degradation profile. This study focuses on the modification of commercially and clinically relevant materials, namely poly(L-lactic acid) (PLLA), poly(L-lactide-hydroxyapatite) (PLLA-HA), poly(L-lactide-glycolide) co-polymer (PLG) and poly(L-lactide-DL-lactide) co-polymer (PLDL). Samples were subjected to irradiation treatments using a 0.5 MeV electron beam with delivered surface doses of 150 and 500 kGy. In addition, an acrylic attenuation shield was used for selected samples to control the penetration of the e-beam. E-beam irradiation induced chain scission in all polymers, as characterized by reduced molecular weights and glass transition temperatures (T-g). Irradiation not only produced changes in the physical properties of the polymers but also had associated effects on surface erosion of the materials during hydrolytic degradation. Moreover, the extent to which both mechanical and hydrolytic degradation was observed is synonymous with the estimated penetration of the beam (as controlled by the employment of an attenuation shield). Adult and 3-week-old juvenile Fasciola hepatica were examined for the presence of the cytoskeletal protein actin. Techniques of direct fluorescence using fluorescein isothiocyanate (FITC)-phalloidin and of indirect immunofluorescence using a monoclonal anti-actin antibody (MAA) demonstrated actin in the testes, sub-tegumental and gut musculature, tegumental cell bodies and tegumental spines. In contrast, polyclonal anti-actin antibody (PAA) revealed immunostaining only in the vitellaria. Effective removal of the tegument with 1 % (w/v) sodium dodecyl sulphate (SDS) was confirmed by scanning electron microscopy (SEM), and this enabled immunoblotting of whole fluke and tegumental fractions with and without spines. Whole fluke fractions produced three bands corresponding to molecules exhibiting relative molecular weights of 43, 28 and 15 kDa, respectively, whereas the tegumental fraction with spines revealed a single band corresponding to 15 kDa in size. The fraction without spines displayed no bands. The present study localised actin in a number of different tissue types within the liver fluke. Using MAA, three forms of actin have been identified in the whole fluke and a single one in the tegumental spines.

The excreted/secreted proteinases of adult and juvenile Fasciola hepatica maintained in vitro were found to hydrolyse the fluorogenic substrates Cbz-Phe-Arg- and Cbz-Arg-Arg-NHMec. This activity was demonstrated to have a classical cysteine proteinase inhibitor profile, with turn-over of both substrates being blocked by pre-incubation with E64 and peptidyl diazomethanes. The Cbz-Arg-Arg-NHMec hydrolysing activity of the mature fluke exhibited an alkaline stability not characteristic of its mammalian lysosomal counterparts. Further, the biotinylated affinity reagents biotin-Phe-Ala CHN2 and biotin-Phe-Cys(SBzyl)-CHN2 were used to label and characterize these cysteine proteinases in terms of apparent molecular weight and subsite specificity. Adult fluke media were found to contain four species of molecular weights 66, 58, 50 and 25-26 kDa; juvenile media contained three species of molecular weights 66, 54 and 25-26 kDa. The major 25-26 kDa cysteine proteinase common to both stages was shown to have a subsite specificity similar to that of mammalian cathepsin B.

Six amphiphilic star copolymers comprising hydrophilic units of 2-(dimethylamino)ethyl methacrylate (DMAEMA) and hydrophobic units of methyl methacrylate (MMA) were prepared by the sequential group transfer polymerization (GTP) of the two comonomers and ethylene glycol dimethacrylate (EGDMA) cross-linker. Four star-block copolymers of different compositions, one miktoarm star, and one statistical copolymer star were synthesized. The molecular weights (MWs) and MW distributions of all the star copolymers and their linear homopolymer and copolymer precursors were characterized by gel permeation chromatography (GPC), while the compositions of the stars were determined by proton nuclear magnetic resonance (H-1 NMR) spectroscopy. Tetrahydrofuran (THF) solutions of all the star copolymers were characterized by static light scattering to determine the absolute weight-average MW ((M) over bar (w)) and the number of arms of the stars. The R, of the stars ranged between 359,000 and 565,000 g mol(-1), while their number of arms ranged between 39 and 120. The star copolymers were soluble in acidic water at pH 4 giving transparent or slightly opaque solutions, with the exception of the very hydrophobic DMAEMA(10)-b-MMA(30)-star, which gave a very opaque solution. Only the random copolymer star was completely dispersed in neutral water, giving a very opaque solution. The effective pKs of the copolymer stars were determined by hydrogen ion titration and were found to be in the range 6.5-7.6. The pHs of precipitation of the star copolymer solutions/dispersions were found to be between 8.8-10.1, except for the most hydrophobic DMA-EMA(10)-b-MMA(30)-Star, which gave a very opaque solution over the whole pH range. An acid-labile dimethaerylate acetal cross-linker,di(methacryloyloxy-l-ethoxy)methane(DMOEM), was synthesized by the reaction of 2-hydroxyethyl methacrylate and paraformaldehyde using p-toluenesulfonic acid and toluene as catalyst and solvent, respectively. Group transfer polymerization was employed to use this cross-linker in the preparation of nine hydrolyzable polymer structures: one neat cross-linker network, one randomly cross-linked network of methyl methacrylate (MMA), and seven star-shaped polymers of MMA. Gel permeation chromatography (GPC) in tetrahydrofuran (THF) confirmed the narrow molecular weight distributions of the linear polymer precursors to the stars and demonstrated the increase in molecular weight upon the addition of cross-linker for the formation of star-shaped polymers. Characterization of the star polymers in THF using static light scattering and GPC showed that the molecular weights and the number of arms of each star polymer increased with an increase in the molar ratio of cross-linker to initiator and with a decrease in the molar ratio of monomer to initiator. The star polymers with DMOEM cores bore a smaller number of arms than those cross-linked with the non-hydrolyzable commercial cross-linker ethylene glycol dimethacrylate due to the bulkier structure of DMOEM. All DMOEM-containing polymer networks and star polymers were completely hydrolyzed within 48 h using hydrochloric acid in THF.

Photodynamic therapy can be used in the treatment of pre-malignant and malignant diseases. It offers advantages over other therapies currently used in the treatment of skin lesions including avoidance of damage to surrounding tissue and minimal or no scarring. Unfortunately, systemic delivery of photosensitising agents can result in adverse effects, such as prolonged cutaneous photosensitivity; while topical administration lacks efficacy in the clearance of deeper skin lesions and those with a thick overlying keratotic layer. Therefore, enhancement of conventional photosensitiser delivery is desired. However, the physicochemical properties of photosensitising agents, such as extreme hydrophilicity or lipophilicity and large molecular weights make this challenging. This paper reviews the potential of microneedles as a viable method to overcome these delivery-limiting physicochemical characteristics and discusses the current benefits and limitations of solid, dissolving and hydrogel-forming microneedles. Clinical studies in which microneedles have successfully improved photodynamic therapy are also discussed, along with benefits which microneedles offer, such as precise photosensitiser localisation, painless application and reduction in waiting times between photosensitiser administration and irradiation highlighted.

Neuropeptides such as neuropeptide Y (NPY) and vasoactive intestinal polypeptide (VIP) have been shown by our research group to be present in human dental pulp tissue. Neuropeptides cannot cross cell membranes and therefore to exert their biological effects they must bind to selected receptors on the surface of target cell membranes. However, the expression of receptor proteins for NPY and/or VIP have yet to be reported in human pulp tissue. The presence of neuropeptide receptors can be conveniently determined by Western blotting using specific anti-receptor antibodies. Objectives: The aim of this work was to identify the presence of the NPY Y1 receptor and the VIP receptor VPAC1 in human dental pulp tissue from both intact and carious teeth using Western blotting. Methods: Pulp tissue was collected from both intact and carious teeth and membrane preparations from these tissues were then subject to sodium dodecyl sulphate gel electrophoresis (SDS-PAGE), transferred to nitrocellulose and probed with specific antibodies to either the NPY Y1 receptor or the VPAC1 receptor. Results: Individual Western blotting experiments revealed the presence of immunoreactive bands corresponding to the known molecular weights of the NPY Y1 and VPAC1 receptor proteins in both intact and carious pulp samples. Conclusions: Demonstration of the presence of NPY Y1 and VPAC1 receptor protein expression in pulpal tissue from intact and carious teeth provides further support for the roles of these neuropeptides in pulpal health and disease.

O interesse crescente das membranas inorgânicas deve-se à potencial aplicação em novas áreas de investigação e da indústria, e em alternativa a operações mais convencionais. Em particular, as membranas de titanossilicatos oferecem vantagens importantes sobre as de zeólitos, pois podem ser sintetizadas sem agentes estruturantes orgânicos, para evitar a calcinação subsequente usualmente responsável por defeitos irreversíveis, exibem novas possibilidades de substituição isomórfica da matriz, permitindo um ajuste mais fino das propriedades catalíticas e de adsorção, e são capazes de separar misturas com base em diferenças de afinidade e tamanho molecular (efeito de peneiro). Os objectivos principais deste trabalho foram: i) a caracterização dinâmica de membranas do tipo zeolítico sintetizadas no Laboratório Associado CICECO, realizando-se experiências de permeação com gases puros e misturas; ii) o desenvolvimento e validação de novos modelos para a transferência de massa multicomponente através de membranas porosas pela abordagem de Maxwell-Stefan, tendo em conta os mecanismos específicos encontrados, particularmente a contribuição por difusão superficial; e iii) a modelação dos pontos experimentais medidos, bem como dados compilados da literatura. De forma a realizar os ensaios de permeação, desenhou-se, montou-se e testou-se uma instalação experimental. Para gases puros, os objectivos principais foram a medição de permeâncias a temperatura constante, por variação da pressão transmembranar r ( ΔP ), e de permeâncias a temperatura programada, conduzidas a ΔP constante. Seguidamente, calcularam-se as selectividades ideais. Em relação a misturas, a determinação de selectividades reais requer as fracções molares no permeado e no retido. Na globalidade, estudaram-se três suportes diferentes (aço inoxidável e α − alumina) e dezanove membranas de AM-3, ETS-10, ZSM-5 e zeólito 4A, utilizando-se H2, He, N2, CO2, e O2. A primeira avaliação exploratória da qualidade das membranas foi feita permeando azoto à temperatura ambiente. Assim, permeâncias superiores a 10−6 mol/m2s.Pa evidenciavam defeitos grosseiros, levando-nos a efectuar cristalizações adicionais sobre as primeiras camadas. Este procedimento foi implementado com oito membranas. Um trabalho experimental mais detalhado foi conduzido com cinco membranas. Membranas com curvas permeância-temperatura ( Π −T ) decrescentes indicam tipicamente transporte viscoso e de Knudsen, i.e. meso e macrodefeitos. Por exemplo, a membrana nº 3 de AM-3 exibiu este comportamento com H2, He, N2 e CO2 puros. A contribuição de Knudsen foi confirmada pela relação linear encontrada entre as permeâncias e o inverso da raiz quadrada da massa molar. O mecanismo viscoso foi também identificado, pois as permeâncias eram inversamente proporcionais à viscosidade do gás ou, atendendo a equações do tipo de Chapman-Enskog, directamente proporcionais a 2 0.5 k d M (onde k d é o diâmetro cinético e M a massa molar). Um comportamento de permeação distinto observou-se com a membrana nº 5 de AM-3. As permeâncias registadas a temperatura programada eram aproximadamente constantes para o N2, CO2 e O2, enquanto com o H2 cresciam significativamente. Conjuntamente elas evidenciam a ocorrência de macro, meso e microdefeitos intercristalinos. O transporte gasoso activado através dos microporos compensa o impacto diminuidor dos meso e macroporos. Ao contrário do N2, CO2 e O2, o pequeno diâmetro do hidrogénio torna-lhe possível permear através dos microporos intracristalinos, o que lhe adiciona um mecanismo de transferência responsável por esse crescimento. No que respeita à difusão superficial, o sistema CO2/ZSM-5 pode ser tomado como um exemplo paradigmático. Uma vez que este zeólito adsorve o CO2, as permeâncias diminuem com o crescimento de ΔP , em virtude de as concentrações no sólido aumentarem de forma não linear e tenderem para a saturação. Os resultados contrastantes obtidos com azoto realçam ainda mais o mecanismo superficial, pois o N2 não é adsorvido e as permeâncias medidas são constantes. Globalmente, as selectividades ideais calculadas ( α* ) variam de cerca de 1 a 4.2. Este parâmetro foi também utilizado para discriminar as melhores membranas, uma vez que baixos valores de α* denotam o escoamento viscoso não-selectivo típico de macrodefeitos. Por exemplo, o H2/CO2 na membrana nº 3 de AM-3 apresentou α* = 3.6 − 4.2 para 40–120ºC, enquanto que na membrana nº 5 de AM-3 originou α* = 2.6 − 3.1. Estes resultados corroboraram as observações anteriores, segundo as quais a membrana nº 5 era melhor do que a nº 3. Alguns ensaios foram realizados com membranas saturadas com água para aumentar a selectividade: as medições mostraram claramente uma melhoria inicial seguida de uma redução consistente de α* com o aumento da temperatura, devido à remoção das moléculas de água responsáveis pela obstrução de alguns poros. Em relação às selectividades reais de misturas contendo hidrogénio, devem ser realizadas mais experiências e a quantificação do hidrogénio deve ser melhorada. No que concerne à modelação, novos factores termodinâmicos de Maxwell- Stefan foram derivados para as isotérmicas mono e multicomponente de Nitta, Langmuir-Freundlich e Toth, tendo sido testadas com dados de equilíbrio e de permeação da literatura. (É importante realçar que só estão publicadas equações para Langmuir e Dual-Site Langmuir de componentes puros e misturas). O procedimento de validação adoptado foi exigente: i) as isotérmicas multicomponente foram previstas a partir das de gás puro; ii) os parâmetros de difusão dos componentes puros foram ajustados a dados de permeação de cada gás; iii) depois, as difusividades cruzadas de Maxwell- Stefan foram estimadas pela relação de Vignes; finalmente, v) as novas equações foram testadas usando-se estes parâmetros, tendo sido capazes de estimar com sucesso fluxos binários. Paralelamente ao enfoque principal do trabalho, derivou-se um novo modelo para permuta iónica em materiais microporosos baseado nas equações de Maxwell-Stefan. Este foi validado com dados experimentais de remoção de Hg2+ e Cd2+ de soluções aquosas usando ETS-4. A sua capacidade preditiva foi também avaliada, sendo possível concluir que se comporta muito bem. Com efeito, conseguiram-se boas previsões com parâmetros optimizados a partir de conjuntos de dados independentes. Este comportamento pode ser atribuído aos princípios físicos sólidos da teoria de Maxwell-Stefan.
