O presente experimento foi realizado com o objetivo de avaliar o desempenho de cordeiros em confinamento alimentados com dieta exclusiva de concentrado com diferentes porcentagens de proteína. Foram utilizados 24 cordeiros mestiços da raça Santa Inês, não castrados, com aproximadamente 180 dias de idade, peso vivo médio de 25kg, confinados, num delineamento experimental inteiramente ao acaso, com quatro tratamentos e seis repetições. O consumo voluntário de matéria seca, matéria orgânica, proteína bruta e nutrientes digestíveis totais, além do ganho de peso diário e total, da conversão e da eficiência alimentar dos cordeiros, não foram influenciados (P>0,05) pela porcentagem de proteína na dieta. As dietas com 18 e 20% de proteína bruta apresentaram menores custos de produção, entretanto o menor custo foi obtido com a ração contendo 18% de PB.

Objetivou-se avaliar a ingestão de matéria seca, o ganho de peso diário, a conversão alimentar, o peso vivo ao abate e o período de confinamento em cordeiros Santa Inês puros e ½Dorset ½Santa Inês, alimentados com dietas isoenergéticas (76,59% de NDT) e isoprotéicas (17,48% de PB) contendo diferentes fontes de óleo vegetal (óleos de soja, canola e linhaça) e uma dieta controle (sem inclusão de óleo vegetal). A relação volumoso:concentrado foi de 30:70 e utilizou-se feno de aveia como volumoso. Realizou-se também um ensaio de digestibilidade, utilizando quatro cordeiros não-castrados, distribuídos em delineamento quadrado latino, avaliando-se ingestão, excreção fecal e digestibilidade total dos nutrientes das rações. A ingestão de matéria seca, expressa em porcentagem do peso vivo, foi menor nos cordeiros que receberam dieta contendo óleo de canola que naqueles que receberam dieta controle. Porém, todas as rações proporcionaram ganhos de peso e conversão alimentar satisfatórios. Os valores de digestibilidade total da matéria seca (76,02%) e matéria orgânica (76,82%) da dieta controle foram superiores aos da dieta contendo óleo de linhaça (72,11% e 72,97%, respectivamente), embora não tenham diferido das dietas contendo óleos de soja (72,94 e 73,71%) e canola (73,45 e 74,25%). A digestibilidade do extrato etéreo foi menor na dieta controle (84,02%), enquanto as demais dietas apresentaram valor médio de 91,98%. Os óleos vegetais reduziram a digestibilidade da matéria seca e da matéria orgânica, não afetando a ingestão e digestão dos demais nutrientes.

Changes in Ca-45 uptake and insulin secretion in response to glucose, leucine, and arginine were measured in isolated islets derived from 4-week-old rats born of mothers maintained with normal protein (NP, 17%) or low protein (LP, 6%) diet during pregnancy and lactation. Glucose provoked a dose-dependent stimulation of insulin secretion in both groups of islets, with basal (2.8 mmol/L glucose) and maximal release (27.7 mmol/L glucose) significantly reduced in LP compared with NP islets. In the LP group the concentration-response curve to glucose was shifted to the right compared with the NP group, with the half-maximal response occurring at 16.9 and 13.3 mmol/L glucose, respectively. In LP islets, glucose-induced first and second phases of insulin secretions were drastically reduced. In addition, insulin response to individual amino acids, or in association with glucose, was also significantly reduced in the LP group compared with NP islets. Finally, in LP islets the Ca-45 uptake after 5 minutes or 90 minutes of incubation (which reflect mainly the entry and retention, respectively, of Ca2+), was lower than in NP islets. These data indicate that in malnourished rats both initial and sustained phases of insulin secretion in response to glucose were reduced. This poor secretory response to nutrients seems to be the consequence of an altered Ca2+ handling by malnourished islet cells. (J. Nutr. Biochem. 10:37-43, 1999) (C) Elsevier B.V. 1999. Purine nucleoside phosphorylase (PNP) catalyzes the phosphorolysis of the N-ribosidic bonds of purine nucleosides and deoxynucleosides. A genetic deficiency due to mutations in the gene encoding for human PNP causes T-cell deficiency as the major physiological defect. Inappropriate activation of T-cells has been implicated in several clinically relevant human conditions such as transplant tissue rejection, psoriasis, rheumatoid arthritis, lupus, and T-cell lymphomas. Human PNP is therefore a target for inhibitor development aiming at T-cell immune response modulation. In addition, bacterial PNP has been used as reactant in a fast and sensitive spectrophotometric method that allows both quantitation of inorganic phosphate (Pi) and continuous assay of reactions that generate P i such as those catalyzed by ATPases and GTPases. Human PNP may therefore be an important biotechnological tool for P i detection. However, low expression of human PNP in bacterial hosts, protein purification protocols involving many steps, and low protein yields represent technical obstacles to be overcome if human PNP is to be used in either high-throughput drug screening or as a reagent in an affordable P i detection method. Here, we describe PCR amplification of human PNP from a liver cDNA library, cloning, expression in Escherichia coli host, purification, and activity measurement of homogeneous enzyme. Human PNP represented approximately 42% of total soluble cell proteins with no induction being necessary to express the target protein. Enzyme activity measurements demonstrated a 707-fold increase in specific activity of cloned human PNP as compared to control. Purification of cloned human PNP was achieved by a two-step purification protocol, yielding 48 mg homogeneous enzyme from 1 L cell culture, with a specific activity value of 80 U mg -1. © 2002 Elsevier Science (USA). The effect of protein or energy restriction during the second week post-hatching on body weight gain and femur development of broiler chickens reared at different environmental temperatures (18°C, 25°C and 33°C) was studied. From 1 to 7 days of age and after a restriction period broilers were fed on a control diet with 2850 kcal ME/kg and 20% crude protein. From 8 to 14 days of age, two groups of broilers were fed on restricted energy and protein diets with 2565kcal ME/kg and 20% of crude protein or 2850kcal ME/kg and 15% of crude protein, respectively. At 14, 21, 28, 35 and 42 days, the bones were weighed and the length and width of bones measured. The protein restriction reduced the body weight gain and the diameter of bone at second week of life. After 21 days of age no differences between treatments for these traits were observed. The body weight gain and femur growth were not affected by energy restriction. The high temperature (33°C) reduced weight gain and femur diameter from 21 to 42 days of age, and femur length at 42 days of age. The protein restriction at second week or the high environmental temperature after 21 st day of life decreased body weight gain and femur growth of broiler chickens.

Nitrogen limitation in the common bean (Phaseolus vulgaris L.) produces reduced productivity and seeds with low protein content and physiological quality. We assessed the effects of nitrogen fertilizer side dressing on the protein content and physiological quality of the seeds of P. vulgaris L. cultivars (cv.) IPR Juriti and Pérola grown in plots (soil type = Dystrophic Red Latosol) using a no-tillage system under a thick mulch of millet residues at the Experimental Station of UNESP-Ilha Solteira campus, located in Selvíria, MS (Köppen climate type = Aw) during autumn (March/June) and winter (June/September) 2005. For each cultivar, a randomized block experimental design was used with four replicates and factorially arranged treatments equivalent to 0, 30, 60, 90 and 120 kg.ha-1 added nitrogen (as urea, containing 45% of N) applied as a side dressing during the V4-3 and V4-6 phenological stage corresponding to the 3rd and 6th completely opened trifoliolate leaf on the main stem. Supplementation with up to 120 kg.ha-1 nitrogen promoted a greater increase in crude protein at V4-3 (unsupplemented = 17.6% and 16.3%; 120 kg.ha-1 N = 24.1% and 22.3% for cv. IPR Juriti and Pérola, respectively) than at V4-6 (unsupplemented = 19.2% and 18.3%; 120 kg.ha-1 N = 21.3% and 20.3% for cv. IPR Juriti and Pérola, respectively). About 90% of the crude protein from cv. IPR Juriti was composed of soluble protein compared to 72% for the cv. Pérola. Albumins and globulins represented about 80% of the soluble protein and prolamins were lower at 0.6%. In conclusion, nitrogen fertilization up to 120 kg.ha-1 applied as a side dressing at the V4-3 phenological stage in no-tillage under a thick mulch of millet promoted a greater increase of crude protein in common bean seeds than at the V4-6 stage. The highest accumulation of soluble protein occurred at 90 kg.ha-1 applied nitrogen without having a significant influence on the physiological quality of the seeds.
