Patterns of spatio-temporal distribution of Brachyura are determined by the interaction among life history traits, inter and intraspecific relationships, as well as by the variation of abiotic factors. This study aimed to characterize patterns of spatio-temporal distribution of Persephona lichtensteinii, Persephona mediterranea and Persephona punctata in two regions of the northern coast of Sao Paulo State, southeastern region of Brazil. Collections were done monthly from July 2001 to June 2003 in Caraguatatuba and Ubatuba, using a shrimp fishery boat equipped with double-rig nets. The patterns of species distribution were tested by means of redundancy analysis (RDA) and generalized linear mixed models (GLMM) in relation to the recorded environmental factors (BT: bottom temperature, BS: bottom salinity, OM: organic matter and granulometry (Phi)). The most influent environmental factor over the species distribution was the Phi, and the ascendant order of influence was P. lichtensteinii, P. punctata and P. mediterranea. The greater abundance of P. mediterranea showed a conservative pattern of distribution for the genus in the sampled region. The greater occurrence of P. punctata and P. lichtensteinii, in distinct transects than those occupied by P. mediterranea, seems to be a strategy to avoid competition among congeneric species, which is related to the substratum specificity.

We investigated the influence of environmental factors in spatial and temporal distribution of the seabob shrimp Xiphopenaeus kroyeri in Santos Bay and São Vicente Estuary, state of São Paulo, Brazil. Monthly samples were obtained, from May 2008 through April 2010, from four locations in the estuary and four in the bay. No individual was collected in the estuary and this was attributed to the low salinity means recorded in this environment. We collected 109,153 individuals in the bay and there was no difference in abundance between the two years comprised by the study period. The similarity in spatial distribution can be related to sediment grain size that in all sampling locations showed great amount of very fine sand. The largest amount of reproductive females was obtained in early 2010, when temperature was high, and this could have increased the juvenile recruitment in April 2010. According to our results, the distribution of X. kroyeri in the study area is influenced by temperature, which is related to reproduction, and salinity, limiting the entrance of individuals in the estuarine region.

New strategies to fulfill craniofacial bone defects have gained attention in recent years due to the morbidity of autologous bone graft harvesting. We aimed to evaluate the in vivo efficacy of bone tissue engineering strategy using mesenchymal stem cells associated with two matrices (bovine bone mineral and α-tricalcium phosphate), compared to an autologous bone transfer. A total of 28 adult, male, non-immunosuppressed Wistar rats underwent a critical-sized osseous defect of 5 mm diameter in the alveolar region. Animals were divided into five groups. Group 1 (n = 7) defects were repaired with autogenous bone grafts; Group 2 (n = 5) defects were repaired with bovine bone mineral free of cells; Group 3 (n = 5) defects were repaired with bovine bone mineral loaded with mesenchymal stem cells; Group 4 (n = 5) defects were repaired with α-tricalcium phosphate free of cells; and Group 5 (n = 6) defects were repaired with α-tricalcium phosphate loaded with mesenchymal stem cells. Groups 2-5 were compared to Group 1, the reference group. Healing response was evaluated by histomorphometry and computerized tomography. Histomorphometrically, Group 1 showed 60.27% ± 16.13% of bone in the defect. Groups 2 and 3 showed 23.02% ± 8.6% (p = 0.01) and 38.35% ± 19.59% (p = 0.06) of bone in the defect, respectively. Groups 4 and 5 showed 51.48% ± 11.7% (p = 0.30) and 61.80% ± 2.14% (p = 0.88) of bone in the defect, respectively. Animals whose bone defects were repaired with α-tricalcium phosphate and mesenchymal stem cells presented the highest bone volume filling the defects; both were not statistically different from autogenous bone.

Luciferid shrimps have short life spans and a rapid turnover of generations, engage in sequential spawning, and protect their eggs during incubation. This study investigates the ecology of Lucifer faxoni Borradaile, 1915 in the littoral zone, Ubatuba region, São Paulo. Sampling was conducted monthly from July 2005 to December 2006 using a Renfro net trawled over a distance of 50 m for a total sampling effort of 50 m² at each station. Nine stations were sampled, ranging from 1 to 15 m deep. Three stations each were grouped into zones 1, 2 and 3 (Z1, Z2 and Z3). Monthly values of salinity, temperature and rainfall were recorded at each station. The pre-buccal somite length (SL) of each specimen was measured. The results showed that in shallower zones (Z1 and Z2), 6306 individuals were captured, whereas in the deeper zone (Z3), 3808 specimens were captured, but no significant differences in SL was detected between the specimens from Z1 and Z2 and those from Z3 (ANOVA, p=0.25). The abundance of shrimps did not differ significantly between seasons (Tukey’s test, p=0.02) except in the spring. The sex ratio differed significantly over the seasons (χ², p<0.05). The results were closely associated with environmental factors with respect to the spatial and seasonal distribution of L. faxoni. Rainfall affected salinity directly, and contributed to the displacement of these shrimps to deeper areas.

Stomach contents were analyzed from 127 Baird’s beaked whales, Berardizls bairdii, taken in coastal waters of Japan. During late July-August of 1985- 1987, 1989, and 1991, 107 samples were collected from off the Pacific coast of Honshu. An additional 20 samples were collected from whales taken in the southern Sea of Okhotsk during late August-September of 1988 and 1989. Prey identification using fish otoliths and cephalopod beaks revealed the whales fed primarily on deep-water gadiform fishes and cephalopods in both regions. Prey species diversity and the percentage of cephalopods and fish differed between the two regions. Off the Pacific coast of Honshu the whales fed primarily on benthopelagic fishes (81.8%) and only 18.0% on cephalopods. Eight species of fish representing two families, the codlings (Moridae) and the grenadiers (Macrouridde), collectively made up 81.3% of the total. Thirty species of cephalopods representing 14 families made up 12.7%. In the southern Sea of Okhotsk, cephalopods accounted for 87.1% of stomach contents. The families Gonatidae and Cranchiidae were the predominant cephalopod prey, accounting for 86.7% of the diet. Gadiform fish accounted for only 12.9% of the diet. Longfin codling, Laernonma longipes, was the dominant fish prey in both regions. Depth distribution of the two commonly consumed fish off the Pacific coast of Honshu indicate the whales in this region fed primarily at depths ranging from 800 to 1,200 m.

The detection of pertinent biomarkers has the potential provide an early indication of disease progression before considerable damage has been incurred. A decrease in an individual’s sensitivity to insulin, which may be quantified as the ratio of insulin to glucose in the blood after a glucose pulse, has recently been reported as an early predictor of insulin-dependent diabetes mellitus. Routine measurement of insulin levels is therefore desirable in the care of diabetes-prone individuals. A rapid, simple, and reagentless method for insulin detection would allow for wide-spread screenings that provide earlier signs of diabetes onset. The aim of this thesis is to develop a folding-base electrochemical sensor for the detection of insulin. The sensor described herein consists of a DNA probe immobilized on a gold disc electrode via an alkanethiol linker and embedded in an alkanethiol self-assembled monolayer. The probe is labeled with a redox reporter, which readily transfers electrons to the gold electrode in the absence of insulin. In the presence of insulin, electron transfer is inhibited, presumably due to a binding-induced conformational or dynamic change in the DNA probe that significantly alters the electron-tunneling pathway. A 28-base segment of the insulin-linked polymorphic region that has been reported to bind insulin with high affinity serves as the capture element of the DNA probe. Three probe constructs that vary in their secondary structure and position of the redox label are evaluated for their utility as insulin-sensing elements on the electrochemical platform. The effects of probe modification on secondary structure are also evaluated using circular dichroism spectroscopy.

In southeastern Brazil, slipper lobsters (Scyllarides deceptor and S. brasiliensis) are caught by fleets trawling for pink shrimp (Farfantepenaeus brasiliensis and F. paulensis) and pots-and-traps fishing for octopuses (Octopus vulgaris). Eight hundred fifty-six landings of shrimp trawlers and 28 of the octopus fleet were monitored in the Santos region from May 2006 to April 2007. Additional analysis was performed using a database covering the period from 1999 onwards. This study seeks to identify the recent patterns of exploitation of these lobsters with the goal of improving the way towards fishery sustainability. Scyllarides deceptor was the dominant lobster species with 1032 specimens collected, while only three specimens of S. brasiliensis were identified. The area known as the `Farol do Boi` (23 degrees 01`S, 45 degrees 00`W to 25 degrees 00`S, 45 degrees 40`W at 60-135 m deep) showed the highest Catch Per Unit Effort (CPUE). A General Linearized Model (GLM) was used to investigate the factors influencing variations in CPUE in trawl fleets and led to the conclusion that year, month and depth were the most important factors. We detected a significant decrease in the relative abundance of lobsters in the fishing zone despite relatively low fishing effort. Recommendations to protect the lobster resources include taking special precautions in the natural refuge area of the `Farol do Boi`, as an exclusion zone for trawl fleets, and controlling the use of traps longlines to catch octopuses. The morphology and anatomy of Vitularia salebrosa, a muricid ectoparasitic on other mollusks, are investigated based on study of specimens from western Panama. Distinctive characters of this species include the small size of the buccal mass and radular apparatus, simplification of the odontophore muscles and diminished lateral teeth of the radula; all elongated, narrow proboscis; narrow digestive tract and a differentiable glandular region at the beginning of the posterior esophagus. These traits are consistent with adaptive specialization for an ectoparasitic life history.

This paper provides a paleoenvironmental reconstruction of a Late Quaternary lagoon system in the Jaguaruna region of Santa Catarina state, southern Brazil. Integrated results of bulk sedimentary organic matter characterization (delta C-13, delta N-15 and C/N), microfossil (pollen and diatom) and grain-size analysis from three shallow cores (similar to 2.5m depth) allowed us to propose an evolving paleogeographic scenario in this coastal region for the last ca. 5500 cal a BP. The lagoonal system in this area was more extensive during the mid-Holocene than today, with a gradual and continuous lagoon-sea disconnection until the present. We add to the debate regarding relative sea-level (RSL) variations for the Brazilian coast during the Holocene and discuss the importance of sedimentary dynamics for interpreting changes in coastal ecosystems. The multi-proxy analysis suggests that changes in coastal ecosystems could be directly related to local sedimentary processes, which are not necessarily linked to RSL fluctuations and/or to climatic variations. Among the organisms acting in the natural biological control of tephritids, members of the family Braconidae are the most active form of natural parasite, and in Neotropical regions, members of Opiinae are the main control agents of Anastrepha. The objective of this work was to discover the percentage of parasitism and the species of braconid associated with fruit trees growing in cities on the southern coast of Bahia. During the period of August, 2005 to March, 2008, hosts fruits of fruit flies from several plant species were collected and from the fruits the following species of Anastrepha were obtained: A. fraterculus, A. obliqua, A. bahiensis, A. serpentina, A. sororcula and A. zenildae. Of the total of 838 specimens of braconids, 21.36% were of the species Utetes anastrephae (Viereck), obtained from yellow mombin, carambola, guava, mango and pitanga; 4.42% were of the species Asobara anastrephae (Muesebeck) obtained from the fruits of the yellow mombin, carambola and guava, and only one example of Opius bellus Gahan (0.12%) that came from a guava sample. The species Doryctobracon areolatus (Szepligeti) (74.10%) was predominant and emerged from puparia from all the host fruits collected, probably due to the greater efficiency of this species in locating tephritid larvae. The mean percentage of parasitism by Anastrepha spp. was 4.45%.

Nuclear abnormalities in erythrocytes (NAE) were taken as biomarkers in the catfish Cathorops spixii (Ariidae) sampled in an estuary little affected by human activity (Cananeia) and in three regions (Santos Channel: SC, Santos Bay: SB and Sao Vicente Channel: SVC) of the Santos-Sao Vicente estuary impacted by various anthropogenic activities. Increases in NAE were observed in fish from SC and SVC sampled in the summer period as compared with specimens from the Cananeia estuary. These results suggest the presence of genotoxic compounds in these regions. However, the absence of significant differences in micronuclei frequency reflects slight mutagenic effects in these individuals. It is possible that the lower NAE frequency in specimens from SB might be associated with the greater remobilization and dilution of chemicals in this region. The low frequency of NAE in C. spixii from the Cananeia estuary is in accordance with the slight anthropogenic influence in this system, and may be suggestive of the absence of genotoxic and mutagenic effects in these organisms.
