Este trabajo es continuación de una serie de estudios sobre la biogeografÃa de Fusarium que se están realizando desde hace 5 años en España. En él se presentan los resultados analÃticos para el género Fusarium de muestras de aguas del cauce del rÃo Andarax y de fondos del mar Mediterráneo en las provincias de Granada y AlmerÃa (Sureste de España). Se analizan un total de 18 muestras de agua del rÃo Andarax. De ellas se aislaron 10 especies de Fusarium: F. anthophilum, F. acuminatum, F. chlamydosporum, F. culmorum, F. equiseti, F. verticillioides, F. oxysporum, F. proliferatum, F. solani y F. sambucinum. De las 23 muestras del mar Mediterráneo se aislaron 5 especies: F. equiseti,F. moliniforme, F. oxysporum, F. proliferatum y F. solani. Sobre el total de muestras analizadas, un 27,45% de las muestras de aguas del rÃo y un 29,41% de muestras de procedencia marina presentaron como mÃnimo una especie de Fusarium a lo largo de casi 12 meses de muestreo. Considerando las muestras según sus orÃgenes se encuentra que en las de origen aguas del rÃo un 77,77% presentaron alguna especie de Fusarium; en el caso de los fondos marinos un 45,45% de las muestras presentó alguna especie de Fusarium. La mayor presencia de especies en las aguas del rÃo puede ser debida a los contenidos en el agua de partÃculas de suelo y materia orgánica, después de los arrastres producidos en las orillas por las lluvias. La presencia de especies encontradas en el mar puede ser consecuencia de las aguas de los cauces que desembocan en éste. Sin embargo, no pueden excluirse otras vÃas.

En este artÃculo se estudia la patogenicidad de las especies de Fusarium aisladas de muestras de fondos marinos del Mediterráneo y de aguas del cauce del rÃo Andarax en las provincias de Granada y AlmerÃa (Sureste de España) sobre plántulas de cebada, colirrábano, melón y tomate. La evaluación del poder patógeno se hizo para 41 aislados de 9 especies de Fusarium aisladas de agus de mar y de rÃo: F. acuminatum, F. chlamydosporum, F.culmorum, F. equiseti, F. verticillioides, F. oxysporum, F. proliferatum, F. sambucinum y F. solani. Todos los aislados de las diferentes especies mostraron patogenicidad tanto en preemergencia como en postemergencia de plántulas. No fue posible distiguir a los aislados según su procedencia: aguas marinas o de rÃo.

Species of Fusarium were isolated from water samples collected from the Andarax River and coastal sea water of the Mediterranean in Granada and AlmerÃa provinces of southeastern Spain. In total, 18 water samples were analyzed from the Andarax River, and 10 species of Fusarium were isolated: Fusarium anthophilum, F. acuminatum, F. chlamydosporum, F. culmorum, F. equiseti, F. verticillioides, F. oxysporum, F. proliferatum, F. solani, and F. solani. When considering the samples by their origins, 77.8% of the river water samples yielded at least one species of Fusarium , with F. oxysporum comprising 72.2% of the total isolates. In the case of marine water, 45.5% of the samples yielded at least one species of Fusarium, with F. solani comprising 36.3% of the total isolates. The pathogenicity of 41 isolates representing nine of the species collected from river an sea water during the study ws evluated on barley, kohlrabe, melon, and tomato. Inoculation with F. acuminatum, F. chlamydosporum, F. culmorum, F. equiseti, F. verticillioides, F. oxysporum, F. proliferatum F. solani, and F. sambucinum resulted in pre-and post-emergence damping off. Pathogenicity of Fusarium isolates did not seem to be related to the origin of the isolates (sea water or fresh water). However, the presence of pathogenic species of Fusarium in river water flowing to the sea could indicate long-distance dispersal in natural water environments

The I-3 gene from the wild tomato species Lycopersicon pennellii confers resistance to race 3 of the devastating vascular wilt pathogen Fusarium oxysporum f. sp. lycopersici. As an initial step in a positional cloning strategy for the isolation of I-3, we converted restriction fragment length polymorphism and conserved orthologue set markers, known genes and a resistance gene analogue (RGA) mapping to the I-3 region into PCR-based sequence characterised amplified region (SCAR) and cleaved amplified polymorphic sequence (CAPS) markers. Additional PCR-based markers in the I-3 region were generated using the randomly amplified DNA fingerprinting (RAF) technique. SCAR, CAPS and RAF markers were used for high-resolution mapping around the I-3 locus. The I-3 gene was localised to a 0.3-cM region containing a RAF marker, eO6, and an RGA, RGA332. RGA332 was cloned and found to correspond to a putative pseudogene with at least two loss-of-function mutations. The predicted pseudogene belongs to the Toll interleukin-1 receptor-nucleotide-binding site-leucine-rich-repeat sub-class of plant disease resistance genes. Despite the presence of two RGA332 homologues in L. esculentum, DNA gel blot and PCR analysis suggests that no other homologues are present in lines carrying I-3 that could be alternative candidates for the gene.

Fusarium wilt of banana is a potentially devastating disease throughout the world. Options for control of the causal organism, Fusarium oxysporum f.sp. cubense (Foc) are limited. Suppressive soil sites have previously been identified where, despite the presence of Foc, Fusarium wilt does not develop. In order to understand some aspects of this disease suppression, endophytic Fusarium oxysporum isolates were obtained from banana roots. These isolates were genetically characterized and compared with an isolate of Fusarium oxysporum previously identified as being capable of suppressing Fusarium wilt of banana in glasshouse trials. Three additional isolates were selected for glasshouse trials to assess suppression of Fusarium wilt in two different cultivars of banana, Cavendish and Lady Finger. One isolate (BRIP 29089) was identified as a potential biocontrol organism, reducing the disease severity of Fusarium wilt in Lady Finger and Cavendish cultivars. Interestingly, one isolate (BRIP 45952) increased Fusarium wilt disease severity on Cavendish. The implications of an isolate of Fusarium oxysporum, non-pathogenic on banana, increasing disease severity and the potential role of non-pathogenic isolates of Fusarium oxysporum in disease complexes are discussed. Fusarium wilt of tomato, caused by the fungal pathogen, Fusarium oxysporum f. sp. lycopersici (Fol), is an economically damaging disease that results in huge losses in Australia and other countries worldwide. The I-3 gene, which confers resistance to Fol race 3, has been described in wild tomato, Lycopersicon pennellii, accessions LA716 and PI414773. We are pursuing the isolation of I-3 from LA716 by map-based cloning. We have constructed a high-resolution map of the I-3 region and have identified markers closely flanking I-3 as well as markers co-segregating with I-3. In addition, construction of a physical map based on these markers has been initiated. This review describes the context of our research and our progress towards isolating the I-3 gene. It also describes some important practical outcomes of our work, including the development and use of a PCR-based marker for marker-assisted selection for I-3, and the finding that the I-3 gene from LA716 is different to that from PI1414773, which we have now designated I-7. Tomato varieties combining I-3 and I-7 have been developed and are currently being introduced into commercial production to further safeguard tomato crops against Fusarium wilt.

Fusarium oxysporum is a diverse, asexual fungal species composed of both saprophytic and pathogenic members. The destructive phytopathogens are classified into formae speciales based on the host species and into vegetative compatibility groups (VCGs) based on the ability of two individuals to form heterokaryons. Parasexuality, a non-sexual mode of genetic exchange unique to some fungi has been demonstrated in the laboratory in Fusarium oxysporum f. sp. cubense (FOC). The goals of this dissertation were threefold: to ascertain whether mitochondrial (mt) markers can distinguish race differences in FOC; to determine genetic relatedness of VCGs in FOC based on a mt marker; and to discover the mode of mt inheritance during a parasexual cycle.^ Band patterns produced by electrophoresis of Hae III digested genomic DNA indicated that VCG differences, not race, could be discerned by mtDNA analysis. Primers were designed to amplify a mt intergenic locus which served as a molecular marker to screen 55 strains of FOC in 16 VCGs using both single strand conformational polymorphism and DNA sequencing. Based on homogeneity of the locus, strains were assigned to seven mitotypes, a classification unit which I introduced and found informative for grouping related VCGs.^ To determine the mode of mt inheritance during a parasexual cycle, strains in different mitotypes were paired. Mitochondrial inheritance in all hybrid progeny was found to be uniparental. I speculated that if a parasexual cycle occurs in nature there would be greater variation in the nuclear genome than the mt. This could produce multiple VCGs within a mitotype, a phenomenon observed in FOC. Based on these data, I concluded that parasexuality in nature may contribute to the diversity observed in Fusarium oxysporum. ^

Muitos métodos rápidos e eficientes de seleção de agentes de biocontrole de fitopatógenos tem sido utilizados, visando reduzir tempo e custo dispendido em testes de campo. Neste trabalho realizou-se uma seleção de isolados endofÃticos com potencial de uso no biocontrole de fitopatógenos em testes de antagonismo in vitro. De um total de 95 isolados de bactérias endofÃticas do milho, seis foram selecionados quanto à inibição a Pythium aphanidermatum. A essa seleção, foram incluÃdos um isolado de Bacillus subtilis 0G, Bacillus lentimorbus e Streptomyces sp., para verificação de antagonismo a Rhizoctonia solani, Fusarium moniliforme, Sclerotium rolfsii e Exserohilum turcicum. Verificou-se que os endofÃticos B. subtilis 0G, B. lentimorbus e Streptomyces sp., apresentaram ação antagônica superior aos demais, com taxas de inibição entre 32,0% e 53,8%. Dentre os endofÃticos do milho, Bacillus agaradhaerens foi o que mais se destacou, com taxas de inibição variando entre 43,7% e 52,3% e indicando uma inespecificidade de ação. Este estudo, embora preliminar, permite vislumbrar a utilização desses endofÃticos na supressão de doenças em diferentes sistemas patógeno-hospedeiro em testes subseqüentes, sob condições de casa-de-vegetação e a campo.

O objetivo deste trabalho foi avaliar o comportamento de açaizeiro (Euterpe precatoria Mart.) em cultivo a pleno sol e submetido ao sombreamento disponibilizado por bananeira em diferentes densidades de plantio, nas condições edafoclimáticas do estado do Acre.

Inclusions of sp-hybridised, trans-polyacetylene [trans-(CH)x] and poly(p-phenylene vinylene) (PPV) chains are revealed using resonant Raman scattering (RRS) investigation of amorphous hydrogenated carbon (a-C:H) films in the near IR – UV range. The RRS spectra of trans-(CH)x core Ag modes and the PPV CC-H phenylene mode are found to transform and disperse as the laser excitation energy ћωL is increased from near IR through visible to UV, whereas sp-bonded inclusions only become evident in UV. This is attributed to ћωL probing of trans-(CH)x chain inhomogeneity and the distribution of chains with varying conjugation length; for PPV to the resonant probing of phelynene ring disorder; and for sp segments, to ћωL probing of a local band gap of end-terminated polyynes. The IR spectra analysis confirmed the presence of sp, trans-(CH)x and PPV inclusions. The obtained RRS results for a-C:H denote differentiation between the core Ag trans-(CH)x modes and the PPV phenylene mode. Furthermore, it was found that at various laser excitation energies the changes in Raman spectra features for trans-(CH)x segments included in an amorphous carbon matrix are the same as in bulk trans-polyacetylene. The latter finding can be used to facilitate identification of trans-(CH)x in the spectra of complex carbonaceous materials.
