This study examines the diaconia work of the Finnish Evangelical Lutheran Church from the standpoint of clients. The role of diaconia work has grown since the early 1990s recession, and since it established itself as one of the actors along with other social organizations. Previous studies have described the changing role of diaconal work, especially from the standpoint of diaconia workers and co-operators. This research goes back to examine, beyond the activities of the diaconia work of everyday practices, its relations of ruling which are determining practices. The theoretical and methodological framework rises from the thinking of Dorothy E. Smith, the creator of institutional ethnography. Its origins are in feminism, Marxism, phenomenology, etnomethodology, and symbolic interactionism. However, it does not represent any school. Unlike the objectivity-based traditional sociology, institutional ethnography has its starting point in everyday life, and people s subjective experience of it. Everyday life is just a starting point, and is used to examine everyday life s experiences of hidden relations of ruling, linking people and organizations. The level of generalization is just on the relations of ruling. The research task is to examine those meanings of diaconia work which are embedded in its clients experiences. The research task is investigated with two questions: how diaconia work among its clients takes shape and what kinds of relations of ruling exist in diaconia work. The meanings of diaconia work come through an examination of the relations of ruling, which create new forms of diaconal work compared with previous studies. For the study, two kinds of data were collected: a questionnaire and ethnographic fieldwork. The first data set was collected from diaconal workers using the questionnaire. It gives background information of the diaconia work process from the standpoint of the clients. In the ethnographic study there were two phases. The first ethnographic material was collected from one local parish by observing, interviewing clients and diaconal workers and gathering documents. The number of observations was 36 customer appointments, and 29 interviews. The second ethnographic material was included as a part of the analysis, in which ruling relations in people s experiences were collected from the transcribed data. Close reading and narrative analysis are used as analysing methods. The analysis has three phases. First, the experiences are identified with close reading; the following step is to select some of the institutional processes that are shaping those experiences and are relevant for the research. At the third stage, those processes are investigated in order to describe analytically how they determine people s experience. The analysis produces another narrative about diaconia work, which provides tools for examining the diaconal work from a new perspective. Through the analysis it is possible to see diaconia as an exchange ratio, in which the exchange takes place between a client and a diaconia worker, but also more broadly with other actors, such as social workers, shop clerks, or with other parishioners. The exchange ratio is examined from the perspective of power which is embedded in the client s experiences. The analysis reveals that the most important relations of ruling are humiliation and randomness in the exchange ratio of diaconia work; valuating spirituality above the bodily being; and replacing official social work. The results give a map about the relations of ruling of diaconia work which gives tools to look at diaconia work s meanings to the clients. The hidden element of humiliation in the exchange ratio breaks the current picture of diaconia work. The ethos of the holistic encounters and empathic practices are shown to be of another kind when spirituality is preferred to the bodily being. Nevertheless, diaconia appears to be a place for a respectful encounter, especially in situations where the public sector s actors are retreating on liability or clients are in a life crisis. The collapse of the welfare state structures imposes on diaconia work tasks that have not previously belonged to it. At the local level, clients receive partners from diaconia workers in order to advocate them in the welfare system. Actions to influence the wider societal structures are not reached because of lacking resources. An awareness of the oppressive practices of diaconia work and their critical reviewing are the keys to the development of diaconia work, since there are such practices even in holistic and respectful diaconia work. While the research raises new information for the development of diaconia work, it also opens up new aspects for developing other kinds of social work by emphasizing the importance of taking people s experiences seriously. Hermansky-Pudlak syndrome (HPS) is a group of disorders characterized by the malformation of lysosome-related organelles, such as pigment cell melanosomes. Three of nine characterized HPS subtypes result from mutations in subunits of BLOC-2, a protein complex with no known molecular function. In this paper, we exploit melanocytes from mouse HPS models to place BLOC-2 within a cargo transport pathway from recycling endosomal domains to maturing melanosomes. In BLOC-2-deficient melanocytes, the melanosomal protein TYRP1 was largely depleted from pigment granules and underwent accelerated recycling from endosomes to the plasma membrane and to the Golgi. By live-cell imaging, recycling endosomal tubules of wild-type melanocytes made frequent and prolonged contacts with maturing melanosomes; in contrast, tubules from BLOC-2-deficient cells were shorter in length and made fewer, more transient contacts with melanosomes. These results support a model in which BLOC-2 functions to direct recycling endosomal tubular transport intermediates to maturing melanosomes and thereby promote cargo delivery and optimal pigmentation.

Reúne depoimentos de servidores da Câmara do Deputados sobre os acontecimentos que marcaram a história do parlamento brasileiro a partir da segunda metade do século XX. Coletados pelo Núcleo de História Oral da Câmara durante as comemorações dos 180 anos do Poder Legislativo em 2003, os textos retratam a realidade dos primeiros anos vividos no Planalto Central: a carência de infra-estrutura, a falta de opção de atividades de lazer e o profundo sentimento de saudade da antiga capital da República. Mas também levam a reminiscências de momentos divertidos, curiosidades e reflexões animadas e esperançosas sobre a mudança da capital para Brasília. O ponto alto são as experiências vivenciadas nos prédios do Congresso Nacional, a lembrança dos amigos, dos colegas e, especialmente, dos fatos marcantes, que sublinham as significativas mudanças nas rotinas de trabalho da Instituição para se adequar à modernização e às novas exigências da sociedade, no contexto de acontecimentos políticos que modificaram seguidas vezes o cenário do país.

In directly, phytoplankton serves as food for all aquatic animals since it is at the base of the food chain in which the phytoplankton-feeding animals are eaten by larger animals and these in turn are consumed by still larger forms. Hence, it becomes evident that the phytoplankton, its presence, and seasonal variations are of great importance. The report at hand is based on a record of the variations in the plankton population of surface waters at a single station, where collections were made biweekly from September 1943 through September 1945. The station chosen was in the channel of the Patuxent River, Maryland, near its entrance into Chesapeake Bay, about midway between the head and the mouth of the Bay. (PDF contains 31 pages)

Cancer chemotherapy has advanced from highly toxic drugs to more targeted treatments in the last 70 years. Chapter 1 opens with an introduction to targeted therapy for cancer. The benefits of using a nanoparticle to deliver therapeutics are discussed. We move on to siRNA in particular, and why it would be advantageous as a therapy. Specific to siRNA delivery are some challenges, such as nuclease degradation, quick clearance from circulation, needing to enter cells, and getting to the cytosol. We propose the development of a nanoparticle delivery system to tackle these challenges so that siRNA can be effective.

Chapter 2 of this thesis discusses the synthesis and analysis of a cationic mucic acid polymer (cMAP) which condenses siRNA to form a nanoparticle. Various methods to add polyethylene glycol (PEG) for stabilizing the nanoparticle in physiologic solutions, including using a boronic acid binding to diols on mucic acid, forming a copolymer of cMAP with PEG, and creating a triblock with mPEG on both ends of cMAP. The goal of these various pegylation strategies was to increase the circulation time of the siRNA nanoparticle in the bloodstream to allow more of the nanoparticle to reach tumor tissue by the enhanced permeation and retention effect. We found that the triblock mPEG-cMAP-PEGm polymer condensed siRNA to form very stable 30-40 nm particles that circulated for the longest time – almost 10% of the formulation remained in the bloodstream of mice 1 h after intravenous injection.

Chapter 3 explores the use of an antibody as a targeting agent for nanoparticles. Some antibodies of the IgG1 subtype are able to recruit natural killer cells that effect antibody dependent cellular cytotoxicity (ADCC) to kill the targeted cell to which the antibody is bound. There is evidence that the ADCC effect remains in antibody-drug conjugates, so we wanted to know whether the ADCC effect is preserved when the antibody is bound to a nanoparticle, which is a much larger and complex entity. We utilized antibodies against epidermal growth factor receptor with similar binding and pharmacokinetics, cetuximab and panitumumab, which differ in that cetuximab is an IgG1 and panitumumab is an IgG2 (which does not cause ADCC). Although a natural killer cell culture model showed that gold nanoparticles with a full antibody targeting agent can elicit target cell lysis, we found that this effect was not preserved in vivo. Whether this is due to the antibody not being accessible to immune cells or whether the natural killer cells are inactivated in a tumor xenograft remains unknown. It is possible that using a full antibody still has value if there are immune functions which are altered in a complex in vivo environment that are intact in an in vitro system, so the value of using a full antibody as a targeting agent versus using an antibody fragment or a protein such as transferrin is still open to further exploration.

In chapter 4, nanoparticle targeting and endosomal escape are further discussed with respect to the cMAP nanoparticle system. A diboronic acid entity, which gives an order of magnitude greater binding (than boronic acid) to cMAP due to the vicinal diols in mucic acid, was synthesized, attached to 5kD or 10kD PEG, and conjugated to either transferrin or cetuximab. A histidine was incorporated into the triblock polymer between cMAP and the PEG blocks to allow for siRNA endosomal escape. Nanoparticle size remained 30-40 nm with a slightly negative ca. -3 mV zeta potential with the triblock polymer containing histidine and when targeting agents were added. Greater mRNA knockdown was seen with the endosomal escape mechanism than without. The nanoparticle formulations were able to knock down the targeted mRNA in vitro. Mixed effects suggesting function were seen in vivo.

Chapter 5 summarizes the project and provides an outlook on siRNA delivery as well as targeted combination therapies for the future of personalized medicine in cancer treatment.

A conservação dos ecossistemas e dos recursos ambientais neles inclusos é uma condição básica e essencial para o desenvolvimento sustentado de uma dada região. A degradação faz com que a possibilidade dele retornar ao seu estado original seja ínfima pois, sua dinâmica de restauração não seria a mesma do que antes foi sua colonização. A recuperação de áreas degradadas é dificultada devido à complexidade estrutural dos ecossistemas, exemplo das restingas ambientes sujeitos as condições bastante adversas (altas temperaturas, períodos de seca, vento constante, alta salinidade e escassez de nutrientes), por isso demandam de alta tecnologia para o desenvolvimento e produção de mudas, além de alto custo associado. O objetivo deste estudo é a criação de uma proposta para uso socioeconômico das áreas degradadas por plantios de cocos, em formações vegetais de restinga, município de Caravelas, e criar um modelo para que essas áreas sejam mais produtivas economicamente, a médio e longo prazo, a partir da geração de trabalho e renda e, conseqüentemente, inclusão social voltada para o uso sustentável de espécies nativas de restinga, através do extrativismo, considerando-se o potencial e a vocação natural do ecossistema de restinga. Nesse contexto, as categorias de análise desta tese basearam-se nos conflitos e vulnerabilidade socioambiental, etnobotânica, fitofisionomias, bens e serviços associados, tecnologia social, desenvolvimento local, gestão costeira, sustentabilidade ambiental e democrática, produtos florestais não madeiráveis e inclusão social. Os procedimentos metodológicos utilizados neste estudo foram apresentados em cada capítulo desta tese, estando inserido em pesquisas qualitativas (técnicas de observação participante e análise do discurso coletivo) associada aos levantamentos bibliográficos (dados secundários) e as pesquisas quantitativas, por entrevistas semiestruturadas. Os resultados deste estudo subsidiaram a formação de uma rede interativa para implantação de empreendimentos sustentáveis no processo produtivo local, no que se refere à utilização de espécies nativas de restingas com reflorestamento de áreas degradadas por plantios de coco, para fins de geração de trabalho e renda com base no movimento de tecnologia social.

O presente estudo teve como objetivo central conhecer a composição florística, a estrutura e a arquitetura das espécies da formação arbustiva fechada pós-praia e investigar possíveis associações com Formicivora littoralis, considerando tática de forrageamento, construção de ninhos e abundância da ave. F. littoralis é endêmica de restingas, e está ameaçada de extinção devido a perda acelerada de habitat. Entretanto, pouco se sabe sobre a vegetação em que ela ocorre, principalmente sobre as áreas de maior abundância da ave localizadas na formação Arbustiva Fechada Pós-praia (AFP) da Restinga da Massambaba, as quais estão inseridas em um Centro de Diversidade Vegetal (CDV) na Região de Cabo Frio. Diante disto, este estudo foi realizado em dois trechos desta formação na Restinga da Massambaba, nos municípios de Araruama e Arraial do Cabo, RJ, Brasil. Foram efetuadas 60 excursões a campo, com coletas aleatórias realizadas ao longo de toda a formação, geração de 20 parcelas de 2x50 m (0,2ha) perpendiculares ao mar, incluindo na amostragem indivíduos com DAP e DAS ≥2,5, estes, foram ainda categorizados em modelos de arquitetura de ramificação considerando número de ramos em dois patamares de altura (DAP e DAS). O levantamento florístico resultou em 327 coletas de 160 espécies, sendo pelo menos 12 espécies vegetais sob algum estado de ameaça, inclusive redescoberta uma Salicaceae (Casearia sessiliflora). Orchidaceae, Leguminosae e Myrtaceae foram as famílias mais ricas. Foram acrescentados 75% mais espécies na lista preliminar da AFP na Massambaba, além de 14 novos registros para o CDV de Cabo Frio. Na estrutura e arquitetura foram analisados 906 indivíduos de 58 espécies. Sendo os maiores valores de importância de Pilosocereus arrabidae (42,30) e Chrysophyllum lucentifolium (23,45). A diversidade de Shannon foi de 3,46 e equabilidade de 0,85. A arquitetura da maior parte dos indivíduos foi complexa, 58% de indivíduos com múltiplas ramificações, e as espécies apresentando variados padrões de ramificação. A densidade populacional de F. littoralis na AFP foi elevada, sendo estimada em 172 ind/km2. A arquitetura da AFP tem influência na ecologia da ave, pois ela foi generalista quanto à espécie utilizada como suporte na construção de ninhos e também para as táticas de forrageamento, mas houve seleção de ramos finos que formavam na horizontal ângulos de até 90 de abertura para construir ninhos. Ramos finos e mais horizontais também foram utilizados com frequência para as táticas de forrageamento. A abundância de F. littoralis esteve correlacionada positivamente à diversidade vegetal e negativamente à altura da vegetação, características marcantes nesta formação. Além disso, elevada taxa de vegetais com síndrome de dispersão zoocórica indica a importância desta formação na oferta de recursos alimentícios para a fauna e atração de pequenos artrópodes, os quais fazem parte da dieta de F. littoralis.
