Although the sunspot-number series have existed since the mid-19th century, they are still the subject of intense debate, with the largest uncertainty being related to the "calibration" of the visual acuity of individual observers in the past. Daisy-chain regression methods are applied to inter-calibrate the observers which may lead to significant bias and error accumulation. Here we present a novel method to calibrate the visual acuity of the key observers to the reference data set of Royal Greenwich Observatory sunspot groups for the period 1900-1976, using the statistics of the active-day fraction. For each observer we independently evaluate their observational thresholds [S_S] defined such that the observer is assumed to miss all of the groups with an area smaller than S_S and report all the groups larger than S_S. Next, using a Monte-Carlo method we construct, from the reference data set, a correction matrix for each observer. The correction matrices are significantly non-linear and cannot be approximated by a linear regression or proportionality. We emphasize that corrections based on a linear proportionality between annually averaged data lead to serious biases and distortions of the data. The correction matrices are applied to the original sunspot group records for each day, and finally the composite corrected series is produced for the period since 1748. The corrected series displays secular minima around 1800 (Dalton minimum) and 1900 (Gleissberg minimum), as well as the Modern grand maximum of activity in the second half of the 20th century. The uniqueness of the grand maximum is confirmed for the last 250 years. It is shown that the adoption of a linear relationship between the data of Wolf and Wolfer results in grossly inflated group numbers in the 18th and 19th centuries in some reconstructions.

Lymphocyte subsets, activation markers and apoptosis were assessed in 20 HIV-exposed noninfected (ENI) children born to HIV-infected women who were or not exposed to antiretroviral (ARV) drugs during pregnancy and early infancy. ENI children and adolescents were aged 6-18 years and they were compared to 25 age-matched healthy non-HIV-exposed children and adolescents (Control). ENI individuals presented lower CD4(+) T cells/mm(3) than Control group (control: 1120.3 vs. ENI: 876.3; t-test, p=0.030). ENI individuals had higher B-cell apoptosis than Control group (Control: 36.6%, ARV exposed: 82.3%, ARV nonexposed: 68.5%; Kruskal-Wallis, p < 0.05), but no statistical difference was noticed between those exposed and not exposed to ARV. Immune activation in CD4(+) T, CD8(+) T and in B cells was comparable in ENI and in Control children and adolescents. Subtle long-term immune alterations might persist among ENI individuals, but the clinical consequences if any are unknown, and these children require continued monitoring.

Objective: To compare and evaluate longitudinally the dental arch relationships from 4.5 to 13.5 years of age with the Bauru-BCLP Yardstick in a large sample of patients with bilateral cleft lip and palate (BCLP). Design: Retrospective longitudinal intercenter outcome study. Patients: Dental casts of 204 consecutive patients with complete BCLP were evaluated at 6, 9, and 12 years of age. All models were identified only by random identification numbers. Setting: Three cleft palate centers with different treatment protocols. Main Outcome Measures: Dental arch relationships were categorized with the Bauru-BCLP yardstick. Increments for each interval (from 6 to 9 years, 6 to 12 years, and 9 to 12 years) were analyzed by logistic and linear regression models. Results: There were no significant differences in outcome measures between the centers at age 12 or at age 9. At age 6, center B showed significantly better results (p = .027), but this difference diminished as the yardstick score for this group increased over time (linear regression analysis), the difference with the reference category (center C, boys) for the intervals 6 to 12 and 9 to 12 years being 10.4% (p = .041) and 12.9% (p = .009), respectively. Conclusions: Despite different treatment protocols, dental arch relationships in the three centers were comparable in final scores at age 9 and 12 years. Delaying hard palate closure and employing infant orthopedics did not appear to be advantageous in the long run. Premaxillary osteotomy employed in center B appeared to be associated with less favorable development of the dental arch relationship between 9 and 12 years.

The role of PPAR-gamma in ciglitazone and 15-d PGJ(2)-induced apoptosis and cell cycle arrest of Jurkat (before and after PPAR gamma gene silencing), U937 (express high levels of PPAR gamma) and HeLa (that express very low levels of PPAR gamma) cells was investigated. PPAR gamma gene silencing, per se, induced a G2/M cell arrest, loss of membrane integrity and DNA fragmentation of Jurkat cells, indicating that PPAR gamma is important for this cell survival and proliferation. Ciglitazone-induced apoptosis was abolished after knockdown of PPAR gamma suggesting a PPAR gamma-dependent pro-apoptotic effect. However, ciglitazone treatment was toxic for U937 and HeLa cells regardless of the presence of PPAR gamma. This treatment did not change the cell cycle distribution corroborating with a PPAR gamma-independent mechanism. On the other hand, 15-d PGJ(2) induced apoptosis of the three cancer cell lines regardless of the expression of PPAR gamma. These results suggest that PPAR gamma plays an important role for death of malignant T lymphocytes (Jurkat cells) and PPAR gamma agonists exert their effects through PPAR gamma-dependent and -independent mechanisms depending on the drug and the cell type. Aims: The ATP-binding cassette transporters, ABCA1 and ABCG1, are LXR-target genes that play an important role in reverse cholesterol transport. We examined the effects of inhibitors of the cholesterol absorption (ezetimibe) and synthesis (statins) on expression of these transporters in HepG2 cells and peripheral blood mononuclear cells (PBMCs) of individuals with primary (and nonfamilial) hypercholesterolemia (HC). Materials & methods: A total of 48 HC individuals were treated with atorvastatin (10 mg/day/4 weeks) and 23 were treated with ezetimibe (10 mg/day/4 weeks), followed by simvastatin (10 mg/day/8 weeks) and simvastatin plus ezetimibe (10 mg of each/day/4 weeks). Gene expression was examined in statin- or ezetimibe-treated and control HepG2 cells as well as PBMCs using real-time PCR. Results: In PBMCs, statins and ezetimibe downregulated ABCA1 and ABCG1 mRNA expression but did not modulate NR1H2 (LxR-beta) and NR1H3 (LXR-alpha) levels. Positive correlations of ABCA1 with ABCG1 and of NR1H2 with NR1H3 expressions were found in all phases of the treatments. In HepG2 cells, ABCA1 mRNA levels remained unaltered while ABCG1 expression was increased by statin (1.0-10.0 mu M) or ezetimibe (5.0 mu M) treatments. Atorvastatin upregulated NR1H2 and NR1H3 only at 10.0 mu M, meanwhile ezetimibe (1.0-5.0 mu M) downregulated NR1H2 but did not change NR1H3 expression. Conclusion: Our findings reveal that lipid-lowering drugs downregulate ABCA1 and ABCG1 mRNA expression in PBMCs of HC individuals and exhibit differential effects on HepG2 cells. Moreover, they indicate that the ABCA1 and ABCG1 transcript levels were not correlated directly to LXR mRNA expression in both cell models treated with lipid-lowering drugs.

The reaction of cis-[RuCl2(dppb)(N-N)], dppb = 1,4-bis(diphenylphosphino)butane, complexes with the ligand HSpymMe(2), 4,6-dimethyl-2-mercaptopyrimidine, yielded the cationic complexes [Ru(SpymMe(2))(dppb)(N-N)]PF6, N-N = bipy (1) and Me-bipy (2), bipy = 2,2`-bipyridine and Me-bipy = 4,4`dimethyl-2,2`-bipyridine, which were characterized by spectroscopic and electrochemical techniques and X-ray crystallography and elemental analysis. Additionally, preliminary in vitro tests for antimycobacterial activity against Mycobacterium tuberculosis H37Rv ATCC 27264 and antitumor activity against the MDA-MB-231 human breast tumor cell line were carried out on the new complexes and also on the precursors cis-[RuCl2(dppb)(N-N)], N-N = bipy (3) and Me-bipy (4) and the free ligands dppb, bipy, Me-bipy and SpymMe(2). The minimal inhibitory concentration (MIC) of compounds needed to kill 90% of mycobacterial cells and the IC50 values for the antitumor activity were determined. Compounds 1-4 exhibited good in vitro activity against M. tuberculosis, with MIC values ranging between 0.78 and 6.25 mu g/mL, compared to the free ligands (MIC of 25 to >50 mu g/mL) and the drugs used to treat tuberculosis. Complexes I and 2 also showed promising antitumor activity, with IC50 values of 0.46 +/- 0.02 and 0.43 +/- 0.08 mu M, respectively, against MDA-MB-231 breast tumor cells. (C) 2008 Elsevier Inc. This work reports the synthesis and characterization of a new copper complex with nadolol, a beta-blocker aminoalcohol. The stoichiometry found was Na[Cu(nadololate)(CO(3))] center dot H(2)O. Electronic and vibrational spectroscopy analysis was performed, and the crystal structure of Na[Cu(nadololate)-(CO(3))] center dot H(2)O was determined by X-ray diffraction.

The aim of this study was to identify a candidate drug for the development of anti-tuberculosis therapy from previously synthesized compounds based on the thiosemicarbazones, semicarbazones, dithio-carbazates and hydrazide/hydrazones compounds. The minimal inhibitory concentration (MIC) of these compounds against Mycobacterium tuberculosis was determined. Their in vitro cytotoxicity to J774 cells (IC(50)) was determined to establish a selectivity index (SI) (SI = IC(50)/MIC). The best compounds were the thiosemicarbazones (2, 3 and 4) and the hydrazide/hydrazones (14, 15, 16 and 18). The results are comparable to or better than those of ""first line"" or ""second line"" drugs commonly used to treat TB, suggesting these compounds as anti-TB drug candidates. The synthesis, characterization and the anti-Mycobacterium tuberculosis (MTB) activities of three ruthenium complexes containing the 2-pyridinecarboxylic acid anion (picolinate), with formulae cis-[Ru(pic)(dppm)(2)]PF(6) (1), Cis- [Ru(pic)(dppe)(2)]PF(6) (2) and [Ru(pic)(2)(PPh(3))(2)] (3) [pic = 2-pyridinecarboxylate; dppm = bis(diphenylphosphino)methane: dppe = 1,2-bis(diphenylphosphino)ethane; PPh(3) = triphenylphosphine] are reported in this article. The complexes were characterized by elemental analysis, spectroscopic and electrochemical techniques. Their in vitro anti mycobacterial activity was determinated as the Minimum Inhibitory Concentration (MIC) for MTB cell growth, measured by the REMA method. The best MICs were found for complexes (1) and (2), with values of 0.78 and 0.26 mu g/mL, respectively. The results are comparable to or better than ""first line"" or ""second line"" drugs commonly used in the treatment of TB. (C) 2009 Elsevier Masson SAS. All rights reserved.

O fantástico incremento do turismo nas últimas décadas, hoje uma das maiores atividades econômicas do mundo, tem despertado o interesse do seu estudo tanto em nível teórico, nas chamadas Ciências Sociais, como nas Ciências Aplicadas, entre elas, o marketing. E em virtude da importância desse fenômeno e com o objetivo de identificar as oportunidades turísticas da região missioneira, em seus aspectos de potencialização e motivação, de forma a encontrar um novo instrumento de desenvolvimento, optou-se por este trabalho. Para isso, o estudo começou por caracterizar sócio-economicamente a região (Missões) onde se localizam as Ruínas Jesuíticas e teve por objetivo identificar as potencialidades e as motivações para este turismo regional porque é uma região de atratividade turística. Por se tratar de uma investigação de natureza exploratória, iniciou-se com uma pesquisa qualitativa através de estudos bibliográficos e entrevistas em profundidade com especialistas em turismo. Posteriormente, tendo como base esse resultado, partiu-se para a fase quantitativa, aprofundando-se, o conhecimento iniciado anteriormente, cuja etapa foi divida em duas partes: partes: a primeira contém três dimensões medindo os atrativos histórico-culturais, lazer e entretenimento; e serviços de infra-estrutura e apoio para o desenvolvimento turístico regional. A segunda medindo fatores motivacionais e onze questões caracterizando os respondentes da pesquisa. As dimensões foram avaliadas através de uma escala de concordância de 5 pontos que variou de 1 “discordo totalmente” a 5 “ concordo totalmente” . Para as análises das dimensões, foram utilizadas os seguintes estudos estatísticos: frequência simples, média, teste de Cronbach, análise fatorial e análise de covariância. Utilizando-se dessas ferramentas estatísticas, procurou-se identificar os fatores de desenvolvimento turístico existentes ou que possam vir a ser potencializados, dentre os quais se destacaram a organização do museu, a arquitetura jesuítico-guaranítica e a divulgação da civilização guarani, no agrupamento atrativos histórico-culturais. Em relação à recreação do turista, o espetáculo de Som e Luz, histórias pitorescas e passeios ecológicos, no grupo de lazer e entretenimento. E plantão médico, farmácia e postos de gasolina 24 horas, segurança policial e facilidade de acesso rodoviário, em termos de serviços de infra-estrutura e apoio. Na dimensão que mediu os fatores de motivação para visitar as Missões, se sobressaíram-se, o desejo de conhecer a história da região missioneira, o conhecimento do Patrimônio Histórico da Humanidade do Rio Grande do Sul, o conhecimento da civilização guarani e possibilidade de contato com a cultura missioneira, os quais têm vínculo com os atributos histórico-culturais. Por outro lado, o fator motivador relacionado a lazer e entretenimento, foi o Espetáculo de Som e Luz. Indentificou-se, também, a possibilidade de crescimento, no sentido de motivação, o turismo místico-religioso que o local oferece. Os resultados demonstraram que o produto turístico “ Ruínas Jesuíticas da Região das Missões “ tem potencial para desenvolver o turismo regional. Os seus atrativos histórico-culturais seriam suas principais atrações, porém as categorias de lazer e entretenimentos aliados aos de infra-estrutura poderão alicerçar melhor os empreendimentos turísticos a serem realizados. Entretanto várias percepções aqui descritas já foram referendadas no levantamento obtido na primeira fase da pesquisa. Por último, são expostas as propostas e caminhos para futuras pesquisas relacionadas ao turismo da Região Missioneira do Rio Grande do Sul, de maneira a contribuir com o crescimento e desenvolvimento regional.

The motivation of this study is based on the expectation of understanding, at least in part, the reason why the brazilian academic production ¿ which is among the largest if compared to the developing countries, has not been translated in a useful way into concrete technology transference for the production sector. The supposition that different cultures and subcultures, in terms of values, behavior, practices among organizations and groups of selected actors involved in that process,- which is the central problem of this study, and peculiar to the technology context, has elected that relationship process the object of this research this study theoretical fundaments rely on culture and technology to analyze the subcultures which manifest themselves through the typified groups of researchers, in the public organizations, small scale employers, acting on technology based companies in the private sector (ebts) and mediators. Who also act in the process the preliminary step was the case study. Several qualitative methods an procedures were used. And the investigation has advanced toward interacting interviews, at the end added by the focal group method. The conclusions reject the existence of incompatibility among the subject groups points of view, however, points out some divergent values, the presence of mutual stereotypes and also confirms the supposition that the research platform is strongly influenced by the financial factor. Although, the study reveals that the presence of common objectives which lay behind the cited divergences make possible to present a core of practical suggestions to implement the studied relationships for the benefit of the production sector as well. The peculiarities perceived while dealing with that singular context suggest futures research to clarify possible new and obscure areas in that complex relationships.
