A black and white German Holstein calf displayed a complex double malformation in shape of a thoracopagus parasiticus. By means of a molecular genetic investigation the genesis of the malformation from one zygote could be demonstrated. Both vertebral columns showed a pronounced lordosis, with the vertebral column of one animal ending in a rudimentary head. Close to this rudiment two derivates of branchial arches were found. The two thoracic cavities merged into one "thorax". In the shared thoracic cavity one heart was found. In its right atrium, a cherry-sized structure was found in which heart- and vascular smooth muscles were demonstrated histologically. The aorta split shortly after its origin to provide both animals with one aorta each. The larger pair of lungs was connected with a trachea leading to the head while the smaller pair of lungs originated from a trachea deriving from the rudimentary head. The diaphragm jejunum and split afterwards. The pedigree of the affected animal showed neither inbreeding nor any other affected animal.

The purpose of the investigation was to determine the most effective means of concentrating the manganese in the ore. The study was that of identification of the minerals present. This showed the presence of oxides of manganese and iron, and carbonates of calcium and manganese. Methods of investigation were planned to determined whether the ore could be concentrated effectively by gravity concentration methods. ­

Chromite occurs in Madison County, Montana, in two areas, one three miles southwest of Silver Star, the other five miles southeast of Sheridan. The ore bodies are small, lenticular masses surrounded by metamorphosed sedimentary rocks of the pre-Cambrian Cherry Creek series. The ore was deposited from the hydrothermal solutions which serpentinized the surrounding metamorphic host rocks.

The minerals sillimanite, kyanite, andalusite, dumortierite, and topaz comprise a group of minerals whose high alumina content and physical properties are particularly desirable in the manufacture of refractory products. Sillimanite is the least plentiful of the minerals of this group, and for this reason it is not used extensively at the present time. However, it would be very desirable to the refractory industry if a suitable domestic source of supply could be established.

Nucleus pulposus (NP) regeneration by the application of injectable cell-embedded hydrogels is an appealing approach for tissue engineering. We investigated a thermo-reversible hydrogel (TR-HG), based on a modified polysaccharide with a thermo-reversible polyamide [poly(N-isopropylacrylamide), pNIPAM], which is made to behave as a liquid at room temperature and hardens at > 32 °C. In order to test the hydrogel, a papain-induced bovine caudal disc degeneration model (PDDM), creating a cavity in the NP, was employed. Human mesenchymal stem cells (hMSCs) or autologous bovine NP cells (bNPCs) were seeded in TR-HG; hMSCs were additionally preconditioned with rhGDF-5 for 7 days. Then, TR-HG was reversed to a fluid and the cell suspension injected into the PDDM and kept under static loading for 7 days. Experimental design was: (D1) fresh disc control + PBS injection; (D2) PDDM + PBS injection; (D3) PDDM + TR-HG (material control); (D4) PDDM + TR-HG + bNPCs; (D5) PDDM + TR-HG + hMSCs. Magnetic resonance imaging performed before and after loading, on days 9 and 16, allowed imaging of the hydrogel-filled PDDM and assessment of disc height and volume changes. In gel-injected discs the NP region showed a major drop in volume and disc height during culture under static load. The RT–PCR results of injected hMSCs showed significant upregulation of ACAN, COL2A1, VCAN and SOX9 during culture in the disc cavity, whereas the gene expression profile of NP cells remained unchanged. The cell viability of injected cells (NPCs or hMSCs) was maintained at over 86% in 3D culture and dropped to ~72% after organ culture. Our results underline the need for load-bearing hydrogels that are also cyto-compatible.

Animal pollination is essential for the reproductive success of many wild and crop plants. Loss and isolation of (semi-)natural habitats in agricultural landscapes can cause declines of plants and pollinators and endanger pollination services.We investigated the independent effects of these drivers on pollination of young cherry trees in a landscape-scale experiment. We included (i) isolation of study trees from other cherry trees (up to 350 m), (ii) the amount of cherry trees in the landscape, (iii) the isolation from other woody habitats (up to 200 m) and (iv) the amount of woody habitats providing nesting and floral resources for pollinators. At the local scale, we considered effects of (v) cherry flower density and (vi) heterospecific flower density. Pollinators visited flowers more often in landscapes with high amount of woody habitat and at sites with lower isolation from the next cherry tree. Fruit set was reduced by isolation from the next cherry tree and by a high local density of heterospecific flowers but did not directly depend on pollinator visitation. These results reveal the importance of considering the plant’s need for con-specific pollen and its pollen competition with co-flowering species rather than focusing only on pollinators’ habitat requirements and flower visita-tion. It proved to be important to disentangle habitat isolation from habitat loss, local from landscape-scale effects, and direct effects of pollen availability on fruit set from indirect effects via pollinator visitation to understand the delivery of an agriculturally important ecosystem service.

BACKGROUND Clinical prognostic groupings for localised prostate cancers are imprecise, with 30-50% of patients recurring after image-guided radiotherapy or radical prostatectomy. We aimed to test combined genomic and microenvironmental indices in prostate cancer to improve risk stratification and complement clinical prognostic factors. METHODS We used DNA-based indices alone or in combination with intra-prostatic hypoxia measurements to develop four prognostic indices in 126 low-risk to intermediate-risk patients (Toronto cohort) who will receive image-guided radiotherapy. We validated these indices in two independent cohorts of 154 (Memorial Sloan Kettering Cancer Center cohort [MSKCC] cohort) and 117 (Cambridge cohort) radical prostatectomy specimens from low-risk to high-risk patients. We applied unsupervised and supervised machine learning techniques to the copy-number profiles of 126 pre-image-guided radiotherapy diagnostic biopsies to develop prognostic signatures. Our primary endpoint was the development of a set of prognostic measures capable of stratifying patients for risk of biochemical relapse 5 years after primary treatment. FINDINGS Biochemical relapse was associated with indices of tumour hypoxia, genomic instability, and genomic subtypes based on multivariate analyses. We identified four genomic subtypes for prostate cancer, which had different 5-year biochemical relapse-free survival. Genomic instability is prognostic for relapse in both image-guided radiotherapy (multivariate analysis hazard ratio [HR] 4·5 [95% CI 2·1-9·8]; p=0·00013; area under the receiver operator curve [AUC] 0·70 [95% CI 0·65-0·76]) and radical prostatectomy (4·0 [1·6-9·7]; p=0·0024; AUC 0·57 [0·52-0·61]) patients with prostate cancer, and its effect is magnified by intratumoral hypoxia (3·8 [1·2-12]; p=0·019; AUC 0·67 [0·61-0·73]). A novel 100-loci DNA signature accurately classified treatment outcome in the MSKCC low-risk to intermediate-risk cohort (multivariate analysis HR 6·1 [95% CI 2·0-19]; p=0·0015; AUC 0·74 [95% CI 0·65-0·83]). In the independent MSKCC and Cambridge cohorts, this signature identified low-risk to high-risk patients who were most likely to fail treatment within 18 months (combined cohorts multivariate analysis HR 2·9 [95% CI 1·4-6·0]; p=0·0039; AUC 0·68 [95% CI 0·63-0·73]), and was better at predicting biochemical relapse than 23 previously published RNA signatures. INTERPRETATION This is the first study of cancer outcome to integrate DNA-based and microenvironment-based failure indices to predict patient outcome. Patients exhibiting these aggressive features after biopsy should be entered into treatment intensification trials. In early pregnancy, abortion can be induced by blocking the actions of progesterone receptors (PR). However, the PR antagonist, mifepristone (RU38486), is rather unselective in clinical use because it also cross-reacts with other nuclear receptors. Since the ligand-binding domain of human progesterone receptor (hPR) and androgen receptor (hAR) share 54% identity, we hypothesized that derivatives of dihydrotestosterone (DHT), the cognate ligand for hAR, might also regulate the hPR. Compounds designed and synthesized in our laboratory were investigated for their affinities for hPRB, hAR, glucocorticoid receptor (hGRα) and mineralocorticoid receptor (hMR), using whole cell receptor competitive binding assays. Agonistic and antagonistic activities were characterized by reporter assays. Nuclear translocation was monitored using cherry-hPRB and GFP-hAR chimeric receptors. Cytostatic properties and apoptosis were tested on breast cancer cells (MCF7, T-47D). One compound presented a favorable profile with an apparent neutral hPRB antagonistic function, a selective cherry-hPRB nuclear translocation and a cytostatic effect. 3D models of human PR and AR with this ligand were constructed to investigate the molecular basis of selectivity. Our data suggest that these novel DHT-derivatives provide suitable templates for the development of new selective steroidal hPR antagonists.

Herein we provide a detailed molecular analysis of the spatial heterogeneity of clinically localized, multifocal prostate cancer to delineate new oncogenes or tumor suppressors. We initially determined the copy number aberration (CNA) profiles of 74 patients with index tumors of Gleason score 7. Of these, 5 patients were subjected to whole-genome sequencing using DNA quantities achievable in diagnostic biopsies, with detailed spatial sampling of 23 distinct tumor regions to assess intraprostatic heterogeneity in focal genomics. Multifocal tumors are highly heterogeneous for single-nucleotide variants (SNVs), CNAs and genomic rearrangements. We identified and validated a new recurrent amplification of MYCL, which is associated with TP53 deletion and unique profiles of DNA damage and transcriptional dysregulation. Moreover, we demonstrate divergent tumor evolution in multifocal cancer and, in some cases, tumors of independent clonal origin. These data represent the first systematic relation of intraprostatic genomic heterogeneity to predicted clinical outcome and inform the development of novel biomarkers that reflect individual prognosis.

Corn, Zea mays L., is the most abundant field crop in Iowa, and there are many insect pests associated with this field crop. Although aphids are not typically economically important in corn, recent observations have indicated several aphid species developing heavy populations in northwest Iowa and southwest Minnesota. Historically, the corn leaf aphid, Rhopalosiphum maidis (Hemiptera: Aphididae), has been the most abundant aphid species in corn; however, the bird cherry oat aphid, R. padi (Hemiptera: Aphididae), and several other species have also been detected. Recent observations show a shift to populations peaking later in the summer. Damage potential and management guidelines for aphids in corn are not well defined and this research is aimed at developing economic threshold and sampling protocols.

En las últimas temporadas se ha relacionado la presencia de trips con daños observados en cerezas en Mendoza, Argentina. Los daños serían producidos por la oviposición de estos insectos sobre frutos pequeños. El objetivo del trabajo fue comparar la evolución de poblaciones de las especies de trips en dos montes frutales con diferente manejo de malezas. Se tomaron muestras de malezas y de flores de cerezo en los dos montes y se analizó la evolución de las poblaciones de trips. Tres especies de trips fueron frecuentes en flores de cerezo en ambos montes: Frankliniella australis, F. occidentalis y Thrips tabaci. F. australis es la de mayor reproducción de las tres. No se observaron diferencias consistentes en cuanto al número de trips por flor de cerezo en ambas fincas y esto puede atribuirse a que la abundancia de dichos insectos en las flores de cerezo depende de otras variables tales como densidad de plantación, aporte por migración de áreas vecinas además de los niveles de infestación de malezas en el cultivo.

En cerezos plantas con excesivo vigor son poco precoces, a menudo poco productivas y de difícil manejo en el cultivo. El exceso de vigor puede ser controlado con el uso de estrategias de riego deficitario controlado (RDC). Para contribuir a la racionalización del uso del recurso hídrico, controlar el crecimiento vegetativo vigoroso y estimular la producción precoz en plantaciones jóvenes de cerezo, se estableció un ensayo de RDC en un monte frutal comercial de la variedad Bing regado por goteo en la localidad de Agua Amarga, Mendoza, Argentina, Se evaluó la respuesta a distintos regímenes de riego poscosecha sobre parámetros de crecimiento vegetativo (crecimiento de brotes y tronco, área y peso seco foliar), reproductivo (densidad de floración, rendimiento y calidad de frutos) y estado nutricional (nutrimentos foliares y reservas de carbohidratos no estructurales). Los tratamientos de riego poscosecha fueron: riego a demanda plena (T1= Etc 100 %) y RDC reponiendo el 75 % (T2= Etc 75 %) y 50 % (T3= Etc 50 %) respecto de T1. Se midió el estado hídrico de la planta a través del potencial agua del tallo a mediodía y del suelo con sonda de capacitancia y gravimetría. En T3 disminuyó la longitud de brotes, número y longitud de entrenudos, número de hojas, área foliar y peso seco foliar, y área de tronco. En T2 disminuyó la longitud de brotes y de entrenudos. En T3 la intensidad del déficit hídrico impuesta aumentó la calidad de los ramilletes y la producción de yemas de flor, flores y frutos en el ciclo vegetativo siguiente. La calidad y madurez de frutos no fue afectada por los tratamientos de RDC, aunque en T3 aumentó levemente la proporción de frutos dobles. Luego del primer año de RDC en las plantas del T3 hubo una disminución significativa, aunque leve, del contenido de Ky P foliares y de almidón en raíces, El potencial hídrico del tallo a mediodía resultó un buen indicador del estado hídrico de las plantas. En cerezos un ajuste preciso del nivel de restricción hidrica poscosecha puede ser una estrategia de manejo para controlar el vigor y estimular la producción precoz, Al mismo tiempo se ahorran importantes cantidades de agua.

Durante dos años se realizó un ensayo de RDC en un monte frutal comercial joven y de alto vigor de cerezos Bing, plantado en suelo árido poco profundo y regado por goteo. Se evaluó la respuesta a distintos regímenes de riego sobre el crecimiento de brotes terminales y vigorosos, área y peso seco foliar, y crecimiento de tronco. Los tratamientos de riego fueron: T1 = 100%, T2 = 75% y T3 = 50% de la evapotranspiración máxima (ETc full), respectivamente. Se midió periódicamente el estado hídrico de la planta a través del potencial agua del tallo a mediodía y el estado hídrico del suelo mediante gravimetría. En T3 disminuyó la longitud de brotes, número y longitud de entrenudos, número de hojas, área foliar y peso seco foliar, y área de tronco. En T2 disminuyó la longitud de brotes y entrenudos y el área de sección de tronco. El potencial hídrico del tallo a mediodía fue un buen indicador del estado hídrico de las plantas. En cerezos, un ajuste preciso del nivel de restricción hídrica puede ser una estrategia de manejo para controlar vigor y para ahorrar importantes cantidades de agua.
