In natural streptavidin, tryptophan 120 of each subunit makes contacts with the biotin bound by an adjacent subunit through the dimer-dimer interface. To understand quantitatively the role of tryptophan 120 and its intersubunit communication in the properties of streptavidin, a streptavidin mutant in which tryptophan 120 is converted to phenylalanine was produced and characterized. The streptavidin mutant forms a tetrameric molecule and binds one biotin per subunit, as does natural streptavidin, indicating that the mutation of tryptophan 120 to phenylalanine has no significant effect on the basic properties of streptavidin. However, its biotin-binding affinity was reduced substantially, to approximately 10(8) M-1, indicating that the contact made by tryptophan 120 to biotin has a considerable contribution to the extremely tight biotin binding by streptavidin. The mutant retained bound biotin over a wide pH range or with the addition of urea up to 6 M at neutral pH. However, bound biotin was efficiently released by the addition of excess free biotin due, presumably, to exchange reactions. Electrophoretic analysis revealed that the intersubunit contact made by tryptophan 120 to biotin through the dimer-dimer interface is the major interaction responsible for the biotin-induced, tighter subunit association of streptavidin. In addition, the mutant has weaker subunit association than natural streptavidin even in the absence of biotin, indicating that tryptophan 120 also contributes to the subunit association of tetramers in the absence of biotin.

Os poemas homéricos integram no tecido narrativo evidências do processo oral de transmissão poética a que as suas origens estão associadas. Seria, pois, a voz hábil e poderosa do aedo o meio através do qual os heróis do passado alcançavam renome, passando à póstuma memória como protagonistas das mais notáveis façanhas. Contudo, enquanto a epopeia celebriza em tom solene e grave a memória de um passado glorioso, contrariamente a poesia herói-cómica, que se caracteriza pelo contraste entre a expressão elevada e o tema vulgar, torna objecto de canto incidentes triviais. Neste capítulo, reflectirei sobre a subversão do valor encomiástico da poesia épica antiga n’ O Hissope de Cruz e Silva, poema herói-cómico português, que celebra, num estilo alto e sublimado, próprio da epopeia, uma das mais frívolas questiúnculas ocorridas na Sé de Elvas, na segunda metade de setecentos, entre o Deão José Lara e o Bispo D. Lourenço de Lancastro. Demonstrarei que o poema em causa glorifica, de forma paródica e satírica, acções grosseiras e insignificantes, inserindo a realidade social, não num passado heróico, mas num presente vicioso e ordinário. Assiste-se, assim, neste texto, a uma imitação dos recursos épicos não para imortalizar feitos gloriosos, mas para preservar a memória de bagatelas. Significativo é o episódio (cuja análise dou especial atenção) em que Vidigal entretém o banquete em casa do deão, no canto VII de O Hissope. O padre cantor afirma categórico que deixará em silêncio os grandes sucessos “em mais remotos tempos celebrados”, optando por glorificar a memoranda Elvas com o relato musical de três incidentes triviais nela ocorridos.
