Seedborne peanut viruses pose important constraints to peanut production and safe movement of germ plasm. They also pose a risk of accidental introduction into previously disease-free regions. We have developed reverse transcription-polymerase chain reaction (RT-PCR) assays based on identical cycling parameters which identified peanut stripe, Peanut mottle, Peanut stunt, and Cucumber mosaic viruses through production of specific DNA fragments of 234 bp, 327 bp, 390 bp, and 133 bp, respectively. Assay sensitivity in the picogram range was achieved. The two potyviruses and two cucumoviruses could be differentiated using duplex RT-PCR assays. These assays should be useful for testing peanut leaves or seeds for virus identification in epidemiological studies, seed testing or in post-entry quarantine.

Two putative promoters from Australian banana streak badnavirus (BSV) isolates were analysed for activity in different plant species. In transient expression systems the My (2105 bp) and Cv (1322 bp) fragments were both shown to have promoter activity in a wide range of plant species including monocots (maize, barley, banana, millet, wheat, sorghum), dicots (tobacco, canola, sunflower, Nicotiana benthamiana, tipu tree), gymnosperm (Pinus radiata) and fern (Nephrolepis cordifolia). Evaluation of the My and Cv promoters in transgenic sugarcane, banana and tobacco plants demonstrated that these promoters could drive high-level expression of either the green fluorescent protein (GFP) or the beta -glucuronidase (GUS) reporter gene (uidA) in vegetative plant cells. In transgenic sugarcane plants harbouring the Cv promoter, GFP expression levels were comparable or higher (up to 1.06% of total soluble leaf protein as GFP) than those of plants containing the maize ubiquitin promoter (up to 0.34% of total soluble leaf protein). GUS activities in transgenic in vitro-grown banana plants containing the My promoter were up to seven-fold stronger in leaf tissue and up to four-fold stronger in root and corm tissue than in plants harbouring the maize ubiquitin promoter. The Cv promoter showed activities that were similar to the maize ubiquitin promoter in in vitro-grown banana plants, but was significantly reduced in larger glasshouse-grown plants. In transgenic in vitro-grown tobacco plants, the My promoter reached activities close to those of the 35S promoter of cauliflower mosaic virus (CaMV), while the Cv promoter was about half as active as the CaMV 35S promoter. The BSV promoters for pregenomic RNA represent useful tools for the high-level expression of foreign genes in transgenic monocots.

Transgenic tobacco plants expressing a phenylalanine ammonia-lyase cDNA (ShPAL), isolated from Stylosanthes humilis, under the control of the 35S promoter of the cauliflower mosaic virus were produced to test the effect of high level PAL expression on disease resistance. The transgenic plants showed up to eightfold PAL activity and were slowed in growth and flowering relative to non-transgenic controls which have segregated out the transgene. The expression of the ShPAL transgene and elevated PAL levels were correlated and stably inherited. In T-1 and T-2 tobacco plants with increased PAL activity, lesion expansion was significantly reduced by up to 55% on stems inoculated with the Oomycete pathogen Phytophthora parasitica pv. nicotianae, Lesion area was significantly reduced by up to 50% on leaves inoculated with the fungal pathogen Cercospora nicotianae. La familia Rhabdoviridae incluye varios patógenos económicamente importantes de cultivos, entre los más de 70 virus que afectan plantas. Estos últimos se clasifican en los géneros Cytorhabdovirus y Nucleorhabdovirus, dependiendo de si producen inclusiones en el espacio perinuclear, o si desarrollan viriones citoplasmáticos. Los integrantes de esta familia infectan gran cantidad de monocotiledóneas y dicotiledóneas y la mayoría son dependientes de transmisión por insectos. Las interacciones virus-vector son altamente específicas, y se ha registrado la replicación en insectos, del rhabdovirus que transmiten a las plantas. Cada especie de rhabdovirus induce un amplio espectro de síntomas en sus plantas huéspedes, y estos van desde la falta de efectos discernibles hasta la muerte total de la planta. El maíz (Zea mays L.) es el cultivo más ampliamente distribuido a nivel mundial y uno de los principales cultivos de cereales, ubicándose tercero en el ranking de producción en el mundo. En maíz se ha citado la presencia de varios rhabdovirus, entre estos American wheat striate mosaic virus (AWSMV), Cereal chlorotic mottle virus (CCMV), Maize mosaic virus (MMV), Maize sterile stunt virus (strains of Barley yellow striate virus), Northern cereal mosaic virus (NCMV) y Maize fine streak virus (MFSV). Ninguno de ellos reportado en Argentina. Desde 2001 un rhabdovirus es observado, por sintomatología y microscopía electrónica, en plantas de maíz de diferentes localidades de la provincia de Córdoba. Esta virosis pudo ser transmitida en dos oportunidades a plantas de maíz sanas mediante Peregrinus maidis y logró amplificarse mediante RT-PCR con iniciadores degenerados, el gen de la polimerasa L. Nuestra hipótesis es que el agente causal de la sintomatología de mosaico estriado amarillo en maíz sería un rhabdovirus emergente en Argentina, diferente de Maize mosaic virus (MMV), transmitido por delfácidos, que puede aislarse y mantenerse en condiciones controladas. El objetivo del presente trabajo es generar conocimientos biológicos, moleculares y epidemiológicos sobre el agente causal de la sintomatología en maíz de mosaico estriado amarillo. Para ello se colectarán plantas de maíz con sintomatología de mosaico estriado amarillo, en distintas localidades donde se presente la sintomatología. Las muestras se observarán al microscopio electrónico en cortes ultrafinos y en “leaf dip". Los viriones se purificarán, extraerá el RNA de los mismos, y obtendrá la secuencia de nucleótidos, para compararla con otras publicadas de virosis vegetales y se obtendrán homologías. Se realizarán transmisiones experimentales de esta virosis, por incisiones vasculares y mediante el empleo de diferentes especies de insectos vectores. Importancia del proyecto El avance de patógenos tropicales hacia zonas templadas es una de las causas de la aparición de las virosis emergentes, que se caracterizan por producir epifítias al ingresar a nuevos ecosistemas. El Maize mosaic virus (MMV) es un rhabdovirus que produce una de las virosis más importantes del maíz en el continente americano. Determinar la identidad del agente etiológico del mosaico estriado amarillo y establecer su relación con MMV es fundamental para desarrollar medidas proactivas y diseñar estrategias de manejo de esta nueva enfermedad.

We conducted a study of the patterns and dynamics of oxidized fatty acid derivatives (oxylipins) in potato leaves infected with the late-blight pathogen Phytophthora infestans. Two 18-carbon divinyl ether fatty acids, colneleic acid and colnelenic acid, accumulated during disease development. To date, there are no reports that such compounds have been detected in higher plants. The divinyl ether fatty acids accumulate more rapidly in potato cultivar Matilda (a cultivar with increased resistance to late blight) than in cultivar Bintje, a susceptible cultivar. Colnelenic acid reached levels of up to approximately 24 nmol (7 microgram) per g fresh weight of tissue in infected leaves. By contrast, levels of members of the jasmonic acid family did not change significantly during pathogenesis. The divinyl ethers also accumulated during the incompatible interaction of tobacco with tobacco mosaic virus. Colneleic and colnelenic acids were found to be inhibitory to P. infestans, suggesting a function in plant defense for divinyl ethers, which are unstable compounds rarely encountered in biological systems.

The constitutive Cauliflower Mosaic Virus 35S promoter (CaMV 35S) is widely used as a tool to express recombinant proteins in plants, but with different success. We previously showed that the expression of an F-actin marker, GFP-talin, in Physcomitrella patens using the CaMV 35S promoter failed to homogenously label moss tissues. Here, we show a significant diminution of the GFP fluorescence in dark grown old moss cells and complete lack of labelling in newly differentiated cells. Furthermore, we demonstrate that stable moss lines harbouring a resistance cassette driven by the CaMV 35S are unable to grow in darkness in the presence of the antibiotic. In contrast to the CaMV 35S, the heat inducible promoter, hsp17.3B showed uniform expression pattern in all cells and tissues following a mild heat shock.

Arabidopsis thaliana (L.) Heynh. expressing the Crepis palaestina (L.) linoleic acid delta12-epoxygenase in its developing seeds typically accumulates low levels of vernolic acid (12,13-epoxy-octadec-cis-9-enoic acid) in comparison to levels found in seeds of the native C. palaestina. In order to determine some of the factors limiting the accumulation of this unusual fatty acid, we have examined the effects of increasing the availability of linoleic acid (9cis, 12cis-octadecadienoic acid), the substrate of the delta12-epoxygenase, on the quantity of epoxy fatty acids accumulating in transgenic A. thaliana. The addition of linoleic acid to liquid cultures of transgenic plants expressing the delta12-epoxygenase under the control of the cauliflower mosaic virus 35S promoter increased the amount of vernolic acid in vegetative tissues by 2.8-fold. In contrast, the addition to these cultures of linoelaidic acid (9trans, 12trans-octadecadienoic acid), which is not a substrate of the delta12-epoxygenase, resulted in a slight decrease in vernolic acid accumulation. Expression of the delta12-epoxygenase under the control of the napin promoter in the A. thaliana triple mutant fad3/fad7-1/fad9, which is deficient in the synthesis of tri-unsaturated fatty acids and has a 60% higher level of linoleic acid than the wild type, was found to increase the average vernolic acid content of the seeds by 55% compared to the expression of the delta12-epoxygenase in a wild-type background. Together, these results reveal that the availability of linoleic acid is an important factor affecting the synthesis of epoxy fatty acid in transgenic plants.

O objetivo deste trabalho foi avaliar a incidência de viroses e enfezamentos e estimar as perdas causadas por enfezamentos na cultura do milho safrinha. Os diagnósticos baseados em sintomas foram confirmados por PCR ou RTPCR. Em todas as lavouras, foram identificadas plantas com sintomas de enfezamentos, em incidência de 6,2% a 49,9% (média de 20,7%). Na identificação de insetos vetores desses patógenos, a cigarrinha Dalbulus maidis foi detectada em 20 lavouras das 24 amostradas, constituindo 66,6% do total de espécimens de cigarrinhas coletadas. A perda potencial causada pelos enfezamentos no período foi estimada em cerca de 16,5 milhões de dólares. A ocorrência de plantas com sintomas de "Maize rayado fino virus" e "Maize dwarf mosaic virus" foi baixa e o diagnóstico confirmado por RTPCR. A análise de 441 amostras suspeitas de infecção por "Mal de Río Cuarto virus", por DASELISA, mostrou ausência desse vírus. Resultados de PCR indicaram a presença de um possível fitoplasma distinto de "Maize bushy stunt phytoplasma" em duas plantas apresentando nanismo acentuado, folhas estreitas, enrijecidas, com deformações, e grãos na inflorescência, havendo necessidade de mais estudos para a confirmação da identidade desse possível novo fitoplasma.

O objetivo deste trabalho foi elucidar a atividade e a expressão isoenzimática das esterases, das peroxidases e das aspartato aminotransferases em função da infecção de plantas de trigo pelo Soil-borne wheat mosaic virus (SBWMV). Foram analisadas, aos 45 dias após a emergência, quatro cultivares e uma linhagem de trigo, com diferentes níveis de resistência ao SBWMV: BRS Guabiju, BRS 194, BRS 179, BR 23 e PF 980524. De modo geral, ocorreram diferenças qualitativas e quantitativas intra e interpopulacional, quando comparadas plantas assintomáticas e sintomáticas ao SBWMV. Para o sistema esterase, nove padrões de bandas foram determinados e para peroxidase e aspartato aminotransferase foram detectados três padrões de bandas, para ambas as condições. Padrões eletroforéticos foram observados para plantas infectadas, quando comparadas com as não infectadas, destacando-se a atividade da esterase, o que permitiu identificar com maior precisão o estado metabólico e diferenciado das células.

O objetivo deste trabalho foi avaliar a base genética da resistência de Lycopersicon hirsutum ao potyvírus Pepper yellow mosaic virus (PepYMV). Foram avaliadas 540 plantas, inclusive os parentais 'Santa Clara' (suscetível) e 'BGH 6902' (resistente), e as gerações F1, F2, RC1:1 e RC1:2, derivadas do cruzamento desses parentais. As plantas receberam inoculações mecanicamente, e a concentração viral de PepYMV em cada planta foi determinada por ELISA indireto. Foram realizadas as análises quantitativa e qualitativa. A primeira, baseada na concentração viral de cada planta, indicou herança oligogênica com herdabilidade de 99%. Os mesmos dados, quando analisados de forma qualitativa, indicaram herança governada por dois genes, com interação epistática dominante e recessiva. Entretanto, quando foi analisada a geração F2:3, oriunda da autofecundação de plantas F2 resistentes, a hipótese de dois genes foi descartada e a de um gene, com dominância completa entre os alelos, foi a que melhor se ajustou aos dados. A análise qualitativa, pela sintomatologia observada, demonstrou que a herança da resistência ao PepYMV é determinada por um gene recessivo, com ausência de dominância entre os seus alelos.

O objetivo deste trabalho foi selecionar famílias F4 de alface, oriundas do cruzamento entre as cultivares Verônica e Salinas 88, para o cultivo no verão, com relação ao tipo de folha, e à resistência ao Lettuce mosaic virus (LMV) e ao nematóide-das-galhas Meloidogyne javanica. Primeiramente, avaliaram-se a coloração da folha, tipos de borda e limbo foliares, além da tolerância ao calor no campo, em blocos ao acaso compostos pelas 15 famílias F4 previamente selecionadas, pelas cultivares parentais e pela cultivar testemunha Regina 71 (folhas lisas e tolerante ao calor), com cinco repetições e oito plantas por parcela. Na segunda etapa, as famílias foram avaliadas quanto à resistência ao LMV e ao nematóide-das-galhas, em bandejas de 128 células acondicionadas em estufa. As médias das notas atribuídas a cada família foram comparadas às médias de cada cultivar parental pelo teste de Dunnet (5%). A família AFX007B-140-21, homozigota resistente aos nematóides e ao LMV e, também, tolerante ao calor, foi a mais promissora. O cruzamento entre uma cultivar de folhas crespas e soltas com uma de folhas crespas e repolhuda, pode originar linhagens promissoras tanto de folhas crespas quanto de folhas lisas.

Salicylic acid (SA) plays a central role as a signalling molecule involved in plant defense against microbial attack. Genetic manipulation of SA biosynthesis may therefore help to generate plants that are more disease-resistant. By fusing the two bacterial genes pchA and pchB from Pseudomonas aeruginosa, which encode isochorismate synthase and isochorismate pyruvate-lyase, respectively, we have engineered a novel hybrid enzyme with salicylate synthase (SAS) activity. The pchB-A fusion was expressed in Arabidopsis thaliana under the control of the constitutive cauliflower mosaic virus (CaMV) 35S promoter, with targeting of the gene product either to the cytosol (c-SAS plants) or to the chloroplast (p-SAS plants). In p-SAS plants, the amount of free and conjugated SA was increased more than 20-fold above wild type (WT) level, indicating that SAS is functional in Arabidopsis. P-SAS plants showed a strongly dwarfed phenotype and produced very few seeds. Dwarfism could be caused by the high SA levels per se or, perhaps more likely, by a depletion of the chorismate or isochorismate pools of the chloroplast. Targeting of SAS to the cytosol caused a slight increase in free SA and a significant threefold increase in conjugated SA, probably reflecting limited chorismate availability in this compartment. Although this modest increase in total SA content did not strongly induce the resistance marker PR-1, it resulted nevertheless in enhanced disease resistance towards a virulent isolate of Peronospora parasitica. Increased resistance of c-SAS lines was paralleled with reduced seed production. Taken together, these results illustrate that SAS is a potent tool for the manipulation of SA levels in plants.

The objective of the present work was to determine the inheritance and stability of transgenes of a transgenic bean line expressing the genes rep-trap-ren from Bean golden mosaic virus and the bar gene. Crosses were done between the transgenic line and four commercial bean cultivars, followed by four backcrosses to the commercial cultivars. Progenies from each cross were evaluated for the presence of the transgenes by brushing the leaves with glufosinate ammonium and by polymerase chain reaction using specific oligonucleotides. Advanced generations were rub-inoculated with an isolate of Bean common mosaic necrosis virus (BCMNV). The transgenes were inherited consistently in a Mendelian pattern in the four crosses studied. The analyzed lines recovered close to 80% of the characteristics of the recurrent parent, as determined by the random amplified DNA markers used, besides maintaining important traits such as resistance to BCMNV. The presence of the transgene did not cause any detectable undesirable effect in the evaluated progenies.

O objetivo deste trabalho foi determinar a presença do alelo Pvr4, que confere resistência contra o PepYMV (Pepper yellow mosaic virus), em genótipos de pimentão comunmente encontrados no mercado brasileiro, com uso de um marcador molecular codominante tipo CAPS. A resistência ao PepYMV, nos genótipos CM-334-INRA, Myr-29 e em genótipos derivados do híbrido comercial Mônica-R, foi detectada como associada à banda de 444 pb, ligada ao alelo de resistência Pvr4. As plantas resistentes homozigotas (pvr4/pvr4) mostraram uma banda de 444 pb, as suscetíveis (Pvr4+/Pvr4+) uma banda de 458 pb e as resistentes heterozigotas (Pvr4+/Pvr4) mostraram as duas bandas. No entanto, no acesso resistente CM-334-UFV, nos híbridos Magali-R e Martha-R, assim como em populações derivadas desse acesso e desses híbridos, a resistência ao PepYMV não esteve associada ao marcador CAPS. O acesso CM-334-UFV ('Criollo de Morelos-334', de Viçosa, MG) distinguiu-se do CM-334-INRA ('Criollo de Morelos-334', da França); embora ambos os acessos tenham sido resistentes ao PepYMV, apenas em CM-334-INRA foi encontrada a associação da resistência com a banda de 444 pb.
