Atomic force microscopy (AFM) and lateral force microscopy (LFM) were used simultaneously to analyze a model membrane bilayer structure consisting of a phospholipid outer monolayer deposited onto organosilane-derivatized mica surfaces, which were constructed by using painting and self-assembly methods. The phospholipid used as outer monolayer was dimyristoylphosphatidylcholine (DMPC). The hydrocarbon-covered substrate that formed the inner half bilayer was composed of a self-assembly monolayer (SAM) of octadecyltrichloroorganosilane (OTS) on mica. SAMs of DMPC were formed by exposing hydrophobic mica to a solution of DMPC in decane/isobutanol and subsequently immersing into pure water. AFM images of samples immersed in solution for varying exposure times showed that before forming a complete monolayer the molecules aggregated into dense islands (2.2-2.6 nm high) on the surface. The islands had a compact and rounded morphology. LFM, coupled with topographic data obtained with the atomic force mode, had made possible the distinction between DMPC and OTS. The rate constant of DMPC growth was calculated. This is the first systematic study of the SAM formation of DMPC by AFM and LFM imaging. It reveals more direct information about the film morphology than previous studies with conventional surface analytical techniques such as infrared spectroscopy, X-ray, or fluorescence microscopy.

Multilayers of Y-type bilayers of pure and mixed erbium palmitate(EP), nonadecanate(Er) and behenate(EB) on CaF2 substrates were prepared by conventional Langmuir-Blodgett (LB) method. II is demonstrated that two systems composed of alternating bilayer of different fatty acid salts are unidimensional superlattices. These LB films were characterized by means of x-ray photoelectronic Spectrometry (XPS), FTIR and x-ray diffraction (XRD) measurements.

The ion channel sensor is reviewed. The concept and sensing principle of this kind of sensor are briefly discussed. The fabrication of the sensing membrane and the application of the ion channel sensor in electroanalytical chemistry are evaluated. The future developing direction is also anticipated.

Monolayers of biological compounds including redox proteins and enzymes, and phospholipids have been immobilized on a gold electrode surface through self-assembling. These proteins and enzymes, such as cytochrome c, cytochrome c oxidase and horseradish peroxidase (HRP), immobilized covalently to the self-assembled monolayers (SAMs) of 3-mercaptopropionic acid on a gold electrode, communicate directly electrons with the electrode surface without mediators and keep their physiological activities. The electron transfer of HRP with the gold electrode can also be mediated by the alkanethiol SAMs with electroactive group viologens on the gold electrode surface. All these direct electrochemistries of proteins and enzymes might offer an opportunity to build a third generation of biosensors without mediators for analytes, such as H2O2, glucose and cholesterol. Monensin and valinomycin have been incorporated into the bilayers on the gold electrode consisting of the SAMs of alkanethiol and a lipid monolayer, which have high selectivity for monovalent ions, and the resulting Na+ or K+ sensor has a wide linear range and high stability. These self-assembly systems provide a good mimetic model for studying the physiological function of a membrane and its associated enzyme. (C) 1997 Elsevier Science S.A.

Supported lipid membranes consisting of self-assembled alkanethiol and lipid monolayers on gold substrates could be produced by three different deposition methods: the Langmuir-Blodgett (L-B) technique, the painted method, and the paint-freeze method, By using cyclic voltammetry, chronoamperometry/chronocoulometry and a.c. impedance measurements, we demonstrated that lipid membranes prepared by these three deposition methods had obvious differences in specific capacitance, resistance and thickness. The specific capacitance of lipid membranes prepared by depositing an L-B monolayer on the alkanethiol alkylated surfaces was 0.53 mu Fcm(-2), 0.44 mu Fcm(-2) by the painted method and 0.68 mu Fcm(-2) by the paint-freeze method. The specific conductivity of lipid membranes prepared by the L-B method was over three times lower than that of the painted lipid membranes, while that of the paint-freeze method was the lowest. The difference among the three types of lipid membranes was ascribed to the influence of the organic solvent in lipid films and the changes in density of the films. The lipid membranes prepared by the usual painted method contained a trace amount of the organic solvent. The organic solvent existing in the hydrocarbon core of the membrane reduced the density of the membrane and increased the thickness of the membrane. The membrane prepared by depositing an L-B monolayer containing no solvent had higher density and the lowest fluidity, and the thickness of the membrane was smaller. The lipid membrane prepared by the paint-freeze method changed its structure sharply at the lower temperature. The organic solvent was frozen out of the membrane while the density of the membrane increased greatly. All these caused the membrane to exist in a ''tilted'' state and the thickness of this membrane was the smallest. The lipid membrane produced by the paint-freeze method was a membrane not containing organic solvent. This method was easier in manipulation and had better reproducibility than that of the usual painting method and the method of forming free-standing lipid film. The solvent-free membrane had a long lifetime and a higher mechanical stability. This model membrane would be useful in many areas of scientific research.

Cyclic voltammetry was employed to study the influence of sterols on the lipophilic ion transport through the BLM. The mole fraction of the sterols (cholesterol, oxidized cholesterol). as referred to total lipid, was varied in a range of 0-0.8. Data demonstrate that a thin-layer model is suitable to this BLM system. By this model, the number of charges transported per lipophilic ion, the concentration of the ion in the membrane bulk phase and the aqueous/membrane phase partition coefficient could be calculated. These parameters proved that sterols had an obvious influence on the lipophilic ion transport. Cholesterol had a stronger influence on the ion transport than oxidized cholesterol. Its incorporation into egg lecithin membranes increased the partition coefficient beta of the ion up to more than 3-fold. Yet, oxidized cholesterol incorporated into egg lecithin membranes only increased the beta up to less than 2-fold, and the beta had no great variation at different oxidized cholesterol mole fractions. The higher beta obtained was partly due to the trace amount of solvent existing in the core of the lipid bilayers. At the different sterol mole fractions, combining the change of beta with the change of peak current, we also concluded that sterols had somewhat inhibiting effect on the ion transport at the higher sterols mole fraction (>0.4). These results are explained in terms of the possible change of dipole potential of the membrane produced by sterols and the decrease of the membrane fluidity caused by the condensation effect of sterols and the thinning effect caused by sterols. The substituting group (in the oxidized cholesterol) had some inhibiting effects on the ion transport at higher mole fractions (oxidized cholesterol mole fraction >0.4).

Long-range ordered stripes domain structures were observed in Dipalmitoylphosphatidylcholine (DPPC) Langmuir-Blodgett monolayer film which was spread on the subphase of lanthanide ion (Eu3+) solution and transferred to a freshly cleaved mica substrate by vertical deposition. This novel phenomenon was discussed in terms of the competitive interaction of dipole-dipole and electrostatic interactions of the DPPC molecules combined with lanthanide ions with those DPPC molecules free of lanthanide ions.

With the wide application of rare earth in agriculture, medicament, especially the application of Gd-DTPA as nuclear magnetic resonance image reagent in clinical practice([1]), the studies on the toxicology in biological body, as well as the study on the use as informative probes instead of divalent calcium ion in biological and biochemical research have attracted intensive concern([2]). Phospholipids bilayers have served as a model of biomembrane in the last two decades. The effects of metal ions on the conformation of polar headgroup of dipalmitoylphosphatidylcholine (DPPC) bilayers have been reported([3]). Sphingomyelin is major component of several biological tissues such as brain and nerve cells and has identical polar headgroup to DPPC. The interaction of metal ions with sphingomyelin bilayer remains nonrevealed. This note presents the results of the study on this aspect.

The interaction of silica with dipalmitoylphosphatidylcholine (DPPC) liposomes has been studied by detecting H-1 and 2D NOESY NMR spectra and measuring proton spin-lattice relaxation time (T-1). The antagonistic effect of aluminium citrate on silica has also been studied by measuring the proton T-1 values, The results show that silica particles mainly bind to the phosphate moiety of the polar headgroup of DPPC bilayers through hydrogen bonds between Si-OH and O-P groups, but they have no significant effect on the conformation of the choline group of the polar headgroup, The addition of silica particles decreases the mobilities of the hydrocarbon chains slightly, Proton T-1 data also indicate that aluminium citrate reduces the effect of silica, A possible mechanism for the toxicity of silica and the antagonistic effect of aluminium titrate on silica is suggested. The effects of lanthanide ions and their complexes of citrate and DTPA ligands on the fluidity of dipalmitoylphosphatidylethanolamine (DPPE) bilayers have been studied by FT-Raman spectroscopy. the results show that lanthanide ions of lower concentrationn decrease the fluidity of acyl chains of DPPE bilayers and change the conformation of C C-C backbone from gauche to the trans lanthanide ions of higher concentration, however, increase the fluidity of acyl chains and increase the gauche population of C-C-C backbone. Lanthanide complex of citrate have no effect on the fluidity of acyl chains of DPPE bilayers in the region of experimental concentration, but La-DTPA complex increase slightly the fluidity of acyl chains. the results also indicated that lanthanide ion of lower concentration changed the lattice packing of hydrocarbon chains from hexagonal form to orthorhombic form, but it is still in hexagonal or distorted hexagonal lattice cell in the gel state in the presence of metal ions and lanthanide complexes of higher concentration

A key element in the rational design of hybrid organic-inorganic nanostructures, is control of surfactant packing and adsorption onto the inorganic phase in crystal growth and assembly. In layered single crystal nanofibers and bilayered 2D nanosheets of vanadium oxide, we show how the chemisorption of preferred densities of surfactant molecules can direct formation of ordered, curved layers. The atom-scale features of the structures are described using molecular dynamics simulations that quantify surfactant packing effects and confirm the preference for a density of 5 dodecanethiol molecules per 8 vanadium attachment sites in the synthesised structures. This assembly maintains a remarkably well ordered interlayer spacing, even when curved. The assemblies of interdigitated organic bilayers on V2O5 are shown to be sufficiently flexible to tolerate curvature while maintaining a constant interlayer distance without rupture, delamination or cleavage. The accommodation of curvature and invariant structural integrity points to a beneficial role for oxide-directed organic film packing effects in layered architectures such as stacked nanofibers and hybrid 2D nanosheet systems.

Experiments are reported which show that currents of low energy ("cold") electrons pass unattenuated through crystalline ice at 135 K for energies between zero and 650 meV, up to the maximum studied film thickness of 430 bilayers, indicating negligible apparent trapping. By contrast, both porous amorphous ice and compact crystalline ice at 40 K show efficient electron trapping. Ice at intermediate temperatures reveals metastable trapping that decays within a few hundred seconds at 110 K. Our results are the first to demonstrate full transmission of cold electrons in high temperature water ice and the phenomenon of temperature-dependent trapping.

O objectivo deste trabalho é a produção de novos eléctrodos modificados com polioxotungstatos (POMs) do tipo Keggin, incluindo POMs lacunares e substituídos por metais de transição. A preparação e caracterização dos polioxotungstatos encontram-se descritas no Capítulo 2. No Capítulo 3 descreve-se a produção de eléctrodos de carbono vítreo funcionalizados com sais híbridos de tetra-n-butilamónio de vários silicotungstatos pelo método de evaporação da gota. As propriedades electroquímicas dos polioxotungstatos imobilizados foram comparadas com as das espécies solúveis correspondentes. A morfologia dos depósitos foi avaliada por microscopia óptica e por microscopia electrónica de varrimento. No capítulo 4 descreve-se a preparação de novos eléctrodos compósitos de carbono e poli(hexilmetacrilato) com fosfotungstatos. Os estudos electroquímicos revelaram que as principais características dos POMs são mantidas e que os processos de redução são controlados por difusão, dependendo da difusão dos protões da solução. O Capítulo 5 descreve a construção de filmes em multicamadas ultrafinos contendo POMs e polietilenimina, preparados pelo método de auto-montagem camada-sobre-camada em eléctrodos de carbono vítreo. Os filmes em multicamada foram caracterizados por voltametria cíclica e por microscopia electrónica de varrimento e foi usada a espectroscopia de absorção de UV-Vis em placas de quartzo para monitorar o crescimento de filme. Os resultados voltamétricos revelaram que os processos de redução dos POM são confinados à superfície. Alguns destes eléctrodos modificados revelaram propriedades electrocatalíticas relativamente à redução dos aniões nitrito, bromato e/ou iodato. A espectroscopia de impedância electroquímica também foi usada na caracterização destes filmes e os resultados revelaram que a resistência à transferência de carga aumenta com o aumento do número de bicamadas para ambas as espécies redox, indicando que a espessura do filme tem um efeito importante sobre a cinética de reacções de transferência de carga. No capítulo 6 descreve-se a síntese de filmes híbridos orgânicos/inorgânicos compostos por poli(3,4-etilenodioxitiofeno) (PEDOT) e por silicotungstatos do tipo Keggin através da polimerização electroquímica, em condições aquosas, na superfície de electrodos de carbono vítreo. A voltametria cíclica revelou que as características principais dos POMs são mantidas nos filmes. Verificou-se que estes filmes são muito estáveis, possivelmente devido a fortes interacções electrostáticas entre os POMs aniónicos e o polímero positivamente carregado. A espectroscopia de impedância electroquímica foi também utilizada e os resultados mostraram que a resistência de transferência de carga aumenta com o aumento do pH e para valores de potenciais mais elevados. O capítulo 7 apresenta as conclusões finais e possíveis trabalhos futuros.

This thesis applies x-ray diffraction to measure he membrane structure of lipopolysaccharides and to develop a better model of a LPS bacterial melilbrane that can be used for biophysical research on antibiotics that attack cell membranes. \iVe ha'e Inodified the Physics department x-ray machine for use 3.'3 a thin film diffractometer, and have lesigned a new temperature and relative humidity controlled sample cell.\Ve tested the sample eel: by measuring the one-dimensional electron density profiles of bilayers of pope with 0%, 1%, 1G :VcJ, and 100% by weight lipo-polysaccharide from Pse'udo'lTwna aeTuginosa. Background VVe now know that traditional p,ntibiotics ,I,re losing their effectiveness against ever-evolving bacteria. This is because traditional antibiotic: work against specific targets within the bacterial cell, and with genetic mutations over time, themtibiotic no longer works. One possible solution are antimicrobial peptides. These are short proteins that are part of the immune systems of many animals, and some of them attack bacteria directly at the membrane of the cell, causing the bacterium to rupture and die. Since the membranes of most bacteria share common structural features, and these featuret, are unlikely to evolve very much, these peptides should effectively kill many types of bacteria wi Lhout much evolved resistance. But why do these peptides kill bacterial cel: '3 , but not the cells of the host animal? For gramnegative bacteria, the most likely reason is that t Ileir outer membrane is made of lipopolysaccharides (LPS), which is very different from an animal :;ell membrane. Up to now, what we knovv about how these peptides work was likely done with r !10spholipid models of animal cell membranes, and not with the more complex lipopolysa,echaricies, If we want to make better pepticies, ones that we can use to fight all types of infection, we need a more accurate molecular picture of how they \vork. This will hopefully be one step forward to the ( esign of better treatments for bacterial infections.
