Ao adotar como foco inicial a referência, feita por Horkheimer e Adorno em "Elementos do Anti-Semitismo: limites do esclarecimento", ao texto de Freud denominado "O Inquietante", o presente trabalho pretende estabelecer alguns pontos de contato entre si no que se refere ao importante campo de estudos voltado ao preconceito. Nestes termos, propõe uma ética e uma estética da alteridade que, voltadas à dissonância de uma estranha-familiaridade, aparecem pautadas pela valorização da negatividade em detrimento de uma filosofia positiva cujas luzes podem conduzir à mais absoluta cegueira totalitária. A aposta aqui reside na possibilidade de que, remetendo às fraturas do contato do homem com o que lhe parece real, tal movimento traga consigo o resgate de uma tragicidade que resista em se deixar apropriar pelos anseios de domínio e comodidade representativa tão caros à razão instrumental e, com ela, aos discursos de intolerância para com as diferenças.

Pancreatic ductal adenocarcinoma (PDAC) represents the fourth most common cause of cancer-associated death in the United States. Little progress has been made in understanding how proteotoxic stress affects rapidly proliferating pancreatic tumor cells. Endoplasmic reticulum (ER) stress occurs when protein homeostasis in the ER lumen is perturbed. ER stress activates the unfolded protein response (UPR) to reduce the protein load in the ER. Under conditions of moderate ER stress, the UPR promotes cell cycle arrest which allows time for successful protein load reduction and enables cell survival. However, under conditions of high levels of ER stress the UPR induces cellular apoptosis. In this dissertation, I investigated the role of endoplasmic reticulum (ER) stress and its effects on the cell cycle in pancreatic cancer cells. Activation of the unfolded protein response after ER stress induction was determined by comparing expression of key UPR mediators in non-tumorigenic pancreatic ductal cells to pancreatic cancer cells. Two arms of the UPR were assessed: eIF2α/ATF4/CHOP and IRE1α/XBP1s. Pancreatic cancer cells exhibited altered UPR activation characterized by a delay in both phosphorylation of eIF2α and induction of spliced XBP1. Further evaluation of the UPR-mediated effects on cell cycle progression revealed that pancreatic cancer cells showed a compromised ability to inhibit G1 to S phase progression after ER stress. This reduced ability to arrest proliferation was found to be due to an impaired ability to downregulate cyclin D1, a key gatekeeper of the G1/S checkpoint. Abrogation of cyclin D1 repression was mediated through a slow induction of phosphorylation of eIF2α, a critical mediator of translational attenuation in response to ER stress. In conclusion, pancreatic cancer cells demonstrate a globally compromised ability to regulate the unfolded protein response. This deficiency results in reduced cyclin D1 repression through an eIF2α-mediated mechanism. These findings indicate that pancreatic cancer cells have increased tolerance for elevated ER stress compared to normal cells, and this tolerance results in continued tumor cell proliferation under proteotoxic conditions.

Wilms' tumor (WT) is a childhood embryonic tumor of the kidney. In some cases, WT has been associated with a chromosome deletion in the region 11p13. The majority of WT cases, however, have normal karyotypes with no discernable deletions or rearrangements of chromosome 11.^ To study the genetic events predisposing to the development of WT, I have used a number of gene markers specific for chromosome 11. Gene probes for human catalase and apolipoprotein A1 were localized to chromosome 11 by in situ hybridization. A number of other probes previously mapped to chromosome 11 were also used. Nine WT patients who were heterozygous for at least one 11p marker were shown to lose heterozygosity in their tumor DNA. Gene dosage experiments demonstrated that two chromosomes 11 were present although loss of heterozygosity had occurred in all but two cases. By using gene probes from the short and long arms of chromosome 11, I discerned that loss of heterozygosity was due to somatic recombination in four cases, chromosome deletion in two cases, and chromosome loss and reduplication or somatic recombination in these cases. Examination of DNAs from the parents of six of these patients indicated that the alleles that were lost in tumor tissues were alleles inherited from the mother. In sporadic WT cases one would expect the loss of alleles to be random. These data suggest that the loss of alleles resulting in the development of WT is not a random event, however, the significance of this is not known. ^

Clinical trials have shown a beneficial effect of mechanical thrombectomy in acute ischemic stroke patients treated within six up to even 12 h after symptom onset. This treatment was already performed in selected hospitals in Belgium before completion of the randomized controlled trials. Outcome data on these procedures in Belgium have not been published. We performed a retrospective multicenter study of all patients with acute ischemic stroke treated with mechanical endovascular therapy in four hospitals in Belgium. Clinical outcomes, as measured by the modified Rankin Scale (mRS), site of arterial occlusion, reperfusion and the association between these variables were studied. The study included 80 patients: 65 patients with an occlusion in the anterior circulation and 15 with an occlusion in the posterior circulation. Good functional outcome (GFO) rates, defined as mRS 0-2 at 90 days, were 42 % in all patients, 44 % in anterior circulation stroke and 34 % in posterior circulation stroke. Reperfusion was achieved in 78 % of patients; more (100 %) in patients with posterior compared to patients with anterior circulation stroke (72 %; p = 0.02). The rate of GFO was greater in patients with reperfusion versus patients in whom reperfusion was not achieved (adjusted OR 8.2, 95 % CI 2.0-34.2). Symptomatic intracerebral hemorrhage was documented in 5 % of all patients. Endovascular treatment with mechanical devices for acute ischemic stroke in Belgium results in GFO and reperfusion rates similar to recently published results in the endovascular-treated arms of randomized clinical trials. Rates of symptomatic intracranial hemorrhage are low and comparable to other cohort studies and clinical trials.

Differential Phase Shift Keying (DPSK) modulation format has been shown as a robust solution for next-generation optical transmission systems. One key device enabling such systems is the delay interferometer, converting the signal phase information into intensity modulation to be detected by the photodiodes. Usually, Mach-Zehnder interferometer (MZI) is used for demodulating DPSK signals. In this paper, we developed an MZI which is based on all-fiber Multimode Interference (MI) structure: a multimode fiber (MMF) located between two single-mode fibers (SMF) without any transition zones. The standard MZI is not very stable since the two beams go through two different paths before they recombine. In our design the two arms of the MZI are in the same fiber, which will make it less temperature-sensitive than the standard MZI. Performance of such MZI will be analyzed from transmission spectrum. Finally such all-fiber MI-based MZI (MI-MZI) is used to demodulate 10 Gbps DPSK signals. The demodulated signals are analyzed from eye diagram and bit error rate (BER).

The function of the small-Mr Ras-like GTPase Rap1 remains largely unknown, but this protein has been demonstrated to regulate cortical actin-based morphologic changes in Dictyostelium and the oxidative burst in mammalian neutrophils. To test whether Rap1 regulates phagocytosis, we biochemically analyzed cell lines that conditionally and modestly overexpressed wild-type [Rap1 WT(+)], constitutively active [Rap1 G12T(+)], and dominant negative [Rap1 S17N(+)] forms of D. discoideum Rap1. The rates of phagocytosis of bacteria and latex beads were significantly higher in Rap1 WT(+) and Rap1 G12T(+) cells and were reduced in Rap1 S17N(+) cells. The addition of inhibitors of protein kinase A, protein kinase G, protein tyrosine kinase, or phosphatidylinositide 3-kinase did not affect phagocytosis rates in wild-type cells. In contrast, the addition of U73122 (a phospholipase C inhibitor), calphostin C (a protein kinase C inhibitor), and BAPTA-AM (an intracellular Ca2+ chelator) reduced phagocytosis rates by 90, 50, and 65%, respectively, suggesting both arms of the phospholipase C signaling pathways played a role in this process. Other protein kinase C–specific inhibitors, such as chelerythrine and bisindolylmaleimide I, did not reduce phagocytosis rates in control cells, suggesting calphostin C was affecting phagocytosis by interfering with a protein containing a diacylglycerol-binding domain. The addition of calphostin C did not reduce phagocytosis rates in Rap1 G12T(+) cells, suggesting that the putative diacylglycerol-binding protein acted upstream in a signaling pathway with Rap1. Surprisingly, macropinocytosis was significantly reduced in Rap1 WT(+) and Rap1 G12T(+) cells compared with control cells. Together our results suggest that Rap1 and Ca2+ may act together to coordinate important early events regulating phagocytosis.
