L'objectiu d'aquest estudi és el d'investigar sobre l'ús de matèria orgànica per part dels fongs i bacteris que colonitzen diferents substrats bentònics en rius Mediterranis i analitzar l'efecte dels factors ambientals i antròpics sobre l'estabilitat estructural i funcional de les comunitats del biofilm. La metodologia emprada en aquest estudi consisteix en: i) anàlisi de la biomassa bacteriana i fúngica, ii) anàlisi de la composició de les comunitats bentòniques (identificació d'hifomicets aquàtics i anàlisi del 16S rDNA bacterià), i iii) anàlisi de l'activitat enzimàtica extracel·lular relacionada amb el reciclatge de matèria orgànica en rius.

La gestió de xarxes és un camp molt ampli i inclou molts aspectes diferents. Aquesta tesi doctoral està centrada en la gestió dels recursos en les xarxes de banda ampla que disposin de mecanismes per fer reserves de recursos, com per exemple Asynchronous Transfer Mode (ATM) o Multi-Protocol Label Switching (MPLS). Es poden establir xarxes lògiques utilitzant els Virtual Paths (VP) d'ATM o els Label Switched Paths (LSP) de MPLS, als que anomenem genèricament camins lògics. Els usuaris de la xarxa utilitzen doncs aquests camins lògics, que poden tenir recursos assignats, per establir les seves comunicacions. A més, els camins lògics són molt flexibles i les seves característiques es poden canviar dinàmicament. Aquest treball, se centra, en particular, en la gestió dinàmica d'aquesta xarxa lògica per tal de maximitzar-ne el rendiment i adaptar-la a les connexions ofertes. En aquest escenari, hi ha diversos mecanismes que poden afectar i modificar les característiques dels camins lògics (ample de banda, ruta, etc.). Aquests mecanismes inclouen els de balanceig de la càrrega (reassignació d'ample de banda i reencaminament) i els de restauració de fallades (ús de camins lògics de backup). Aquests dos mecanismes poden modificar la xarxa lògica i gestionar els recursos (ample de banda) dels enllaços físics. Per tant, existeix la necessitat de coordinar aquests mecanismes per evitar possibles interferències. La gestió de recursos convencional que fa ús de la xarxa lògica, recalcula periòdicament (per exemple cada hora o cada dia) tota la xarxa lògica d'una forma centralitzada. Això introdueix el problema que els reajustaments de la xarxa lògica no es realitzen en el moment en què realment hi ha problemes. D'altra banda també introdueix la necessitat de mantenir una visió centralitzada de tota la xarxa. En aquesta tesi, es proposa una arquitectura distribuïda basada en un sistema multi agent. L'objectiu principal d'aquesta arquitectura és realitzar de forma conjunta i coordinada la gestió de recursos a nivell de xarxa lògica, integrant els mecanismes de reajustament d'ample de banda amb els mecanismes de restauració preplanejada, inclosa la gestió de l'ample de banda reservada per a la restauració. Es proposa que aquesta gestió es porti a terme d'una forma contínua, no periòdica, actuant quan es detecta el problema (quan un camí lògic està congestionat, o sigui, quan està rebutjant peticions de connexió dels usuaris perquè està saturat) i d'una forma completament distribuïda, o sigui, sense mantenir una visió global de la xarxa. Així doncs, l'arquitectura proposada realitza petits rearranjaments a la xarxa lògica adaptant-la d'una forma contínua a la demanda dels usuaris. L'arquitectura proposada també té en consideració altres objectius com l'escalabilitat, la modularitat, la robustesa, la flexibilitat i la simplicitat. El sistema multi agent proposat està estructurat en dues capes d'agents: els agents de monitorització (M) i els de rendiment (P). Aquests agents estan situats en els diferents nodes de la xarxa: hi ha un agent P i diversos agents M a cada node; aquests últims subordinats als P. Per tant l'arquitectura proposada es pot veure com una jerarquia d'agents. Cada agent és responsable de monitoritzar i controlar els recursos als que està assignat. S'han realitzat diferents experiments utilitzant un simulador distribuït a nivell de connexió proposat per nosaltres mateixos. Els resultats mostren que l'arquitectura proposada és capaç de realitzar les tasques assignades de detecció de la congestió, reassignació dinàmica d'ample de banda i reencaminament d'una forma coordinada amb els mecanismes de restauració preplanejada i gestió de l'ample de banda reservat per la restauració. L'arquitectura distribuïda ofereix una escalabilitat i robustesa acceptables gràcies a la seva flexibilitat i modularitat.

The INtegrated CAtchment (INCA) model has been developed to simulate the impact of mine discharges on river systems. The model accounts for the key kinetic chemical processes operating as well as the dilution, mixing and redistribution of pollutants in rivers downstream of mine discharges or acid rock drainage sites. The model is dynamic and simulates the day-to-day behaviour of hydrology and eight metals (cadmium, mercury, copper, zinc, lead, arsenic, manganese and chromium) as well as cyanide and ammonia. The model is semi-distributed and can simulate catchments, sub-catchment and in-stream river behaviour. The model has been applied to the Roia Montan Mine in Transylvania, Romania, and used to assess the impacts of old mine adits on the local catchments as well as on the downstream Aries and Mures river system. The question of mine restoration is investigated and a set of clean-up scenarios investigated. It is shown that the planned restoration will generate a much improved water quality from the mine and also alleviate the metal pollution of the river system.

The aim of this three year project funded by the Countryside Council for Wales (CCW) is to develop techniques firstly, to refine and update existing targets for habitat restoration and re-creation at the landscape scale and secondly, to develop a GIS-based model for the implementation of those targets at the local scale. Landscape Character Assessment (LCA) is being used to map Landscape Types across the whole of Wales as the first stage towards setting strategic habitat targets. The GIS habitat model uses data from the digital Phase I Habitat Survey for Wales to determine the suitability of individual sites for restoration to specific habitat types, including broadleaf woodland. The long-term aim is to develop a system that strengthens the character of Welsh landscapes and provides real biodiversity benefits based upon realistic targets given limited resources for habitat restoration and re-creation.

Flavonoids are a diverse class of polyphenolic compounds that are produced as a result of plant secondary metabolism. They are known to play a multifunctional role in rhizospheric plant-microbe and plant-plant communication. Most familiar is their function as a signal in initiation of the legume-rhizobia symbiosis, but, flavonoids may also be signals in the establishment of arbuscular mycorrhizal symbiosis and are known agents in plant defence and in allelopathic interactions. Flavonoid perception by, and impact on, their microbial targets (e.g. rhizobia, plant pathogens) is relatively well characterized. However, potential impacts on 'non-target' rhizosphere inhabitants ('non-target' is used to distinguish those microorganisms not conventionally known as targets) have not been thoroughly investigated. Thus, this review first summarizes the conventional roles of flavonoids as nod gene inducers, phytoalexins and allelochemicals before exploring questions concerning 'non-target' impacts. We hypothesize that flavonoids act to shape rhizosphere microbial community structure because they represent a potential source of carbon and toxicity and that they impact on rhizosphere function, for example, by accelerating the biodegradation of xenobiotics. We also examine the reverse question, 'how do rhizosphere microbial communities impact on flavonoid signals?' The presence of microorganisms undoubtedly influences the quality and quantity of flavonoids present in the rhizosphere, both through modification of root exudation patterns and microbial catabolism of exudates. Microbial alteration and attenuation of flavonoid signals may have ecological consequences for below-ground plant-microbe and plant-plant interaction. We have a lack of knowledge concerning the composition, concentration and bioavailability of flavonoids actually experienced by microbes in an intact rhizosphere, but this may be addressed through advances in microspectroscopic and biosensor techniques. Through the use of plant mutants defective in flavonoid biosynthesis, we may also start to address the question of the significance of flavonoids in shaping rhizosphere community structure and function.

An isolate of Gliocladium virens from disease affected soil in a commercial tomato greenhouse proved highly antagonistic to Fusarium oxysporum f.sp. lycopersici, used together with an isolate of the nematophagus fungus Verticillium chlamydosporium. Significant disease control was obtained when young mycelial preparation (on a food-base culture) of the G. virens together with V. chlamydosporium was applied in potting medium. Similar results were observed when a Trichoderma harzianum isolate was treated in combination with the V. chlamydosporium isolate. Most promising, in terms of minimizing the Fusarium wilt of tomato incidence, was also the effect of the bacteria associated with entomopathogenic nematodes (Steinernema spp.), Pseudomonas oryzihabitans and Xenorhabdus nematophilus.

Polygalacturonase-inhibiting proteins (PGIPs) are extracellular plant inhibitors of fungal endopolygalacturonases (PGs) that belong to the superfamily of Leu-rich repeat proteins. We have characterized the full complement of pgip genes in the bean (Phaseolus vulgaris) genotype BAT93. This comprises four clustered members that span a 50-kb region and, based on their similarity, form two pairs (Pvpgip1/Pvpgip2 and Pvpgip3/Pvpgip4). Characterization of the encoded products revealed both partial redundancy and subfunctionalization against fungal-derived PGs. Notably, the pair PvPGIP3/PvPGIP4 also inhibited PGs of two mirid bugs (Lygus rugulipennis and Adelphocoris lineolatus). Characterization of Pvpgip genes of Pinto bean showed variations limited to single synonymous substitutions or small deletions. A three-amino acid deletion encompassing a residue previously identified as crucial for recognition of PG of Fusarium moniliforme was responsible for the inability of BAT93 PvPGIP2 to inhibit this enzyme. Consistent with the large variations observed in the promoter sequences, reverse transcription-PCR expression analysis revealed that the different family members differentially respond to elicitors, wounding, and salicylic acid. We conclude that both biochemical and regulatory redundancy and subfunctionalization of pgip genes are important for the adaptation of plants to pathogenic fungi and phytophagous insects.

Biochars are biological residues combusted under low oxygen conditions, resulting in a porous, low density carbon rich material. Their large surface areas and cation exchange capacities, determined to a large extent by source materials and pyrolysis temperatures, enables enhanced sorption of both organic and inorganic contaminants to their surfaces, reducing pollutant mobility when amending contaminated soils. Liming effects or release of carbon into soil solution may increase arsenic mobility, whilst low capital but enhanced retention of plant nutrients can restrict revegetation on degraded soils amended only with biochars; the combination of composts, manures and other amendments with biochars could be their most effective deployment to soils requiring stabilisation by revegetation. Specific mechanisms of contaminant-biochar retention and release over time and the environmental impact of biochar amendments on soil organisms remain somewhat unclear but must be investigated to ensure that the management of environmental pollution coincides with ecological sustainability.

The objective of the present study was to evaluate the mycoflora and occurrence of aflatoxins in stored peanut samples (hulls and kernels) from Tupa, State of Sao Paulo, Brazil. The samples were analyzed monthly over a period of one year. The results showed a predominance of Fusarium spp. (67.7% in hulls and 25.8% in kernels) and Aspergillus spp. (10.3% in hulls and 21.8% in kernels), and the presence of five other genera. The growth of Aspergillus flavus was mainly influenced by temperature and relative humidity. Analysis of hulls showed that 6.7% of the samples were contaminated with AFB(1) (mean levels 15-23.9 mu g/kg) and AFB(2) (mean levels = 3.3-5.6 mu g/kg); in kernels, 33.3% of the samples were contaminated with AFB(1) (mean levels 7.0-116 mu g/kg) and 28.3% were contaminated with AFB(2) (mean levels = 3.3-45.5 mu g/kg). Analysis of the toxigenic potential revealed that 93.8% of the A. flavus strains isolated were producers of AFB(1) and AFB(2). mu 2007 Elsevier Ltd. All rights reserved.

Metal cation toxicity to basidiomycete fungi is poorly understood, despite its well-known importance in terrestrial ecosystems. Moreover, there is no reported methodology for the routine evaluation of metal toxicity to basidiomycetes. In the present study, we describe the development of a procedure to assess the acute toxicity of metal cations (Na(+), K(+), Li(+), Ca(2+), Mg(2+), Co(2+), Zn(2+), Ni(2+), Mn(2+), Cd(2+), and Cu(2+)) to the bioluminescent basidiomycete fungus Gerronema viridilucens. The method is based on the decrease in the intensity of bioluminescence resulting from injuries sustained by the fungus mycelium exposed to either essential or nonessential metal toxicants. The assay described herein enables LIS to propose a metal toxicity series to Gerronenia viridilucens based on data obtained from the bioluminescence intensity (median effective concentration [EC50] values) versus metal concentration: Cd(2+) > Cu(2+) > Mn(2+) approximate to Ni(2+) approximate to Co(2+) > Zn(2+) > Mg(2+) > Li(+) > K(+) approximate to Na(+) > Ca(2+), and to shed some li-ht on the mechanism of toxic action of metal cations to basidiomycete fungi. O objetivo deste trabalho foi quantificar variáveis microbiológicas e produtividade do feijoeiro em razão do manejo do solo e da calagem. Parte de uma área cultivada há 20 anos em plantio direto (PD) foi submetida ao cultivo mínimo (CM), com escarificação a 0,25 m de profundidade, para incorporação do calcário. Os tratamentos consistiram de manejo do solo (PD e CM) e 0 e 2 t ha-1 de calcário dolomítico aplicadas na superfície, com quatro repetições. Os maiores valores de C da biomassa microbiana foram verificados no CM e os de C de CO2 liberado no PD sem calagem. A menor colonização micorrízica e a maior esporulação foram observadas no CM sem calagem. Não foram detectadas diferenças entre os tratamentos em relação à produção de matéria seca, enquanto na produtividade de grãos, o maior valor foi verificado no CM com calagem. É possível evitar a interrupção do PD com aplicação de calcário na superfície, visto que a incorporação do material de cobertura e a calagem pouco alteraram a estabilidade do sistema, conforme comprovado pelos valores estatísticos semelhantes de C do CO2 liberado e pelas pequenas diferenças observadas nas demais variáveis entre manejos.

Ensaios foram conduzidos, em casa de vegetação, com solos de pastagem degradada reflorestada e cerrado preservado (controle) visando avaliar a contribuição de fungos micorrízicos arbusculares (FMA) autóctones no crescimento de mutambo (Guazuma ulmifolia Lamb.). As mudas foram transplantadas para sacos de plástico (2 kg) com substratos esterilizados na proporção 4:1 (solo:areia), e o tratamento inoculado recebeu 300 esporos de FMA por saco. A inoculação não proporcionou aumento significativo na produção da matéria seca da parte aérea, matéria fresca das raízes e altura da planta, sugerindo que a G. ulmifolia não é responsiva à micorrização.

O trabalho foi desenvolvido em casa de vegetação com o objetivo de comparar o crescimento e a micorrização por fungos micorrízicos arbusculares (FMA) em nove genótipos de milho, além de verificar o potencial de inóculo de FMA do solo. Coletado de uma área de cerrado sensu stricto ocupada por pastagem degradada, o solo foi adubado, misturado com areia de rio, fumigado e colocado em sacos plásticos (3 kg). Foram semeadas cinco sementes de cada genótipo de milho em 20 sacos, mas apenas 10 receberam cerca de 300 esporos de FMA, coletados do solo de pastagem. Cada saco constituiu-se em uma repetição, com apenas uma planta. As avaliações de altura e diâmetro do caule foram realizadas aos 15, 30, 45 e 60 dias após a emergência, além de massa seca do sistema radicular (MSR) e parte aérea (MSPA), colonização micorrízica (COL) e dependência micorrízica (DM). Concomitantemente, um segundo experimento foi realizado para avaliar o potencial de inóculo de FMA do solo de pastagem, o qual passou por uma diluição seriada de 90%, 80%, 70%, 60%, 50%, 40%, 30%, 20% e 10%, incluindo apenas o solo não esterilizado (100%) e somente solo esterilizado (0%), em sacos plásticos, semeado o híbrido Tractor. Após 60 dias, as raízes foram colhidas para quantificar a colonização micorrízica. A inoculação de FMA acarretou incrementos na produção de matéria seca de forma diferenciada entre genótipos; com Condá, F0, D1 e F8 exibindo os maiores valores de MSPA, enquanto Tractor e D7 os menores valores de MSR. Os genótipos não responsivos ou pouco responsivos quanto à dependência micorrízica tiveram comportamento diferente quanto à COL, com Condá, Sol da Manhã, F0 e D1 proporcionando média de 60%. Verificou-se que a área de pastagem, mesmo degradada, propiciou alto potencial de inóculo, revelado pela alta porcentagem de colonização das raízes por FMA.

In Brazil, the upland rice Culture system is predominant, but its water supply depends of precipitation and its distribution. Sol-lie practices or disturbances on soil conditions may cause alterations that call be detected by microorganisms, which are quite sensitive. This experiment was developed to study microbiological alterations (microbial biomass carbon (MBC), released CO2 (C-CO2), metabolic quotient (qCO(2)) and mycorrhization), as well as alterations in soil fertility and productivity of upland rice, cultivated under different soil and water managements. Cultivar BRS Talento was used in the experiment. The experimental design was a completely randomized block design, with four replications, using three soil managements: no-tillage (NT), heavy disk + leveling disk harrowing (HL), and chisel plowing + leveling disk harrowing (CL), plus three water managements: no irrigation (WD0); water depth 1 (WD1), with irrigation at the reproductive and maturation periods; and water depth 2 (WD2), with irrigation throughout the rice cycle. Autochthones arbuscular mycorrhizal fungi proved to be sensitive to soil and water management. The NT presented the highest values for MO, Ca, SB and V% and the lowest for H+A1. This management, together with irrigation at the reproductive and maturation periods of BRS Talento cultivar, promoted goods results for crop yield and microbiology characteristics.
