Prostate cancer is the second leading cause of cancer death in the country. Due to this factor, the interest of showing what relationship exists between vasectomy surgery and the incidence of prostate carcinoma has started. Some epidemiological studies showed an increased risk of prostate cancer in vasectomized men (Emard et al., 2001). On the other hand other authors have argued that there is no correlation (Patel et al., 2005) and there are those who said that vasectomy is linked to reduced risk of prostate cancer (Ross, 1983). Faced with an analysis of published works, such discussion remains today. The vasectomy, or deferentectomia, is a contraceptive male method, which is the section of the vas deferens of man by preventing the sperm from being expelled along with the seminal fluid during ejaculation, and is one of the most simple, economical, uncomplicated post- operative. This study aims to evaluate the process of cell proliferation and to evaluate immuno-histochemically the expression of specific markers of PCNA (Proliferating Cell Nuclear Antigen) and Ki-67, in the prostate of the gerbil after chemical induction by intraperitoneal injection of the carcinogen N-methyl -N-nitrosourea (MNU), because the regulation of the functional balance between cell proliferation and apoptosis is associated with hyperplasia and prostatic carcinoma. Nos últimos anos vem aumentando o reconhecimento mundial sobre a relevância da nutrição como pilar básico para o desenvolvimento econômico e social. Milhares de crianças têm seu crescimento retardado pela má nutrição, no entanto poucos trabalhos relacionam a desnutrição proteica materna e o desenvolvimento do sistema reprodutor masculino. Dessa forma, o presente trabalho tem por objetivo investigar os possíveis efeitos da restrição proteica gestacional e durante a lactação sobre a morfogênese do complexo prostático, tendo em vista que a etiologia das doenças prostáticas relaciona-se com fatores epigenéticos, e dentre eles, fatores toxicológicos e nutricionais. Após o acasalamento e diagnóstico da prenhês, dois grupos de mães foram constituídos: Normoproteico (NP, 17% proteína, n=7) e Hipoproteico (LP, 6% proteína, n=7). No segundo dia pós-nascimento (DPN2), as ninhadas foram inspecionadas e reduzidas para no máximo oito animais (preferencialmente machos), a distância anogenital foi mensurada e, 2 machos por ninhada foram mortos por decapitação para coleta do broto prostático no dia do nascimento. Após o nascimento dos filhotes, um grupo de mães do grupo hipoproteico continuou recebendo a ração hipoproteica (n=3), enquanto as demais passaram a receber a ração normoproteica (n=3). No dia 22, dia do desmame, 2 machos de cada ninhada foram pesados, mortos por decapitação e o complexo prostático foi coletado para as análises morfológicas e imunoistoquímicas. Foram investigados a morfologia geral e o padrão de distribuição dos elementos celulares e da matriz extracelular por métodos citoquímicos e morfométricos, e a imunorreatividade para os seguintes marcadores: receptor de andrógeno (AR), Ki67, α-actina, EGF-R e P63. Os índices de proliferação e morte celular (TUNEL) foram calculados. A prole LP apresentou uma redução significativa do peso corporal e da distância anogenital no DPN2. ...

Adequate testosterone levels are necessary for the development, growth and maintenance of the male reproductive system. Testosterone deficiency is common in men with diabetes in whom it may contribute to impaired performance, with consequent reduction of the activity of the androgen regulated organs, such as the prostate. However, little attention has been given to the plasma dihydrotestosterone (DHT) level, the most potent androgen, nor to the expression of the androgen receptor (AR), insulin-like growth factor type I (IGF-1) and receptor (IGF-1R) in target tissues. Here, we investigated the effect of type I diabetes mellitus on DHT plasma levels and on prostate AR, IGF-1 and IGF-1R expression during rat pubertal growth. Diabetes was induced in prepubertal male rats through administration of streptozotocin (STZ; 40 mg/kg). Diabetic, diabetic treated with insulin, and age-matched control animals were killed by overdoses of pentobarbital. The ventral prostatic lobe (VP) was dissected, weighed and processed for immunohistochemistry for AR, IGF-1 e IGF-1R; plasma T and DHT levels were also determined. Hyperglycemia at puberty reduced VP weight gain to about 50% and plasma T level to about 80% of the control levels. In contrast there were no changes in plasma DHT levels. Insulin replacement restored the VP weight gain, but not the plasma T levels, which remained 90% below the ones of controls. Immunohistochemistry showed that AR, IGF-1 and IGF-1R expression in the prostate epithelial cells did not change with hyperglycemia or insulin replacement. Thus, the AR expression in the prostate epithelial cells appears to be regulated by DHT, and to a minor extent it also controls glandular growth

A hiperplasia fibroadenomatosa mamária felina é uma enfermidade não neoplásica que ocorre, principalmente em gatas jovens, devido ao estímulo excessivo de progesterona endógena ou exógena. Embora não seja relatada com frequência, sua incidência tem aumentado, especialmente no Brasil, em consequência do uso de anticoncepcionais de longa duração como, por exemplo, acetato de medroxiprogesterona. O objetivo desta revisão sistemática é avaliar as características clínicas e comparar os diferentes tratamentos preconizados para esta patologia. O uso de antiprogestágenos, como o aglepristone, mostrou-se muito eficiente no tratamento da hiperplasia fibroadenomatosa mamária felina por ser uma alternativa pouco invasiva e com resposta clínica mais rápida que os demais métodos

Tumors of the Harderian gland occurred frequently in red-backed voles, Clethrionomys rutilus (Pallas), in a captive colony maintained since 1957. Although the tumors became obvious only in animals about a year old and older, early stage changes were histologically evident in glands from younger, clinically normal voles. The tumors ranged in type from poorly differentiated adenoma to adenocarcinoma, with invasion of adjacent tissues and characteristic deformation of the zygomata. Electron micrographs disclosed viral particles in the cytoplasm of glands from clinically normal animals as well as in glands showing malignant changes. The tumor was not observed in arvicoline rodents of other species maintained in the same rooms with the red-backed voles.

Purpose: Reporting on the laparoscopic technique for adrenal disease in children and adolescents has been limited. We review here our experience with laparoscopic adrenal surgery in children. Patients and methods: 19 laparoscopic unilateral adrenalectomies were performed in 10 girls and 7 boys (mean age 3.9 years) during 1998-2011. The clinical diagnosis before surgery was virilizing tumor (n = 8), pheochromocytoma (n = 3), nonfunctioning solid adrenal tumor (n = 3), mixed adrenocortical tumor (n = 2), cystic adrenal mass (n = 1). Unilateral adrenal lesions were 20-65 mm at the longest axis on computerized tomography (12 right side, 7 left side). Results: The final clinicopathological diagnosis was cortical adenoma (n = 9), pheochromocytoma (n = 3, bilateral in two), neuroblastoma (n = 1), ganglioneuroblastoma (n = 1), ganglioneuroma (n = 1), adrenocortical carcinoma (n = 1), benign adrenal tissue (n = 1). Average operative time was 138.5 min (range 95-270). Blood transfusion was required in one case (5%). No conversion to open surgery was required and no deaths or postoperative complications occurred. Average hospital stay was 3.5 days (range 2-15). Average postoperative follow-up was 81 months (range 2-144). Two contralateral metachronic pheochromocytomas associated with von Hippel-Lindau syndrome occurred, treated with partial laparoscopic adrenalectomy (one without postoperative need of cortisone replacement therapy). Conclusions: Laparoscopic adrenalectomy is a feasible procedure that produces good results. It can be used safely to treat suspected benign and malignant adrenal masses in children with minimal morbidity and short hospital stay. (C) 2011 Journal of Pediatric Urology Company. Dysregulation of the WNT and insulin-like growth factor 2 (IGF2) signaling pathways has been implicated in sporadic and syndromic forms of adrenocortical carcinoma (ACC). Abnormal beta-catenin staining and CTNNB1 mutations are reported to be common in both adrenocortical adenoma and ACC, whereas elevated IGF2 expression is associated primarily with ACC. To better understand the contribution of these pathways in the tumorigenesis of ACC, we examined clinicopathological and molecular data and used mouse models. Evaluation of adrenal tumors from 118 adult patients demonstrated an increase in CTNNB1 mutations and abnormal beta-catenin accumulation in both adrenocortical adenoma and ACC. In ACC, these features were adversely associated with survival. Mice with stabilized beta-catenin exhibited a temporal progression of increased adrenocortical hyperplasia, with subsequent microscopic and macroscopic adenoma formation. Elevated Igf2 expression alone did not cause hyperplasia. With the combination of stabilized beta-catenin and elevated Igf2 expression, adrenal glands were larger, displayed earlier onset of hyperplasia, and developed more frequent macroscopic adenomas (as well as one carcinoma). Our results are consistent with a model in which dysregulation of one pathway may result in adrenal hyperplasia, but accumulation of a second or multiple alterations is necessary for tumorigenesis. Background: Although the molecular pathogenesis of pituitary adenomas has been assessed by several different techniques, it still remains partially unclear. Ribosomal proteins (RPs) have been recently related to human tumorigenesis, but they have not yet been evaluated in pituitary tumorigenesis. Objective: The aim of this study was to introduce serial analysis of gene expression (SAGE), a high-throughput method, in pituitary research in order to compare differential gene expression. Methods: Two SAGE cDNA libraries were constructed, one using a pool of mRNA obtained from five GH-secreting pituitary tumors and another from three normal pituitaries. Genes differentially expressed between the libraries were further validated by real-time PCR in 22 GH-secreting pituitary tumors and in 15 normal pituitaries. Results: Computer-generated genomic analysis tools identified 13 722 and 14 993 exclusive genes in normal and adenoma libraries respectively. Both shared 6497 genes, 2188 were underexpressed and 4309 overexpressed in tumoral library. In adenoma library, 33 genes encoding RPs were underexpressed. Among these, RPSA, RPS3, RPS14, and RPS29 were validated by real-time PCR. Conclusion: We report the first SAGE library from normal pituitary tissue and GH-secreting pituitary tumor, which provide quantitative assessment of cellular transcriptome. We also validated some downregulated genes encoding RPs. Altogether, the present data suggest that the underexpression of the studied RP genes possibly collaborates directly or indirectly with other genes to modify cell cycle arrest, DNA repair, and apoptosis, leading to an environment that might have a putative role in the tumorigenesis, introducing new perspectives for further studies on molecular genesis of somatotrophinomas.

Carcinosarcoma is a rare malignant disease with aggressive behaviour rarely producing oral manifestations. This article reports a case of an intraoral carcinosarcoma affecting a 71-year-old black male; the diagnosis was made by histopathological and immunohistochemical analyses. Computed tomography scanning showed metastatic masses in the lungs. The patient was underwent a chemotherapy protocol regimen, but died as a consequence of the disease within 10 months of diagnosis. Distinctive characteristics of this presentation were the location of the lesion (floor of the mouth) and its clinical features resembling a benign lesion. A brief review of intraoral carcinosarcoma cases in the literature is also presented.
