In the present study, a detailed investigation on the alterations of muscarinic M1, M3, α7 nicotinic acetylcholine receptor (α7 nAchR), GABA receptors and its subtypes; GABAAα1 and GABAB in the brain regions of streptozotocin induced diabetic and insulin induced hypoglycemic rats were carried out. Gene expression of acetylcholine esterase (AChE), choline acetyltransferase (ChAT), GAD, GLUT3, Insulin receptor, superoxide dismutase (SOD), Bax protein, Phospholipase C and CREB in hypoglycemic and hyperglycemic rat brain were studied. Muscarinic M1, M3 receptors, AChE, ChAT, GABAAα1, GABAB, GAD, Insulin receptor, SOD, Bax protein and Phospholipase C expression in pancreas was also carried out. The molecular studies on the CNS and PNS damage will elucidate the therapeutic role in the corrective measures of the damage to the brain during hypoglycemia and hyperglycemia.

The present study deals with the Cholinergic Receptor subtypes functional regulation in spinal cord injured monoplegic rats: Effect of 5-HT GABA and bone marrow cells.Spinal cord injury causes permanent and irrevocable motor deficits and neurodegeneration. Disruption of the spinal cord leads to diminished transmission of descending control from the brain to motor neurons and ascending sensory information. Behavioural studies showed deficits in motor control and coordination in SCI rats. Cholinergic system plays an important role in SCI, the evaluation of which provides valuable insight on the underlying mechanisms of motor deficit that occur during SCI. The cholinergic transmission was studied by assessing the muscarinic and nicotinic receptors; cholinergic enzymes- ChAT and AChE; second messenger enzyme PLC; transcription factor CREB and second messengers - IP3, cAMP and cGMP. We observed a decrease in the cholinergic transmission in the brain and spinal cord of SCI rats. The disrupted cholinergic system is the indicative of motor deficit and neuronal degeneration in the spinal cord and brain regions. SCI mediated oxidative stress and apoptosis leads to neuronal degeneration in SCI rats. The decreased expression of anti oxidant enzymes – SOD, GPx and neuronal cell survival factors - BDNF, GDNF, IGF-1, Akt and cyclin D2 along with increased expression of apoptotic factors – Bax, caspase-8, TNFa and NF-kB augmented the neuronal degeneration in SCI condition. BMC administration in combination with 5-HT and GABA in SCI rats showed a reversal in the impaired cholinergic neurotransmission and reduced the oxidative stress and apoptosis. It also enhanced the expression of cell survival factors in the spinal cord region. In SCI rats treated with 5-HT and GABA, the transplanted BMC expressed NeuN confirming that 5-HT and GABA induced the differentiation and proliferation of BMC to neurons in the spinal cord. Neurotrophic factors and anti-apoptotic elements in SCI rats treated with 5-HT and GABA along with BMC rendered neuroprotective effects accompanied by improvement in behavioural deficits. This resulted in a significant reversal of altered cholinergic neurotransmission in SCI. The restorative and neuro protective effects of BMC in combination with 5-HT and GABA are of immense therapeutic significance in the clinical management of SCI.

The toxic effects of chemicals in the WAF on the organisms ; their tissues and eventually on rate functions have been elucidated. Marine oil pollution not only affects productivity and quality of marine organisms but also eventually affects the health of human population due to a possible health risk by way of consumption of oil contaminated seafood

Im Rahmen dieser Dissertation wurde an der Darstellung stabiler, amphiphiler Silantriole gearbeitet. Es ist gelungen eine kontinuierliche Reihe sukzessive um CH2-Einheiten verlängerter Silantriole des Typs H3C(CH2)nC(CH3)2Si(OH)3 (n = 1-5) durch eine vierstufige Synthesesequenz ausgehend von n-Alkylbromiden herzustellen und zum Teil röntgenographisch zu untersuchen. Ihre oberflächenaktiven Eigenschaften in wässrigen Lösungen konnten erstmals mittels Oberflächenspannungsmessungen belegt werden. Durch gezielte Kondensationsreaktionen mit Trifluoressigsäure wurden ausgehend von den oben beschriebenen Silantriolen selektiv die entsprechenden Disiloxan-Tetrole erhalten und ebenfalls zum Teil durch Röntgenstrukturanalysen charakterisiert. Als weitere Kondensationsprodukte der Silantriole konnten die ersten Octasilsesquioxane mit tertiären Kohlenstoff-Substituenten durch Umsetzungen mit n-Bu4NF selektiv und in hohen Ausbeuten erhalten und ebenfalls röntgenographisch identifiziert werden. Es wurde an der Darstellung Aryl-substituierter Silantriole RSi(OH)3 (R = Mesityl-, Xylyl- und Tetramethyl-phenyl-) gearbeitet. Diese zeichnen sich jedoch durch ihre hohe Instabilität in Lösung und im Festkörper aus und konnten zum Teil nur zusammen mit ihren primären Kondensationsprodukten erhalten werden. Darüber hinaus wurden stabile Silandiole des Typs R(t-Bu)Si(OH)2 (R = n-Butyl und n-Pentyl) sowie das (o-CF3C6H4)2Si(OH)2 in hohen Ausbeuten und selektiv synthetisiert. Die Festkörpereigenschaften des (o-CF3C6H4)2Si(OH)2 sowie seiner Vorstufe (o-CF3C6H4)2SiCl2 konnten durch Kristallstrukturanalysen genauer untersucht werden. In der Arbeit wurden die ersten Anwendungen von Silanolen als Silan-Kupplungsreagenzien bei der Oberflächenmodifizierung von Glas beschrieben. Im Gegensatz zu gängigen säureassistierten Beschichtungen zeichnen sich diese Silanol-Beschichtungen durch eine deutlich höhere Hydrophobizität aus. Dies konnte durch Kontaktwinkel- und Zeta-Potential-Messungen bestätigt werden. Durch Röntgenreflektivitäts- und Sarfus-Messungen ist die Ausbildung von Monolagen im Falle von t-BuSi(OH)3-beschichteten Oberflächen plausibel. Die Oberflächenmorphologie der Silantriol-Beschichtungen wurde mittels AFM-Messungen untersucht. Die Si(OH)3-Funktion konnte in der vorliegenden Arbeit als ein neues Pharmakophor etabliert werden. Die Silantriole CySi(OH)3, TerSi(OH)3 und CH3CH2C(CH3)2Si(OH)3 (Cy = Cyclohexyl, Ter = Terphenyl) sind in der Lage das Enzym Acetylcholinesterase reversibel zu hemmen. Dabei zeigt das CySi(OH)3 mit 45 % relativ zur Kontrolle die höchste Inhibition.

La butirilcolinesterasa humana (BChE; EC 3.1.1.8) es una enzima polimórfica sintetizada en el hígado y en el tejido adiposo, ampliamente distribuida en el organismo y encargada de hidrolizar algunos ésteres de colina como la procaína, ésteres alifáticos como el ácido acetilsalicílico, fármacos como la metilprednisolona, el mivacurium y la succinilcolina y drogas de uso y/o abuso como la heroína y la cocaína. Es codificada por el gen BCHE (OMIM 177400), habiéndose identificado más de 100 variantes, algunas no estudiadas plenamente, además de la forma más frecuente, llamada usual o silvestre. Diferentes polimorfismos del gen BCHE se han relacionado con la síntesis de enzimas con niveles variados de actividad catalítica. Las bases moleculares de algunas de esas variantes genéticas han sido reportadas, entre las que se encuentra las variantes Atípica (A), fluoruro-resistente del tipo 1 y 2 (F-1 y F-2), silente (S), Kalow (K), James (J) y Hammersmith (H). En este estudio, en un grupo de pacientes se aplicó el instrumento validado Lifetime Severity Index for Cocaine Use Disorder (LSI-C) para evaluar la gravedad del consumo de “cocaína” a lo largo de la vida. Además, se determinaron Polimorfismos de Nucleótido Simple (SNPs) en el gen BCHE conocidos como responsables de reacciones adversas en pacientes consumidores de “cocaína” mediante secuenciación del gen y se predijo el efecto delos SNPs sobre la función y la estructura de la proteína, mediante el uso de herramientas bio-informáticas. El instrumento LSI-C ofreció resultados en cuatro dimensiones: consumo a lo largo de la vida, consumo reciente, dependencia psicológica e intento de abandono del consumo. Los estudios de análisis molecular permitieron observar dos SNPs codificantes (cSNPs) no sinónimos en el 27.3% de la muestra, c.293A>G (p.Asp98Gly) y c.1699G>A (p.Ala567Thr), localizados en los exones 2 y 4, que corresponden, desde el punto de vista funcional, a la variante Atípica (A) [dbSNP: rs1799807] y a la variante Kalow (K) [dbSNP: rs1803274] de la enzima BChE, respectivamente. Los estudios de predicción In silico establecieron para el SNP p.Asp98Gly un carácter patogénico, mientras que para el SNP p.Ala567Thr, mostraron un comportamiento neutro. El análisis de los resultados permite proponer la existencia de una relación entre polimorfismos o variantes genéticas responsables de una baja actividad catalítica y/o baja concentración plasmática de la enzima BChE y algunas de las reacciones adversas ocurridas en pacientes consumidores de cocaína.

La asignatura de Competitividad Internacional Urbana (ciu) del programa de Gestión y Desarrollo Urbanos (gdu) de la Universidad del Rosario ha sido desde 2009, cuando asumí su dirección y orientación, un reto permanente de aprendizajes tan estimulantes y variados cuantas ciudades y atributos hay por descubrir en el inmenso mundo de lo urbano-rural-regional. Si bien la competitividad es un asunto urbano-regional antes que nacional, la mayor parte de los enfoques y de las consiguientes referencias bibliográficas abordan la competitividad a nivel nacional siendo relativamente escasas las publicaciones sobre la competitividad urbana. Así, los documentos abordan una descripción general de las ciudades, las causas de las crisis y las consecuencias para la ciudad y su estructura económica, analizadas a partir de los impactos sobre el mercado laboral, los precios de la vivienda, el desarrollo del turismo, entre otros, y las diversas estrategias que adoptaron para afrontar la crisis y convertirla en una oportunidad de desarrollo.

La tesi de Crous (2009) descriu les característiques de les estructures gramaticals del català i del mandinga que expressen malalties, signes i símptomes, trets físics i psíquics, etc. (com ser diabètic, estar refredat, tenir febre, tenir un bony (a la cama), tenir mal de panxa o tenir mal a la panxa, venir un atac de tos, agafar febre o fer mal el cap). La tesi demostra que la conceptualització i la categorització dels estats patològics, la manera com els parlants perceben alguns aspectes dels estats (com la temporalitat, l'abast corporal, la causalitat o el grau d'afectació) i les relacions entre aquests estats i els altres dos participants de la situació d'afectació (una persona i les seves parts del cos), no només es posa de manifest en els mots d'una llengua sinó que també intervé en la construcció i en la distribució dels diferents participants dins d'una oració.

As esponjas marinhas são organismos ubíquos possuindo muitas características que lhes conferem um elevado potencial como organismos bioindicadores. Dado que se reveste de enorme importância e premência o estabelecimento de um grande número de organismos que possam actuar como bioindicadores de exposição a poluentes, neste trabalho investigamos a presença do biomarcador acetilcolenesterase nas esponjas marinhas Spongia officianalis e Spongia agaricina. Os exemplos foram recolhidos em locais pré-seleccionados ao longo da costa oeste portuguesa em locais considerados não sujeitos a poluição. Escolheram-se também alguns pontos de amostragem onde pode ocorrer alguma actividade antropogénica. Para o estudo foi usado um método padrão de detecção da actividade de acetilcolinesterase - a produção do ião 5-tio-2-nitrobenzoato. Estabeleceu-se a presença de acetilcolinesterase nestas espécies e validou-se o método em termos de repetibilidade e reprodutibilidade para estes organismos. Foi também possível determinar o intervalo normal de valores de actividade específica de AChE para as espécies em estudo [0,000; 1,270] mU of AChE/mg of proteina para S. officinallis e [0,000; 1,439] mU of AChE/mg de proteina para S. agaricina.

Nos últimos séculos perdeu-se o interesse por uso de plantas medicinais nos países desenvolvidos, devido à evolução da síntese química de fármacos. Ressurgiu recentemente este uso para desenvolvimento de novos fármacos de patologias com múltiplos mecanismos fisiopatológicos associados devido às actividades simultâneas dos extractos. Dos potenciais usos medicinais explorou-se a inibição da acetilcolinesterase (AChE) para problemas gastrointestinais e para terapêutica da doença de Alzheimer com menos efeitos secundários, e menor toxicidade associada. Realizou-se um estudo de actividades antioxidante e inibitória de AChE onde as duas espécies estudadas de plantas tiveram actividade. A decocção de A. cherimola apresentou capacidade de actividade mais elevada de inibir Ache com um IC50 de 0,627±0,028 mg/mL e antioxidante com um EC50 de 15,435±0,184 mg/mL. Nenhum dos extractos perdeu actividade com o metabolismo in vitro gástrico e pancreático. Na biodisponibilidade verificou-se que a maior parte dos compostos permeia até determinada extensão a monocamada de células Caco-2 com excepção do glucósido da quercetina. A via de transporte predominante dos compostos estudados parece ser difusão paracelular passiva.

The lymph heart is a sac-like structure on either side of avian tail. In some adult birds, it empties the lymph from the copulatory organ; however, during embryonic development, it is thought to circulate extra-embryonic lymph. Very little is known about the origin, innervation and the cellular changes it undergoes during development. Using immunohistochemistry and gene expression profiling we show that the musculature of the lymph heart is initially composed solely of striated skeletal muscle but later develops an additional layer composed of smooth myofibroblasts. Chick-quail fate-mapping demonstrates that the lymph heart originates from the hypaxial compartments of somites 34-41. The embryonic lymph heart is transiently innervated by somatic motoneurons with no autonomic input. In comparison to body muscles, the lymph heart has different sensitivity to neuromuscular junction blockers (sensitive only to decamethonium). Furthermore, its abundant bungarotoxin-positive acetylcholinesterase receptors are unique as they completely lack specific acetylcholinesterase activity. Several lines of evidence suggest that the lymph heart may possess an intrinsic pacing mechanism. Finally, we assessed the function of the lymph heart during embryogenesis and demonstrate that it is responsible for preventing embryonic oedema in birds, a role previously thought to be played by body skeletal muscle contractions.

Daphnia magna is a key invertebrate in the freshwater environment and is used widely as a model in ecotoxicological measurements and risk assessment. Understanding the genomic responses of D. magna to chemical challenges will be of value to regulatory authorities worldwide. Here we exposed D. magna to the insecticide methomyl and the herbicide propanil to compare phenotypic effects with changes in mRNA expression levels. Both pesticides are found in drainage ditches and surface water bodies standing adjacent to crops. Methomyl, a carbamate insecticide widely used in agriculture, inhibits acetylcholinesterase, a key enzyme in nerve transmission. Propanil, an acetanilide herbicide, is used to control grass and broad-leaf weeds. The phenotypic effects of single doses of each chemical were evaluated using a standard immobilisation assay. Immobilisation was linked to global mRNA expression levels using the previously estimated 48h-EC(1)s, followed by hybridization to a cDNA microarray with more than 13,000 redundant cDNA clones representing >5000 unique genes. Objective. Most snakebite deaths occur prior to hospital arrival; yet inexpensive, effective, and easy to administer out-of-hospital treatments do not exist. Acetylcholinesterase inhibitors can be therapeutic in neurotoxic envenomations when administered intravenously, but nasally delivered drugs could facilitate prehospital therapy for these patients. The architecture of the amygdaloid complex of a marsupial, the opossum Didelphis aurita, was analyzed using classical stains like Nissl staining and myelin (Gallyas) staining, and enzyme histochemistry for acetylcholinesterase and NADPH-diaphorase. Most of the subdivisions of the amygdaloid complex described in eutherian mammals were identified in the opossum brain. NADPH-diaphorase revealed reactivity in the neuropil of nearly all amygdaloid subdivisions with different intensities, allowing the identification of the medial and lateral subdivisions of the cortical posterior nucleus and the lateral subdivision of the lateral nucleus. The lateral, central, basolateral and basomedial nuclei exhibited acetylcholinesterase positivity, which provided a useful chemoarchitectural criterion for the identification of the anterior basolateral nucleus. Myelin stain allowed the identification of the medial subdivision of the lateral nucleus, and resulted in intense staining of the medial subdivisions of the central nucleus. The medial, posterior, and cortical nuclei, as well as the amygdalopiriform area did not exhibit positivity for myelin staining. On the basis of cyto- and chemoarchitectural criteria, the present study highlights that the opossum amygdaloid complex shares similarities with that of other species, thus supporting the idea that the organization of the amygdala is part of a basic plan conserved through mammalian evolution.
