Com o objetivo de identificar a tolerância à dessecação de sementes de Euterpe espiritosantensis, dois lotes de frutos foram colhidos na coleção de palmeiras do Instituto Agronômico de Campinas, no município de Ubatuba, estado de São Paulo e transportados em embalagem impermeável até a Faculdade de Ciências Agronômicas/UNESP, município de Botucatu, estado de São Paulo, onde foram despolpados. As sementes foram submetidas à secagem em sílica gel a partir das testemunhas não desidratadas (51,4% e 46,6% de teor de água), retirando-se amostras a cada 12 ou 24 horas. O efeito da desidratação foi avaliado através da porcentagem de germinação, e da velocidade de protrusão de embrião, do botão germinativo, da plúmula, e do comprimento das plântulas. Avaliaram-se ainda a matéria seca e o teor de água das sementes. Concluiu-se que as sementes de E. espiritosantensis são recalcitrantes, apresentando porcentagem alta de germinação (90,0% a 87,5%) quando não desidratadas (teor inicial de água de 51,4% a 46,6%). Abaixo de uma faixa de teor de água situada entre 40,7% e 51,4 % os valores de germinação e vigor foram significativamente reduzidos. A mortalidade total das sementes foi observada, com a redução do teor de água das sementes a uma faixa situada entre 13,4% e 15,8%.

O objetivo deste trabalho foi avaliar a sensibilidade à dessecação de sementes de Tapirira obtusa (Benth.) J. D. Mitchell provenientes de três ambientes e submetidas a duas velocidades de secagem. As sementes foram coletadas em área de campo rupestre, Cerrado e em mata ciliar, da região de Lavras, MG. As sementes foram submetidas à secagem com cloreto de magnésio (secagem lenta) ou sílica gel (secagem rápida), em ambiente fechado, até os graus de umidade de 40%, 30%, 20% e 10%, considerando como controle o percentual de germinação na umidade inicial em cada ambiente, a qual variou de 47% a 50%. Foram avaliados os percentuais de germinação e de plântulas normais e também o índice de velocidade de germinação. Nos três ambientes analisados, não houve efeito da secagem lenta sobre a germinação das sementes. Sementes oriundas do Cerrado apresentaram pequena redução da germinação, quando submetidas à secagem rápida. Sementes oriundas de áreas rupestres apresentaram aumento na germinação quando submetidas à secagem rápida. Sementes oriundas de mata ciliar não apresentaram redução na germinação, independente da velocidade de secagem. Os resultados sugerem que sementes de T. obtusa não apresentam sensibilidade à dessecação.

The physicochemical properties and morphology of spongolite, a fibrous hollow material from Mato Grosso do Sul State (Brazil) have been studied. The results of thermal analysis, scanning electron microscopy (SEM), X-ray diffraction and NMR spectroscopy indicated that external and internal surfaces of silica spicules are covered by silica gel layers. The water evolved in the range 120-350degreesC is the result of silanol groups condensation to siloxane bonds. Total homogenization of the needles is achieved by heating spongolite over 900degreesC. This mineral may be considered as a natural composite material containing surface-immobilized reactive species. The presence of active silica gel layers opens the possibilities of attaching functional groups to spongolite surface. (C) 2002 Elsevier B.V. B.V. All rights reserved.

Several clean-up procedures which included the use of glass chromatography columns (silica gel, alumina, Florisil, silanized Celite-charcoal), Sep-Pak cartridges and standard solutions were compared for the determination of the following N-methylcarbamate (NMC) insecticides: aldicarb, carbaryl, carbofuran, methomyl and propoxur. According to recovery results of the compounds after elution in a glass column, the most efficient systems employed 4.6% deactivated alumina and a silanized Celite-charcoal (4:1) as adsorbents, using dichloromethane-methanol (99:1) and toluene-acetonitrile (75:25) mixtures, respectively, as binary eluents. The recoveries of the compounds studied varied from 84 to 120%. Comparable recoveries (75-100%) for Sep-Pak cartridges in normal phase (NH2, CN) and reversed phase (C-8) were observed. Different temperatures were tested during the concentration step in a rotary evaporator, and we verified a strong influence of this parameter on the stability of some compounds, such as carbofuran and carbaryl. Recovery studies employing the best clean up procedures were performed at the Brazilian agricultural level in potato and carrot samples; Validation methodology of the US Food and Drug Administration was adapted for the N-methylcarbamate analysis. Their recoveries ranged between 79 and 93% with coefficients of variation of 2.3-8%. (C) 1998 Elsevier B.V. B.V.

The presence of trace neutral organonitrogen compounds as carbazole and indole in derivative petroleum fuels plays an important role in the car's engine maintenance. In addition, these substances contribute to the environmental contamination and their control is necessary because most of them are potentially carcinogenic and mutagenic. For those reasons, a reliable and sensitive method was proposed for the determination of neutral nitrogen compounds in fuel samples, such as gasoline and diesel using preconcentration with modified silica gel (Merck 70-230 mesh ASTM) followed by differential pulse voltammetry (DPV) technique on a glassy carbon electrode. The electrochemical behavior of carbazole and indole studied by cyclic voltammetry (CV) suggests that their reduction occurs via a reversible electron transfer followed by an irreversible chemical reaction. Very well resolved diffusion controlled voltammetric peaks were obtained in dimethylformamide (DMF) with tetrabutylammonium tetrafluoroborate (TBAF(4) 0.1 mol L-1) for indole (-2.27 V) and carbazole (-2.67 V) versus Ag vertical bar AgCl vertical bar KClsat reference electrode. The proposed DPV method showed a good linear response range from 0.10 to 300 mg L-1 and a limit of detection (L.O.D) of 7.48 and 2.66 mu g L-1 for indole and carbazole, respectively. The results showed that simultaneous determination of indole and carbazole presents in spiked gasoline samples were 15.8 +/- 0.3 and 64.6 +/- 0.9 mg L-1 and in spiked diesel samples were 9.29 +/- 1 and 142 +/- 1 mg L-1, respectively. The recovery was evaluated and the results shown the values of 88.9 +/- 0.4 and 90.2 +/- 0.8% for carbazole and indole in fuel determinations. The proposed method was also compared with UV-vis spectrophotometric measures and the results obtained for the two methods were in good agreement according to the F and t Student's tests. Scanning electron microscopy (SEM) was used to investigated the effects of volatile solvents (such as water, propanone, ethanol, methanol or ethyl ether), treatment and drying processes, microwave ovens, drying ovens, and vacuum desiccators or freeze driers, on silica morphology. Silica gel was obtained from diluted sodium silicate (1:5 w/w SiO2:H2O). The results showed that the drying process based on freeze drying is more efficient for structural conservation of the precipitate. Treatment with volatile solvents does not change the shape of the aggregates, but has an important role in the determination of aggregate surface roughness.

The brown alga Pilayella littoralis was used as a new biosorbent in an on-line metal preconcentration procedure in a flow-injection system. Al, Co, Cu and Fe were determined in lake water samples by inductively coupled plasma optical emission spectrometry (ICP-OES) after preconcentration in a silica-immobilized alga column. Like other algae, P. littoralis exhibited strong affinity for these metals proving to be an effective accumulation medium. Metals were bound at pH 5.5 and were displaced at pH < 2 with diluted HCl. The enrichment factors for Cu-II, Fe-III, Al-III and Co-II were 13, 7, 16 and 11, respectively. Metal sorption efficiency ranged from 86 to 90%. The method accuracy was assessed by using drinking water certified reference material and graphite furnace atomic absorption spectrometry (GFAAS) as a comparison technique. The column procedure allowed a less time consuming, easy regeneration of the biomaterial and rigidity of the alga provided by its immobilization on silica gel. (C) 2003 Elsevier B.V. A comparative study is reported between C-18 bonded silica gel and powdered polyethylene (PE) as sorbent for Cd, Cu, and Pb determination using ammonium diethyldithiophosphate (ADTP) as the complexing agent in a flow injection system. The complexes were formed in 0.14 mol L-1 HNO3 and processed in a simple flow system comprising a peristaltic pump, a manual injector-commutator, and a sorbent-packed minicolumn. Ethanol was selected as the eluent and analytes in the eluate were determined by flame atomic absorption spectrometry. The optimum concentration of the complexing agent was 0.1% (m/v) ADTP for Cu and Pb determination using either C-18 or PE, and 0.25% (m/v) ADTP for Cd determination using PE. The sample loading flow rates were 5.0, 3.6, and 3.0 mL min(-1) for Cu, Pb, and Cd, respectively. The best elution flow rate was 6.5 mL min(-1). For a 60-sec preconcentration time, the sampling rate was 40 h(-1) and the enrichment factors of 33, 36, and 11 times (C-18) or 18, 22, and 23 times (PE) were obtained for Cu, Pb, and Cd, respectively. The limits of detection (LOD) were 1.6 mug L-1 Cu, 11 mug L-1 Pb, and 2.0 mug L-1 Cd using C-18 or 2.9 mug L-1 Cu, 19 mug L-1 Pb, and 1.0 mug L-1 Cd using PE, respectively. The relative standard deviations (n = 12) were typically <2%, <2%, and <6% for Cd, Cu, and Pb, respectively. The recoveries of Cd, Cu, and Pb added to wine samples varied from 96-99%, 97-102%, and 90-99%, respectively, using C-18 or PE. Accuracy was checked for Cd, Cu, and Pb determination in six wine samples digested by block digestor and open-vessel microwave-assisted digestion systems. The results revealed that C-18 was more efficient for Cu and Pb determination, while PE was the best sorbent for Cd.

The present paper describes the one-pot procedure for the formation of self-assembled thin films of two silanes on the model oxidized silicon wafer, SiO2/Si. SiO2/Si is a model system for other surfaces, such as glass, quartz, aerosol, and silica gel. MALDI-TOF MS with and without a matrix, XPS, and AFM have confirmed the formation of self-assembled thin films of both 3-imidazolylpropyltrimethoxysilane (3-IPTS) and 4-(N- propyltriethoxysilane-imino)pyridine (4-PTSIP) on the SiO2/Si surface after 30 min. Longer adsorption times lead to the deposition of nonreacted 3-IPTS precursors and the formation of agglomerates on the 3-IPTS monolayer. The formation of 4-PTSIP self-assembled layers on SiO2/Si is also demonstrated. The present results for the flat SiO2/Si surface can lead to a better understanding of the formation of a stationary phase for affinity chromatography as well as transition-metal-supported catalysts on silica and their relationship with surface roughness and ordering. The 3-IPTS and 4-PTSIP modified SiO2/Si wafers can also be envisaged as possible built-on-silicon thin-layer chromatography (TLC) extraction devices for metal determination or N-heterocycle analytes, such as histidine and histamine, with on-spot MALDI-TOF MS detection. © 2005 Elsevier Inc. The aim of this paper was to evaluate the antimicrobial activity of 2% chlorhexidine gel (CLX) associated with various intracanal medicaments against Candida albicans and Enterococcus faecalis inoculated in root canals. Thirty six human single-rooted teeth were contaminated with C.albicans and E.faecalis. The canals were instrumented using 2% CLX gel and were divided into three groups according to the intracanal medicaments (ICM) used. Group 1: calcium hydroxide paste [Ca(OH)], Group 2: 2% chlorhexidine gel (CLX) and Group 3: 2% CLX gel + Ca(OH). The root canal collections were performed after 21 days of contamination (control collection), after instrumentation (1st collection), after 14 days of intracanal medicament (2nd collection) and 7 days after medicament removal (3rd collection). The microbiological samples were plated in culture media and incubated for 48 hours. The results were submitted to Kruskal-Wallis test (P ≤ 0.05). It was verified that the instrumentation with CLX reduced the number of CFU/ml significantly when compared with the confirmation collection (control). However, the use of the ICM was only capable to eliminate completely the microorganisms in the root canals without difference statistics between them. Although the use of 2% chlorherixidine gel reduces the number of microorganisms significantly, only the ICM calcium hydroxide and calcium hydroxide associated with chlorhexidine are able to eliminate these microorganisms completely.
