In 1966, when the annual damage survey was initiated, Dr. C. R. Weaver, Statistician at the Ohio Agriculture Research and Development Center, Wooster, Ohio, drew up a sampling plan, balancing costs against desired precision. The plan included three combinations of fields to be sampled and stations per field for high damage areas, three combinations for moderate damage areas, and two combinations for light damage areas. Alternatives for the high damage area included (1) 497 fields with two stations per field (± .48), (2) 775 fields with two stations per field (± .26), and (3) 235 fields with ten stations per field (— .68). For the moderate damage areas, the alternatives were (1) 441 fields with three stations per field (± .26), (2) 155 fields with three stations per field (± .50), and (3) 235 fields with ten stations per field (± .32). The light dam¬age area alternatives were (1) 297 fields with three stations per field (- .26), and (2) 81 fields with three stations per field (± .50). The original survey in 1966 sampled eight counties in three regions. In 1967, 14 counties in the same three regions were sampled. Two new counties were added to one region and two new regions with two counties each (treated as one region for sampling purposes) were added to the 1968 survey. The 1968 survey was of sufficient size to be representative of the corn damage picture in Ohio and Southeast Michigan. The 1969 survey was identical to the 1968 survey.

In the Amazon Basin, within a landscape of infertile soils, fertile Anthrosols of pre-Columbian origin occur (Amazonian Dark Earths or terra preta de Indio). These soils are characterized by high amounts of charred organic matter (black carbon, biochar) and high nutrient stocks. Frequently, they were considered as sign for intensive landscape domestication by way of sedentary agriculture and as sign for large settlements in pre-Columbian Amazonia. Beyond the archaeological interest in Amazonian Dark Earths, they increasingly receive attention because it is assumed that they could serve as a model for sustainable agriculture in the humid tropics (terra preta nova). Both questions lack information about the pre-Columbian practices which were responsible for the genesis of Amazonian Dark Earths. It has often been hypothesized that deposition of faeces could have contributed to the high nutrient stocks in these soils, but no study has focussed on this question yet. We analyzed the biomarkers for faeces 5 beta-stanols as well as their precursors and their 5 alpha-isomers in Amazonian Dark Earths and reference soils to investigate the input of faeces into Amazonian Dark Earths. Using Amazonian Dark Earths as example, we discuss the application of threshold values for specific stanols to evaluate faeces deposition in archaeological soils and demonstrate an alternative approach which is based on a comparison of the concentration patterns of 5 beta-stanols with the concentration patterns of their precursors and their 5 alpha-isomers as well as with local backgrounds. The concentration patterns of sterols show that faeces were deposited on Amazonian Dark Earths. (C) 2011 Elsevier Ltd. All rights reserved.

Susceptibility to acute lymphoblastic leukemia can be highly influenced by genetic polymorphisms in metabolizing enzyme genes of environmental carcinogens. This study aimed to evaluate the impact of the CYP3A5 and NAT2 metabolizing enzyme polymorphisms on the risk of childhood acute lymphoblastic leukemia. The analysis was conducted on 204 ALL patients and in 364 controls from a Brazilian population, using PCR-RFLP. The CYP3A5*3 polymorphic homozygous genotype was more frequent among ALL patients and the *3 allele variant was significantly associated with increased risk of childhood ALL (OR = 0.29; 95% CI, 0.14-0.60). The homozygous polymorphic genotype for the *6 allele variant was extremely rare and found in only two individuals. The heterozygous frequencies were similar for the ALL group and the control group. No significant differences were observed between the groups analyzed regarding NAT2 variant polymorphisms. None of the polymorphisms analyzed was related to treatment outcome. The results suggest that CYP3A5*3 polymorphism may play an important role in the risk of childhood ALL.

It is already known that progressive degeneration of cholinergic neurons in brain areas such as the hippocampus and the cortex leads to memory deficits, as observed in Alzheimer's disease. This work verified the effects of the infusion of amyloid-beta (A beta) peptide associated to an attentional rehearsal on the density of alpha 7 nicotinic cholinergic receptor (nAChR) in the brain of male Wistar rats. Animals received intracerebroventricular infusion of A beta or vehicle (control - C) and their attention was stimulated weekly (Stimulated A beta group: S-A beta and Stimulated Control group: SC) or not (Non-Stimulated A beta group: N-SA beta and Non-Stimulated Control group: N-SC), using an active avoidance apparatus. Conditioned avoidance responses (CAR) were registered. Chronic infusion of A beta caused a 37% reduction in CAR for N-SA beta. In S-A beta, this reduction was not observed. At the end, brains were extracted and autoradiography for alpha 7 nAChR was conducted using [I-125]-alpha-bungarotoxin. There was an increase in alpha 7 density in hippocampus, cortex and amygdala of SA beta animals, together with the memory preservation. In recent findings from our lab using mice infused with A beta and the alpha 7 antagonist methyllycaconitine, and stimulated weekly in the same apparatus, it was observed that memory maintenance was abolished. So, the increase in alpha 7 density in brain areas related to memory might be related to a participation of this receptor in the long-lasting change in synaptic plasticity, which is important to improve and maintain memory consolidation.

Electronic polarization induced by the interaction of a reference molecule with a liquid environment is expected to affect the magnetic shielding constants. Understanding this effect using realistic theoretical models is important for proper use of nuclear magnetic resonance in molecular characterization. In this work, we consider the pyridine molecule in water as a model system to briefly investigate this aspect. Thus, Monte Carlo simulations and quantum mechanics calculations based on the B3LYP/6-311++G (d,p) are used to analyze different aspects of the solvent effects on the N-15 magnetic shielding constant of pyridine in water. This includes in special the geometry relaxation and the electronic polarization of the solute by the solvent. The polarization effect is found to be very important, but, as expected for pyridine, the geometry relaxation contribution is essentially negligible. Using an average electrostatic model of the solvent, the magnetic shielding constant is calculated as -58.7 ppm, in good agreement with the experimental value of -56.3 ppm. The explicit inclusion of hydrogen-bonded water molecules embedded in the electrostatic field of the remaining solvent molecules gives the value of -61.8 ppm.

Common bean, one of the most important legumes for human consumption, may have drastic reduction in yield due to anthracnose, a disease caused by the fungus Colletotrichum lindemuthianum. Rapid induction of the plant defense mechanisms is essential to establish an incompatible interaction with this pathogenic fungus. In this study, we evaluated spatial (leaves, epicotyls and hypocotyls) and temporal (24, 48, 72 and 96 hours after inoculation [HAI]) relative expression (RE) of 12 defense-related transcripts selected from previously developed ESTs libraries, during incompatible interaction between the resistant common bean genotype SEL 1308 and the avirulent anthracnose pathogen race 73, using real time quantitative RT-PCR (RT-qPCR) analysis. All selected transcripts, including the ones coding for pathogenesis-related (PR) proteins (PR1a, PR1b, PR2, and PR16a and PR16b) were differentially regulated upon pathogen inoculation. The expression levels of these transcripts were dependent on the tissue and time post inoculation. This study contributes to a better understanding of the kinetics of induced defenses against a fungal pathogen of common bean and may be used as a base line to study defenses against a broad range of pathogens including bacteria as well as non-host resistance. São diversificadas as recomendações referentes à temperatura (T°) e umidade relativa do ar (UR) no armazenamento de materiais esterilizados em Centrais de Material e Esterilização (CME), sem que essas recomendações estejam embasadas em referenciais teóricos ou experimentos. A prática mostra dificuldades em controlar esses parâmetros, suscitando dúvidas quanto ao risco para a manutenção da esterilidade dos materiais. Este artigo propôs, por meio de uma revisão bibliográfica integrativa, identificar e analisar as recomendações referentes à T° e UR indicadas para o setor de guarda dos materiais na CME. Não foi encontrada literatura que justifique tais recomendações. Foram incluídas sete publicações que analisaram as variáveis T° e UR da área de armazenagem como fatores que podem afetar a manutenção do material esterilizado, e apresentaram resultados contraditórios quanto à interferência desses fatores na manutenção da esterilidade dos materiais.

The present PhD project was focused on the development of new tools and methods for luminescence-based techniques. In particular, the ultimate goal was to present substantial improvements to the currently available technologies for both research and diagnostic in the fields of biology, proteomics and genomics. Different aspects and problems were investigated, requiring different strategies and approaches. The whole work was thus divided into separate chapters, each based on the study of one specific aspect of luminescence: Chemiluminescence, Fluorescence and Electrochemiluminescence. CHAPTER 1, Chemiluminescence The work on luminol-enhancer solution lead to a new luminol solution formulation with 1 order of magnitude lower detection limit for HRP. This technology was patented with Cyanagen brand and is now sold worldwide for Western Blot and ELISA applications. CHAPTER 2, Fluorescescence The work on dyed-doped silica nanoparticles is marking a new milestone in the development of nanotechnologies for biological applications. While the project is still in progress, preliminary studies on model structures are leading to very promising results. The improved brightness of these nano-sized objects, their simple synthesis and handling, their low toxicity will soon turn them, we strongly believe, into a new generation of fluorescent labels for many applications. CHAPTER 3, Electrochemiluminescence The work on electrochemiluminescence produced interesting results that can potentially turn into great improvements from an analytical point of view. Ru(bpy)3 derivatives were employed both for on-chip microarray (Chapter 3.1) and for microscopic imaging applications (Chapter 3.2). The development of these new techniques is still under investigation, but the obtained results confirm the possibility to achieve the final goal. Furthermore the development of new ECL-active species (Chapter 3.3, 3.4, 3.5) and their use in these applications can significantly improve overall performances, thus helping to spread ECL as powerful analytical tool for routinary techniques. To conclude, the results obtained are of strong value to largely increase the sensitivity of luminescence techniques, thus fulfilling the expectation we had at the beginning of this research work.

In der murinen kutanen Leishmaniose ist die Ausheilung der Infektion mit dem Auftreten von schützenden CD4+ Th1- sowie CD8+ Tc1-Immunantworten assoziiert. Daher sollte eine wirksame Vakzine beide T-Zell-Populationen induzieren. Im Rahmen dieser Dissertation konnte gezeigt werden, daß mit TAT-LACK Fusionsproteinen effektiv gegen eine progressiv verlaufende Infektion mit Leishmania major in empfindlichen BALB/c sowie resistenten C57BL/6 Mäusen vakziniert werden kann. TAT-LACK konnte hierbei sowohl im DC- als auch im proteinbasierten Ansatz protektive Immunität verleihen. Das TAT-Peptid ist in der Lage, Proteine direkt in das Zytosol von DC zu schleusen und so den für die CD8+ T-Zell-Antworten erforderlichen MHC Klasse I Präsentationsweg einzuschlagen. Stammesspezifische Untersuchungen des inflammatorischen Infiltrates in Läsion und Lymphknoten bestätigten die physiologische Relevanz von DC und CD8+ T-Zellen im Verlauf einer etablierten Leishmania-Infektion in beiden Mausstämmen und rechtfertigten somit den Einsatz einer DC basierten Vakzine, die vermehrt CD8+ T-Zellen induzieren sollte. Tatsächlich konnte mit TAT-LACK transduzierten DC in beiden Mausstämmen effektiv gegen eine progressiv verlaufende Leishmania-Infektion vakziniert werden. Der Vakzinierungserfolg ließ sich anhand verringerter Läsionsvolumina, reduzierter Parasitenlasten und einer Verschiebung des Zytokinprofils in Richtung einer Th1 dominierten Immunantwort bestätigen. In allen Ansätzen war die i.d. Vakzinierung mit TAT-LACK transduzierten DC der Injektion von LACK gepulsten DC überlegen. Experimente mit DC aus IL-12p40 defizienten Mäusen belegten die IL-12 Abhängigkeit der Vakzine. Mit Hilfe von in vitro Restimulierungsexperimenten konnte nachgewiesen werden, daß nach Applikation TAT-LACK transduzierter DC in beiden Mausstämmen vermehrt CD8+ T-Zellen induziert werden. Des weiteren waren TAT-LACK transduzierte DC in Restimulationsexperimenten stärkere Aktivatoren der CD8+ T-Zell-Proliferation als LACK gepulste DC. Depletionsexperimente bestätigten die T-Zell-Abhängigkeit der Vakzine. Weitere in vivo Versuche belegten zudem die protektive Wirkung von TAT-LACK Fusionsproteinen im direkten, proteinbasierten Vakzinierungsansatz. Floreszenzmikroskopische Analysen der Epidermis bestätigten hierbei Aktivierung und Auswanderung von LC nach i.d. TAT-LACK Applikation.

The spectroscopic investigation of the gas-phase molecules relevant for the chemistry of the atmosphere and of the interstellar medium has been performed. Two types of molecules have been studied, linear and symmetric top. Several experimental high-resolution techniques have been adopted, exploiting the spectrometers available in Bologna, Venezia, Brussels and Wuppertal: Fourier-Transform-Infrared Spectroscopy, Cavity-Ring-Down Spectroscopy, Cavity-Enhanced-Absorption Spectroscopy, Tunable-Diode-Laser Spectroscopy. Concerning linear molecules, the spectra of a number of isotopologues of acetylene, 12C2D2, H12C13CD, H13C12CD, 13C12CD2, of DCCF and monodeuterodiacetylene DC4H, have been studied, from 320 to 6800 cm-1. This interval covers bending, stretching, overtone and combination bands, the focus on specific ranges depending on the molecule. In particular, the analysis of the bending modes has been performed for 12C2D2 (450-2200 cm-1), 13C12CD2 (450-1700 cm-1), DCCF (320-850cm-1) and DC4H (450-1100 cm-1), of the stretching-bending system for 12C2D2 (450-5500 cm-1) and of the 2nu1 and combination bands up to four quanta of excitation for H12C13CD, H13C12CD and 13C12CD2 (6130-6800 cm-1). In case of symmetric top molecules, CH3CCH has been investigated in the 2nu1 region (6200-6700 cm-1), which is particularly congested due to the huge network of states affected by Coriolis and anharmonic interactions. The bending fundamentals of 15ND3 (450-2700 cm-1) have been studied for the first time, characterizing completely the bending states, v2 = 1 and v4 = 1, whereas the analysis of the stretching modes, which evidenced the presence of several perturbations, has been started. Finally, the fundamental band nu4 of CF3Br in the 1190-1220 cm-1 region has been investigated. Transitions belonging to the CF379Br and CF381Br molecules have been identified since the spectra were recorded using a sample containing the two isotopologues in natural abundance. This allowed the characterization of the v4 = 1 state for both isotopologues and the evaluation of the bromine isotopic splitting.
