Carrageenin-induced inflammatory responses in the hindpaws of rats were quantitated by measuring: (1) alterations in volumes of the paws; and (2) alterations in concentration of dye, previously injected intravenously, which was recovered in perfusates from the paws. The inflammatory response in one paw was attenuated by previously inducing an inflammatory response in the contralateral paw. The effect was abolished by pretreatment with insulin. Indexes of adrenal activity were increased after the induction of the inflammatory response and they were not attenuated by pretreatment with insulin. Adrenal hyperactivity was characterized by increased serum corticosterone concentration, decreased adrenal ascorbic acid content, and reduced number of circulating eosinophils. It is concluded that inflammatory stimuli which lead to alterations in microvessels depend on a facilitatory effect of insulin. This effect is antagonized by glucocorticoids released in enhanced concentrations after the application of noxious stimuli. Therefore, endogenous insulin and glucocorticoids act as modulators of inflammatory responses.

This article describes the structures and functions of the erythrocyte membrane and its importance in transfusional medicine. The erythrocyte membrane is one of the best known membranes in terms of structure, function and genetic disorders. As any other plasma membrane, it mediates transport functions. It also provides the erythrocytes with their resilience and deformability. According to the International Society of Blood Transfusion (ISBT), more than 500 antigens are expressed in the erythrocyte membrane, and around 270 are involved in transfusion reaction cases and hemolytic diseases of the fetus and newborn. In the ISBT classification, the high frequency series is represented by antigens in more than 99% of population (high prevalence antigen). In transfusion, the absence of these antigens determines severe problems as for example, one woman without the P antigen suffered 6 repetitive miscarriages due to placental insufficiency, which was caused by an antibody formed against the absent P antigen. Some important erythrocyte membrane proteins are described here including Band 3, Glycophorins and spectrin. The most abundant integral membrane protein is Band 3 and its main function is to mediate exchange of chloride and bicarbonate anions across the plasma membrane. The second most abundant integral membrane protein in the human erythrocyte is sialoglycoprotein glycophorin A (GPA). With its high sialic acid content, GPA is the main contributor to the net negative cell-surface charge and is thus critical for minimizing cell-cell interactions and preventing red cell aggregation. Glycophorin C (GPC) is the receptor for PfEBP-2 (baebl, EBA-140), the newly identified erythrocyte binding ligand of Plasmodium falciparum. The ternary complex of spectrin, actin and 4.1R defines the nodes of the erythrocyte membrane skeletal network, and is inseparable from membrane stability when under mechanical stress. This erythrocyte membrane review is important for a better understanding of transfusion reactions, where the antibody formation against high prevalence antigens makes compatible transfusions difficult. The study of antigen diversity and biochemical characterization of different proteins will contribute to healthcare, as well as diagnosis, development of technology such as monoclonal antibody production and the therapeutic conduct of many diseases.

The shelf life of the organic passion fruit pulp, both fresh and pasteurized at 70C and 90C and stored under refrigeration, was evaluated. The heat treatment did not affect the sensory and physicochemical characteristics of the pasteurized pulps when compared with the fresh pulp, except for the ascorbic acid content. The pulps were also microbiologically safe. The pulps pasteurized at 70 and 90C were suitable for consumption for a minimum shelf-life period of 207 days of storage under refrigeration and for the fresh pulp it was attributed a shelf-life period of 60 to 90 days. The pulp pasteurized at 70C showed higher acceptance scores for all the attributes and purchase intention scores, suggesting a more stable behavior and higher sensory quality. Practical Applications: This work intended to evaluate the influence of the minimum pasteurization on the sensory acceptance and microbiological and physicochemical characteristics of the organic passion fruit pulp stored under refrigeration, with the aim to identify the shelf life. Heat treatment is one of the processes used for food preservation. Lower pasteurization temperature than that used by the Brazilian industries and storage under refrigeration showed to be appropriate for passion fruit pulp quality. In this way, the microbiological, physicochemical and sensory features of passion fruit were preserved. This work can be used as a reference for passion fruit pasteurization, which is able to increase the shelf life of this fruit while preserving its desirable original features. Glycomacropeptide is a glycosilated fraction of bovine kappa-casein that remains soluble when milk is clotted by rennin. Determinations of milk sialic acid content are useful because its concentration reflects the amount of free GMP of milk. In normal milk these amounts are very low, 12 to 16 times lower than in sweet whey. Therefore, its determination may be applied to verify possible frauds with whey addictions, since it works as a fingerprint. With the description of a new spectrophotometric method for determination of free GMP (ANSM) occurred a simplification of procedures, being faster than others (HPLC method), without loss of accuracy. However, due to variations of glycosilation in kappa-casein between animals, during the lactation period, due to mastitis and yet due to proteolysis on milk, it was necessary to know these variations to interpret correctly the analytical results. It was analyzed 1,703 samples of producer's raw milk and 1,189 samples of processed milk (HTST and UHT). The results showed that normal milk from herd (producer's milk) have only small amounts of free GMP, with A470nm = 0.232±0.088 or 3.89±1.25 mg of sialic acid/L. The upper limit of this distribution was A = 0.496; thus every bigger value may represent a problem, being outside of normal distribution.

Crambe is an important biofuel crop and its oil has unique traits such as high erucic acid content which can be used as industrial lubricant, corrosion inhibitor as well as ingredient in synthetic rubber manufacturing. Genetic diversity among 70 progenies of Crambe abyssinica Hochst selected from a population of FMS Brilhante cultivar was quantified by multivariate analysis for traits related to germination, thousand grain weight and oil content. There were significant differences among progenies for all traits studied. Estimation of genetic variance and heritability coefficients showed that the variability found in the progeny is more genetic than environmental which enables genetic gains with selection. Heritability coefficient varied from 68 to 79%, except for oil content and number of dead seedlings. Simple correlation analysis showed that germination and vigor were positively correlated, and thousand grain weight and oil content were not correlated with any of the seed traits. Based on multivariate analysis, the progenies could be grouped into 26 clusters. Clusters 1, 2 and 3 had the highest number of progeny with 7, 8 and 6 lineages, respectively. Clusters 21-26 had higher dissimilarity within the cluster with one in each progeny. The trait that most contributed to the cluster was the germination (36.2%) and less contributed was the number of seedlings killed (1.1%). The progenies indicate genetic diversity for seed traits and the selection of superior progenies is possible considering the studied traits. © 2013.

Four different assays (the Folin-Ciocalteu, DPPH, enzymatic method, and inhibitory activity on lipid peroxidation) based on radically different physicochemical principles and normally used to determine the antioxidant activity of food have been confronted and utilized to investigate the antioxidant activity of fruits originated from Brazil, with particular attention to more exotic and less-studied species (jurubeba, Solanum paniculatum; pequi, Caryocar brasiliense; pitaya, Hylocereus undatus; siriguela, Spondias purpurea; umbu, Spondias tuberosa) in order to (i) verify the correlations between results obtained by the different assays, with the final purpose to obtain more reliable results avoiding possible measuring-method linked mistakes and (ii) individuate the more active fruit species. As expected, the different methods give different responses, depending on the specific assay reaction. Anyhow all results indicate high antioxidant properties for siriguela and jurubeba and poor values for pitaya, umbu, and pequi. Considering that no marked difference of ascorbic acid content has been detected among the different fruits, experimental data suggest that antioxidant activities of the investigated Brazilian fruits are poorly correlated with this molecule, principally depending on their total polyphenolic content. © 2013 Elena Gregoris et al.

Neste trabalho foi estudado o processo de craqueamento termocatalítico do óleo de fritura nas escalas de bancada e piloto, variando-se o percentual do catalisador carbonato de sódio de 5 e 10% m/m em relação a matéria prima utilizada e temperatura de 440 ºC. O objetivo foi obter misturas de hidrocarbonetos ricas na fração diesel. O óleo de fritura neutralizado e seco foi caracterizado em relação ao Índice de Acidez, Índice de saponificação, Viscosidade Cinemática, Densidade e Índice de Refração. Após o craqueamento, o produto líquido obtido foi purificado por decantação da fase aquosa e filtração simples em escala de bancada. Esse produto foi fracionado por destilação fracionada e os condensados foram coletados em um funil de decantação de acordo a faixa de destilação da gasolina (40ºC-175ºC), querosene (175ºC-235ºC), diesel leve (235°C-305ºC) e diesel pesado (305ºC-400 ºC). Foi realizada a caracterização tanto físico química quanto da composição dos produtos líquidos e suas respectivas frações. Também foi realizada a evolução do processo de craqueamento em escala piloto, acompanhando o comportamento das características físico químicas e de composição do produto formado no decorrer do processo de craqueamento. Os resultados mostraram que o catalisador carbonato de sódio forneceu produtos de baixa acidez e com boas características para uso como combustível. A variação do percentual de catalisador influencia significamente as propriedades físico químicas e composição tanto do produto quanto de suas frações. Verificou-se, ainda, que o craqueamento termocatalítico do óleo de fritura propicia a formação de hidrocarbonetos ricos na fração do diesel (19,16% diesel leve e 41,18% diesel pesado para o teste com 10% de Na2CO3 e de 13,53% leve e 52,73% diesel pesado para o teste com 5% de Na2CO3 ). Os intervalos de tempos finais do craqueamento geram um combustível com baixo teor de acidez e com propriedades físico químicas em conformidade a norma especificada para o diesel mineral.
