During meiosis in Saccharomyces cerevisiae, the first chemical step in homologous recombination is the occurrence of site-specific DNA double-strand breaks (DSBs). In wild-type cells, these breaks undergo resection of their 5' strand termini to yield molecules with 3' single-stranded tails. We have further characterized the breaks that accumulate in rad50S mutant stains defective in DSB resection. We find that these DSBs are tightly associated with protein via what appears to be a covalent linkage. When genomic DNA is prepared from meiotic rad50S cultures without protease treatment steps, the restriction fragments diagnostic of DSBs selectively partition to the organic-aqueous interphase in phenol extractions and band at lower than normal density in CsCl density gradients. Selective partitioning and decreased buoyant density are abolished if the DNA is treated with proteinase K prior to analysis. Similar results are obtained with sae2-1 mutant strains, which have phenotypes identical to rad50S mutants. The protein is bound specifically to the 5' strand termini of DSBs and is present at both 5' ends in at least a fraction of breaks. The stability of the complex to various protein denaturants and the strand specificity of the attachment are most consistent with a covalent linkage to DSB termini. We propose that the DSB-associated protein is the catalytic subunit of the meiotic recombination initiation nuclease and that it cleaves DNA via a covalent protein-DNA intermediate.

It has been proposed that the depolarizing responses of chromaticity horizontal cells (C-HCs) to red light depend on a feedback signal from luminosity horizontal cells (L-HCs) to short-wavelength-sensitive cones in the retinas of lower vertebrates. In this regard we studied the C-HCs of the Xenopus retina. C-HCs and L-HCs were identified by physiological criteria and then injected with neurobiotin. The retina then was incubated with peanut agglutinin, which stains red-but not blue-sensitive cones. Electron microscopic examination revealed that L-HCs contact all cone classes, whereas C-HCs contact only blue-sensitive cones. Simultaneous recordings from C-HC/L-HC pairs established that when the L-HC was saturated by a steady bright red light, C-HCs alone responded to a superimposed blue stimulus. In response to red test flashes, the C-HC response was delayed by approximately 30 msec with respect to the L-HC response. Isolated HCs of both subtypes were examined by whole-cell patch clamp. Both responded to kainate with sustained inward currents and to quisqualate or alpha-amino-3-hydroxy-5-methyl-4-isoxazolepropionic acid (AMPA) with desensitizing currents from a negative holding potential; i.e., both have AMPA-type glutamate receptors. gamma-Aminobutyric acid or glycine opened a chloride channel in the L-HC, whereas the C-HC was unresponsive to either inhibitory amino acid. Since glycine has been shown to abolish selectively the depolarizing response of the C-HC, this finding and other pharmacological data strongly implicate the L-HC in the underlying circuit. Moreover, because the C-HC does not respond to gamma-aminobutyric acid, the neurotransmitter of the L-HC, by elimination, a feedback synapse from L-HC to blue cone is the most plausible mechanism for the creation of depolarizing responses in C-HCs.

Fossil manganese nodules and encrustations from seamount' and basin' localities in the Transdanubian Central Mountains of Hungary are lithologically, mineralogically and chemically similar to some modern marine ferromanganese oxide deposits, and show no evidence of postdepositional changes other than cementation. Five groups of deposits were encountered: Fe/Mn nodules, encrusted shells, pavements, stains, and Fe oxide encrusted intraclasts, the first three of which are specific to the 'seamount' environment and the last to the basins'. Optical and electron microprobe investigation of the samples shows them to exhibit many similarities with modern ferromanganese oxide deposits, and that many of the nodules are surrounded by a halo of dispersed ferromanganese oxides, strongly suggesting that they continued to accrete metals through the pore waters of unlithified sediments for a period after burial. By contrast, pavements which appear to have grown on hardgrounds at the sea floor show little or no evidence of derivation of metals from underlying sediments. Geochemical investigations on the deposits show the seamount' varieties to be closer in composition to most modern nodules and crusts than the basin' varieties, and that the latter are essentially manganese and trace-element-poor ferruginous deposits. Nevertheless, all can be more or less compositionally equated with modern ferromanganese oxide deposits forming in marginal Atlantic environments, which would be in accord with the proposed depositional environment of the Transdanubian Central Mountains based on other evidence.
